분리균주에 의한 좁쌀주의 양조특성

  • 제주민속주인 좁쌀약주의 제조를 위하여 전국에서 수집된 35종의 누룩으로부터 분리하여 선발한 균주를 사용하여 균주와 원료배합비율에 따른 누룩을 제조한 후 양조과정에서의 발효특성을 검토하였다. 좁쌀주의 유기산 및 유리당의 분석결과 유기산은 lactic acid와 acetic acid가 대부분이었고, 이외에 citric acid, oxalic acid, tartaric acid, malic acid, fumaric acid도 일부 검출되었다. 유리당은 glucose와 arabinose, maltose가 많은 함량을 나타내었고, 이외에 xylose도 일부에서 검출되었다. 향기성분은 i-amyl alcohol, i-butyl alcohol, n-propyl alcohol 등이 주를 이루고 있었으며, 이외에 ethylacetate, acetaldehyde가 검출되었다. i-buthyl alcohol과 n-propyl alcohol은 대조구인 Dl이 다른 처리구보다 높게 나타났고, i-amyl alcohol은 K5 처리구가 높게 나타났다. 본 연구를 통하여 우수균주로 분리하여 제조한 누룩을 사용하여 좁쌀주를 양조하는 경우, 품질을 유지하면서 수율을 향상시킬 수 있었다.

제주민속 좁쌀약주 생산을 위한 균주선발

  • 제주민속주인 좁쌀약주의 제조를 위하여 전국에서 수집된 35종의 누룩으로부터 우수 곰팡이와 효모를 분리하였다. 수집된 누룩의 균수는 곰팡이가 6.4$\times$105~107개, 효모는 1.4$\times$104~107개로 나타났고, 이 중에서 곰팡이 169균주, 효모 103균주를 분리하였으며 전분당화력이 좋은 곰팡이 16균주와 효모 1균주를 선발하였다. 이 중에서 전분당화 균주의 효소활성을 분석한 결과 Aspergillus속으로 동정된 A8-3이 glucoamylase 활성, 액화력, xylanase 활성이 가장 높았고, Rhizopus속으로 동정된 B23-3은 당화력이 가장 우수하였다. 우수효모를 선발하기 위하여 pH, 무게 감량, 내당성, 내알코올성 등을 측정한 결과, Saccharomyces속인 A10-4가 가장 우수하였다. 같은 원료비율로 만든 누룩에 우수균주를 접종하였을 때, 단일 균주를 처리할 때보다 A8-3과 B23-3인 두 균주를 혼합하여 처리한 경우가 당화력이 높게 나타났다. 누룩을 원반형의 누룩과 팰릿(개량형) 형태로 만들어 혼합종균 배양액을 접종한 후 당화력을 측정한 결과, 비슷한 당화력을 나타내었다. 개량형 누룩을 사용하여 양조하는 경우, 좁쌀주 양조에 발효효율을 높일 수 있을 것으로 판단되었다. 수집된 누룩은 수분이 10~13%, 총당은 55~70%, 조단백질은 10~18% 조지방은 0.2~l.0%, 회분은 1.8~2.1%이었다. 본 연구에서 제조한 누룩은 수분이 12~15%, 총당은 61~7l%, 조단백질은 15~20%, 조지방은 0.4~1.5%, 회분은 1.1~1.5%이었다.

Fermentation Characteristics of Jeju Foxtail Millet-wine by Isolated Alcoholic Yeast and Saccharifying Mold (제주좁쌀약주의 품질개선을 위한 선발균주에 의한 양조특성)

  • In order to improve foxtail millet wine, a traditional Jeju cereal wine, fermentation characteristics of millet Yakju with different types of Nuruks prepared using isolated Aspergillus sp. and Rhizopus sp. were investigated. When the millet wine was brewed with the Nuruk prepared in this study, the combination ratio of wheat flour: barley : wheat bran : millet = 8 : 1 : 1 : 0 (pellet) showed the highest level of alcohol concentration, and a more favorable score than Kuksundang Nuruk in sensory evaluation. The main organic acids in millet wine were lactic and acetic acids, and the minor organic acids were fumaric, oxalic, citric and malic acids. Analysis of sugar compositions showed that glucose, arabinose, and maltose were present in decreasing order, and that xylose was also detected. Flavor components of millet wine were mainly iso-amyl, iso-butyl and n-propyl alcohols. Ethylacetate and acetadehyde were also detected. The contents of iso-butyl and n-propyl alcohols were higher in the millet wine prepared with Kuksundang Nuruk than those prepared with other Nuruks.

Zymological Properties of Foxtail Millet Wine-making by Isolated Strains from Nuruk (누룩에서 분리한 우수균주에 의한 좁쌀주의 양조특성)

  • In order to improve the quality of foxtail millet wine, zymological properties by isolated strains from Nuruk were investigated. Saccharomyces sp Y5-1 as brewing yeast, Aspergillus sp. M6-3, Aspergillus awamori 6970, and Aspengillus usamii mut. shirousamii 6959 (KCTC) as saccharifying molds were used, respectively. Acid content, soluble solids, color (b) and alcohol contents were increased during fermentation. Ethanol concentration of millet wine made with Nuruk by Aspergillus awamori 6970 and Aspergillus usamii mut. shirousamii 6959 were higher than the other, 10.6 and 10.1% respectively. Citric acid was only detected on $1{\sim}2$ day starting fermentation. Oxalic acid, lactic acid and acetic acid of millet wine were high in the wine made of Nuruk by Aspergillus usamii mot. shirousamii 6959, Aspergillus awamori 6970 and traditional Nuruk, respectively. During fermentation, glucose and xylose was higher than the others. Xylose was increased, but most of other sugar were decreased during fermentation. Acetone, ethyl acetate, methanol, ethanol, n-propanol, iso-buthanol and iso-amyl alcohol were detected In the wine made with Nuruk by Aspergillus awamori and Aspergillus usamii mut. shirousamii, iso-imyl alcohol and ethanol were high. On sensory evaluation, the wine made with Nuruk by Aspergillus usamii mut. shirousamii was the best on color and taste.

Diffuse Large B-Cell Lymphoma Manifesting as Miliary Nodules in the Lung: A Case Report (폐의 좁쌀 결절로 발현된 림프종 사례: 증례 보고)

  • Malignant lymphoma has various pulmonary manifestations on chest CT, including nodules, masses, areas of consolidation, and ground-glass opacity. These presentations can pose a diagnostic challenge, as they mimic other disease patterns. Herein, we report a case of diffuse large B-cell lymphoma (DLBCL) manifesting as miliary nodules in a 67-year-old male initially presenting with dyspnea and fever. Radiologic findings included diffuse, bilateral, multiple tiny nodules consistent with metastasis, miliary tuberculosis, and fungal infection. However, further investigations, including laboratory tests, imaging, and biopsies, led to the diagnosis of DLBCL involving the lungs. Herein we reported a rare case of lymphoma involvement of the lung presenting as miliary nodules. Accurate diagnosis relies on a comprehensive evaluation of the clinical history, physical features, laboratory test results, and imaging findings.

Screening of Brewing Yeasts and Saccharifying Molds for Foxtail Millet-Wine Making (제주민속 좁쌀약주 발효를 위한 우수균주의 선발)

  • In order Nuruk to improve the quality of millet wine, a traditional Jeju cereal wine, yeasts and molds were isolated from 35 kinds of Nuruk collected nationwide. Isolated strains were screened for saccharification of starch and brewing of millet wine. Fermentation characteristics of millet wine with different types of Nuruk were also investigated. The average number of microbial populations in the Nuruk were $6.4{\times}10^5{\sim}4.5{\times}10^7\;cfu/g$ for molds and $1.4{\times}10^4{\sim}7.7{\times}10^7\;cfu/g$ for yeasts. Among the 169 strains of molds and 103 strains of yeasts, 16 strains were screened for saccharifying activity on starch as a substrate, and one yeast strain was screened for the brewing of millet wine. A8-3, supposed as Aspergillus sp., showed the highest enzyme activities of glucdamylase, ${\alpha}-amylase$ and xylanase while B23-3 strain, supposed as Rhizopus sp., showed the highest saccharifying activity. A10-4, supposed as Saccharomyces sp., showed the highest level of weight loss from $CO_2$ evolution, sugar and alcohol tolerance during fermentation. When the Nuruk was made after inoculation with the selected strains, saccharifying activity was higher for the co-cultivation of A8-3 and B23-3 than individual cultivation of each strain. Similar saccharifying activities were shown in both disc-type and pellet-type Nuruk. It was suggested that pellet-type Nuruk could improve fermentation yield. The collected Nuruk consisted of $10{\sim}13%$ moisture, $55{\sim}70%$ total sugar, $10{\sim}18%$ crude protein, $0.2{\sim}1.0%$ crude fat and $1.8{\sim}2.1%$ ash. The Nuruk made in this study was composed of $12{\sim}15%$ moisture, $61{\sim}71%$ total sugar, $15{\sim}20%$ crude protein, $0.4{\sim}1.5%$ crude fat and $1.1{\sim}1.5%$ ash.

Flavor Analysis of Commercial Korean Distilled Spirits using an Electronic Nose and Electronic Tongue (전자코와 전자혀를 이용한 시판 증류주의 향미특성 분석)

  • The objective of this study was to analyze the flavor patterns of commercial Korean distilled spirits using a mass spectrometry-based electronic nose and an electronic tongue. Thirteen types of domestic distilled spirits were used for analysis. The flavor patterns of the distilled spirits were clearly identified through discriminant function analysis of the raw materials. The obtained data of the electronic nose analysis showed that the flavor pattern of the distilled spirits aged in oak barrels was not significantly different from that of the general distilled spirits. According to the results of the electronic tongue, the distilled spirits made with rice had a stronger flavor than those made with foxtail millet. Compared to the sensitivity value of each taste sensor, the distilled spirits showed various intensities of sourness and umami. Therefore, an electronic nose and electronic tongue could be successfully used for quality evaluation and new product development of distilled spirits.

Molluscan Death Assemblages and Their Ecological Implications on a Tidal Flat, Inchon, Korea (인천연안 간석지산 연체동물 유해집단(遺骸集團)의 구조와 생태학적 의미)

  • Temporal changes, density, calcimass, mode of occurrence, size-frequency histogram and survivorship curves were studied for the molluscan death assemblages on a macrotidal flat, Inchon, Korea. The living and death assemblages were compared on the basis of the taxonomic compositions and their numerical abundances. A total of 28 species (16 taxa in gastropods and 12 in bivalves) were identified. Most of the dead shells were the species inhabiting that intertidal mud flat. Species diversity was higher in gastropods than in bivalves. Seasonal variation of the calcimass was influenced by the mactrid bivalve, Mactra veneriformis. Shell-boring naticid gastropods preferred selecting the umbo areas of various valves. Size-frequency distribution and size-specific survivorship curves were analyzed for the dominant species. Survivorship curve of the tellinid bivalve, Morella rutila was convex-up in shape, which is congruent with the expected equilibrium condition but indicates higher mortality in winter. Whereas the survivorship curves of Mactra veneriformis and Reticunassa festiva were semi convex-up with dual modes, suggesting a disequilibrium due to the changes in recruitment and seasonal mortality. This study suggests that the dead shell assemblages may be useful in getting population information like live molluscan assemblages, if solved for several problems related to taphonomic processes.

Macrobenthos Assemblages on the Rocky Shores of Wando and Jodo islands in Dadohaehaesang National Park in Summer (다도해해상국립공원 완도·조도 암반조간대 하계에 출현한 대형저서무척추동물)

  • To determine the composition of benthic species on the southwest coast, a macrobenthos study was conducted at 18 sites on the islands of Wando and Jodo for seven summers (2014-2021) from June to September. Two hundred and twenty species(645 ind./m2) were collected on the rocky shores of Wando and Jodo. A similar number of species was observed in Wando (165 species) and Jodo (167 species). The mean density was higher in Wando (726 ind./m2) than in Jodo (564 ind./m2). The number of species was the highest at Site 15 (116 species), and the density was highest at Site 7 (1,664 ind/m2). The dominant species were Nodilittorina radiata with 175 ind./m2 (27.1%), Littorina brevicula with 97 ind./m2 (15.1%), and Heminerita japonica with 41 ind./m2 (6.3%). The climate change indicator species, Tetraclita japonca, appeared in 15 sites, and the endemic species, Ptilohyale bisaeta, appeared in some sites. Fusinus spectrum, which appears on the Red List as a Near Threatened (NT) species, the lowest level of extinction risk, appeared in Jodo. The macrobenthos of Wando and Jodo demonstrated the general characteristics of rocky shores in Korea. Based on the tides and substrate, the rocky shores revealed a vertical distribution trend where Nodilittorina radiata, Littorina brevicula, Chthamalus challengeri, and Reishia clavigera were dominant. Wando and Jodo exhibited similar habitat distribution patterns.

A Study on Food Resource and Utilization of Artificial Nest of Wild-birds in Urban Woodland (도시공원내 야생조류의 먹이자원 및 인공새집 이용에 관한 연구)

  • Preference of food plants of wild birds was shown in the order of Pinus densiflora, Celtis sinensis, Celastrus orbiculatus, Rosa multiflora, Taxus cuspidata, and Euonymus japonica, etc. The wild birds preferred the animal food(92.2%) rather than the vegetable food(32.7%) in breeding season(May through June), but preferred both animal and vegetable foods in non-breeding season(November through December). The rates utilzing artficial nest were 77.5%, 50.3%, 44.1% and 42.2% in Nosan, Sanho, Chinju-castle and Bibong parks, respectively. The wild birds used more the 3cm hole than 5cm hole.
