Isolation and Identification of Pathogenic Bacteria of Grapevine Crown Gall in Korea (우리나라 포도나무 줄기혹병 병원세균의 분리 및 동정)

  • 우리나라 포도재배 지역인 포천, 남양주 및 수원의 포도나무 줄기혹병에 이병된 포장으로부터 채집한 줄기혹(gall), 수액(sap) 및 근권토양으로부터 56균주의 유사 Agrobacterium를 분리하였으며, 이들 중 7균주가 포도나무 유묘의 줄기에 혹ㅇ르 형성시키는 병원성 Agrobacterium biovar의 선택배지에서의 생장, oxidase의 생성, Na L-tartrate로부터 알칼리의 성장, 2% NaCl 첨가 배지에서 생장, 3-ketolactose의 비생산, mesoerythritol과 ethanol로부터 산을 생성하지 못하는 등 Agrobacterium biovar 3인 A. vitis로 동정되었다.

BIOLOGICAL ACTIVITIES OF PRODUCTS FROM SUGAR CANE BAGASSE FERMENTATION BY Pleurotus sajor-caju (사탕수수 찌꺼기를 이용한 여름느타리 발효부산물의 생물활성)

  • In order to evaluate the biological activities of some fractions from the fungal(Pleurotus sajorcaju) fermentation products of sugar cane bagasses, the antimutagenicity, the glyceollin elicitor activity on soybean and the effect on the stem elongation in pea were observed. The alcohol extract fraction and DMSO soluble fraction had excellent antimutagenicity even though it is weaker than that of the extracts from the fruiting bodies. All of the extracts had the ability to elicit glyceollins in soybean cotyledons and these extracts could be helpful for plants to protect themselves from pathogenic contaminations. IAA and the extracts had shown synergistic effects on pea stem elongation in all experimental groups positively determined.

Comparison of Neural Cell Differentiation of Human Adipose Mesenchymal Stem Cells Derived from Young and Old Ages (연령별 지방 중간엽 유래 줄기세포의 신경세포로의 분화 능력 비교)

  • Recently, adipose mesenchymal stem cells (AdMSC) that are similar to bone marrow MSC and blood derived MSC are thought to be another source for stem cell therapy. However, the diseases that can be applied for stem cells therapy are age-dependent degenerative diseases. Accordingly, the present study investigated the growth and differentiation potential to neural cells of human AdMSC (hAdMSC) obtained from aged thirty, forty and fifty. The growth of cells and cell viability were measured by passage and neural differentiation of hAdMSC was induced in neural differentiation condition for 10 days. Our results demonstrated that cell number, viability and morphology were not different from hAdMSC by age and passage. Immunofluorescence analysis of neural cell marker (TuJ1, NSE, Sox2, GFAP or MAP2) demonstrated no significant differences in neural cell differentiation by age and passage. As the number of passage was increased, the mRNA level of MAP2 and Sox2 was decreased in hAdMSC from age of 50 compared to hAdMSC from age of 30. In conclusion, the present study demonstrated that ability of neural cell differentiation of hAdMSC was maintained with ages, suggesting that autologous stem cells from aged people can be applied for stem cell therapy with age-dependent neural disease with the same stem cell quality and ability as stem cell derived from young age.

Effect of Ursolic Acid on the Development of Mouse Embryonic Stem Cells under Hypoxia (저산소 상태에서 우르솔산이 배아줄기세포 성장에 미치는 효과)

  • Ursolic acid (UA) a bio-active ingredient found in a variety of fruits and vegetables, and it has potent antioxidant activity. However, the role of UA in mouse embryonic stem (ES) cells is poorly understood. This study investigated the functional role of UA in regulating the development of mouse ES cells under hypoxia. Hypoxia did not exert a significant effect on the undifferentiated state of mouse ES cells. However, it induced reactive oxygen species (ROS) generation and increased the level of lactate dehydrogenase (LDH) production at 48 h of hypoxic exposure. Conversely, oxidative stress induced by hypoxia was significantly inhibited by UA ($30{\mu}M$) pretreatment. Hypoxia significantly decreased cell survival and the level of [$^3H$] thymidine incorporation, both of which recovered following pretreatment of UA. In addition, UA decreased the apoptotic effect of hypoxia by attenuating caspase-3 cleavage or by recovering cellular inhibition of the apoptotic protein (cIAP)-2 and Bcl-2 expression. We further found that UA decreased senescence-associated beta-galactosidase activity. We suggest that UA is a natural antioxidant and one of the functional modulators of hypoxia-induced survival, apoptosis, proliferation, and aging in mouse ES cells.

Interaction of Bone Marrow Stromal Stem Cells with Adhesive Protein and Polypeptide-adsorbed Poly(lactide-co-glycolide) Scaffolds (골수유래 간엽줄기세포와 점착성 단백질 및 폴리펩타이드가 흡착된(락티이드/글리콜라이드) 공중합체 지지체와의 상호작용)

  • The interaction of cell adhesive protein and polypeptide with bone marrow stromal stem cells (BMSCs) grown in tissue engineered films and scaffolds were examined. Several proteins or polypeptide known as cell-adhesive were coated adsorption on poly(lactide-co-glycolide) (PLGA) films and scaffolds and adhesion and proliferation behavior of BMSC on those surfaces were compared. The protein and polypeptide used include collagen IV, fibrinogen, laminin, gelatin, fibronectin, and poly(L-lysine). The protein and polypeptide were adsorbed on the PLGA film surfaces with almost monolayer coverage except poly(L-lysine). BMSCs were cultured for 1, 2, and 4 days on the protein- or polypeptide-adsorbed PLGA films and scaffolds. The cell adhesion and proliferation behaviors were assessed by sulforho damine B assay. It was observed that the protein- or polypeptide-adsorbed surfaces showed better cell adhesion and proliferation than the control.

Cancer Stem Cells and the Tumor Microenvironment (암줄기세포와 종양 미세환경에 대한 고찰)

  • Solid tumors are heterogeneous populations of multiple cell types. While the majority of the cells that comprise cancer are unable to divide, cancer stem cells have self-renewal and differentiation properties. Normal stem cell pathways that control self-renewal are overactivated in cancer stem cells, making cancer stem cells important for cancer cell expansion and progression. Dick first proposed the definition of cancer stem cells in acute myeloid leukemia, according to which cancer stem cells can be classified based on the expression of cell surface markers. Cancer stem cells maintain their potential in the tumor microenvironment. Multiple cell types in the tumor microenvironment maintain quiescent cancer stem cells and serve as regulators of cancer growth. Since current cancer treatments target proliferative cells, quiescent state cancer stem cells that are resistant to treatment increase the risk of recurrence or metastasis. Various signals of the tumor microenvironment induce changes to become a tumor-supportive environment by remodeling the vasculature and extracellular matrix. To effectively treat cancer, cancer stem cells and the tumor microenvironment must be targeted. Therefore, it is important to understand how the tumor microenvironment induces reprogramming of the immune response to promote cancer growth, immune resistance, and metastasis. In this review, we discuss the cellular and molecular mechanisms that can enhance immunosuppression in the tumor microenvironment.

Antimicrobial Activities of Ethanol Extracts from Korean Bamboo Culms and Leaves (국내산 대나무 줄기와 잎의 에탄올 추출물의 항균활성)

  • Antimicrobial activities in 70% ethanol extracts of Korean bamboo trees were investigated. Among the antimicrobial activities against ten major strains causing food poisening, Wangdae (Phyllostachys bambusoides S. et Z.) showed the strongest activity among five major bamboo trees cultivated in Korea. Although all extracts showed relatively strong antimicrobial activities against Staphylococcus aureus and Streptoccus mutans, the extracts of bamboo culms showed stronger activities than that of bamboo leaves. MICs (minimal inhibitory concentration) of the extracts of Wangdae culm against Staphylococcus aureus and Streptococcus mutans were over $10\;and\;20\;{\mu}L/disc$, respectively. The extract of Wangdae culm inhibited the growth of Staphylococcus aureus and Streptococcus mutans at over 0.5% concentration, and almost inhibited the growth of these strains at 1% level. The inhibitory effect of the extract of Wangdae culm was not destroyed by heating at 60, 80, and $100^{\circ}C$ for 60 min and at $121^{\circ}C$ for 15 min, suggesting that the component in the bamboo tree was very heat-stable. These results indicate that 70% ethanol extract of Wangdae culm could be useful as a natural antimicrobial agent.

Biological Evaluation of Bone Marrow-Derived Stem Cells onto Different Wettability by RT-PCR (역전사 중합효소 연쇄반응을 이용한 표면 적심성에 따른 골수유래 줄기세포의 생물학적 평가)

  • The adhesion and proliferation of mammalian cells on polymeric biomaterials depend on the surface characteristics such as wettability, chemistry, charge and roughness. In order to recognize the correlation between the adhesion and proliferation of human bone marrow derived stem cells (BMSCs) and surface property, radio frequency generated plasma treatment on low density polyethylene (LDPE) has been carried out. The modified LDPE surfaces were characterized by measuring the static water contact angle. The adhesion and proliferation of cells on LDPE films were characterized by cell counting and reverse transcription-polymerase chain reaction (RT-PCR). The water contact angle of the film surface decreased with plasma treatment time. Proto-oncogenes (c-myc, c-fos) and tumor suppressor gene (p153) showed maximum expression with contact angle of 60 ∼ 70$^{\circ}$ range of LDPE film. By cell counting, we confirmed that the rate of cell proliferation appeared the higher on the film surface of the contact angle of 60∼70$^{\circ}$ We concluded that the surface wettability is an important role for the growth and differentiation of BMSCs.

Protective Effect of Rhus Semialata M. extract on Epidermal Stem Cells against UV Irradiation (자외선 조사된 상피 줄기세포에 대한 붉나무 추출물의 보호 효과)

  • Human epidermal stem cells(ESCs) residing in the basement membrane of the skin have an important role in maintenance of skin homeostasis of epidermal layer. Although, ESCs provide new cells to repair damaged tissue in response to tissue injury, subsets of stem cells remain in the quiescent state protected from differentiation and senescence for prolonged survivals. In this perspective, the stem cell niche, which is specific microenvironment composed of niche cells and an extracellular matrix(ECM), supplies the relevant signal to save stem cells from microenvironmental damages. The expression of stemness marker on the surface of ESCs contributes to the attachment on their ECM of the basement membrane, which lead to growth potential and apoptotic resistance against environmental stimuli. In this study, we observed that UV irradiation, a major factor of environmental stimuli, reduced the expression of α2, β1 and α6 integrin in ESCs. Rhus Semialata M extract(RSE) showed inhibitory effect on the UVB-induced reduction of integrin expression. Furthermore, RSE could upregulate the expression of Col-IV and Laminin, which contribute to the attachment of ESCs. These results indicated that RSE could be a potent ingredient for the protection of ESCs from UV irradiation by increasing the expression of integrin and substrate ECM components at their niche.

an Influence of Stem Cell Study on Life Insurance Industry (줄기세포연구와 생명보험산업에 대한 고찰)

  • 생명공학의 시대로 일컬어 지고 있는 오늘날, 재생의학 분야에서는 난치성 질환 치료를 목적으로 활발한 연구가 진행되고 있다. 특히, 줄기세포를 이용한 세포 대체치료 관련 연구는 최근 국내 황우석 박사의 체세포 핵이식 배아세포주 확립에 이르기 까지 괄목할 만한 발전을 보여 주고 있다. 이와 관련해 생명보험산업에 적잖은 파장이 예상되며, 생명보험사 내부적으로 기존에 판매된 상품의 사차손 관리와 함께 급속도로 발전하는 줄기세포 연구에 직접적으로 대응하는 상품개발, 언더라이팅, 지급 심사 등 보험사 내외에서의 전방위적인 변화가 필요하다는 문제 제기가 있다. 줄기세포란 조직 분화 과정에서 볼 수 있는 세포이며 근육 뼈 뇌 피부 등 신체의 어떤 기관으로도 전환할 수 있는 만능세포로서, 간 폐 심장 등 구체적 장기를 형성하기 이전에 분화를 멈출 배아 단계의 세포를 말한다. 한편, 성체줄기세포는 조직이나 기관의 분화된 세포들 사이에서 발견되는 미분화 세포로써, 자기 스스로 증식할 수 있으며, 조직이나 기관의 특수한 기능을 가지고 있는 세포로 분화할 수 있는 능력을 가진 신체줄기세포를 말한다. 배아줄기세포와 생체줄기세포를 통한 장기이식 등 난치병 정복은 윤리적, 사회적으로 많은 논란이 예상되며, 기술적으로도 해결해야 할 문제점들이 산적해 있기 때문에 아직은 요원한 것이 사실이다. 현재 유럽 대부분의 나라와 미국에서는 인간 배아의 복제가 금지되어 있으며, 일본 정부는 연구용 배아 복제를 제한적으로 허용하고 있다. 하지만, 우리 나라의 경우 2005년 1월에 '생명윤리 및 안전에 관한 법'이 발효되었지만 정부는 관련 부작용에 대한 깊은 고찰 없이 전폭적인 지원들 약속하고 있는 실정이다. 줄기세포 연구의 발달로 인해 인류가 난치병 치료의 첫 장을 열었다고 하더라도 그 영향이 당장 보험사에 미친다고 할 수는 없다. 왜냐하면 앞으로 이러한 신기술이 실제 의료행위에 적용되기 위해서는 여러 단계의 안정화 작업과 임상시험이 필요한데 이러한 작업이 기술적으로 어렵고 그 시간도 만만치 않게 걸리기 때문이다. 또한, 보험사의 보장은 크게 사망/수술/입원/암/기타보장으로 구별할 수 있는데, 줄기세포 연구의 발달과 관련이 있는 보장이 제한되어 있어 보험사에 미치는 영향이 당장 우려할 만한 수준이라 할 수 없다. 하지만 만약 치료용 줄기세포 배양으로 인한 장기 기관의 이식이나 손상세포의 대체 등과 같은 의학신기술의 예상 외로 급격하게 발전한다면 보험사의 Risk 관리에 상당한 저해요인으로 작용할 것으로 판단된다. 특히 진단 입원 수술로 대표되는 생존보장에 대한 사차 Risk 및 사차손의 급증이나 역선택 증가는 보험사의 경영수지 악화를 유발하여 보험산업 전반에 위험으로 작용할 수도 있다. 따라서, 장기적인 안목으로 업계 공동의 대응이 필요하고, 각 사에서도 상품개발, 언더라이팅, 지급심사 간의 긴밀한 협조가 요구된다. 생명보험산업의 Risk 관리는 기존의 시장환경에 영향을 받는 비차, 이차중심에서 보험회사가 어느 정도 관리를 통해 적정규모를 유지할 수 있는 사차로 그 중심축이 이동하고 있다. 보험산업이 계속 활력을 갖고 성장하기 위해서는 체계적인 Risk나 관리가 핵심일 것이며, 보험사의 사차 Risk의 중요성이 더욱 커져 가고 있는 현실에서 거시적으로 의학신기술 발달 등 위험요인에 대해 미리 분석하고 이에 대한 대비책 마련이 필요할 것으로 판단된다.
