A Study on the Gelation of Starch (전분의 젤화에 관한 연구 -강남콩 조전분 및 정제전분의 이화학적 특성-)

  • The physicochemical properties of kidney bean crude and refined starch were investigated. The results were as follows : Amylose content of refined starch was 77%. Blue values of crude and refined starch were 0.375 and 0.410, respectively. Ferricyanide numbers of crude and refined starch were 1.00 and 1.94, respectively, and alkalinumbers of crude and refined starch were 8.67 and 6.90, respectively. Amylose had molecular weight of 18067 and degree of polymerization was 112. Amylopectin had degree of branching of 3.7 per 100 glucose units and glucose units of 27 per segment of amylopectin. Water binding capacities of crude and refined starch were 202.1% and 169.4%, respectively. Both swelling powers of crude and refined starch were increased rapidly from $70^{\circ}C$ to $90^{\circ}C$ and their curves showed a single-stage pattern. The optical transmittance of 0.2% crude starch suspension was increased rapidly from $80^{\circ}C$ to $83^{\circ}C$ and that of 0.2% refined starch suspension was increased rapidly from $77^{\circ}C$ to $83^{\circ}C$. Brabender hot-paste viscosities of crude and refined starch at 6% and 8% concentation (solid basis) showed the similar amylogram patterns of c type with no peak vircosity.

Isolation and Partial Characterization of Cysteine Proteinase from Sparganum (스파르가눔 총체에서 분리한 cysteine proteinase의 정제 및 부분 특성)

  • A proteolytic enzyme was purified from the tissue extract of spargana (plerocercoids of Spirometra erinacei) by DEAE-Trisacryl M ion exchange chromatography and thiopropyl-sepharose affinity chromatography resulted in a 21-fold purification. The proteinase activity was assayed with a synthetic fluorescent substrate, carbobensoxy-phenylalanyl-7-amiso-4-trifluoromethyl-coumarin. SDS-polyacplamide gel electrophoresis of the purified materials revealed a single 28,000 dalton band. Inhibitor profiles of the band indicated that it belonged to cysteine endopeptidases. It exhibited identical pH curves with optimum at pH 5,5, and 50% activity from pH 4.7 to 8. It could completely degrade collagen chains to three identical products. It also showed some activity on hemoglobin. Furthermore, the band on immunoblots was reactive to the sera of sparganosis patients. These results suggest that the proteolytic enzyme belongs to cysteine proteinase which plays a role in the tissue penetration. Also it may be used as the antigen for diagnosis of active sparganosis.

  • This experiment was carried out to optimize the condition for the enzyme production by selected strain in the basal medium, to purify the enzyme and to characterize the purified enzyme. The results obtained were as follows. 1. The optimal conditions for the $\beta$-galactosidase production were initial pH 7.0 and temperature $65^{\circ}C$. 2. Enzyme was induced by the addition of lactose and galactose, and it was intracellular enzyme. 3. The purified enzyme was obtained with the increased level of activity approximately 28.5 folds as compared with crude enzyme and the yield of 15.2% by means of DEAE-Cellulose column chromatography, Sephadex G-150 gel filtration 4. $\beta$-galactosidase from final step of purification showed a sing1e protein band on polyacrylamide gel disc electrophoresis. 5. The optimal temperature and pH of the purified enzyme were $65^{\circ}C$, pH 6.5 for the hydrolysis of lactose.

Alkaline and Antioxidant Effects of Bamboo Salt (죽염의 알칼리성 및 항산화 효과)

  • Mineral contents of various salts were determined by the ICP-OES method. Bamboo salt (baked 9 times) contained more potassium, calcium, magnesium, and manganese, compared to purified and solar salts. Bamboo salt had a pH of 11.04, higher than those of purified (6.29) and solar (9.13) salts. Contents of [$OH^-$] were measured by using the FT-IR spectra. Bamboo salt exhibited higher reduction potential and contained more OH groups than purified and solar salts. The reduction peak of bamboo salt was observed to be about three times broader than that of solar salt in terms of redox potential amperometry. At a salt concentration of 25%, bamboo salt showed higher radical scavenging activities (81.4%) than solar (5.0%) and purified (2.0%) salts, as evaluated by DPPH assay. Bamboo salt revealed alkaline property, more OH groups and antioxidative activity.

The Separation of Transglutaminase Produced from Streptomyces mobaraensis and Its Application on Model Food System (Streptomyces mobaraensis로부터 생산되는 transglutaminase 분리 및 모델식품 적용)

  • Transglutaminase (TG) was prepared from Streptomyces mobaraensis to improve texture and self-life of food. In preliminary experiments, texture of the dough was not improved due to the interference in microbial TG reaction by proteases present in the crude enzyme. Among the cation exchange resins tested for the removal of proteases, MonoPlus S 100 was the most efficient. Further purification steps with a quaternary ammonia salt resin and gel permeation chromatography effectively removed proteases from crude enzyme. Molecular weight of purified enzyme was about 38,000 on SDS-polyacrylamide gel electrophoresis. Farinograph data showed the addition of purified enzyme to wheat flour gave higher stability and lower weakness values those that of crude enzyme.

Purification and Characterization of Transglutaminase from a Newly Isolated Streptomyces platensis YK-2 (토양 방선균 Streptomyces platensis YK-2가 생산하는 Transglutaminase의 정제 및 효소학적 특성)

  • A species producing transglutaminase (EC 2.3.2.13) was isolated from forest soil and identified as Streptomyces platensis YK-2. The transglutaminase was purified from culture broth by 50% methanol precipitation, followed by successive chromatography on DEAE-Sephadex. The yield and purification-fold was 63.4% and 2.2-fold, respectively. The purified microbial transglutaminase (MTG) migrated as a single band of approximately 45 kDa upon sodium dodecyl sulfate polyacrylamide gel eletrophoresis. The isoelectric point determined by multichambered electrofocusing was pH $6.0{\sim}7.0$. The enzyme was strongly inhibited by $Hg^{++}$, but was activated by $Cd^{++}$, $Mg^{++}$, $Mn^{++}$, $Pb^{++}$ and reducing agents such as dithiothreitol and mercaptoethanol.

  • The authentic hGH converted from met-hGH by immobilized ApM was purified by successive chromatographic processes based on the differences in isoelectric points, hydrophobicities and charges. The final recovery yield was about 14.1% and the specific activity of the purified hGH was 2.75IU per mg when assayed by enzyme immunoassay. The purified hGH was verified to be authentic hGH through the analysis of amino acid composition, amino-terminal amino acid sequence, carboxy-terminal amino acid and tryptic peptide map. The purity of purified hGH was higher than that of commercial hGH when assessed by SDS-PAGE, PAGE, IEF and HSGF. In weight-gain assay and tibia test with hypophysectomized rats, the hGH produced in this study showed the same growth effect as the commercial hGH.

  • 세계의 정제능력이 계속 감소되고 있다. 현재 많은 석유회사들이 손실을 줄이기 위해 애써 확보해 놓은 시장을 포기할 태세를 갖추고 있는 가운데 서구와 미국에서는 정유공장폐쇄율이 높아지고있다. 그러나, 주요시장에서 정제능력과 수요가 균형을 이룰 전망은 아직도 요원하며 이로인해 정유산업은 계속 높은 고정비와 낮은 제품가격의 2중고에 시달릴것으로 보인다.

  • 태양전지 산업의 급속한 발전에 따라 고순도 실리콘을 경제적으로 제조하려는 노력이 경주되고 있다. 이중금속 실리콘(MG)의 정제에 의한 태양전지 실리콘(SoG) 제조에관한 연구가 더욱 더 주목받고 있다. 본 연구에서는 고순도 금속 실리콘 정제 연구의 일환으로 금속 실리콘에 대한 플라즈마 용해시 불순물 정제 가능성에 대한 기초연구를 수행하였다. 먼저 4N 급 Ar 금속 실리콘을 출발물질로 하여 반복 용해횟수(용해시간)를 달리하여 200g 급 Si button을 제조하였다. 이후 화학적 분석법으로 B, P 및 미량 금속 불순물 평가를 실시하였으며, 이로부터 Ar 플라즈마의 정제 효과를 분석하고자 하였다.
