Effects of Mustard Seeds and Powder on In vitro Ruminal Fermentation Characteristics and Methane Production (겨자종자와 겨자분의 첨가가 반추위 발효성상과 메탄생성에 미치는 영향)

  • The purpose of this study was to investigate the effects of mustard, which contains allyl isothiocyanate, on ruminal fermentation and methane emission in vitro. To this end, diluted ruminal fluid(30ml) was incubated anaerobically at $39^{\circ}C$ or 6, 12, and 24 h with or without seeds or powdered mustard. Either mustard seed or powdered mustard was weighed and serially (0, 3.33, 5.00, 6.67, and 8.33 g/L) mixed with ruminal fluid. Ammonia-N was increased (P < 0.05) by mustard treatment in a dose dependent manner. Regardless of concentration or form, mustard increased (P < 0.05) total VFA content but decreased (P < 0.01) pH compared to control group. Molar proportion of acetate (A) was decreased (P < 0.05) whereas propionate (P) was increased (P < 0.05) by mustard treatment, thereby A:P ratio was decreased (P < 0.05) compared to control group. Total gas production was increased (P < 0.01) in a linear manner by mustard treatment compared to control group. There was no effect of mustard powder, except 8.33 g/L level at 6 h, on methane emission. However, at 24 h, methane emission was reduced (P < 0.05) by 4.77% and 11.54% with 6.67 g/L and 8.33 g/L of mustard seeds supplementation, respectively. Altogether, these results suggest that mustard seeds containing allyl isothiocyanate may reduce methane production without disturbing ruminal fermentation.

Effects of Temperature, Seed Water Content and Osmoconditioning on Germination and Seedling Elongation of Soybeans (온도.종자수분함량 및 삼투처리가 콩의 발아 및 묘신장에 미치는 영향)

  • Germination and seedling elongation of soybeans [cult. ‘Williams’] were measured at two temperatures (15 and 3$0^{\circ}C$), three seed water contents (30, 50, and 70%), and with and without polyethylene glycol-8000 (PEG) treatments. A split-plot design was used with four replications. Observations were made from two hours to eight days for each treatment combination. Seeds were soaked with 30% PEG solution with 0.2% thiram at 15 C for six days, rinsed with deionized water and dried at 25.5$^{\circ}C$ for three days. Ten treated or untreated seeds were placed on Whatman No. I (9 cm) filter paper in plastic petridishes to which sufficient deionized water was added to adjust seed moisture content to the desired level. The dishes were then sealed with parafilm and placed in a continuous programmed temperature chamber under dark condition. Seedling growth did not occur at either temperature when moisture content was 50 percent or less. Osmo-conditioning with PEG showed positive effects on seedling moisture uptake and seedling growth at 15$^{\circ}C$ but little effect at 3$0^{\circ}C$. Seedling moisture content increased rapidly early in the germination period and reached 60% at 15 C for 56 hours and at 3$0^{\circ}C$ for 28 hours. Seedling growth started when seed moisture reached a critical point of 60% at 15$^{\circ}C$, however, growth started after 20 hours of germination at 3$0^{\circ}C$. Seedlings of soybeans elongated more than four times faster at 3$0^{\circ}C$ than at 15$^{\circ}C$. Water uptake during germination was characterized by two phases in this experiment.

Effect of Hot Water Treatment Times on Moisture Absorption and Germination of Albizzia julibrissin Seeds (열탕처리시간이 자귀나무 종자의 수분흡수 및 발아에 미치는 영향)

  • This study was carried out to examine optimal hot water treatment time in Albizzia julibrissin seeds. Germination and moisture absorption characteristics among intact seeds, immersed seeds for 24 hours in distilled water and hot water treated seeds were surveyed. As result, treated seeds showed a highly significant difference with intact and immersed seeds (p<0.0l). Especially, treated seed for 2.0 minutes represented the highest percent of germination (PG). But mean germination time (MGT) did not have significant difference between non-treatment and treatments (p=0.502). Germination speed and germination performance index showed similar tendency with PG. In percent of moisture absorption (PMA) and moisture absorption rate constant (MARC) treatments had higher values than non-treatment whereas 2.0 minutes treatment was lower than non-treatment in initial moisture absorption rate (IMAR). In relation between germination properties and moisture absorption characteristics, all properties except MGT among germination properties had high correlations with PMA and MARC ($r=0.854{\sim}0.931$,p<0.01) whereas IMAR didn't have correlation.

  • This study has intended to disclose the change of some chemical compositions of Ginkgo seeds which were acquired the treatment of cold-moist-stratification after collection. As check sample, the room-stored seeds were used. With the reasons that when the seeds not stratified were sown the delay of field germination has usually been resulted, the effectiveness of stratificaation in respect to alteration of chemical composition is to be investigated. The increase and decrease of growth promoting and inhibiting substances were investigated by means of chromatography method followed by rice seedling test or wheat coleoptile straight-growth test. The results obtained are summarized as follows; 1. In the untreated seeds, the zone of growth inhibitors on paper chromatograph were observed without regard to the tissue differences, embryo, endosperm and seedcoat. 2. Due to stratification, the amount of inhibitor has decreased in the embryo and seed coat, but growth promoters was decreased as compared with the check materials 3. The indications of results appear that each portion of the embryo, endosperm, and seedcoats of Ginkgo biloba L. contains the growth in hibitor taking part in germination dormancy. 4. It was presumed that hastening germination was influenced by decreasing of inhibitors in the embryo and seed coats rather than by increasing of promoters. 5. Gibberellin was detected at Rf 0.26 under the UV-lamp and the abscisic acid was detected at Rf 0.62, Rf 0.70, and Rf 0.78 and showed purple, gray, blue fluorescence respectively under the UV-lamp.

An Etiological Study on the Anthracnose Fungus of Pepper Caused by Colletotrichum dematium in Korea (고추에서 분리(分離)한 탄저병균(炭疽病菌) Colletotrichum dematium에 대한 병원학적(病原學的) 연구(硏究))

  • In order to investigate an etiological characteristics of the pepper anthracnose fungus infected with Collectotrichum dematium, this experiment was undertaken. Isolation percent of the fungus was 55.3% from the seed sample of Jewon and 0.3% from that of Eumsung Kun, according to seed health blotter method. Although the acervulus was similar to the known two Colletotricum species, C. acutatum and C. gloeosporioides, setae was not only straightly extended above the acervulus, but also shaped as falcate conidia ranging $12{\sim}31.2{\times}2.4{\sim}4.8{\mu}m$. Acervului of the fungus were evenly scattered on the surface of pepper seed coat, and caused seedling blight after seed germination. Pre-and post-emergence seedling blight as well as foliar lesion and fruit rot was confirmed by inoculating conidial suspension. No typical anthracnose sysmptom was obtained from soybean seedlings, welsch onion and spinach seedlings upon inoculation. Therefore, this fungus is said to the undescribed form species of the fungus of pepper in Korea (Colletotricum dematium f. sp. capsicum).

  • 복주머니난(Cypripedium)속 식물은 우리나라에 광릉요강꽃을 비롯하여 털복주머니난, 흰털복주머니란, 복주머니란, 노랑복주머니란등 5종이 분포하는 것으로 알려져 있다. 광릉요강꽃과 털복주머니란 두 종은 환경부에서 지정한 멸종위기 식물 1급과 2급에 지정되어 보호를 받고 있고 나머지 3종은 제도적으로도 보호를 받지 못하는 실정이다. 본 연구에서는 충분히 성숙한 털복주머니난과 복주머니난의 종자에 NaOCl처리를 하여 발아율을 향상시킬수 있었는데 이러한 전처리가 발아에 미치는 원인에 대한 연구를 실시하였다. 털복주머니란의 무균적인 종자발아는 1.0% NaOCl 처리구에서 70% 이상의 종자발아율을 보였으며, POM배지가 MS배지보다 신초분화가 양호했다. GA3와 활성탄(Activated charcoal)의 혼합첨가는 신초증식에 효과적이었다. 신초분화 후 저온처리는 신초의 증식 율을 증가시켰다. 한편 NaOCl 농도(0, 1, 3, 5, 10%)와 NaOCl 처리시간(0, 5, 15, 30, 45, 60분)에 따라서 복주머니란의 종자발아를 확인한 결과, NaOCl 1%를 30분간 처리하였을 때 가장 높은 발아율을 나타냈다. NaOCl을 처리하여 종자의 종피상태를 SEM과 TEM으로 관찰한 결과 NaOCl의 처리는 종피 세포벽의 부분적 해리 및 작은 구멍을 만들게 하였는데 이러한 종피의 물리화학적 변화가 물과 양분의 이동을 원활히 하여 종자의 발아를 촉진하는 것으로 사료된다. 복주머니난의 신초분화에 미치는 casein과 활성탄의 효과를 알아본 결과 casein 200 mg/L와 활성탄 200 mg/L를 혼합 첨가한 실험구에서 가장 높은 신초분화율을 보였다. 토양순화 후 생존률은 극히 저조했으며 30 개체중에 5 개체만이 다음해 어린동아를 싹틔우는 것을 확인 하였다. 본 결과들을 종합하여 보면 멸종위기식물, 특히 털복주머니란과 복주머니란의 조직배양을 통해서 서식지외 보존방안(기내증식)에 관해 가능성을 제시하였다고 생각되어 진다.

Studies on the method for promoting the germination of sod seed (잔디종자의 발아촉진에 관한 연구)

  • For the study of method for promoting the germination of Korean sod seeds harvseted newly some physical and chemical treatments were tested in this experiment. The summarized results of these tests are as follows: 1. Storage of wet seed in low temperature of 0~$2^{\circ}C$ during 2~30 days was seemed the most effective treatment for promoting the germination of sod seed. 2. The treatment that Soaking and mixing up the seeds in cone sulphuric acid about one and half minutes and then washing off in the water promoted fairly the germination of sod seeds. 3. Better germination of sod seeds was found in the light than in the dark or soil. 4. Cutting the top of seed and crushing of seed coat with sand seemed to promote slightly the germination of sod seeds. 5. No promoting effect for germination of sod seeds was found with treatments of Gibberellin, $NH_4$ $NO_3$ and NAA.

Evaluation of Nutritional Characteristics of Platycodon grandiflorum Seeds (도라지 종자의 영양학적 특성 평가)

  • In this study, proximate composition, crude fiber, reducing sugar, free sugars, organic acids, minerals and amino acids of Platycodon grandiflorum seeds were analyzed to evaluate its nutritional value. Moisture, crude protein, crude fat, crude ash and carbohydrate contents of seeds were 6.97, 26.05, 27.46, 3.78 and 35.74%, respectively. Crude fiber of 6.31% and reducing sugars of 1.54% were also determined. Sucrose(1,661 mg/100 g) and lactic acid(1,224 mg/100 g) were most abundant free sugar and organic acid, respectively. Both phosphorus and potassium were main minerals that contained more than 700 mg in 100 g seeds. Amino acids analysis of 100 g seeds showed that glutamic acid(3.45 g), arginine(2.51 g), aspartic acid(1.66 g), leucine(1.29 g), lysine(1.10 g), alanine(1.05 g) and glycine(1.04 g) were abundantly contained in order, while others were less than 1 g.

Morphological Characteristics and Germination Conditions of Seeds in Trollius chinensis (큰금매화(Trollius chinensis) 종자의 형태적 특성 및 발아조건 분석)

  • 기온, 강수량 등 기후변화에 따른 식물 멸종이 증가함에 따라 기후변화에 민감한 북방계식물의 중요성이 증대되고 있는 실정이다. 본 연구 대상종의 큰금매화(Trollius chinensis)는 북한 및 북방계 식물로 고산 초지에서 자생하는 미나리아재비과 여러해살이풀이며, 기후변화에 직·간접적인 영향을 받고 있는 실정이다. 이에, 큰금매화의 발아조건을 규명하여 종 복원과 대량증식법 개발을 위한 기초자료를 확보하는 데 목적이 있다. 종자의 형태학적 특성은 원형~계란형(circular~obovate)에 작은 돌기가 겹겹이 있는(Colliculate) 형태였으며, 종피는 진갈색이다. 크기는 길이 1.37±0.741mm, 너비 0.81±0.531mm이었다. 종자의 단면을 확인한 결과, 배는 작은 선형태(Axial-miniature)이었으며, 종자는 91%의 충실도를 보였으며 TZ 테스트에서 모두 배가 전체적으로 붉은색으로 염색되어 양호한 활력을 보였다. 처리구별 발아실험 전 종자 소독은 파종 전 70% 에탄올 처리 후 2%의 NaOCl을 넣어 10분 동안 소독 후 멸균수로 5회 수세하고 멸균수를 제거하여 침지처리에 사용하였다. 발아실험 결과 종자의 최종발아율(FGP)은 15/6℃ GA3 1000mg·L-1 처리에서 40.0%로 가장 높았다. 평균발아일수(MGT)는 15/6℃ 무처리에서 22.8일로 가장 높았으며, 최종발아일의 평균은 약 9.5일로 확인되었다. 대부분 GA3 처리에서 최종발아율(FGP)이 20% 이상 높게 나타났으며, GA3 500mg·L-1 보다는 GA3 1000mg·L-1의 조건에서 3.3~5.5% 최종발아율(FGP)이 향상된 것으로 확인되었다. 한편, 온도 조건 25/15℃ 무처리에서는 발아가 확인되지 않았다.

Plant Regeneration from Unpollinated Ovary Culture in Allium tuberosum Rottl (부추의 미수분 자방배양에 의한 식물체 재분화)

  • This study was carried out to determine the optimal conditions for the production of plants derived from the unpollinated ovary culture of Chinese chive (Allium tuberosum Rottl.). The Chinese chive collected from Korea showed much higher frequency of plantlet formation than those from Japan in the culture of unpollinated ovaries. Among the collections, 'Youngiljaerae' showed the highest frequency of plantlet formation. The MS basal medium was superior to B/sub 5/ in plantlet formation. The ovaries inoculated on the 2,4-D-free medium were directly induced plantlets without callus formation. Floral parts inoculated as a unit played important roles in callus formation and plant production. The frequency of callus and plantlet formation was higher in the culture of ovary with anthers than that of ovary alone.
