Changes of Nitrogen Utilization Ratio , Protein Solution Ratio , free Sugars in Defatted Soybeans During the Manufacturing of Amino Acid Soysauce by a Low Hydrochloric Acid, Temperature (저염산(低鹽酸)으로 저온분해(低溫分解)한 아미노산(酸)간장 제조중(製造中) 질소이용률(窒素利用率), 단백분해율(蛋白分解率) 및 유리당(遊離糖)의 동향(動向))

  • The changes of nitrogen utilization ratio (NUR), Protein solution ratio (PSR) and free sugar contents during the amino acid soysauce manufacturing process by a low hydrochloric acid, temperature were investigated. On hydrolysis by 6%-HCI (3 liquor rate of defatted soybean weight, 3LR) at $85^{\circ}C$, NUR and PSR were 74.51%, 56.49% at 65 hours. At the same time free sugars were detected glucose, galactose, arabinose, fructose, xylose. on hydrolysis at $95^{\circ}C$, NUR and PSR were 77.72%, 64.04% at 50 hours, and 5 free sugars of the above statement were detected at 5 hours. Remarkable decreases in the levels of free sugars, only glucose were observed after 80 hours of the hydrolysis. On hydrolysis by 12%-HCI(3LR) at $95^{\circ}C$, NUR and IRA were 88.41%, 69.47% at 50 hours, free sugar were nearly disappeared after 20-35hours. On hydrolysis, galactose's disappearence rate was faster than glucose's.

  • Free fatty acid and total saponin, which were extracted from the spent media and colony of the strain Phizopus delemar Rh-1, were qualitatively examined by GLC and spectroscopic methods and compared with these of intact ginseng media. The 18, 18, and 14 free fatty acids were respectively identified from the control, spent media and colony. Among them, $iC_{12:0},\;C_{16:2},\;C_{17:0},\;C_{18:3}\; and\;C_{20:0}$ acids were not detectable from the colony. In add ition, $C_{16:2}\;and\;C_{20:0}$ peaks were rot found in the spent media and control, respectively. The isible absorption spectra of free fatty acids and total saponins in the control, spent media and colony were invariable. In contrast, however, the infrared and ultraviolet absorption spectra showed significant spectral variations, particularly in the it finger-print region, demonstrating that the ginseng components were considerably utilized by the strain Rhizopus delemar Rh-1.

Characterization of Volatile Components according to Fermentation Periods in Gamdongchotmoo Kimchi (발효기간에 따른 감동젓무 김치의 휘발성 향기 성분 특성)

  • The volatile components in Gamdongchotmoo kimchi, unfermented and fermented for 3 or 25 days, were extracted via solvent-assisted flavor evaporation (SAFE), and then analyzed via gas chromatography/mass spectrometry (GCMS). A total of 57 components, including 14 S-containing compounds, 22 terpene hydrocarbons, 13 aliphatic hydrocarbons, 4 alcohols, and 4 miscellaneous components, were detected in Gamdongchotmoo kimchi. Among them, the S-compounds were quantitatively dominant. The aroma-active compounds were also determined via gas chromatography-olfactometry (GC-O), using aroma extract dilution analysis (AEDA). A total of 16 aroma-active compounds were detected via GC-O. The most intense aroma-active compounds in Gamdongchotmoo kimchi included 4-isothiocyanato-1-butene ($Log_3$ FD factor 7, rancid), an unknown($Log_3$ FD factor 7, spicy) and another unknown ($Log_3$ FD factor 7, seasoning-like). In addition, other aroma-active compounds, including dimethyldisulfide ($Log_3$ FD factor 6, rotten onion-like/sulfury), 2-vinyl-[4H]-1,3-dithiin ($Log_3$ FD factor 5, spicy/garlic-like), and an unknown ($Log_3$ FD factor 5, rancid/cheese-like) might be crucial to the flavor characteristics of Gamdongchotmoo kimchi.

The Antiproliferative Effects of Compounds Isolated from Schisandra chinensis (오미자로부터 분리된 화합물의 암세포 증식 억제 효과)

  • We isolated twelve lignans and three terpenoids were isolated from the n-hexane fraction of Schisandra chinensis extract. Using spectroscopic data and comparison with available literature, the following compounds were identified: (1) wuweizisu C, (2) gomisin N, (3) deoxyschisandrin, (4) gomisin A, (5) schisandrin, (6) chamigrenal, (7) schisanlactone D, (8) methylgomisin O, (9) angeloylgomisin O, (10) (-)-gomisin $L_2$, (11) schisandronic acid, (12) (-)-gomisin $L_1$, (13) (+)-gomisin $K_3$, (14) gomisin J, and (15) tigloylgomisin H. Notably, this was the first finding that compound (8) was isolated from this plant. Each compound was evaluated for its in vitro cytotoxic activities toward HL-60 (human leukemia), HeLa (human cervical carcinoma), and MCF-7 (breast cancer) cell lines. Compounds (7), (8), and (9) exhibited strong cytotoxic effects on HL-60 ($IC_{50}$ 7.37, 6.60, and $8.00{\mu}M$, respectively), whereas compound (6) exhibited weak cytotoxicity towards MCF-7 ($IC_{50}$ $30.50{\mu}M$). In addition, compound (8) showed the strongest activity towards HeLa cells ($IC_{50}$ $1.46{\mu}M$).

Changes in Endogenous Gibberellin Contents during Bulb Development Period in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육 과정에서 내생 지베렐린류의 함량변화)

  • This study was performed to investigate the role of phytohormones in the bulbing of garlic in order to assess the yield and quality. The effect on endogenous plant hormones such as gibberellin (GA) content was also examined during growth stage i.e. clove differentiation to bulbing in garlic. More than 18 gibberellins in garlic were identified with extensive gas chromatograph-mass spectrometry-selected ion monitoring (GC-MS-SIM) quantitative analysis. The results showed that GAs were biosynthesized by both non C-13 hydroxylation pathway (NCH) and early C-13 hydroxylation pathway (ECH) in garlic plant. It was also revealed that NCH pathway leading to synthesis of bioactive $GA_4$ was the more prominent GA biosynthesis pathway than ECH pathway in which bioactive $GA_1$ was synthesized. Total GAs level was gradually increased from clove differentiation to bulbing and later decreased, which portrays the active role of GA in differentiation. The biosynthesis ratio of bioactive $GA_4$ and $GA_1$ concentration was similar to that of total GAs content, which was closely related with bulb development in garlic.

Diversity and Plant Growth-Promotion of Endophytic Fungi Isolated from the Roots of Plants in Dokdo Islands (독도의 자생식물 뿌리에서 분리한 내생진균의 다양성과 생장촉진활성)

  • Endophytic fungi were isolated from the roots of plants growing naturally on the island of Dokdo. Plant samples, such as Miscanthus sinensis, Achyranthus japonica and Echinochloa crusgali were isolated from Dongdo, and those such as Honkenya peploides and Artemsia koidzumii were isolated from Seodo. Twenty one strains of endophytic fungi were isolated from these plants. To identify the strains, PCR (polymerase chain reaction) amplification of the partial ITS (Internal Transcribed Spacer) regions was done with universal primers ITS-1 and ITS-4 to determine the nucleotide sequence of the ITS regions. Of the strains isolated from Miscanthus sinensis, 75% were Penicillium sp. and 25% were Aspergillus sp. Fifty five percent of strains isolated from Achyranthus japonica were Penicillium sp., 30% were Aspergillus sp. and 15% were Zygorhynchus sp. Strains isolated from Echinochloa crusgali were Penicillium sp. (50%), Aspergillus sp. (12%), Giberella sp. (13%), Talaromyces sp. (9%) and Umbelopsis sp. (8%). Of the strains isolated from Honkenya peploides, 76% were Penicillium sp. and 24% were Pestalotiopsis sp. Strains isolated from Artemisia koidzumii were Penicillium sp. (81%) and Mucor sp. (19%). As a result of bioassay, Ec-3-1 strain isolated from Echinochloa crusgalli showed plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp. was the most abundantly distributed fungal strain in all plants used in this study.

Optimal Condition to Produce Protease by Strain Separated from the Intestine of Reticulitermes speratus (흰개미(Reticulitermes speratus)의 장에서 분리된 세균에 의한 protease생성 최적조건)

  • We separated the bacteria showing protease activity from Reticulitermes speratus which is known as the only termite species in Korea. Then, we collected the best activated strain and studied the optimal culture condition for producing the enzyme. According to the results of observing morphological and physiological characteristics, and the type of 16S rRNA of the strain, it was identified as Serratia marcescens and named S. marcescens strain TM-3215. This strain showed the best activity under conditions of 0.8% (w/v) starch, 0.4% (w/v) peptone, 0.08% (w/v) $MgSO_4{\cdot}7H_2O$, $30^{\circ}C$ and pH 8.0. After being cultivated under optimal conditions for 9 hr, the strain produced 19.8 U/ml of enzyme, an amount 1.8 times greater than the control.

Flavor Composition of Garlic from Different Area (국내산 마늘의 향미성분)

  • Garlic from 4 main growing area was analyzed for the composition related to garlic flavor. Twenty-eight volatile compounds in garlic were separated by GC and twenty five were identified by GC/MS. Total peak area was high in garlic from $S\u{o}san$, and low from $Hampy\u{o}ng$. However, no significant difference was found in total peak area of sulfur compounds for garlic from Namhae, $S\u{o}san$, and $Uis\u{o}ng$. The results of free sugars analysis showed that relatively large amount of 1-kestose, 1-nystose, and 1-F-fructosyl nystose known as fructooligosaccharide were found in garlic in addition to glucose, fructose, and sucrose. Garlic from $S\u{o}san$ contained high amount of 1-nystose and 1-F- fructosyl nystose. Total free amino acid content in garlic was in a range of $2036.1{\sim}2704.0mg%$, and it was higher in garlic from $S\u{o}san$ and $Uis\u{o}ng$. Lactic acid, pyruvic acid, oxalic acid, malonic acid, fumaric acid, levunic acid, succinic acid, malic acid, citric acid and pyroglutamic acid were found in garlic. Total organic acid content in garlic was $1905.7{\sim}2359.2mg$, and garlic from $Hampy\u{o}ng$ had relatively low organic acid contents.

The Effect of S. thermophilus Isolated from Saliva Treated with Phytoncide on P. gingivalis (피톤치드 처리 후 구강 내 잔존 S. thermophilus의 P. gingivalis에 대한 효과)

  • The antibacterial effect of phytoncide on Porphyromonas gingivalis, which is the main causative agent of periodontal disease and halitosis, has been reported. However, little is known about its effect on normal oral microflora. The present study was performed to observe the effect of phytoncide on oral normal microflora and the inhibitory effect of surviving resident oral bacteria on P. gingivalis. In this study, saliva from each of 20 healthy subjects was treated with 1% phytoncide from Japanese Hinoki (Chamaecyparis obtusa Sieb. et Zucc.). Surviving salivary bacteria were isolated on blood agar plates and identified by 16S rDNA sequencing. In order to select inhibitory isolates against P. gingivalis, the isolates from the phytoncide-treated saliva were cultured with P. gingivalis. The results were as follows: 1. In general, the number of bacteria in saliva from periodontally healthy subjects was decreased when the saliva was treated with 1% phytoncide. 2. The majority of the salivary bacteria surviving the treatment of phytoncide were S. thermophilus (53%). 3. Most of the surviving salivary bacteria (72.5%) inhibit the growth of P. gingivalis A7A1-28 and P. gingivalis W83 on blood agar plates. 4. Among the surviving S. thermophilus, 85.8% of them were observed to inhibit P. gingivalis strains and 75.8% of the surviving S. sanguinis were inhibitory. Taken together, oral resident bacteria surviving phytoncide, which has been shown to inhibit P. gingivalis, may exert an additional inhibitory activity against the periodontopathic bacterium. Therefore, phytoncide can be used for preventing and ceasing the progress of periodontal disease and halitosis, and thus is expect to promote oral health.

A Study on Discriminative Criteria of 6 Kinds of Achyranthis Radix Using HPLC/DAD;Isolation and Identification of 20-hydroxyecdysone from Aclryranthes japonica $N-{AKAI}$ and Comparison of Patterns of Achyranthis Radix from Different Locations by HPLC (HPLC/DAD를 이용한 6종(種) 우슬(牛膝)의 분류기준 연구;우슬(牛膝)(쇠무릎, Achyranthes japonica $N_{AKAI}$)로부터 20-hydroxyecdysone 분리.동정 및 산지별 우슬의 HPLC 패턴 비교)

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.
