Effects of Multipore Spawn Inoculation on Productivity of Lentinus edodes (Berk) Sing (다공 접종재배가 표고 생산성에 미치는 영향)

  • The effects of multipore spawn inoculation of Lentinus edodes on the mycelial growth stage and on the quantitative and qualitative yields of fruit-bodies for 5 years were investigated at natural environmental conditions. The spawn inoculation quantity did not affect the mycelial rooting at a range of 3.0kg-6.0kg per $m^3$ of bed-logs while the increase of spawn quantity by multipore inoculation increased the mycelial growth and the inner erosion of bed-logs, resulting in the increased yields of total fruit-bodies of L. edodes per $m^3$ of bed-logs for 5 years but resulting in the shortened major production period of fruit-bodies to the the first three years. Major production of fruit-bodies occurred during May-June and August-September, which was not affected by the spawn inoculation quantity. No significant differences in dry weight of a fruit-body were observed as increasing the spawn quantity by multipore inoculation; however the heaviest dry weight of a fruit-body was 1.63g at the spawn inoculation of 4.0kg per $m^3$/TEX> of bed-logs.

Impact of shiitake mushroom (Lentinula edodes) spawn imports on fresh shiitake mushroom import volumes -Focus on the Korea-China FTA- (표고버섯 접종배지 수입이 신선 표고버섯 수입량 변화에 미친 영향 -한중 FTA를 중심으로-)

  • This study was conducted to investigate the reasons for the decreased importation of fresh Shiitake mushrooms into Korea after implementation of the Korea-China Free Trade Agreement (FTA). Monthly time-series data from January 2009 to December 2022 were analyzed using regression analysis and vector autoregression (VAR) models to determine the relationship between the amounts of fresh and spawn Shiitake mushrooms imported. The analysis revealed that a major reason for the decreased importation of fresh Shiitake mushrooms was an increase in mushroom spawn imports after Korea-China FTA implementation. The same results were obtained from the VAR model analysis. However, in terms of the dynamic changes in amount of fresh shiitake mushrooms imported, it was confirmed that the impact of the change in mushroom spawn imports could increase the amount of Shiitake mushrooms imported.

Effect of Amounts of Pisolithus tinctorius Spores and Fertilizer on the Growth of Potted Pinus densiflora Seedlings (모래밭 버섯 포자접종량(胞子接種量)과 시비량(施肥量)에 따른 소나무 화분파종묘(花盆播種苗)의 생장촉진효과(生長促進効果))

  • The effect of different inoculation amounts of Pisolithus tinctorius (Pt) spores and fertilizers on the growth of Pinus densiflora seedlings grown in fumigated or nonfumigated soils were tested in the: polyethylene pots. To infest the pot soil with Pt spores, the mixture of the spores with sterilized nursery soil was used after seeding. Eighteen months after inoculation, mycorrhizal formation was 42-70% in fumigated plus Korean-Pt inoculated pots, 60-70% in fumigated plus U.S.-Pt inoculated pots, and less than 1% in non-fumigated, Korean-Pt inoculated pots. Growth enhancement effect of Pt spore inoculation was shown on only fumigated soil and the inoculation increased the seedlings height (8-38%), stem diameter (9-40%) and dry weight (6-73%). Especially 0.4g per pot application rate increased the height (30-31%), stem diameter (23-28%) and dry weight (56-69%), while the 0.2g U.S. Pt spore per pot rate increased 26-38, 17-20 and 58-60%, for height, stem diameter, and dry weight respectively. At 1X fertilizer application rate (urea 2g, fused superphoshate 4g, and potassium chloride 1g per pot), the 0.4g per pot rate resulted in more dry weight by 18% than that at 1/2X rate. The result indicated that there is a need for further research on inoculating nonfumigated soil with Pt and that appropriate application rates of fertilizers and Pt spores are 1X and 0.2-0.4g per pot on fumigated soil, respectively.

  • We have researched on methods which can reduce fruit-body malformations of king oyster mushroom(Pleurotus eryngii). We collected many pathogens from diseased fruit-body or malformated fruit-body and identified with chemicobiological test and microscope. The factors of fruit-body malformations and increment of contamination during the pin-heading induction time were researched with ventilation amounts in growing room. When the pathogens having high pathogenicity were inoculated in spawn running bottle and at pin-heading induction time, symptom appeared or not appeared in according to air ventilation amounts in growing room. During the pin-heading induction time, humidity degree in growing room have kept of high level and air ventilation amounts were so little that fruit-body malformations ratio was high. But, even though pathogens were inoculated at the surface of bottle factitiously, if air ventilation amounts were enough, fruit-body malformation ratio was low.

Culture Condition and Inoculum Volume of Liquid Spawn on the Bottled Cultivation of Agrocybe cylindracea (버들송이(Agrocybe cylindracea)의 액체종균 배양조건과 접종량)

  • In order to shorten cultivation periods of Agrocybe cylindracea, liquid spawn was used in place of sawdust spawn. Optimum culture condition of liquid spawn was PSB (potato extract sugar broth) medium, $25^{\circ}C$ temperature and pH 7.0. And, optimum inoculum volume was $10{\sim}15ml$. Two grams per liter of dry weight of the fungal mycelia were obtained with 10l of culture bottle with liquid spawn after 10 days. In 850 ml-bottle of sawdust media, cultivation period using liquid spawn (30 days) was five days shorter compared with the cultivation period in sawdust spawn (35 days). Also, complete growth of sawdust media was increased to 92% (liquid spawn) from 75% (sawdust spawn) and yield of fruiting bodies was increased to about 10%.

Production of Pleurotus ostreatus and Flammulina velutipes Using Liquid Spawn Inoculation System (느타리 팽나무버섯 재배에서 액체종균 배양 및 접종시스템 적용방법의 구명)

  • This research aimed at developing the efficient method and device applicable to the inoculation of mushroom using (spawn was intended to utilize) liquid spawn for the stable production of mushroom. For the mass production of liquid culture, optimal inoculum volume and cultural period were $5{\sim}6%$ and 4 days for Flammulina velutipes and 4% and 5 days for Pleurotus ostreatus. Fruit body weight in 850 ml polythene bottle was highest at 6% or 15 ml liquid inoculum for P. ostreatus and $4{\sim}6%$ or $10{\sim}15ml$ for F. velutipes. Weight of fruit body by the application of liquid spawn inoculation system increased up to 33.7% for P. ostreatus and 32.8% for F. velutipes, respetively, compared to conventional spawn making system. The system of liquid spawn inoculation was successfully operated without malfunction in opening or closing the lid, and it took 26 min to inoculate 1200 bottles.

Comparison of Shiitake Productivity in Sawdust Media According to the use of Sawdust, Plug-shaped and Liquid Spawn (톱밥종균, 성형종균 및 액체종균을 사용한 표고톱밥배지에서의 생산성 비교)

  • This study was made to investigate the difference of the shiitake productivity according to the use of sawdust, plug-shaped and liquid spawn. Contamination of medium was confirmed by eye. There was no contamination in the liquid spawn treatment. And, contamination rates in the sawdust spawn and plug-shaped spawn were 6.7%, respectively. Productivity of fruit-bodies was 725.1 g from sawdust spawn, 726.2 g from plug-shaped spawn and 573.9 g from liquid spawn until 3rd flushing of fruit-bodies. The number of fruit-bodies was 57.4 per medium of sawdust spawn, 64.3 plug-shaped spawn and 45.1 liquid spawn. At yield per medium, the percentage of media producing over 1,000 g was 13% in plug-shaped spawn and 7% in liquid spawn. However, there was no medium producing over 1,000 g of fruit-bodies in sawdust spawn. Fruit-bodies of $600{\sim}900g$ were yielded from 79% of sawdust spawn treatment, 53% plug-shaped spawn and 33% liquid spawn treatment. When compared the productivity of fruit-bodies, liquid spawn treatment appeared lower than sawdust spawn and plug-shaped spawn treatment at 1st flushing, but appeared higher than the others at 2nd and 3rd flushing.

Bioleaching of Heavy Metals from Shooting Range Soil Using a Sulfur-Oxidizing Bacteria Acidithiobacillus thiooxidans (황산화균 Acidithiobacillus thiooxidans를 이용한 사격장 토양 내 중금속 용출)

  • Applicability of bioleaching techniques using a sulfur-oxidizing bacteria, Acidithiobacillus thiooxidans, for remediation of shooting range soil contaminated with toxic heavy metals was investigated. The effects of sulfur concentration, the amount of bacterial inoculum and operation temperature on the efficiency of heavy metal solubilization were examined as well. As sulfur concentration and the amount of bacterial inoculum increased, the solubilization efficiency slightly increased; however, significant decrease of heavy metal extraction was observed with no addition of sulfur or bacterial inoculum. Bacteria solubilized the higher amount of heavy metals at $26^{\circ}C$ than $4^{\circ}C$. Lead showed the highest removal amount from the contaminated soil but the lowest removal efficiency when compared with Zn, Cu and Cr. It was likely due to formation of insoluble $PbSO_{4(s)}$ as precipitate or colloidal suspension. Sequential extraction of the microbially treated soil revealed that the proportion of readily extractable phases of Zn, Cu and Cr increased by bacterial leaching, and thus additional treatment or optimization of operation conditions such as leaching time were required for safe reuse of the soil. Bioleaching appeared to be a useful strategy for remediation of shooting range soil contaminated with heavy metals, and various operating conditions including concentration of sulfur input, inoculum volume of bacteria, and operation temperature exerted significant influence on bioleaching efficiency.
