Quality Characteristics of Fermented Soybean Products by Bacillus sp. Isolated from Traditional Soybean Paste (전통장류 유래 Bacillus sp.를 이용한 콩 발효물의 품질 특성)

  • This study evaluated the quality characteristics of soybean fermented with several Bacillus sp., which were selected based on their high enzymatic and antimicrobial activities. Total aerobic bacterial counts of fermented soybeans with HJ5-2 ($3.00{\times}10^9CFU/mL$) were the highest among all strains. Lactic acid bacteria numbered $2.50{\times}10^2{\sim}7.30{\times}10^4CFU/mL$ in soybeans fermented with isolates. Amylase and protease activities of the RD7-7 sample were the highest among all strains. Reducing sugar and amino-type nitrogen contents of fermented soybeans with HJ18-4 (2.35%) and RD7-7 (227.96 mg%) were the highest. Total amino acid contents of the samples were 16.62~18.38%, and glutamic acid, aspartic acid, leucine, lysine, arginine were major amino acids. Oxalic acid (36.51~63.57 mg/100 g) and succinic acid (429.49~600.15 mg/100 g) were the predominant organic acid. These results provide useful information for development starter (single and complex) as well as for the production of high quality fermented soybean foods.

Effect of Omija(Schizandra chinensis) Extract on the Growth Inhibition of Food Borne Pathogens in Yoghurt (오미자 추출액 첨가 요구르트의 식중독균 증식 억제 효과)

  • Escherichia coli O157:H7, Staphylococcus aureus and Salmonella enteritidis are food borne pathogens involved in food poisoning in numerous countries. This study aimed to obtain knowledges on the survival of Esc coli O157:H7, Sta aureus and Sal. enteritidis in the yoghurt added with water extract of Omija(Schizandra chinensis). The growth inhibition of Schizandra chinensis extract on the food borne pathogens were measured by total microbial count and effect of growth inhibition was correspondent to the concentration of Schizandra chinensis extract. The highest growth inhibition effect of Schizandra chinensis extract was shown on the Sta aureus followed by Sal. enteritidis and Esc. coli O157:H7. The number of surviving Esc. coli O157:H7 cell(3.55${\times}$10$\^$5/ CFU/mL) was decreased to 1.00${\times}$10$^1$∼3.00${\times}$10$^1$ CFU/mL after 24 hours incubation by the addition of 0.4∼l.0% of Schizandra chinensis extract in the yoghurt. And also the viable cell counts of surviving Sta. aureus cells (initial inoculum 1.24${\times}$10$\^$5/ CFU/mL) were decreased gradually to 4.00${\times}$10$^2$∼8.50${\times}$10$^2$ CFU/mL after 48 hours of incubation, but the viable cells of Sal. enteritidis were not detected after 24 hours of incubation. Growth of the food borne pathogens was strongly inhibited by the addition and incubation of Schizandra chinensis extract for 48 hours in the yoghurt.

  • Varietal resistance of 49 strawberry varieties to two isolates of Mycosphaerella fragariae was investigated by incoulation experiments and its chemical control was also studied. 1. The susceptibility of varieties and pathogenecity of the isolates were significantly different. 2. in most instances isolate $S_3$ and the reactions of varieties to the isolates were different. 3. Among the varieties tested, Tioga, Donner, Marhall, Northwest, Red star, Senga, sengana, Shasta, Torrey, Hokowase and Daehak No. 1 were found highly susceptible to isolate $K_2$, and America, Dabreak, Takanae, Kurumae No. 103, Horida's wander, Benizuru, Hukuba and Himiko were found as highly resistant. 4. It was apparent that America, Morioka No. 17, Takanae, Kurumae No. 13, Horida's wander, Benizuru and Hukuba were resistant to isolate $S_3$ and Marshall, Tioga were very susceptible to it. 5. As for the susceptibility of varieties by old End new leaf to isolate $K_2$, old leaves were evaluated as highly resistant. 6. Under the conditions of artificial infection Captan was the most effective in controlling for the leafspot and followed by Zineb and Difolatan.

Response of Turnip to Botrytis cinerea Infection and Their Relationship with Glucosinolate Profiles (잿빛곰팡이병에 대한 순무잎의 반응과 Glucosinolate 함량변화)

  • In order to determine the response of turnip leaves to various races of Botrytis cinerea, fourteen different races were infected to the leaf surface and measured the lesion size. Based on lesion size of the pathogen on the leaves, turnip showed susceptible response to four races, moderate to seven races and resistant to three races. Four glucosinolate (GLS) compounds, such as butenyl GLS, indole-3ylmethyl GLS (I3M), 4-methoxy-indole-3-ylmethyl GLS (4MOI3M) and normal methoxy-indole-3ylmethyl GLS (NMOI3M) were identified in turnip leaves infected with Botrytis cinerea. Leaves infected with resistant races showed higher GLS contents as compared with the leaves infected with susceptible races. Contents of I3M in the leaves with resistant races were 2.5 times as high as those in uninfected leaves, whereas I3M in the leaves with susceptible races showed lower contents than those in untreated leaves. Leaves infected with resistant races showed 4MOI3M and NMOI3M contents 2.3 and 2.7 times as high as those in uninfected leaves, respectively. GLSs in the infected leaves were most abundant at 5~10 mm area from center of the lesion. However, the GLSs in 5~10 mm area were rapidly degradated at leaves with susceptible races which resulted in continuous expansion of the lesion on the leaves, whereas no degradation was obseveved at leaves with resistant races. These results suggest that accumulation and degradation of GLS compounds in turnip leaves closely related to the susceptibility and resistance of turnip leaves to Botrytis races.

Isolation and Characteristics of Acetic Acid Bacteria for Persimmon Vinegar Fermentation (감식초 발효용 초산균의 분리 및 특성)

  • This study was carried out to investigate characteristics of acetic acid bacteria for persimmon vinegar fermen-tation. Acetic acid bacteria were isolated from statically fermented vinegar. Microscopically the cells appeared as non-motile and non-flagellated, preferentially occuring in pairs. Isolated strains were able to grow in the presence of acetic acid, ethanol, and glucose. Four of them produced 2-ketogluconic acid but did not produce 5-ketogluconic acid. The four strains isolated from statically fermented vinegar were considered to be grouped into Acetobacter. The strains were inoculated into persimmon juice for persimmon vinegar fermentation. They produced acetic acid in the range of 5.25$\∼$5.68$\%$.

Isolation and Characterization of Acetic Acid Bacteria for Producing "Makgeolli Seed-Vinegar" (막걸리 종초 제조에 적합한 초산균의 분리 및 발효특성)

  • Acetic acid bacteria strains were isolated from commercial natural vinegar. Ten isolated strains were identified using 16S rRNA gene sequencing data after evaluating the ethanol- and sulfur-tolerance. Eight of the strains isolated were identified as Acetobacter pasteurianus. A. pasteurianus JAC002, JAC005, and JAC008 strains, which showed a high ethanol tolerance, were selected for making "Makgeolli seed-vinegar". Rice wine vinegars were manufactured with the selected strains through fermentation, and their physicochemical properties and antimicrobial activities were evaluated. A. pasteurianus JAC002 strain showed the highest oxidation ability to acetic acid from ethanol on the twentieth day of fermentation, resulting in 4.21% total acidity, 3,791.77 mg% acetic acid content, and 2,931.78 mg% ethanol consumption content. Rice wine vinegar manufactured with the A. pasteurianus JAC002 strain showed increased antimicrobial activities against Staphylococcus aureus (KACC1927) and Escherichia coli (KACC10115). As a result, A. pasteurianus JAC002 strain was found to be the most suitable strain for "Makgeolli seed-vinegar".

Effect of Heterofermentative Lactic Acid Bacteria on the Quality of Italian Ryegrass and Whole-crop Barley Silage (Hetero 발효 유산균주 첨가가 사일리지용 이탈리안 라이그라스와 총체보리의 품질에 미치는 효과)

  • This study was conducted to determine the quality of italian ryegrass (IRG) and whole- crop barley (WCB) silage combined with heterofermentative lactic acid bacteria (LAB) during fermentation. Six strains of LAB (L. plantarum IMAU 70164, L. acidophilus KACC 12419, L. casei KACC 12413, L. reuteri KCTC 3594, L. buchneri KACC 12416 and L. diolivorans KACC 12385) were used in this study. L. casei and L. reuteri had the highest propionic acid production and were therefore used for fermenting the forages. The forages were fermented using monoculture and co-culture of L. casei and L. reuteri for 30, 45 and 60 days of ensiling. Addition of LAB lowered the pH of the IRG silage (p<0.05). Moisture content, lactic acid and acetic acid contents were higher (p<0.05) after addition of LAB. Water soluble carbohydrate was significantly lower (p<0.05) in WCB with a co-culture containing L. casei and L. reuteri. Propionic acid production was comparatively higher after addition of LAB to WCB on days 30, 45 and 60 while butyric acid was only detected in the IRG control on day 60. Fungi was not detected within 60 days after addition of LAB in IRG and WCB. Through this experiment, improved forage preservation was achieved using a co-culture containing L. casei and L. reuteri. WCB silage had higher propionic acid concentration and thus, it was a better forage for ensiling using co-culture of L. casei and L. reuteri.

Synergistic effect of phosphate solubilization by Burkholderia strains isolated from button mushroom bed (양송이배지로부터 분리한 Burkholderia균의 인산가용화 공조효과)

  • This study investigated the synergistic effect of single inoculation and co-inoculation of phosphate-solubilizing bacteria (PSB) Burkholderia metallica JH-7 and Burkholderia contaminans JH-15. Phosphate-solubilizing abilities of these strains were assessed by measuring phosphorus content in culture media that were singly inoculated or co-inoculated with these strains for 7 days. B. metallica JH-7 was found to release the highest content of soluble phosphorus ($140.80{\mu}g\;mL^{-1}$ ) into the medium, followed by single inoculation of B. contaminans JH-15 ($135.95{\mu}g\;mL^{-1}$ ) and co-inoculation of two strains ($134.84{\mu}g\;mL^{-1}$ ). The highest pH reduction, organic acid production, and glucose consumption were observed in the medium inoculated with B. metallica JH-7 alone compared with that in the medium co-inoculated with both the strains. Results of a plant growth promotion bioassay showed 17.4% and 7.48% higher leaf and root growth, respectively, in romaine lettuce inoculated with B. metallica JH-7 alone than in romaine lettuce inoculated with a control strain. However, no significant difference was observed between single inoculation and co-inoculation of these strains with respect to phosphorus release and plant growth. Although the results of the present study did not show the synergistic effect of phosphate solubilization by the PSB strains examined, these results indicate that treatment with PSB exerts a beneficial effect on crop growth.

The effect of local rifampicin instillation on the treatment of suppurative BCG lymphadenitis (BCG 접종에 따른 화농성 림프절염의 rifampicin 국소투여 효과)

  • Purpose : The purpose of this study was to evaluate the types of lymphadenitis after BCG vaccination and the effect of local rifampicin instillation on the treatment of suppurative BCG lymphadenitis. Methods : A total of 32 otherwise healthy infants with suppurative BCG lymphadenitis, who visited the Department of Pediatrics of Chonbuk National University Hospital, from March 2002 through June 2004, were enrolled in this study. They were treated with needle aspiration and local rifampicin instillation. We investigated the time the lymphadenitis took to be suppurative, accompanying clinical manifestations, and the treatment effects. Results : Of the 32 infants, 19 were male and 13 were female. They were full term babies and one preterm baby with a gestational age of 30 weeks. They received intradermal administration, with the BCG vaccine of $Pasteur^{(R)}$(French) strain mostly on the left deltoid area(96.9 percent). Regional lymphadenitis occurred in 1 to 11 months after BCG vaccination, mostly 1-5 months after vaccination (78.1 percent). Among the infants, 87.5 percent had unilocular lesion but 12.5 percent had more than one enlarged lymph node cares. Most of the lymphadenitis presented in the left axillary area(77.8 percent), and the left supuraclavicular area(11.1 percent). After one to three times of needle aspiration with rifampin instillation, all infants recovered completely without surgical excision or severe complication. Conclusion : The regional lymphadenitis is the most common complication in infants who receive intradermal BCG vaccination. This study supports that in suppurative BCG lymphadenitis the needle aspiration and local rifampicin instillation is very effective and can be a more economical treatment modality.

Identification and Pathogenicity of Rhizoctonia species Isolated from Turfgrasses (잔디에서 분리한 Rhizoctonia spp.의 동정과 병원성)

  • Morphological characteristics and pathogenicity of Rhizoctonia species causing blight diseases of turfgrasses were studied. The species were identified as Rhizoctonia cerealis Van der Hoeven, R. oryzae Ryker et Gooch, and R. solani $K{\ddot{u}hn}$ based on their morphological and cultural characteristics. Isolates of R. solani were assigned to anastomosis groups (AG) with cultural type 1 (1A), 2-2 (IIIB), and 2-2 (IV). R. cerealis, R. oryzae and R. solani induced sheath rot and foliar blight symptoms on creeping bentgrass (Agrostis palustris) and zoysiagrass (Zoysia japonica). Inoculation tests showed that disease severity with isolates of R. cerealis and R. oryzae were more serious to creeping bentgrass than zoysiagrass. AG 1(1A) isolates of R. solani were strongly pathogenic on creeping bentgrass, but moderate to zoysiagrass. AG 2-2 (III) isolates were moderately pathogenic to zoysiagrass, but weakly to creeping bentgrass. AG 2-2 (IV) isolates from zoysiagrass were moderately pathogenic to zoysiagrass, but weakly to creeping bentgrass.
