Streptomyces platensis YK-2, newly isolated from forest soil, produces transglutaminase (TGase), which catalyses an acyl transfer reaction between the primary grade amine and protein or $\gamma$-carboxyamide group of peptide bound glutamine residues. For a molecular genetic study of S. platensis, an effective transformation method was established by using a conjugal transfer of DNA from Escherichia coli to spores of actinomycetes. The highest transconjugation frequency of S. platensis was obtained on an MS medium containing 50 mM $MgCl_2$, using $5{\times}10^7\;E$. coli as a DNA donor and $1{\times}10^8$ spores without heat treatment as a host. We also identified that S. platensis contains a single attB site within an ORF encoding a pirin-homolog, and that its attB site sequence shows high homology to that of S. logisporoflavus. In addition, it was confirmed by phenotypic analyses of exconjugants that the introduction of heterologous DNA into the attB site of the S. platensis chromosome does not affect its morphological differentiation and TGase production.
One of the important considerations in structural designing the flat plate system is ensuring the resistance to punching shear caused by axial loads and the ductile ability to follow horizontal deformation under earthquake. In this study, the ECC (Engineered Cementitious Composite) has been placed in the critical section zone of punching shear at slab-column joint to improve ductility and the advanced details of shear reinforced area nearby critical section zone has been developed using stud and steel fiber. The shear performance tests were performed on the specimens with parameters of fiber type mixed with ECC, stud and steel fiber set into the shear reinforced area in which the failure pattern, joint strength, displacement and strain of the specimen were compared and analyzed. The test results showed that the strength and ductility of specimens with ECC applied to joint were better than those of RC flat plate system. Also, the shear reinforcement effect of stud and the ductility improvement of steel fiber concrete were confirmed in the shear reinforcement area.

This study investigates the horizontal shear behavior of an interface between precast concrete (PC) and topping concrete(RC), and evaluates the horizontal based on the investigations by the experimental. Horizontal shear strength in connected surface is determined by the roughness an interface and the shear reinforcement or not. In this study, the main experimental parameters are the shear reinforcement types in the shape of loop-type and lattice-type, rebar spacing. A total of four specimens were shear strength tested and manufactured. As a result, the horizontal shear strength of reinforced connected surface was found to be controlled by deformation in vertical direction. Comparison of reinforcement shape, the mean initial crack load loop type specimens, the average maximum load and the junction of the average in terms of initial stiffness, respectively 33.7%, 45.9% and 55.2% were large enough. Evaluation results for shear strength equation of existing standard domestic, the loop-type reinforced 2.32 to 4.23 times, lattice-type reinforced 1.65 to 3.06 times appears to be higher. Behavior of interface or strength of structural design criteria was fairly safe side. It does not have any problems in the applied field is considered.

A steel modular unit structural system has been used increasingly for mid and high-rise buildings, since the building can be easily constructed by assembling the pre-made modular unit structures. For this structural system, each modular unit structures have to be properly connected to the foundation to transfer the axial force and the bending moment that are generated from external load to the ground. In this study, a new type of the embedded steel column-to-foundation connection was proposed, and its flexural behavior was evaluated through a series of experimental study. Five full scale specimens for the proposed connections were constructed and tested. The effect of the main parameters that affect the flexural behavior of the proposed connection, such as embedment length and shape of end plate, were studied. From the results, it was found that the flexural stiffness of the proposed connection was higher than that of the semi-rigid connection for all test specimens, and 200 mm of embedment length was proper for the given test specimens in this study.
Precast concrete (PC) beam-column connections using ductile rods are proposed for earthquake zone. An existing beam-column connection, two PC specimens designed by considering failure modes and a conventional RC specimen were tested under cyclic loading to evaluate the seismic performance. The specimens were designed to satisfy the requirements of current design code. The variables are the yield strength of longitudinal reinforcing bars of PC beams. The test results showed that the proposed system applying smaller yield strength of the longitudinal reinforcing bars at the PC beams than the ductile rods was satisfied with seismic criteria. The deformation capacity and energy dissipation capacity of the proposed PC beam-column connections were greater than those of the existing DDC system.
The objective of this research is to provide better knowledge on the behavior of strong axis SRC column-RC beam joint, supported by experimental results, that can be broadly applicable to many structures. For this purpose, firstly literature reviews and field survey were made to classify the most commonly used for these types of joints. Then, experimental program was designed and performed including 6 SRC column-RC beam joint specimens designed with various joint details. Using the experimental results obtained from the quasi-static cyclic tests, structural performances of the joints such as hysteretic curves, maximum strength capacities, strength degradation beyond the maximum strength, ductilities, and energy dissipation capacities were investigated. Test results showed that specimens with wide beam shape (RCW-P, RCW-W, RCW-F) and T beam shape (RCT-W) showed better structural performances than the bracket type specimens (HBR-L, HBR-S). These specimens also revealed to have higher strength capacities than the nominal design strength. However, H beam bracket type specimens (HBR-L, HBR-S) need further study both analytically and experimentally to verify the reason for unexpected structural performances.

The purpose of this study was to investigate the performance of precast concrete segmental bridge columns with shear resistance connecting structure. The system can reduce work at a construction site and makes construction periods shorter. A model of precast concrete segmental bridge columns with shear resistance connecting structure was tested under a constant axial load and a cyclically reversed horizontal load. A computer program, RCAHEST (Reinforced Concrete Analysis in Higher Evaluation System Technology), for the analysis of reinforced concrete structures was used. An bonded or unbonded tendon element based on the finite element method, that can represent the interaction between tendon and concrete of prestressed concrete member, is used. A joint element is newly modified to predict the inelastic behaviors of segmental joints. The proposed numerical method gives a realistic prediction of performance throughout the loading cycles for several test specimens investigated.

The application of bonded dissimilar materials to industries as automobiles, aircraft, rolling stocks, electronic devices and engineering structures is increasing gradually because these materials, compared to the homogeneous materials, have many advantages for material properties. In spite of such wide applications of bonded dissimilar materials, the evaluation method of quantitative strength considering the stress singularities for its bonded interface has not been established clearly. In this paper, the stress singularity for Bctors and the stress intensity factors were analyzed by boundary element method(BEM) for the scarf joints of Al/Epoxy with and without a crack, respectively. From static fracture experiments of the bonded scarf joints, a fracture criterion and a evaluation method of interface strength in bonded dissimilar materials were proposed and discussed.
Experimental studies were carried out with test parameters: diaphragm yield type and beam yield type, the opening hole size of inner steel diaphragm, and the existence of slab in order to understand the behavior of beam-to-concrete filled steel tube column rigid connections under cyclic loading condition. Test results show that the connections have good rotational capacity when the beam yields first and the joints should be designed such that the beam yields prior to the inner diaphragms.

This study is to evaluate the shear performance of joint between existing and new slab in apartment remodeling construction for enlarging existing slab. The horizontal joint parameters are consisted by steel pipe cotter, shear reinforcement, H-steel, stud bolt, and round shear key by concrete. And joint specimens will be tested to evaluate the shear performance of these parameters. If the joint detail have sufficient strength, it will be proposed the basic form on the design of joint parts.
