Cytogenetic Analysis of Reciprocal Hybrids Reveals a Robertsonian Translocation between Mud Loach ($Misgurnus$ $mizolepis$) and Cyprinid Loach ($M.$ $anguillicaudatus$) (미꾸라지($Misgurnus$ $mizolepis$)와 미꾸리($M.$ $anguillicaudatus$) 및 유도된 종간 잡종의 세포유전학적 연구)

  • Reciprocal hybrids between the mud loach ($Misgurnus$ $mizolepis$) and cyprinid loach ($M.$ $anguillicaudatus$) were produced by artificial fertilization. The chromosome number of mud loach was 2n=48, consisting of 12M+4SM+32A chromosomes. The cyprinid loach has 2n=50, consisting of 10M+4SM+36A chromosomes. The chromosome numbers of the diploid reciprocal hybrids were 2n=49, consisting of 11M+4SM+34A chromosomes. All the karyotypes documented in this study had the same arm number of 64. There was no evidence of chromosomal polymorphisms or sex-related heteromorphism. The cytogenetic traits of the hybrid genotypes were intermediate between those of the parent species. In all genotypes, the chromosomal NORs localized to the terminal short arms of the same metacentric chromosome pair. These results suggest that Robertsonian translocation occurred between metacentric chromosome 1 of mud loach and acrocentric chromosome of cyprinid loach.

Differential Response of Soybean Cultivars to Alachlor, Linuron, and Metribuzin (Alachlor, Linuron과 Metribuzin의 약해반응에 대한 대두품종간 차이)

  • Forth soybean (Glycine max (L.) Merr.) cultivars were evaluated to determine the extent of crop injury caused by differential sensitivity of cultivars to two rates of alachlor, linuron, and metribuzin. Most cultivars were relatively tolerant of alachlor at 120 and 240g/10a and also linuron at 62.5 and 125g/10a. However, many cultivars were highly sensitive to metribuzin at 50 and 100g/10a. Some of the most sensitive cultivars were ‘Kyungnam 2’, ‘Toyosuzu’, ‘Noki 1’, ‘Iwade 2’, and ‘Hampton’. Metribuzin also showed a very narrow margin of safety to soybean cultivars.

한국산 검정망둑(Tridentiger obscurus)과 민물검정망둑(T.brevispinis)의 분류학적 고찰

  • Morphological and isozyme analyses were used to examine closely related species of Tridentiger obscurus and \ulcorner: brevispinis, a pair of cryptic species which has intricate problem with regard to species boundary, and to clarify the taxonomic position. In this study, it was revealed from their 17 allopatric streams that there are differences of band ornamentation on the lateral side and three genetic markers between two species. However, genetic relationship between them (S = 0.813, D = 0.192) was within the intraspecific level of similarity. However, at the sympatry of two species no hybrid was found except at Bangjuk stream where the electrophoretic hybrids with slight deficiency of heterozygosity were observed. It is shown that at sympatry there is a finer microenvironmental subdivision between two species at all populations including Bangjuk stream. To manifest the extent of assortative mating, four syrnpatric subpopulations at Bangjuk stream were analyzed for departure from Hardy-Weinberg equillibrium by F-statistics. All subpopulations were deviated significantly from the expectation under random mating. The evidences presented here supports the recognition of Tridentiger obscurus and T. brevispinis as semispecies in the late stage of reproductive isolation

Genetic Relationship within the melanogaster Complex of the Genus Drosophila (Drosophila melanogaster complex내의 유전적 유연관계에 관한 연구)

  • Four species belonging to the Drosophila melanogaster complex were examined genetically and morphologically to analyze interspecific relationships. Insemination rates ranged from 96% to 99% within species crosses, but interspecific crosses among the four species exhibited a great variations in the frequency of successful matings. D. melanogaster females mated relatively well with males of other species and D. sechellia males were more successful in mating with females of other species. In the crosses among D. simulans, D. mauritiana and D. sechellia, hybrid flies were fertile in females, but sterile in males regardless of reciprocal matings. The phenogenetically relationship between this complex and their hybrids were investigated by the comparison of sex comb tooth number and genital arch of male. They were controlled by polygenic factors on the chromosome of both parents. The effects of temperature on viability of hybrids between D. melanogaster females and D. simulans males were investigated for detection of genes concerning the speciation. The temperature sensitivity of the hybrid was mainly controlled by genes located on the X chromosome of D. simulans males.

  • In order to find the physiology of responess In order to find the physiology of responses and mechanisms of the actumlation of free proline at low temperatures and water stress. the temperature range, the effect of prolonged exposure to low temperatures, the light-dependence of proline accumulation in barle was significantly lower than in wheat. Proline accumulation at 5\circ C without water stress was shown to be light-dependent. Proline accumulation by water stress was not light-dependent at 2$0^{\circ}C$ but at 5$^{\circ}C$.

  • Sorghum seedlings lacking one of the phytochromes, phyB, have elongated internode, suggesting that they may have an alteration in gibberellin physiology. To test the possibility that phyB mutations affect seedling gibberellin perception and metabolism, the responsiveness of wild-type and phyB-1 seedlings to exogenous $GA_3$ was investigated. The phyB-1 showed higher internode elongation rate than the wild type in response to lower concentrations of exogenous $GA_3$ application, showing that the mutation causes an increase in responsiveness to GA. However, at the higher concentrations of $GA_3$ application, phyB-l and wild-type showed similar elongation rate, impling that responsiveness to higher concentrations of GA is not controlled by phytochrome. These results suggest that, although GAs are required for internode elongation, phyB does not act primarily by changing absolute levels of GAs but rather by decreasing seedling responsiveness to GAs at lower concentrations.

  • Comparative studies on external morphology and anatomical characters were conducted understand the interspecific relationships and possibly to identify the position of Hanabusaya asiatica as an endemic genus among 4 genera and 9 species in H. asiatica and its allied groups. Such morphological characteristics as root shape, leaf size, presence or absence of wings in petiole, corolla and calyx shape, and inflorescence and stigma shape were useful qualitative characters. The morphology of stem, peduncle, calyx lobe, leaf, petiole, and ovary were also good characters to be included in this identification study. In the anatomical observations in these organs, projecting degree of epidermis and number of projecting area rather than cross-sectioned internal structures were found to be valuable index for the classification among species. External morphology and anatomical characters support the position of H. asiatica as an endemic genus very well. Known as the most important external characters to taxonomists(Fedorov, 1957: Nakai, 1952: Hong, 1983), leaf arrangement, inflorescence, and corolla shape were also preven to be very useful characters to identify the genera and species in the H. asiatica and its allied groups.

Variation of Peroxidase Isoenzymes in Rhododendron plants (Rhododendron에 있어서 Peroxidase 동위효소(同位酵素)의 변이성(變異性))

  • Peroxidase banding patterns were obtained by disc electrophoresis of leaf extractions from flowering plants, R. mucronulatum, R. yedoense var. poukhanese, and R. schlippenbachii. A total of 21 peroxidase isoenzymes were resolved from these species, R. mucronulatum, P. yedoense var. poukhanese, and R. schlippenbachii. Five identical peroxidase isoenzymes were resolved in R. yedoense var. poukhanese and R. schlippenbachii. It means that there is a close relationships between two species. The peroxidase banding patterns of all three Rhododendron species were not correlated with each other on percentage total density of each band. The correlation coefficients for peroxidase band densities compared with leaf areas and leaf width/length ratios of Rhododendron plants were not significant at the 5% level. Wide variation was observed in R. yedoense var. poukhanese and R. schlippenbachii compared with R. mucronulatum.

  • This study was carried out to identify the genetic characteristics among isolates of Paecilomyces spp.and Cordyceps spp. by URP-PCR analysis. Twenty URP (universal rice primer) primers of 20 mer which were designed from repetitive sequence of rice, were used for producing PCR DNA fingerprints of the mushrooms. Of them, 5 URP primers, URP2F, URP2R, URP9F, URP4R, and URP17R amplified genomic DNA of the mushrooms with polymorphic PCR patterns. On isolates of Cordyceps militaris, primers URP1F, URP2R, URP6R and URP17R produced PCR polymorphic bands of 4 types. Isolates of Cordypces sp. that are isolated from different area of Korea were identical to isolate of C. militaris, while other species of Cordypces were different to the PCR profiles. However, the URP primers did not identify the polymorphism of PCR profile on isolates of P. japonica.

  • To evaluate the intraspecific variations among the Kasiogalpi(Eleutherococcus senticosus Max.) collections. randomly amplified DNA polymorphisms were examined. Twenty primers from 90 primers applied were selected. The range of polymorphism was $7.1{\sim}90.9%$ in 113 randomly and specifically amplified DNA fragments. Collections were divided into two major groups at the similarity coefficient value of 0.65. A considerable degree of genetic diversity was also detected among plants within the same collections. Deokyu (1, 2, 3, 4, 6), Bukhaedo (7, 8) and Odae (9, 10) col1ections showed higher degree of genetic similarity with a value of $0.65{\sim}0.86$, while Deokyu 5 showed much lower genetic similarity than other col1ections.
