Sulfonylurea (SU)-resistant weeds of eight annual weeds, Monochoria vaginalis, Scirpus juncoides and Cyperus difformis, etc., and four perennial weeds, Scirpus planiculmis, Sagittaria pigmaea, Eleocharis acicularis and Sagittaria trifolia as of 2013 since identification Monochoria korsakowii in the reclaimed rice field in 1998. And the resistant Echinochloa oryzoides to ACCase and ALS inhibitors has been confirmed in rice fields of the southern province, Korea in 2009. In the beginning, the M. vaginalis, S. juncoides and C. difformis of these SU-resistant weeds were rapidly and individually spreaded in different fields, however, these resistant weeds have been occurring simultaneously in the same filed recently. The resistant biotype by weed species demonstrated about 10-to 1,000-fold resistance, based on $GR_{50}$ values of the SU herbicides tested. And the resistant biotype of E. oryzoides to cyhalofop-butyl, pyriminobac-methyl, and penoxsulam was about 14, 8, and 11 times more resistant than the susceptible biotype base on $GR_{50}$ values. The products and applied area of SU-included herbicides have been increased rapidly, and have accounted for about 69% and 96% in Korea, respectively. In Korea, the main cause of SU-resistant weed is extensive use of these herbicides. The top ten herbicides by applied area were composed of all SU-included herbicides by 2003. The concentrated and successive treatment of ACCase and ALS inhibitors for control of barnyardgrass in rice led up to the resistance of E. oryzoides. Also, SU-herbicides like pyrazosulfuron-ethyl and imazosulfuron which effective to barnyardgrass can be bound up with the resistance of E. oryzoides. The ALS activity isolated from the resistant biotype of M. korsakowii to SU-herbicides tested was less sensitive than that of susceptible biotype. The concentration of herbicide required for 50% inhibition of ALS activity ($I_{50}$) of the SU-resistant M. korsakowii was 14-to 76-fold higher as compared to the susceptible biotype. No differences were observed in the rates of [$^{14}C$]bensulfuron uptake and translocation. Acetolactate synthase (ALS) genes from M. vaginalis resistant and susceptible biotypes against SU-herbicides revealed a single amino acid substitution of proline (CCT), at 197th position based on the M. korsakowii ALS sequence numbering, to serin (TCT) in conserved domain A of the gene. Carfentrazone-ethyl and pyrazolate were used mainly to control SU-resistant M. vaginalis by 2006 in Korea. However, the alternative herbicides such as benzobicyclone, to be possible to control simultaneously the several resistant weeds, have been developing and using broadly, because the several resistant weeds have been occurring simultaneously in the same fieled. The top ten herbicides by applied area in Korea have been occupied by products of 3-way mixture type including herbicides with alternative mode of action for the herbicide resistant weeds. Mefenacet, fentrazamide and cafenstrole had excellent controlling effects on the ACCase and ALS inhibitors resistant when they were applied within 2nd leaf stage.
Tissue-type plasminogen activator (t-PA) is a thrombolytic agent important in fibirn clot lysis. T-PA causes fibirn-specific plasminogen activation. Six binary vectors harboring t-PA and its derivative genes were cloned and expressed in transgenic alfalfa plants. The insertion of the t-PA and its derivative genes in genomic DNA of alfalfa plants was confirmed by PCR. The presence of the t-PA and its derivative transcripts in total RNAs of the transgenic alfalfa leaves was verified by RT-PCR. ELISA experiments demonstrated that the highest level of recombinant t-PA expression was $75.1{\mu}g$/ total soluble protein (mg) in alfalfa plants. The amount of recombinant t-PA and its derivative proteins in transgenic plants was estimated to range from 9.7 to $39.5{\mu}g$/ total soluble proteins (mg). Western blot analysis of the transformed alfalfa leaves revealed bands of approximately 68-kDa recombinant t-PA and its derivative proteins. The fibrinolysis of recombinant t-PA and its derivative proteins was confirmed by a fibrin plate assay (range from 3.2 to 8.1 cm). The results presented provide information for the development of an additional production of recombinant human proteins having pharmaceutical applications using transgenic plants.
Papaya (Carica papaya L.) is one of the crops widely planted in tropical and subtropical areas. The papaya fruit has low calories and are plentiful in vitamins A and C and in minerals. A major problem in papaya production is a plant disease caused by the papaya ringspot virus (PRSV). The first PRSV-resistant GM papaya expressing a PRSV coat protein gene was developed by USA scientists in 1992. The first commercial GM papaya cultivars derived from the event was approved by the US government in 1997. Development of transgenic papayas has been focused on vaccine production and limited agricultural traits, including insect and pathogen resistance, long shelf life, and aluminum and herbicide tolerance. Approximately 17 countries, including the USA and China, produced transgenic papayas and/or commercialized them, which provoked studies on biosafety assessment and development of GM-detection technologies. For the biosafety assessment of potential effects on human health, effects of long-term feeding to model animals have been studied in terms of toxicity and allergenicity. Studies on environmental safety assessment include influence on soil-microbial biodiversity and transfer to soil bacteria of GM selection markers. Many countries, such as Korea, the European Union, and Japan, that have strict regulations for GM crops have serious concerns about unintended introduction of GM cultivars and food commodities using unauthorized GM crops. Transgene- and/or GM event-specific molecular markers and technologies for genomics-based detection of unauthorized GM papaya have been developed and have resulted in the robust detection of GM papayas.
For developing standard method for diease, pest and weed control in environmental friendly 'Ssam' vegetable cultivation, this study was carried out to investigating agriculture material use in organic agriculture and no pesticide cultivation for lettuce, kale, leafy perilla and korean cabbage. The 28.6% of investigated farmer carried out seed sterilization by seed selection with salt solution and soaking in chitosan that not validated. For raising seedling periods, the 55.6% of farmer did not use environmental-friendly agriculture material for, diease control and the 50% of farmer used one time for. pest control. Therefore, the control of disease and pest could be achieved with one or two times use of environmental-friendly agriculture material. Seed sterilization was carried out by soil solar sterilization, one time per year in 71.4% of farmer. Weed was controled by black PE film for weed germination of furrow in many farmer, by man-power weeding for weed of ridge in 85% of farmer and by machine weeding and mulching in some farmer. During cultivation period, the major pest were Aphis gossypii in lettuce, Plutella xylostella in kale, Plutella xylostella and Phyllotreta striolata (Fabricius) in korean cabbage and Pyrausta panopealis (Walke) in feat perilla. The many farmers used environ-mental-friendly agriculture material for control of pest over 10 times for spring season, and more used sold materials in market than home-made materials. In result, it needs to develop standardized method and validate cultivation methods for control of disease and pest, and seed sterilization treatment environmental-friendly 'Ssam' vegetable.
To elucidate occurrence patterns of $C_4$ plants in agroecosystems, we surveyed flora inside fields and embankments in 222 sites in farming lands in summer and fall, 2013. We also observed Kranz anatomy and analyzed carbon stable isotope to classify photosynthesis types. Our study observed total 54 species of $C_4$ plants in 33 genera and 9 families of vascular plants besides cultivated crops occurring in farming fields. The most common species was Portulaca oleracea L. followed by Digitaria ciliaris and Echinochloa crusgalli (L.) P. Beauv. var. crusgalli among the $C_4$ plants. The ratios of $C_3$ and $C_4$ species number to total number of species tended to maintain constant regardless of survey times and habitat types. The proportion of $C_4$ species to the total number of species were more influenced by the number of $C_3$ species than those of other types in the surveyed sites. Sizes of cultivation areas did not affect the proportions of $C_4$ plants. Increasing number of simultaneous weeding methods, especially with cutting method, appeared to decrease the number of $C_3$ plants. On the other hand, as the number of cultivated crops increased, both $C_3$ and $C_4$ plants increased but the proportions of $C_4$ species remained. We interpret our results that proportion of $C_4$ species in agroecosystems, appear to be dependent on the number of $C_3$ species, which in turn, appear to be affected by cultivation methods.
Creeping bentgrass (Agrostis palustris Huds.) had been the problematic weed for Kentucky bluegrass (Poa pratensis L.) fairway since it shows light green color all year. Experiment was carried out to determine the best herbicides combination to control creeping bentgrass in Kentucky bluegrass. fairway without injury. To investigate the efficacy of herbicides, five post-emergence herbicides of asulam WG ($87.6\%$), imazaquin SL ($20\%$), fenoxaprop-P-ethyl EC ($7\%$), mecoprop SL ($50\%$), triclopyr-TEA SL ($30\%$) and one pre-emergence herbicide pendimethalin EC ($31.7\%$) treated on 21 Sept. and 10 Nov. 2003. Kentucky bluegrass visual quality evaluated 30 and 50 days after application for phytotoxic effects of the herbicides. As a result, asulam WG (0.2g/$m^{2}$) and imazaquin SL (0.3ml/$m^{2}$) showed approximately $90\%$ of control in creeping bentgrass, but visual quality of Kentucky bluegrass significantly decreased from 20 to 50DAT (day after treatment). However, creeping bentgrass was acceptably controlled(over $80\%$) by fenoxaprop-P-ethyl EC (0.4ml/$m^{2}$)+triclopyr-TEA SL(0.3 ml/$m^{2}$) applied twice on 21 Sept. and 1 Oct. 2003 without serious injury on Kentucky bluegrass. Therefore, it is suggested that an application of fenoxaprop-P-ethyl EC (0.4ml/ $m^{2}$)+triclopyr-TEA SL (0.3 ml/$m^{2}$) may be more effective to control creeping bentgrass in Kentucky bluegrass with the least phytotoxicity by herbicides.

Efficient regeneration methods and transformation system are a priority for successful application of genetic engineering to vegetative propagated plants such as grape (Vitis labrusca L.). This research is to establish shoot regeneration system from plant explants for 'Campbell Early', 'Tamnara', 'Heukgoosul', 'Heukbosek' using two types of plant growth regulators supplemented to medium. The highest adventitious shoot regeneration rate of 5% was achieved on a medium containing of Murashige and Skoog (MS) inorganic salts and Linsmaier and Skoog (LS) vitamins, 2 mg/L of TDZ and 0.1 mg/L of IBA. Leaf tissue of 'Campbell Early', was co-cultivated with Agrobacterium strains, LBA4404 containing the vector pBI121 carrying with CaMV 35S promoter, gus gene as reporter gene and resistance to kanamycin as selective agent, the other Agrobacterium strains, GV3101 containing the vector pB7 WG2D carrying with mPAP1-D gene. mPAP1-D is a regulatory genes of the anthocyanin biosynthetic pathway. 'Campbell Early' harboring mPAP1-D gene was readily able to be selected by red color due to anthocyanin accumulation in the transformed shoot. These results might be helpful for further studies to enhance the transformation efficiency in grape.

This study quantified the storage and annual uptake of carbon by apple trees in orchards as a production-type greenspace, and computed the annual carbon emissions from apple cultivation. Tree individuals in the study orchards were sampled to include the range of stem diameter sizes. The study measured biomass for each part including the roots of sample trees through a direct harvesting method to compute total carbon storage per tree. Annual carbon uptake per tree was quantified by analyzing the radial growth rates of stem samples at ground level. Annual carbon emissions from management practices such as pruning, mowing, irrigation, fertilization, and use of pesticides and fungicides were estimated based on maintenance data, interviews with managers, and actual measurements. Regression models were developed using stem diameter at ground level (D) as an independent variable to easily estimate storage and annual uptake of the carbon. Storage and annual uptake of carbon per tree increased as D sizes got larger. Apple trees with D sizes of 10 and 15 cm stored 9.1 and 21.0 kg of carbon and annually sequestered 1.0 and 1.6 kg, respectively. Storage and annual uptake of carbon per unit area in study orchards were 3.81 t/ha and 0.42 t/ha/yr, respectively, and annual carbon emissions were 1.30 t/ha/yr. Thus, the carbon emissions were about 3 times greater than the annual carbon uptake. The study identified management practices to reduce the carbon footprint of production-type greenspace, including efficient uses of water, pesticides, fungicides, and fertilizers. It breaks new ground by including measured biomass of roots and a detailed inventory of carbon emissions.

This study was designed to assess molinate fate in the lysimeter by measuring the total radioactivity in the leachate, evolved $^{14}CO_2$, and $^{14}C$-residues in soil and rice plant. The amounts of applied $^{14}C$ in the leachate from the lysimeter for 20 weeks were 1.05% in 2.31 pore volume (217,465 mL) at the first and 0.34% in the second year, respectively. The amount of $^{14}CO_2$ evolved from the lysimeter accounted for 6.47% and 0.03% of applied $^{14}C$ in the first and second year. The $^{14}C$-activities in the soil layer of the lysimeter were distributed 18.0% (1st) and 13.3%(2nd) in the depth of 0 to 10 cm, 4.3 (1st) and 1.1% (2nd) in the depth of 10 to 20 cm. Most of the applied $^{14}C$ was detected in the top 20 cm soil layer. Total $^{14}C$ in rice plants grown at lysimeter were detected 11.46% of applied $^{14}C$. 11.11% in straw, 0.24% in brown rice grain, 0.08% in chaff and 0.03% in ears were distributed in the first year. Consequently, environmental fate of molinate using lysimeter simulating a paddy rice field were investigated 25.24% in soil, 11.64% in rice plant, 1.05% in leachate, 6.74% in evolved $^{14}CO_2$ and 0.02% in volatilized organic chemicals in the first year.
The main objective of this topic is to establish strategies and to plan biotechnology researches which are related to the agricultural improvements especially focusing on the crop breeding in Korea. From 1960's to 1980's government policy had been emphasized to develope high yielding cultivars for the self sufficient supply of the staple food crops. As a result, considerable increase of rice production has been made with accumulating technology and man's powers. Recently genetically modified crops harboring useful characteristics have been developed using biotechnology and released in the developed countries. National research institutes and private companies have been developed biotechnology researches to establish competitive capabilities, however they have not been successfully used in commercialization. Therefore it is necessary to promote the practical. application by connecting molecular technology with conventional breeding. Proposed research projects are; (1) basic researches including plant genome studies, (2) developing new cultivars through gene transformation, (3) screening and producing antioxidants, secondary metabolite substances and edible vaccines. To set a government policy, both domestic and international research trends were reviewed and possibility of success based on the economic view point were discussed. The intellectual property and preservation of environment play a key role to decide the research priority. It is also necessary for us to make one step system for the distribution of research resources such as microorganisms, genes cloned, plant seeds and research informations for promoting research activities.
