Identification of Meiotic Recombination Intermediates in Saccharomyces cerevisiae (효모 감수분열과정에서의 유전자 재조합 기전 특이적 DNA 중간체의 구조 변화)

  • During meiosis, genetic recombinants are formed by homologous recombination accompanying with the programmed double-strand breaks (DSBs) and strand exchanges between homologous chromosomes. The mechanism is generated by recombination intermediates such as single-end invasions (SEIs) and double-Holliday junctions (dHJs), and followed by crossover (CO) or non-crossover (NCO) products. Our study was focused on the analysis of meiotic recombination intermediates (DSBs, SEIs, and dHJs) and final recombination products (CO and NCO). We identified these meiotic recombination intermediates using DNA physical analysis under HIS4LEU2 "hot spot" system in budding yeast, Saccharomyces cerevisiae. For DNA physical analysis, when the hot spot locus is recognized by restriction enzyme from synchronous meiotic cells, the fragmented DNA that are forming recombination intermediates can be detected and quantified through Southern hybridization analysis. Our study suggests that this system can analyze the structural change of recombination intermediates during DSB-SEI transition, double-Holiday junctions and crossover/non-crossover products in meiosis.

An Analysis on Weighing the Decision Making Factors of Ship Investments for Korean Shipping Companies (우리나라 해운기업의 선박확보 투자 의사결정요인에 관한 연구)

  • Korean shipping industry is ranked the fifth largest in the world in terms of deadweight tonnage after Greece, Japan, Germany and China with 55 million DWT as of year 2011, and its size of foreign exchange earning marked 30 billion US dollars. In respect of volume of seaborne trade, it has handled 99% of import and export cargoes. Korean shipping fleets have increased from 420 to 979 ships between year 2003 to year 2011. By reviewing through the relating literatures, it has been found that Shipping Funds under Ship Investment Company Act, and Tonnnage Tax System, worked as positive influences to increase the Korean shipping fleets. However, there is scant of research to examine the following two points: 1) weighing the decision making factors of ship investments for Korean shipping companies, and 2) weighing the influential factors of government shipping policies. In this respect, the aim of this study is to evaluate 8 decision making factors of ship investments for Korean shipping companies, and 8 influential factors of government shipping policies. For weighing the factors, the fuzzy methodology was adopted. As the results, for the side of decision making factors of ship investments, 'shipping market conditions and future prospects', 'ship's price and future prospects, and 'securing cargoes and future prospects' are ranked as top 3 factors. For government shipping policies side, 'shipping finance provided by lease companies', 'establishment of Korea Shipping Guarantee Fund', and 'establishment of Korea Shipping Finance Corporation' are verified as the important factors.

Development of Signal Control Strategy for Oversaturated Intersections Using Wayside Video Equipment (노변영상장비를 활용한 과포화 신호제어전략 개발)

  • The conventional real-time signal control strategy for oversaturated situation generally requires a number of detectors at the intersection in order to identify the queue length at each approach. Also, existing strategy for the spillback has limited effect due to the temporal spillback control which only reduce the green split at the approach. In this study, a signal control system utilizing the imagery information from ITS roadside equipment is developed for operation of oversaturated intersections. The strategy calculates the saturation ratio based on the queue length extracted from ITS RSE, and designs the signal control variables according to the saturation ratio. The signal control strategy is divided into two phases: oversaturated and supersaturated. In oversaturated conditions, timing plan for main approach is optimized by the queue length. In oversaturated conditions where spillback might occur, the signal timing is designed in order to avoid the spillback. To increase field adaptability, the strategy is designed to adjust the split length, all-red-time, and clearance time, and keep the major signal control variables intact. The result of the simulation shows that in oversaturated conditions, the improvement is similar to the real-time signal control system. In case of, oversaturated conditions, however, the effect of the strategy is superior to that of a real-time system.

  • 영광 3/4호기 mid-loop 운전중 잔열제거(RHR) 기능 상실사고시 열수력적 현상을 최적 전산코드인 CATHARE2를 이용하여 해석하였다. 이러한 사고시 열수력적 현상은 일,이차측 냉각재 방출유로와 계통내 비응축성 가스의 거동에 의해 크게 영향을 받는다. 본 연구에서는 2개의 경우를 모의하였는데, 하나는 계통내 방출유로가 있는 경우이며 다른 하나는 방출유로가 없는 경우를 계산하였다. 이 때 사용된 가정은 다음과 같다. (가) 계통은 부분충수 운전 상태로 상부에 비응축성 가스나 증기로 가득 차 있다. (나) 증기발생기는 1대만이 이용 가능하고 이차측은 습식보관 상태이며, 보조급수는 공급되지 않고 이차측 압력은 대기압 상태이다 (다) 사고는 원자로 정지후 2일후 발생한다. 이와같은 조건하에서 사고시 계통 최대압력은 방출유로가 있는 경우 사고후 6,000 초에 0.27 MPa이며, 방출유로를 통한 유량은 총 2.4 kg/s이다. 이 방출유량을 외삽하여 계통수위가 고온관 바닦까지 도달하는데 걸린 시간은 사고후 약 5.67시간이다. 증기발생기 U-튜브를 통한 열전달에 의해 이차측 증기 발생으로 이차측 수위가 하락하면 증기발생기 reflux cooling은 제한을 받을 수 있다. 이 경우 이차측 수위가 U-튜브의 active 영역 상부까지 도달하는데 걸리는 시간은 사고후 약 10시간으로 계산되었다. 그러므로 이 경우 보조급수 공급 여유시간보다 노심 노출시간이 더 빨리 도달하여 노심을 손상시킨다. 사고시 수위지시계는 계통감압에 큰 영향을 주지 못하기 때문에 가능한 빨리 닫아 계통 inventory를 유지하는 것이 이차측 보조급수공급보다 우선한다.합한 설계방안으로 분석되었다.크다는 단점이 있다.TEX>$_2$O$_3$ 흡착제 제조시 TiO$_2$ 함량에 따른 Co$^{2+}$ 흡착량과 25$0^{\circ}C$의 고온에서 ZrO$_2$$Al_2$O$_3$의 표면에 생성된 코발트 화합물을 XPS와 EPMA로 부터 확인하였다.인을 명시적으로 설명할 수 있다. 둘째, 오류의 시발점을 정확히 포착하여 동기가 분명한 수정대책을 강구할 수 있다. 셋째, 음운 과 정의 분석 모델은 새로운 언어 학습시에 관련된 언어 상호간의 구조적 마찰을 설명해 줄 수 있다. 넷째, 불규칙적이며 종잡기 힘들고 단편적인 것으로만 보이던 중간언어도 일정한 체계 속에서 변화한다는 사실을 알 수 있다. 다섯째, 종전의 오류 분석에서는 지나치게 모국어의 영향만 강조하고 다른 요인들에 대해서는 다분히 추상적인 언급으로 끝났지만 이 분석을 통 해서 배경어, 목표어, 특히 중간규칙의 역할이 괄목할 만한 것임을 가시적으로 관찰할 수 있 다. 이와 같은 오류분석 방법은 학습자의 모국어 및 관련 외국어의 음운규칙만 알면 어느 학습대상 외국어에라도 적용할 수 있는 보편성을 지니는 것으로 사료된다.없다. 그렇다면 겹의문사를 [-wh]의리를 지 닌 의문사의 병렬로 분석할 수 없다. 예를 들어 누구누구를 [주구-이-ν가] [누구누구-이- ν가]로부터 생성되었다고 볼 수 없다. 그러므로 [-wh] 겹의문사는 복수 의미를 지닐 수 없 다. 그러면 단수 의미는 어떻게 생성되는가\

Analysis of the Effectiveness of Computational Thinking-Based Programming Learning (컴퓨팅 사고력 기반 프로그래밍 학습의 효과성 분석)

  • It is necessary for us to be equipped with problem-solving ability, logical thinking ability, and convergence thinking ability in the digital information age. To have these abilities, modern people in the $21^{st}$ century should be educated to develop the computational thinking ability. However, it is difficult to cultivate the computational thinking ability in current computer education curriculum. Therefore, this paper aims to propose a programming-based computer educational program to make students better understand the principles of computer science and enhance student's computational thinking ability escaping from current fragmentary and limited computer education. In order to verify the effectiveness of the educational program, students in total 42 middle school (22 students in 1st and 2nd grades and 20 in 3rd grade) were applied to the educational program for twenty-four hours on 12 weeks and then we obtained a positive result that might bring improvement on the logical thinking ability and creative problem-solving skills. As a result, this study presents the possibility of its field applications of computational thinking-based programming learning and analyzed the effectiveness. therefore, it has a notable point of presenting the development direction of a variety of education programs for enhancing the computational thinking in the future.

  • Portions of chloroplast genes(psbD and rbcL) were amplified from Pinus thunbergii(Japanese black pine : black pine) and Pinus densiflora(Japanese red pine : red pine) by PCR and digested by a restriction enzyme, HaeIII, respectively. Two species specific cpDNA markers were identified. With the observed cpDNA markers, paternal inheritance of cpDNA in pine hybrids was verified in an artificial hybrid family between black pine(Chollanam 37) and red pine(Chungchongbuk 3). On the basis of paternal inheritance of chloroplast genome in a hybrid, 2 portions of cpDNA amplified from 115 individuals of Pinus densiflora for. erecta were screened to detect any traces of black pine specific cpDNA markers in P. densiflora for. erecta which has been postulated as an introgressive hybrid between red pine and black pine(Hyun el al., 1967). All the analyzed individuals of Pinus densiflora for. erects revealed the identical profiles of HaeIII digested psbD and rbcL genes to red pine. This result suggests that there is no introduced chloroplast genome of black pine in Pinus densiflora for. erecta and that there is no concrete evidence of treating P. densiflora for, erecta as an introgressive hybrid between red pine(♀) and black pine(♂).

Summary on the Dinoflagellate Cyst Assemblages of Modern Sediments from Korean Coastal Waters and Adjoining Sea (한국연안해역 와편모조류 시스트 연구에 관한 고찰)

  • To investigate the research state and characteristics of dinoflagellate cyst along the Korean coastal waters, this work analyzed 45 studies conducted in the Korean coasts and basin, the south of Jeju, East China Sea including some part of the Yellow Sea. It was found that the research on dinoflagellate cyst began in the later half of the 1980s in Korea, and that the research areas focused on eutrophied regions where a red tide occurred or on the seas where coastal development occurred. In other words, no research on the East Sea was found. In terms of research contents, there were various studies on analysis of cyst assemblage, tracking of the changes in marine environment like process of eutrophication, roles as a seed population in occurrence of a red tide, creation of cyst by use of sediment trip and tracking of its changes, morphological changes by the change of pH concentration in the hypoxia zone of eutrophied region, germination of a specific species, and other international-level studies. Species composition and cell density also varied in the Korean coastal waters. However, much research on dinoflagellate cyst in Korea is not systematic, and is fragmentary and individual and hencesuffers from great limitations. The research results so far have focused only on specific regions, and sporadic research had been conducted by specific research groups. Dinoflagellate cyst of surface sediments represents the accumulation index of pelagic environment in the sea and is a useful index to understand the marine environment efficiently. In conclusion, Korea requires organized human resources and collaborative research on dinoflagellate cyst which in turn should be considered as a component of marine ecosystem and as an essential aspect of marine biology.

  • Phytopathogenic Erwinia carotovora subsp. carotovora (Ecc) LY34 causes plant tissue maceration by secretion of pectinolytic enzymes such as pectate Iyase (PL) existed as multiple isoenzyme form. Genomic DNA from Ecc LY34 was digested with Sau3Al and ligated into the BamHI site of pBluescript ll $SK^+$. Among them, a clone hydrolyzing polypectate was selected and its DNA was digested with BamHI. Through the subsequent subcloning the resulting 3.1 kb fragment, corresponding to a peICI, was subcloned into pLYPA 100. The structural organization of a peICI gene encoding a 374 amino acid residues consists of an open reading frame (ORF) of 1,122 bp commencing with a ATG start codon and followed by a TAA stop codon. PeICI contained a typical prokaryotic signal peptide of 22-amino acid. Since the deduced amino acid sequences of PeICl protein was very similar to those of PelIII of Erwinia carotovora subsp. carotovora, and to those of Pel3 of Erwinia carotovora subsp. atroseptica, and to those of PeIC of Erwinia carotovora subsp. carotovora, it belong to the same family PLbc group. The 374-amino acld PeICI had a calculated Mr of 40,507 and pI of 7.60.

  • Favorable policies for freight vehicles (i.e., trucks), such as exclusive "Truck-ways," have been proposed, and at the same time restricted policies for freight vehicles have been suggested because transportation of goods by truck aggravates traffic congestion and environmental conditions in urban areas. This study discusses the limitations of assessment processes for freight vehicle route management systems which have been carried out from limited perspectives such as physical distribution efficiency and traffic circulation. The objective of this study is to propose an assessment method of truck route management systems in urban areas which includes more varied criteria. This research modified the two existing assessment indices of physical distribution efficiency and traffic circulation. In addition two new assessment indexes are proposed: (i) exposure indices for population directly influenced by nitrous oxide, a representative pollutant, and (ii) a friction index between trips for attending elementary school and freight vehicle trips. It was found that from the perspective of physical distribution efficiency and traffic circulation, the "before" alternative is better than "after". However, from the aspect of traffic safety and environmental impacts the "after" alternative is better than "before". It is expected that the proposed evaluation method in this study would be useful when identifying reasonable policies for truck route management in urban areas.

  • 본 연구는 한국의 도시재생과 스마트 물 관리의 정책 분석을 위하여 도시재생과 스마트 물 관리의 등장 배경, 주요 현안 및 연계성을 모색하고 도시재생방안으로서 스마트 물 관리체계의 가능성을 검토하는 것을 목적으로 한다. 1950년대의 도시재건(Urban Reconstruction)과 1970~80년대의 도시재개발(Urban Renewal, Urban Redevelopment) 등의 정비 사업은 물리적 환경정비에 초점을 맞추었다. 그러나 1990년대 환경문제가 세계적 이슈로 등장하면서 교외지역 난개발 문제에 대한 대응책이 필요하게 되었고 도시의 물리 환경적, 산업 경제적, 사회 문화적 측면을 부흥시키는 도시재생 접근법이 출현하였다. 한국 정부는 2017년부터 시작한 '도시재생 뉴딜사업'의 일환으로 스마트 기술을 적용한 도시재생사업을 통해 스마트도시 선도국가 도약과 세계적 흐름에 부합하는 도시성장을 기대하고 있다. 1980년대 초 등장한 스마트 기술은 2000년대 들어와 스마트 도시, 스마트 인프라, 스마트 그리드 등의 분야로 확대, 진보하였다. 물 분야의 스마트 기술은 2009년 스마트워터그리드 이니셔티브(Smart Water Grid Initiative)의 발족과 함께 IBM, CISCO, Intel 등의 IT 기반 물 관리 워킹그룹 형성, Suez, Veolia, Siemens 등 수처리 기업의 스마트워터그리드 분야 진출 모색과 함께 발전하기 시작하였다. 이후 2012년 유엔 스마트 물 관리 포커스 그룹(ITU-T SG 5)의 스마트 물 관리 표준화 연구가 착수되었고 한국은 국토교통부 건설교통기술 연구 개발사업 중 하나로 스마트 물 관리 장기 연구 사업을 시작하였다. 스마트 물 관리는 수자원 및 상하수도 관리의 효율성 제고를 위하여 스마트 미터, 센서, 디지털지도제작 등 ICT를 이용한 차세대 물 관리시스템이라고 정의할 수 있다. 구체적인 대상 분야를 고려한다면 하천수, 우수, 지하수, 하폐수처리수, 해수담수 등 다양한 수자원의 관리, 물의 생산과 수송, 사용한 물의 처리 및 재이용 등 물 관리 전 분야를 포함한다. 그러나 스마트 물 관리의 용어와 개념을 처음으로 도입한 미국 등 선진국과 관련기업들은 스마트 물 관리를 '스마트 워터 미터, 센서, 첨단 모델링, 수문 지도제작, 스마트 관개농업, 자동화 로봇 등 다양한 기술을 통합적으로 운영하는 지능적인 수자원 관리를 위한 정보네트워크'로 정의한다. 일찍이 도시재생으로의 패러다임 전환을 실시한 영국 및 일본과 달리 한국의 도시재생은 개념, 구성요소, 범위, 사업방식 등의 여러 가지 측면에서 아직 형성단계에 있다. 또한 한국의 스마트 물 관리 논의는 개념정립 측면에서 심층적 논의가 거의 부재하였다. 기존의 논의들은 수자원 혹은 상하수도서비스 분야에서의 연구결과와 기술개발성과를 기계적으로 적용하고 확대하는 측면만을 부각시켰다. 그러나 이와 같은 스마트 물 관리에 대한 논의는 정보통신기술과 물 관리 서비스를 단편적으로 연결하고 적용범위를 제한할 수도 있다는 점에서 한계성이 있다. 본 연구는 국내외 문헌검토를 바탕으로 한국의 도시재생과 스마트 물 관리의 정책을 분석하고 지금까지 별개로 간주된 두 개념의 장점을 융합하여 향후 지속가능한 도시개발 사업으로서의 가능성을 검토하고자 한다.
