Distinct Spatio-temporal Expression Patterns of Patatin Promoter-GUS Gene Fusion in Transgenic Potato Microtubers (형질전환 감자 소괴경의 발달단계에 따른 Patatin Promoter-GUS 유전자의 발현 분석)

  • This study was carried out to investigate the expression patterns of foreign gene that controlled by tuber-specific patatin promoter in transgenic potatoes. Potato leaf disc cultured in vitro were transformed by the Agrobacterium strain LBA4404 containing pBl121 or pATGUS from potato cv. Desiree. In order to select the transgenic lines, gene-specific primers deduced from the NPTII were synthesized and used for polymerase chain reaction. The down part of the putative transgenic potatoes was transplanted weekly onto sucrose-enriched medium to accelerate the microtuber formation. RNA gel blot analysis was performed on the total RNAs obtained from tuber that had been harvested at a week interval. Also, histochemical assay was observed in the explants transformed with either pBI121 or pATGUS. Results showed that the transgenic plant containing pATGUS expressed GUS transcripts mainly at the tuber, not in stem, with the highest expression level in 5 weeks-grown microtubers. In contrast to pATGUS plants, the transformed plants with pBI121 showed an equal expression pattern throughout the whole developing stages. Consistent with RNA gel blot analysis, histochemical GUS staining and enzyme activity exhibited pATGUS transcripts were at the highest level in 5 weeks cultures. From these results, we suggest that the best stage to analyze the foreign gene introduced by patatin promoter into potato plants is at 5 weeks cultures after tuber formation.

Effects of Growth Performance and Carcass Characteristics by Fermented Barley Grain Feeding in Post-fattening Hanwoo Steers (거세한우 비육후기 보리곡실 발효사료 급여에 따른 성장과 도체특성에 미치는 효과)

  • The purpose of this study was to investigate the proper feeding level and duration of fermented barley grain feed before harvesting to improve the availability of barley for feed. Trial 1 was to investigate the proper feeding amount of fermented barley grain fermented feed, and we prepared 32 heads (603.4 ± 42.7kg) of 22-month-old Hanwoo steer (603.4 ± 42.7kg) for 8 heads in 4 treatment groups. 48 heads (625.8 ± 13.1kg) for Trial 2 were used for 12heads per 4 treatments, and were reared for about 9 months until 30 months of age. Trial 1 is treated a Control group that feed 10㎏ of concentrate, replaces 10% fermented barley grain feed(FBGF) of the control by (TRT 1) and 20% (TRT 2) and 30%(TRT 3). Concentrate and FBGF fed 9, 2.1 kg, 8, 4.2 kg, 7 and 7, 6.3 kg on each treatments respectively and 1.5 kg of rice straw for forage. In Trial 2, 10% of alternative diets were judged to be appropriate, and the control of 9kg of diets and barley grain fermented feeds were used to determine the appropriate feeding period. The treatments were 3 months before shipment (TRT 1), 6 months (TRT 2) and 9 months (TRT32). Each treatment group had 8 and 2.1 kg of concentrate and barley grain fermentation, respectively. As a feed, rice straw was fed to 1.5 kg. The daily gains per treatment were higher in TRT 1 and TRT 3 was similar to the Control. Body weight and daily gain during the test period were higher in TRT 1 fed 10% barley grain fermented feed. TRT 2 was the highest at 6.13, and TRT 3 was 6.0, which was higher than 5.63 of TRT 1 and 5.5 of Control.

Effect of Mountain Grazing on Growth Performance and Carcass Characteristics of Hanwoo Steers (산간지 방목이 거세한우의 성장특성 및 도체형질에 미치는 영향)

  • This study was carried out to investigate the effect of mountain grazing on growth performance and carcass characteristics of Hanwoo steers. Thirty two Hanwoo steers were randomly assigned. Control were fed concentrate + forage until 30 month of age. Treatment1 were fed concentrate + forage after mountain grazing from 8 to 13 months. Treatment2 and 3 were fed concentrate + forage after mountain grazing from 8 to 17 months. Hanwoo steers on Treatment 1 and 2 were slaughtered at the same age (30 months) as the control, and Hanwoo steers on Treatment 3 were slaughtered at 31 months. The average daily gain (ADG) were higher in the control than in the other treatments during the growing period (p<0.05). Carcass back fat thickness was thicker in the TRT 3 and marbling score was higher in the TRT 1 than in the other treatments; however, the differences were not statistically significant. There was no difference in the meat composition and shear force of the longissimus muscle according to the mountain grazing. However, the yellowness (b) of fat colors were significantly lower in the control than in the TRT 1 and TRT 2 (p<0.05). The effect of grazing on fatty acid composition was not constant. In conclusion, mountain grazing could reduce meat color without affecting the growth and meat quality of Hanwoo steers, and further research on the quality, yield and intake of grassland would be needed.

Changes in Physio-Chemical Properties of Deer Meat During Storage at -30°C and -60°C (동결 온도와 저장기간에 따른 사슴고기의 이화화적 성질의 변화)

  • A total of 5 female elk deer($220kg{\pm}10kg$) were included in a study on the changes in physico-chemical properties of deer meat during storage at $-4^{\circ}C\;and\;-2^{\circ}C$. The deers were exposed to normal pre-slaughter handling and put under anesthesia before slaughtered. The loin and leg cuts were deboned from the carcass after 24hrs slaughter. The samples weighing approximately 300g were packaged using wrap packaging and stored for 1, 2, 3, 4, 5 and 6 months at $-30^{\circ}C\;and\;-60^{\circ}C$. During the freezing period, the changes of moisture maintenance was slower for the meats kept at $-60^{\circ}C$ than that at $-30^{\circ}C$. The meat softness represented higher value after freezing, and it tended to be decreased with the passage of storage time. Comparing the values between freezing temperature, the value at $-30^{\circ}C$ was higher than at $-60^{\circ}C$. All meats frozen at $-30^{\circ}C\;and\;-60^{\circ}C$ were edible after storage for 6 months. Loin was inedible, and leg was edible after freezing storage for 6 months. The pH values of the meats were increased during freezing period, even though pH value of the meats stored for 6 months was less than 5.70. Luminosity of loin was significantly increased with the passage of storage time, although loin stored at $-60^{\circ}C$ was darker, and leg stored at $-30^{\circ}C$ was darker. The index of red color was rapidly decreased for loin stored at $-30^{\circ}C$, and for leg stored at $-60^{\circ}C$, respectively. There were no changes in the index of yellow color for loin stored at $-60^{\circ}C$, and the index for leg was increased with regardless of the freezing storage temperature after 6 months.

Physicochemical Properties and Sensory Characteristics of Korean Native Beef Loin Fed with Citrus Byproduct (한우등심의 이화학적 및 관능특성에 미치는 감귤박의 영향)

  • The purpose of this research is to study the effects of feeding dietary citrus byproducts TMR (total mixed ration) on physicochemical properties and sensory characteristics of Korean native beef loin (KNBL). The samples for experiment consist of the KNBL not fed with citrus byproducts (TMR-0) and the KNBL fed with citrus byproducts during fattening period (TMR-1). The control (TMR-0) KNBL was fed by general practical feeding (roughages and concentrates were fed separately), while the TMR-1 KNBL was fed by the same as TMR-0 until 17 months yearling but was fed by citrus byproducts feeding for 10 months after that. The $L^*(lightness),\;a^*(redness)\;and\;b^*(yellowness)$ value were not significantly different between TMR-0 and TMR-1. The pH of TMR-1 was lower than that of TMR-0 (p<0.05), the VBN content, TBARS value and EDA were not significantly different between TMR-0 and TMR-1. The water holding capacity, frozen loss and cooking loss were not significantly different between TMR-0 and TMR-1, but thawing loss of TMR-0 was higher than that of TMR-1 (p<0.05). The hardness of TMR-0 was higher than that of TMR-1, and the springiness of TMR-1 was higher than that of TMR-0 (p<0.05), but the cohesiveness, gumminess, chewiness and shear force were not significantly different between TMR-0 and TMR-1. The pH and VBN content during storage were not significantly different between TMR-0 and TMR-1, but the TBARS value of TMR-1 stored during 4 weeks was lower than that of TMR-0 (p<0.05). In case of sensory score, the color and aroma of raw meat, and the taste, juiciness and palatability of cooked meat were not significantly different between TMR-0 and TMR-1. But the flavor and tenderness of TMR-1 were superior than those of TMR-0 (p<0.05)

국제 수학 올림피아드 참가 후보자들을 위한 상황대처훈련에 관한연구

  • Currently Korea encourages gifted high schoolers and junior high schoolers to participate in international achievement contests such as International Olympiads. Participants for these contests are selected nationwide among gifted students in areas of mathematics, physics, chemistry, and others. They go through a series of screening tests and programs. One of the screening processes IS Korean Olympiad School, which provides study programs each summer for student-candidates prior to following year's International Olympiads. Approximately 40 students of high schools and junior high schools, in each subject of study, gather at Korean Olympiad Summer School, and they go through intensive study programs during a short period of time. Out of 40 candidates,' less than 10 students are finally selected to participate in International Olympiads. In this study, a psycho-educational program called "Situation Coping Training Program" was developed to enhance ahievement motivation for these student-candidates. This study was to see if this training program actually improved their cognitive, emotive motivation factors, and to see how this training program affected their achievement level. Training was administered for five days. This training program was found effective for participants to increase self-efficacy, internal locus of control, and anxiety copmg. These cognitive and emotive motivation factors, other than intelligence, were found to have positive relationship with achievement level, of which self-efficacy and attribution style of students were found as two best predictors of achievement. This training program was perceived as necessary. by participants, and helpful for recovering self-confidence and self-control as well as coping pressure. Suggestions were made that this kind of training program be administered as a regular curriculum in preparative study programs such as Korean Olympiads, since cognitive, emotive motivation factors are related with achievement, and furthermore, be utilized in all gifted education programs in Korea. in Korea.

Effect of Rearing Period on Yield Rate, Physical Properties and Fatty Acid Composition of Duck Meats (출하 일령에 따른 오리육의 수율, 물리적 특성 및 지방산 조성 변화)

  • This study was performed to investigate the effect of rearing period(45 and 70 days) on the changes of the yield, physical trait and the composition of fatty acids. The average weight of 100 duck meat at 45 days post hatch was 2.2kg. The yield of meats from prime cuts of 2.1kg, which are the most consumed today, was composed to whole legs($14.1\%$), wings($8.7\%$) and breast(12.5\%$). The ratio of whole legs and breast was similar, In the secondary cuts, the meats was consisted in drumstick($12.5\%$), thigh($7.3\%$), breast($11.7\%$), tender lion($0.8\%$), drum matte($4.9\%$) and wing($2.6\%$). Especially, the drumstick was twice higher than the wing. The content of oleic acid was 50.87 to $51.32\%$ and 49.84 to $50.03\%$ in the breasts and leg, respectively. In the breast, meat at 75 days the oleic acid was slightly increased DHA was not detected in the breast meat, However, it was $0.13\%$ in the leg. The $a^{*}$ value in meat color, ranged was 16.67 to 17.92 in the breast and 15.81 to 17.15 in the leg. Statistically, there was no significantly difference between the breast and leg. The cooking loss of the breast and leg was $26.37\%\;and\;30.32\%$, respectively. The shear force value was $2.20\;kg/0.5inch^2$ at 45 and $2.84\;kg/0.5inch^2$ at 75 days post hatchet respectively. Therefore, the raring period was effect on the shear force value, but not on the cooking loss and meat color.

Effect of Feeding Probiotics on Quality Properties of Pork (생균제 급여가 돈육의 품질 특성에 미치는 영향)

  • A total of 120 pigs were used to investigate the effect of feeding probiotics on quality properties of pork. About 6 kg pigs were randomly alloted into one or three experimental diet groups (C: commercial diet feed; T1: 0.1% KBC1121 feed; T2: 0.1% YC2000+0.1% KBC1121 feed). Pigs were slaughtered at approximately 110 kg live weight and chemical composition and physico-chemical characteristics were measured in pork loin. Moisture, crude protein and crude ash were not differences among the treatments. However, crude fat content of T2 was significantly higher than that of other treatments. All of dietary probiotic groups showed significantly higher pH than control. WHC was significantly higher in T1 than other treatments. Cooking loss, shear force value and cholesterol content were not differences among the treatments. In meat color, $L^*$ value was not difference among the treatments, but $a^*\;and\;b^*$ value were lower in T1 than other treatments. In textrure properties of cooked meat, brittleness, hardness, gumminess and chewiness value were significantly higher in T1 than other treatments. Sensory evaluation was not difference among the treatments. The myristic, stearic and oleic acid content of T2 were significantly higher than those of other treatments. Whereas linoleic acid was significantly lower than other treatments. Unsaturated fatty acid (UFA) was significantly higher in T1 than T2. Essential fatty acid (EFA) and EFA/UFA were higher in the order of T1 > C > T2. In amino acid composition, total and essential amino acid, aspartic acid, threonine, serine, glutamic acid, valine, isoleucine, leucine and lysine level were lower in T2 than other treatments.

Effect of pH Control on Physico-chemical Characteristics of Chicken Breast Surimi (닭가슴살 Surimi의 이화학적 특성에 미치는 pH 조절의 영향)

  • This study was conducted to determine the effect of pH adjustment on physico-chemical characteristics of chicken breast surimi. The chicken breast meat was ground with distilled water, of which pH was then adjusted to 2.5 (T1), 3.0 (T2), 10.5 (T3) and 11.0 (T4) for surimi manufacture, respectively. Water content was higher in order of T4>T1>T3>T2 (p<0.05). Crude protein and crude fat were higher in T3 and T4 compared with T1 and T2 (p<0.05). $L^*$ values, myofibrillar protein and water holding capacity of T2 and T4 were higher than those of T1 and T3 (p<0.05). T4 had the lowest yield among the treatments (p<0.05). T1 was higher in yield and pH, whereas breaking force and deformation were higher in T1 (p<0.05). $a^*$ was higher in order of T3>T2>T4>T1 and $b^*$ was lower in T1 compared with other treatments (p<0.05). In textural properties, the chewiness values of T2 and T3 were higher than those of T1 and T4, the hardness was higher in order of T2>T3>T4>T1 (p<0.05). Cohesiveness and gumminess of T1 showed higher values than those of other treatments (p<0.05). In sensory evaluation, the note for appearance was higher in T2 than other treatments (p<0.05), however other traits were not significantly different (p>0.05). Therefore, the alkaline processing (T4, pH 11.0) would be recommended.

Effect of Stevia and Charcoal as an Alternative to Antibiotics on Carcass Characteristics and Meat Quality in Finishing Pigs (돼지의 도체 및 육질특성에서 스테비아와 숯의 항생제 대체효과)

  • This study was conducted to determine effect of dietary reduced antibiotics supplementation on carcass characteristics and meat quality of finishing pigs fed stevia and charcoal. A total of 180 pigs (LYD) were randomly allocated into 3 treatments with 3 replications. Dietary treatments were 1) T1 (control, basal diet, no stevia and charcoal addition, antibiotics both in early and late fattening periods), 2) T2 (basal diet, 0.3% stevia + 0.3% charcoal, antibiotics both in early and late fattening periods), 3) T3 (basal diet, 0.3% stevia + 0.3% charcoal, antibiotics in early fattening period only). At each marketing day, pigs were conventionally slaughtered, examined the carcass characteristics and loin (Longissimus) muscles were removed for the meat quality traits. In the carcass characteristics, T3 group showed higher incidence of A carcass grade compared to the other treatments. Backfat thickness was higher in T2 group compared to the others (p<0.05). In the meat quality traits, pH was higher in T1 group than T3 group (p<0.05). Cooking loss was higher in T2 group than T1 group (p<0.05). However, WHC (water holding capacity), drip loss and shear force values did not show any significant differences among treatments. In the panel test, there were no significant differences in tenderness, juiciness, flavor, and total acceptability scores among treatments. As a result, dietary supplementation of reduced antibiotics to finishing pigs fed stevia and charcoal showed similar growth performance and meat quality traits compared to conventional method.
