Development of Sandwich ELISA for the Detection of Mackerel in Processed Foods (가공식품 중 고등어의 검출을 위한 ELISA의 개발)

  • There have been few studies related to ELISA for mackerel. In this study we developed a sandwich ELISA for mackerel in processed foods using rabbit polyclonal antibodies against mackerel parvalbumin, the major allergen of mackerel and heat-stable protein. The parvalbumin was purified by ammonium sulfate precipitation and Sephadex G-50 column chromatography. From the standard ELISA curves, the detection limit of parvalbumin was 3 ng/mL and the detection range of mackerel was 5-5,000 ${mu}g/mL$. We further investigated the cross-reactivity of the antibodies toward mackerel, mackerel pike, salmon, flatfish, armorclad rockfish, cod fish, squid, shrimp, blue crab, and lobster. The antibodies were specific for mackerel only. The mean assay recoveries in cooked cream soup, baby food, sausage, and sauce spiked with 0.01 to 0.3% mackerel were 104, 101, 54, and 0%, respectively. In sample tests of 16 commercial items, the qualitative coincidence ratio of assay result and indication was 75%.

Problems and Solutions of Matrix Organization Structure: Focusing on the Case of H-Corp. Research Institute (매트릭스 조직구조의 문제점과 해결 방안: H사 연구소 사례를 중심으로)

  • Even though there have been so much practical interests in industry, the relevant empirical researches are not sufficient. In this study, we try to identify the problems of matrix organization structure in the semiconductor industry and make suggestions for improvements. Also, we try to find out whether there are differences in the perceptions of the problems among ranks and teams. This study was conducted to the researchers in the matrix organization structure of the H-corp. research institute. The problems we found are as follows. The researchers agreed that the matrix organization structure is appropriate when highly professional members for the development of next-generation semiconductors are participated in the projects. They showed strong wills to participate and succeed in projects. However, the researchers felt that the equipments and manpowers were not enough and too much tasks and workloads were assigned to both the managers and members Also, in an open ended question, the researchers pointed out the problems of the matrix organization structure such as 'weak project manager's authority', 'communication and teamwork issues', 'non-obvious work priorities', 'compensation and benefit system', 'lack of research manpower and equipment'. From the strengths and weaknesses of the matrix organization structure of the semiconductor industry, we provide some suggestions for improvements.

A Study on the Perceived Language and Non-verbal Communication and Lapport and Customer Satisfaction of Hospital Users : Focused on the Control Effect of Service Authenticity (병원이용객이 지각한 언어·비언어커뮤니케이션과 라포, 고객만족에 관한 연구 : 서비스진정성 조절효과를 중심으로)

  • This study is a descriptive survey using a self-reported survey method to understand the effect of language and non-verbal communication of medical service providers on customer satisfaction through Lapport and to determine the degree of impact on Lapport by adjusting perceived service authenticity. The data collection period was from April 5 to April 30, 2021. Using the convenience sampling method, data were collected from users of medical institutions located in Busan and Gyeongsangnam-do, and a total of 306 valid questionnaires were used as statistical analysis data. The collected data were analyzed using the IBM SPSS statistics version 25.0 and AMOS 20.0 programs through a coding process. In the results of this study, it was found that the language and non-verbal communication of medical service providers influenced customer satisfaction through Lapport, and the perceived service authenticity in the relationship between each variable acts as a moderating effect. Considering that the language and non-verbal factors of the medical service provider are important in the interaction between the medical service provider and the hospital user, the medical service provider should take full account of the language and non-verbal factors and help the medical institution communicate with the hospital user.

Production of green tea jelly using theanine and its physiochemical characterization (녹차 theanine을 이용한 젤리 제조 및 품질특성 조사)

  • Theanine, the major amino acid and a sweet umami component of green tea, has anti-stress effects in humans. From green tea, theanine was extracted at 80℃ for 2 h using a low temperature, high pressure extractor, and caffeine was removed using an HP-20 column with 80% ethanol. Theanine extracts were applied to produce functional jelly using three kinds of gelling agents (I, II, and III) or various concentrations of theanine extracts (10-50%). Theanine jelly was characterized with respect to its physical properties, product stability, and physiological function. Gelling agent III (tamarind gum, xanthan gum, and locust bean gum=2:3:5, w/w/w) and S3 (35% theanine extracts) jelly exhibited the optimum textural properties with lower hardness and high springiness. Among theanine jellies, S3 exhibited optimum product stability, high 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, and acetylcholinesterase inhibitory activity. These results indicate that the anine extracts could be used as a neuroprotective source in the food industry.

  • The purpose of this study is to analyze the user experience and suggest the necessary improvements, in the blockchain based social platform Steemit. First, I investigated what is the blockchain and the characteristic of social platforms based on blockchain system. The honeycomb model was used to investigate and analyze the Steemit user experience, and the quantitative and qualitative data were analyzed through questionnaires and in-depth interviews. As a result of the research, the voting function, which is core function of the service in use, has high usefulness. Improvements suggestion that the editing tools and the abuse of voting function should be improved. I hope that this study will be used as a basic research material to improve the user experience of blockchain based social platforms.

Isolation of a Bacteriocin - Producing Lactobacillus sakei Strain from Kimchi (김치에서 박테리오신을 분비하는 Lactobacillus sakei균주의 분리)

  • Bacteriocin producing lactic acid bacteria (LAB) were isolated from Kimchi by using spot-on-the-lawn method. Listeria monocytogenes, Staphylococcus aureus, and Lactobacillus plantarum were used as indicators. One isolate (P3-l) produced a bacteriocin efficiently inhibiting the growth of Listeria monocytogenes. 16S rDNA sequence and sugar utilization test identified that P3-1 was a Lactobacillus sakei strain. Accordingly, the isolate was named as Lactobacillus sakei P3-1. L. sakei P3-1 produced a bacteriocin which efficiently inhibited the growth of Listeria monocytogenes but did not inhibit other Gram positive and negative organisms tested. The bacteriocin was stable against heat, organic solvent, and pH variation and it retained 50% of activity after 10 min heat treatment at 10$0^{\circ}C$. The molecular weight of Sakacin P3-1 was estimated to be 4 kDa by SDS-PAGE.

Literature Review on Rheological Properties and Required Performances of 3D Printable Cementitious Materials (3D 프린팅 시멘트계 재료의 유변학적 물성과 요구 성능에 관한 문헌 조사)

  • 3D printing techniques have been recently adopted in the construction industry. It mainly utilizes additive manufacturing which is the fabrication process depositing successive layers of materials without any formworks. Conventional cementitious materials may not be directly applicable to 3D printing because 3D printable cementitious materials is required to satisfy such characteristics as pumpability, extrudability, and buildability in a fresh state. This study aimed to investigate rheological properties and required performances of 3D printable cementitious materials, by reviewing existing studies. Test methods and equipments, evaluation results and characteristics of mixture additives were compared. Based on reviews of existing studies, this study indicates that the viscosity is mainly relevant to the pumpability of 3D printable materials whereas the yield stress and thixotropy are important in securing buildability of the materials.

  • 인터넷 환경의 변화와 관련 제반 기술의 발달로 인하여, 이전과는 달리 콘텐츠의 제작, 활용에 있어서 높은 사용자 참여도를 보여주고 있다. 이러한 변화는 여가 활동의 증가와 엔터테인먼트를 추구하는 인터넷 사용자의 목적과 맞물려 UCC(User Created Contents)가 중요한 요소로 부각되게 되었다. 이미 온라인과 오프라인상에서 사용자가 UCC를 제작할 수 있는 장비들과 다양한 도구들이 보편화 되어있다. 그러나 UCC와 관련된 프로그램 및 장비들의 기능과 플랫폼이 복잡해짐에 따라 사용자가 콘텐츠 제작에 있어 어려움을 겪고 있는 실정이다. 따라서 콘텐츠의 제작과 활용 측면에서 기능과 서비스를 자연스럽게 사용할 수 있도록 하는 통합적인 환경 제안과 발전 방향을 모색하는 것을 연구의 목적으로 삼는다. 본 연구의 방법으로는 콘텐츠를 제작하는 사용자를 대상으로 한 환경 조사 그리고 포커스 그룹 정성 조사를 통하여 현재 UCC제작 환경의 문제점을 도출하였다. 이를 기반으로 사용자의 니즈를 충족시키면서도 사용자 중심적인 통합적 UCC 제작 도구와 개발을 위한 가이드 라인을 제시한다.

무주지역 안정 동위원소 조성의 시공간적 분포

  • 본 연구는 무주군 적상산 부근에 위치하는 소유역에서 지표수와 인접한 천층 지하수의 안정동위원소 조성의 시공간적 변화를 규명하기 위해서 시행되었다. 조사는 2001년 8월과 10월, 2002년 4월 세 차례에 걸쳐 수행되었다. 각 계절에 따라 지표수와 지하수의 안정동위원소 조성이 비슷하게 나타나는 것은 이 지역 지표수와 지하수사이에는 배수와 충진이 지속적으로 생기고 있음을 암시한다. 조사지역의 안정동위원소 조성은 북쪽에 위치하는 한강의 안정동위원소 조성보다는 무거우며, 남쪽에 위치하는 섬진강의 안정동위원소 조성보다는 가벼운 위도효과를 잘 나타내고 있다. 또한 시료를 채취한 가장 높은 지점과 가장 낮은 지점의 고도차가 400m인 작은 소유 역에서도 계절의 변화와 관계없이 안정동위원소의 고도효과는 잘 나타나고 있다. 선행된 연구에서 우리나라 강우가 우량효과를 반영하는 결과를 나타내는 것과 같이 강우의 영향을 직접 받는 8월 지표수의 안정동위원소 조성은 10월과 4월보다 더 가벼운 조성을 나타내고 있다.

A relationship between food environment and food insecurity in households with immigrant women residing in the Seoul metropolitan area (수도권 거주 결혼이주여성 가구의 식품환경과 식품불안정성 간의 관련성)

  • Purpose: Food environmental factors related to food insecurity affect household food intake in several socio-ecological aspects. This study explores the relationship between food environment factors and food insecurity in households with married immigrant women. Methods: From November 2018 to February 2020, a survey was conducted enrolling 249 married immigrant women residing in the metropolitan areas of South Korea. In the final analysis, 229 subjects were divided into 2 groups classified as food security (n = 154) and food insecurity (n = 75), as assessed by the score of food security. Three aspects of food environments were measured: built·natural, political·economic, and socio-cultural Results: Food environments were significantly different between food security and food insecurity groups, as follows: the number of foods market and their distance from the home and food status for the last week at home in the built·natural domain; monthly cost of food purchase and experience for food assistance in the political·economic domain; total score of social support, parenting, and cooking skills in the socio-cultural domain. A stepwise multivariate linear regression model showed a negative association between the food insecurity score with social support from family and food inventory status in the last week. After adjusting for confounders, a positive association was obtained between the experience of a food support program. The final regression model explains about 30% of the relationship obtained in the three food environment domains and food insecurity (p < 0.001). Conclusion: Not only economic factors, which are common determinants of household food insecurity, but socio-cultural factors such as social support also affect household food insecurity. Therefore, plans for implementing a food assistance program to improve food insecurity for households with immigrant women should consider financial support as well as other comprehensive aspects, including socio-cultural domain such as social support from family and community.
