Discrimination of Korean Domestic and Foreign Soybeans using Near Infrared Reflectance Spectroscopy (근적외선분광광도계(NIRS)를 이용한 국내산 콩과 수입콩의 판별분석)

  • Discrimination of geographic origin of agricultural products is a important issue in Korea because the price difference between Korean domestic and imported cereals is a key among some reasons. NIRS (Near Infrared Reflectance Spectroscopy) has been applied to classify the geographical origin of soybeans. Total 135 samples (Korean domestic 92 and foreign 43) were used to obtain calibration equation through 400~2,500 nm wavelength. The math treatment with 1st derivative and 4 nm gap and the modified partial least squares(MPLS)regression was outstanding for calibration equation. The standard error of calibration and determination coefficient in calibration set(n=115) was 6.65 and 0.98, respectively. And it showed that the extra 20 samples for validation equation were identified their authentication correctly. This study describes that the application of NIRS would be possible for discrimination of geographical origin between Korean domestic and imported soybeans.

A Study on Synthetic Method and Material Analysis of Calcium Ammine Chloride as Ammonia Transport Materials for Solid SCR (Solid SCR용 암모니아 저장물질인 Calcium Ammine Chloride의 합성방법 및 물질분석 연구)

  • Solid materials of ammonia sources with SCR have been considered for the application of lean NOx reduction in automobile industry, to overcome complex problems of liquid urea based SCR. These solid materials produce ammonia gas directly with proper heating and can be packaged by compact size, because of high volumetric ammonia density. Among ammonium salts and metal ammine chlorides, calcium ammine chloride was focused on this paper due to low decomposition temperature. In order to make calcium ammine chloride in lab-scale, simple reactor and glove box was designed and built with ammonium gas tank, regulator, and sensors. Basic test conditions of charging ammonia gas to anhydrous calcium chloride are chosen from equilibrium vapor pressure by Van't Hoff plot based on thermodynamic properties of materials. Synthetic method of calcium ammine chloride were studied for different durations, temperatures, and pressures with proper ammonia gas charged, as a respect of ammonia gas adsorption rate(%) from simple weight calculations which were confirmed by IC. Also, lab-made calcium ammine chloride were analyzed by TGA and DSC to clarify decomposition step in the equations of chemical reaction. To understand material characteristics for lab-made calcium ammine chloride, DA, XRD and FT-IR analysis were performed with published data of literature. From analytical results, water content in lab-made calcium ammine chloride can be discovered and new test procedures of water removal were proposed.

Extraction of Ginseng Saponins in Supercritical Ammonia Fluids (초임계 암모니아 유체에서의 인삼 사포닌 추출)

  • The extraction of ginseng saponins with near critical or supercritical ammonia(SCF-NH$_3$) was carried out at 80$^{\circ}C$-160$^{\circ}C$ and ammonia densities of 339.8-525 $mg/cm^3$. In order to evaluate brownish color of white ginseng extracts, a spetrophotometric method was applied in ultraviolet and visible range. The extractibilities of ginseonoside $Rb_1$, -$Rb_2$, -Rc, -Rd, -Re, -Rf, $-Rg_1$, and $-Rg_2$ were determined by high performance liquid choromatography. The best extractability was 7.36% at 133$^{\circ}C$ and 403 $\pm$ 24.605 $mg/cm^3$. In the case of the high extraction temperature, it is thought that extraction times can be reduced and the selectivity of protopanaxatriol can be increased. The brownish color of extracts is affected by temperature and extractability. When extraction temperature is between 132$^{\circ}C$ and 140$^{\circ}C$, it is shown that the range is the retrograde region where extractablility decrease with increasing temperature.

A Study on the Identification of Animal Hair in Food (식품 중 동물 털 이물의 판별법 연구)

  • Foreign materials with a variety of types and sizes are found in food; thus, extraordinary efforts and various analytical methods are required to identify the types of foreign materials and to find out accurate causes of how they unintentionally enter food. In this study, human, cow, pig, mouse, duck, goose, dog, and cat were chosen as various types of animal hairs because they can be frequently incorporated into food during its production or consumption step. We morphologically analyzed them using stereoscopic, optical, SUMP method, and scanning electron microscopes, showing differences in each type. In addition, X-ray fluorescence spectrometer (XRF) was used to analysis chemical compositions ($^{11}Na{\sim}^{92}U$, Mass%) of samples. As a result, we observed that mammalian hairs were mainly composed of sulfur. Organic compounds of samples were further analyzed by fourier transform infrared spectroscopy (FT-IR) that can compare spectra of given materials; however, this method did not show significant differences in each sample. In this study, we suggest a rapid method for the identification of the causes and types of foreign materials in food.

Determination of Barley Grain Components at Different Maturing Stages by Near Infrared Reflectance Spectroscopic Analysis (근적외선분광분석법에 의한 등숙시기별 보리종실의 성분측정)

  • This study was conducted to establish the rapid determination method for major components of maturing covered barley grains, and to improve the efficiency of selection in barley breeding. Near Infrared Reflectance Spectroscopy (NIRS) is an established, economical and nondestructive technique applied widely to the food and feed industry. 34 barley lines were sampled at 5 day-interval from 25 to 35 days after heading. A standard regression analysis for the data obtained by analytical laboratory methods and NIRS method was carried out to get a useful calibration equation. The simple significant correlation between these two methods at 25 days after heading was recognized in starch and $\beta$-glucan contents. At 30 days after heading the data obtained by two methods showed significant correlation in starch, $\beta$-glucan and protein contents. Analyzed data and that from NIRS method at 35 days after heading was significantly correlated in starch and protein contents. It was concluded that the applicability of NIRS method for the components analysis in maturing barley grains was different depending on maturing stages and components.

Establishment of discrimination system using multivariate analysis of FT-IR spectroscopy data from different species of artichoke (Cynara cardunculus var. scolymus L.) (FT-IR 스펙트럼 데이터 기반 다변량통계분석기법을 이용한 아티초크의 대사체 수준 품종 분류)

  • To determine whether FT-IR spectral analysis based on multivariate analysis for whole cell extracts can be used to discriminate between artichoke (Cynara cardunculus var. scolymus L.) plants at the metabolic level, leaves of ten artichoke plants were subjected to Fourier transform infrared(FT-IR) spectroscopy. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700-1,500, 1,500-1,300 and $1,100-950cm^{-1}$, respectively. These spectral regions reflect the quantitative and qualitative variations of amide I, II from amino acids and proteins ($1,700-1,500cm^{-1}$), phosphodiester groups from nucleic acid and phospholipid ($1,500-1,300cm^{-1}$) and carbohydrate compounds ($1,100-950cm^{-1}$). PCA revealed separate clusters that corresponded to their species relationship. Thus, PCA could be used to distinguish between artichoke species with different metabolite contents. PLS-DA showed similar species classification of artichoke. Furthermore these metabolic discrimination systems could be used for the rapid selection and classification of useful artichoke cultivars.

A study on the distribution of glitter particles from an university building (대학 강의실에서 글리터의 분포 연구)

  • A distribution study of glitter was conducted from a local university building. The potential recipient surfaces chosen were the 1,000 chairs kept in 16 separate classrooms of the building. The surface of chairs contacting with buttocks and back of users were tape lifted with commercial adhesive tapes, and the surface of adhesive tapes were examined with a stereomicroscope to locate the glitters. 12 glitter particles were found from 11 chairs kept in 16 classrooms of a building. This result indicates that the possibility of glitter finding from Korean violent crime scene is high. The 12 glitters found from the chairs were analyzed with a stereomicroscope and an FTIR spectrophotometer to find the matching glitter. The stereomicroscope was used for the observation of the shapes, dimensions, colors and holograms of each glitter, and the FTIR spectrophotometer was used for the analysis of the chemical composition of the coating material of each glitter. The examination results revealed that 8 glitters out of 12 glitters were not sharing the common origin each other. Also, two glitters found from two separate classroom chairs share common microscopic and FTIR spectroscopic characteristics which indicating two glitters share common origin. The record of the students who had used the classrooms was traced and the authors could find that the classrooms where the same glitters were found were shared by a group of students who listen to the same class. This result implies that the path of the involved people should be considered while assessing the value of trace evidence.

Preparation and Characterization of O-Carboxymethyl Chitosan Ion-complexed Poly(L-Lysine) for Drug and Gene Delivery System (약물 및 유전자 전달체로 응용하기 위한 Poly(L-Lysine)이 결합된 O-Carboxymethyl Chitosan PEG의 제조와 특성)

  • O-carboxymethyl water-soluble chitosan (OCMCh) prepared for enhance the application of chitosan was modified with mthoxy polyethyleneglycol (mPEG) by ion-complex for long circulation in the blood. OCMCh-PEG-PLLs was prepared by forming ion-complex with OCMCh-PEG and Poly(L-Lysine) (PLL) for drug and gene delivery system. The physicochemcal characterisitcs of OCMCh-PEG-PLLs were investigated by FT-IR, $^1H$-NMR. These results showed that CMCh-PEG-PLLs were successfully syntehsized by ion-complex. Particle size distribution and zeta potential of the OCMCh-PEG-PLLs were determined using dynamic light scattering technique. Transmission electron microscopy (TEM) was also used to observe the morphology of the OCMCh-PEG-PLLs. OCMCh-PEG-PLLs have spherical shapes with particle size 290∼390 nm. OCMCh-PEG-PLLs were showed when the feeding amount of mPEG ratio was increased, particle size and zeta potential were decreased. Based on these results, it is possible to introduction of the OCMCh-PEG-PLLs into various biomedical fields such as drug and gene delivery system.

A Study on the Quality Characteristics of Feedstocks for Power Bio-Fuel Oil (발전용 바이오중유용 원료물질의 품질특성 연구)

  • As it carry out RPS(Renewable Portfolio Standards), power producers are using the power bio-fuel oil to meet their RPS quota. In this study, we test the quality characteristics of raw materials for power bio-fuel oil and the property changes of power bio-fuel oil by the kind of feedstocks. The power bio-fuel oil and feedstocks were analyzed for item of quality standard such as acid number, viscosity and metal contents. And it was analyzed for composition distribution by FT-IT and HPLC. Such as low priced palm oil series has high acid number and ash contents due to high free-fatty acid and metal contents. And by-product of biodiesel have a tendency of high viscosity. The fuel properties of power bio-fuel oil, such as viscosity, acid number and metal contents are correlated with the constituent and the mixing ratio of the feedstocks.

Scientific Investigation of the Clothes Collected at Comfort Station in Nara, Japan (일본 나라현 위안소 수습 의복 조사 및 과학적 분석)

  • The aim of this study was to obtain information about two early-20th Century clothes, for which the "National Memorial Museum of Forced Mobilization under Japanese Occupation" has sought to receive preservation treatment. Optical microscopes and a scanning electron microscope were used to investigate the weaving of the clothes, and Fourier transform infrared spectroscopy(FT-IR) was used to investigate the fibers. Cloth A is believed to be a Japanese half sleeved inner wear(Hanjuban) used by women. Cloth B is believed to be working clothing that was checked by an Osaka plant. This was verified by a book written by the Japanese army. Both of the clothes were made mostly from cotton, although the inner wear also used viscose rayon on the neck collar. The button on the working wear was made of urea formaldehyde resin, an early precursor to plastic.
