Role of Protease Activated Receptor 2 (PAR2) in Aspergillus Protease Allergen Induces Th2 Related Airway Inflammatory Response (Aspergillus 단백분해효소 알러젠에 의해 유도된 Th2 관련 기도염증반응에서 protease activated receptor 2 (PAR2)의 역할)

  • Most allergens have protease activities, suggesting that proteases may be a key link between Th2-type immune reactions in allergic responses. Protease activated receptor (PAR) 2 is activated via the proteolytic cleavage of its N-terminal domain by proteinases. To know the role of PAR2 in Aspergillus protease allergen activated Th2 immune responses in airway epithelial cells, we investigated and compared immune cell recruitment and level of chemokines and cytokines between PAR2 knock out (KO) mice and wild type (WT) mice. There were evident immune cell infiltrations into the bronchial alveolar lavage fluid (BALF) of WT mice, but the infiltrations in PAR2 KO mice were significantly lowered than those of WT mice. The IL-25, TSLP, and eotaxin gene expressions were profoundly increased after Aspergillus protease, but their expression was significantly lowered in PAR2 KO mice in this study. Compared to PAR2 KO mice, OVA specific IgE concentrations in serum of WT mice were quite increased; moreover, the IgE level of PAR2 KO mice was lower than in WT mice. The IL-25 expression by Aspergillus protease stimulation was significantly reduced by p38 specific inhibitor treatment. In this study, we determined that Th2 response was initiated with IL-25 and TSLP mRNA up-regulation in lung epithelial cells via PAR2 after Aspergillus protease allergen treatment.

Effect of Larva Extract of Allomyrina dichotoma on Carbon Tetrachloride-induced Hepatotoxicity in Mice (장수풍뎅이 유충 추출물이 사염화탄소에 의한 마우스의 간 손상에 미치는 영향)

  • This study was conducted to investigate the effect of methanol extract of Allomyrina dichotoma larva (MEAL) on carbon tetrachloride $(CCl_4)-induced$ hepatotoxicity in mice. ICR mice were divided into 5 groups [Vehicle control, $CCl_4\;(10{\mu}g/g)$ alone, $CCl_4$ plus a low dose $(50{\mu}g/g)$ of MEAL, $CCl_4$ plus a high dose $(100{\mu}g/g)$ of MEAL]. Silymarin $(2{\mu}g/g)$ was used as the reference in the experiment. Administration of MEAL tended to decrease the serum alanine transaminase (ALT) activity induced by $CCl_4$ treatment in mice. Hepatic concentration of thiobarbituric acid-reactive substances (TBARS) in a high-dose group of diet decreased to the level of silymarin-treated group. Hepatic activity of glutathione S-transferase in MEAL-treated group was lower than that of $CCl_4-treated$ group. Serum concentration of bilirubin was significantly increased by $CCl_4$ treatment, but MEAL or silymarin recovered the level. These results suggest that MEAL may exert the protective effect against $CCl_4-induced$ hepatotoxicity in mice. However, more intensive studies would be needed to elucidate the protective mechanism of the beetle on hepatotoxicity of mice.

Effect of Danchisoyosangagambang Extract on the Hyperglycemic Mice Induced with Streptozotocin (단치소요산가감방(丹梔逍遙散加減方)이 streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향)

  • This study has been carried out to understand the effect of Danchisoyosangagambang (DC) on the hyperglycemic mice induced with streptozotocin(STZ). Experimental groups were made diabetic mice by intraperitoneal injection of STZ(60 mg/kg of body weight) tw ice by 24 h interval and then 120 mg/kg STZ was injected again 3 days after the earlier treatment. Control group was administered mice with 0.9 % saline(2 mL/kg), and experim ental groups were administered DC extract(DCA group, 10 mg/kg/day; DCB group, 30 mg/kg/day) after hyperglycemic induction for 6 weeks. The body weight of experimental groups was lower than control. The blood glucose concentration increased continuously, rea ching to 298.9 mg/dL after 6 weeks, however, experimental groups of the DCA and DCB groups significantly(p<0.0l) decreased in the 4, 5, and 6 weeks groups. Blood glucose tolerance test was not significant between control and experimental groups. We examined the blood transaminase activities to know the effect of herbal medicine on liver function. The GOT activities were lower in group DCB than in control. The GPT activities were lower in group DCA and DCB than in control. The content of triglyceride was significantly increased in group DCA compared to control. The SOD and catalase activities were higher in the group DCA compared to control. The results of immunohistochemical study, a few of insulin positive cells observed in the control and experimental group. These results suggest that administration of DC extract to the hyperglycemic mice decreased the blood glucose level.

Memory-improving Effects of Fermented Sea Tangle Saccharina japonica in Normal Mice (정상 동물모델에서 다시마(Saccharina japonica) 발효물의 기억력 개선 효과)

  • Marine organisms are sources of many bioactive compounds, such as essential fatty acids, essential amino acids, vitamins, and minerals, making them useful candidates for the production of safe bioactive substances. They also synthesize glutamic acid, which can be used to produce γ-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system (CNS), via fermentation with Lactobacillus brevis BJ-20. This study investigated the degree to which fermented sea tangle (FST) inhibits enzymes such as acetylcholine esterase (AChE) and prolyl endopeptidase (PEP) and affects memory of normal mice using the T-maze test. FST inhibited more than 90% of AChE at 1 mg/mL and 50% of PEP at 8 mg/mL. Oral FST (100 mg/kg) significantly improved performance of normal mice on the T-maze. Therefore, sea tangle fermented with L. brevis BJ20 effectively contributes to memory improvement and might be a useful functional food ingredient.

  • The purpose of this research was to investigate the effect of Sojagangqi-tang(SJGQT) on the immune cell activity. The addition of SJGQT enhanced the proliferation of cultured-mice splenocytes and thymocytes. Administration of SJGQT(250 mg/kg) accelerated the subpopulation of splenic T lymphocytes especially CD/sup 4+/-TH cells in BALB/c mice. But high concentration(500 mg/kg) of SJGQT decreased the splenic T, B lymphocytes and thymic Tc (CD/sup 8+/) lymphocytes. Oral administration of SJGQT(250 mg/kg) significantly enhanced the production of IFN-γ and IL-4 in mice serum. And also, the addition of SJGQT(100 ㎍/ml) inhibited the proliferation of cultured-Jurkat leukemia cells in vitro. These results suggest that SJGQT have a cellular immuno-modulatory effect and anti-cancer property action

Effects of Saengjinyanghyutang-gamibang and Rosa rugosa on the Pancreas in the Streptozotocin Treated Mice for Induction of Diabetes (생진양혈탕가미방과 매괴화가 Streptozotocin으로 유발된 고혈당 생쥐의 췌장에 미치는 영향)

  • Objectives : This study has been carried out to investigate the effects of the Saengjinyanghyutang-gamibang and Rosa rugosa extracts on pancreas of the hyperglycemia mice induced with streptozotocin. Methods : We examined immunohistochemistry for insulin and COX-2, ultrastructural changes of acini by electron microscope, and changes of the blood glucose and BUN levels. Results : The ${\beta}-cells$ on Langerhnan's islet were destructed by administration of streptozotocin, so that few insulin-positive cells were observed in the control group. However, a lot of insulin-positive cells were observed in the experimental groups. These cells had recovered from the damage. As a result of COX-2 immunostain, COX-2 expression were highest in the control group other than the Saengjinyanghyutang-gamibang and Rosa rugosa extracts administered groups. As the electron microscopical observation, the centroacinar cells and acini of pancreas were destructed or damaged by administration of streptozotocin in the control group, but these recovered !Tom the damage in the other experimental groups. The levels of serum glucose were decreased remarkably on the Rosa rugosa and Saengjinyanghyutang-gamibang extracts administered groups compared with control group. Conclusions : These results suggest that administration of the Rosa rugosa and Saengjinyanghyultang-gamibang extracts to the mice reduced the damage induced by streptozotocin.

Effect of High-Fat Diet-induced Obesity on the Incidence and Progression of Prostate Cancer in C57BL/6N Mouse (C57BL/6N 마우스에서 전립선암의 발병률 및 진행에 대한 고지방식이-유도 비만의 영향)

  • Obesity induced by high-fat diet (HFD) is verified as a strong risk factor and negative prognostic factor for prostate cancer in several genetically engineered mice although it was not examined in the normal mice. To investigate whether HFD-induced obesity can affect the development and progression of cancer in the prostate of normal mice, alterations in the weight and histological structure of the prostate as well as the expression of cancer-related proteins were analyzed in obese C57BL/6N mice fed with 60% HFD for 16 weeks. First, HFD-induced obesity, including an increase in organ weight, body weight, fat accumulation, and serum lipid profile, was successfully induced in C57BL/6N mice after HFD treatment. The total weight of the prostate significantly increased HFD-induced obesity in the model mice compared with the control group. Among the four lobes of the prostate, the weight of the ventral prostate (VP) and anterior prostate (AP) were higher in HFD-induced obesity model mice than in the control group, although the weights of the lateral prostate (DLP) and seminal vesicle (SV) were constantly maintained. In addition, the incidences of hyperplasia and non-hodgkin's lymphoma (NHL) in the histological structure were remarkably increased in HFD-induced obesity model mice, while the epithelial thickness was higher in the same group. A significant increase in the phosphorylation levels of key proteins in the AKT (protein kinase B) signaling pathway was detected in HFD-induced obesity model mice. Therefore, these results suggest that HFD-induced obesity can promote hyperplasia and NHL in the prostates of C57BL/6N mice through the activation of the AKT signaling pathway.

Anti-Diabetic Effect of Black Ginseng in C57BLKS/J-$db/db$ Mice (C57BLKS/J-$db/db$ 마우스에서 흑삼의 항당뇨 효과)

  • 본 연구는 흑삼의 항당뇨 효과를 알아보고자 4그룹(정상군, 당뇨 쥐, 당뇨 쥐에게 백삼을 투여한 군, 당뇨 쥐에게 흑삼을 투여한 군)으로 나누어 6주간 실시하였다. 6주 후 식품섭취량, 체중 증가량을 비교하였는데, 식품섭취량, 체중 증가량에는 그룹 간에 유의적인 차이가 나타나지 않았으나, OGTT(oral glucose tolerance test)와 IPITT(intraperitoneal insulin tolerance test) 경우는 흑삼 투여군에서 긍정적인 결과를 나타내었다. 또한 혈청 포도당과 인슐린농도에 미친 영향을 비교, 분석한 결과, 흑삼 투여군에서 공복 시 혈당, 혈청 포도당, 인슐린 농도가 유의적으로 감소하였다. 이 결과를 미루어 볼 때 흑삼이 백삼에 비해 당뇨를 치료하는데 더 효과적일 것으로 판단된다.

Haolthane에 의한 C57BL/6 mice의 세포 매개성 면역부전 실험 모델의 개발과 이에 미치는 Brziline의 영향

  • 임상적으로 많이 노출될수록 간단백과 결합하여 항원을 생성함으로써 면역매개성 간염을 유발하는 Halothane을 사용하여 mice에서의 세포매개성 면역선택하였다. Halothane을 복강투여로 2회, 4회 투여한 후 11, 15, 18일 후 지라세포를 이용하여 ConA유도 임파구 분열능을 검색해본 결과 2회 및 4회 투여 시에 15일까지는 대조군과 비교하여 현저히 저하되나 18일 후 오히려 증가하고 ConA유도 IL-2의 생성능은 2회 투여 시 15일 까지는 정상군에 비해 유의적인 차이가 없었으나 18일 이후 현저히 증가하였다. 4회 투여 시 15일까지는 대조군에 비하여 현저히 저하되었고 18일 이후에는 오히려 증가하는 것으로 관찰되었다. 혈청 ALT, AST는 유의적인 차이가 없었다. 또한 이와 같은 실험 결과가 개체간의 차이를 보이는가 알아보기 위하여 개체별로 검정을 한 결과 개체간의 편차는 없는 것으로 관찰되었다.

  • Footpad swelling having the relationship with arthus-reaction of antibody-mediated hypersensitivity and delayed type hypersensitivity was recovered to the almost normal level. The nethemoglobin induced by aniline showed no significant deviation. PFC/spleen cell and PFC/106 spleen cell were increased slightly, but not in case of RFC. Hemagglutination value was increased slightly, but hemolytic value was not changed significantly. Mice in the administration of the polysaccharide does not show any significant stress factor in the cage for mice admlrustered plasma corticosterone.
