  • Trypsin inhibitor produced by Streptomyces sp. was investigated its reactive characteristics against trypsin. The mode of inhibition against trypsin was mixed type of non-competitive and competitive with casein, and enzyme-inhibitor complex was formed rapidly. The inhibitory activity was increased by the addition of isoleucine and depressed by silver, mercuric or cupric ion. And when egg albumin or hemoglobin was used as substrate for trypsin, the inhibition ratio was changed. The inhibitor inhibited coagulation of blood of bovine.

  • A series of $N^1$-alkylnicotinamide chlorides, $N^1$-methyl-to $N^1$-dodecylnicotinamides inclusive were studied with rabbit muscle L-${\alpha}$-glycerophosphate dehydrogenase to investigate the possibility of reversible and irreversible inactivation of the pyridine-linked dehydrogenases by the coenzyme-competitive inhibitor derivatives. The inhibition of the enzyme by $N^1$-alkylnicotinamide chlorides was demonstrated to be reversible at the dilute concentration of the inhibitors but this reversible inhibition was found to be followed by an irreversible time-dependent inactivation measuable at high concentrations of the inhibitors. The properties of this time-dependent inactivation were discussed on the basis of the denaturation of the enzyme by the binding of small micelle-like structures formed at higher concentrations of the inhibitors.

  • The hot water extract from cinnamon (Cinnamomum cassia Presl) inhibited the activity of alcohol dehydrogenase (ADH) with IC50 value of 45.6 ㎍/mL. The ADH inhibitory components in cinnamon extract were relatively stable to acid and heat, but were found to be volatile. The optimum temperature and time for extracting the ADH inhibitory components from cinnamon were 80 ℃ and 2 h, respectively. Among the essential oils of cinnamon, cinnamaldehyde was the main substance for ADH inhibition. Cinnamaldehyde is considered a competitive inhibitor of ethanol to ADH. Therefore, the cinnamon extract and cinnamaldehyde showed the potential to be used as natural materials for relieving symptoms of a hangover.

Inhibitory Effects of the Solvent Fractions from Persimmon Leaves on Xanthine Oxidise Activity (용매별 감잎 추출물의 Xanthine Oxidase 저해효과)

  • The inhibitory effects of xanthine oxidase by the methanol extract and the solvent fractions obtained from persimmon leaves were investigated. The inhibition ratio of xanthine oxidase was 78% by addition of 2.0mg/ml of the methanol extract. Among the solvent fractions, the ethylacetate fraction showed the strongest inhibitory effect against the xanthine oxidase, followed by the hexane fraction. The effect increased with addition of the ethylacetate fraction. At a concentration of 2.0mg/ml of the ethylacetate fraction, 65% of the enzyme activity decreased within 1.0 min of incubation with xanthine oxidase. But the activity of xanthine oxidise did not decrease significantly by the length of the incubation time.

  • This study was conducted to investigate the antimicrobial activity of Prunus mume extracts. Prunus mume extracts have growth inhibitory activity to various microorganisms. Gram positive bacteria such as Bacillus cereus, Bacillus subtilis, Micrococcus leteus, Staphylococcus epidrimidis were more easily inhibited than Gram negative bacteria tested Vibrio parahaemolyticus, Salmonella typhimurium, Escherichia coli, Proteus vulgaris and Yeasts by Prunus mume extracts. Among several fractions of methanol extract, EtOAc & BuOH fractions were showed strong antibacterial activities, but those fractions were not showed on fungi.

Effect of Extract of Fermented Dropwort on Intestinal Bacteria and Enzymes In Vitro (미나리발효액이 장내 유해세균 및 유익균의 In Vitro 생육 및 효소활성에 미치는 영향)

  • Effect of extract of fermented dropwort (Oenanthe stolonifera) on growth of intestinal harmful/useful bacteria and enzyme activity were investigated in vitro. The extract showed strong inhibition on harmful microbes including Vibrio and Salmonella, but mild inhibition on Bifidobacterium longum in both agar plate and liquid cultivation. Minimum inhibitory concentration (MIC) value of B. longum was the highest among tested microbes. Inhibition effect of fermented extract on harmful microbes increased according to fermentation period. Extract of fermented dropwort showed inhibitory effects on activity of microbial ${\beta}$-glucuronidase and tryptophanase. The inhibitory effects were also proportional to fermentation period. As consequence, it is assumed that the uptake of fermented dropwort might be useful for human intestinal health.

Characterization of Microsomal $Ca^{2+}$ Uptake in Tomato Root Tissues (토마토 뿌리조직에서 분리한 마이크로솜의 $Ca^{2+}$ 흡수 특성)

  • In order to characterize the property of $Ca^{2+}$ transport in plant cell, microsomes were prepared from the roots of tomato and microsomal $^{45}Ca^{2+}$ uptake was measured. When 1 mM vanadate, a selective inhibitor of P-type ATPases, 50 mM $NO_3^-$, a specific inhibitor of vacuolar $H^{+}-ATPase$, and both of these inhibitors were treated, the microsomal $^{45}Ca^{2+}$ uptakes were inhibited by 20, 33 and 47%, respectively. The inhibitory effects of these two inhibitors were investigated by using a protonophore, gramicidin. When the chemical gradient of $H^{+}$ was relieved by gramicidin, the uptake was decreased by 30%, implying the presence of $Ca^{2+}/H^+$ antiporter in the microsomal membrane. In the $^{45}Ca^{2+}$ uptake experiment, the effect of gramicidin was independent of vanadate-induced inhibition. However, when the activity of vacuolar $H^{+}-ATPase$ was inhibited by $NO_3^-$, the effect of gramicidin was severely decreased. Meanwhile, thapsigargin, a specific antagonist of ER/SR-type $Ca^{2+}-ATPase$, inhibited the microsomal $^{45}Ca^{2+}$ uptake and the maximum inhibitory effect was obtained at $10\;{\mu}M$. The effect of thapsigargin was blocked by $NO_3^-$ and gramicidin, but not by vanadate. These results imply that vanadate directly inhibits the activity of $Ca^{2+}-ATPase$; however, $NO_3^-$ and thapsigargin block the activity of $Ca^{2+}/H^+$ antiporter by inhibiting the vacuolar $H^{+}-ATPase$. In conclusion, the microsomal $^{45}Ca^{2+}$ uptakes are mediated by two major enzymes, $Ca^{2+}-ATPase$ and $Ca^{2+}/H^+$ antiporter in tomato root tissue.

  • The bioactivity of garlic extract was evaluated, based on the inhibition of soybean lipoxygenase. While the inhibition of lipoxygenase by the chloroform extract (1$_{50}$ value after 10min precirculation, 55mg/$m\ell$) of garlic homogenate shows the property as irreversible inhibitors, the aqueous extract (1$_{50}$ value, 65mg/$m\ell$) appeared to contain mainly reversible inhibitors. In the related study, diallyldislufide and dimethyldisulfide inhibited the enzyme with 1$_{50}$ value of 1.3mM and 18mM, respectively. These disulfides demonstrated both irreversible and reversible patterns of inhibition. In addition, synthetic alliin(allylcysteine sulfoxide) was found to inhibit the enzyme at high concentration (approximately 22%, at 10mM), and its decomposition products showed the irreversible property in the inhibition, in contrast to S-ethyl cysteine sulfoxide which expressed no significant inhibition. Thus, it is suggested that the garlic macerate contains both irreversible and reversible sulfur inhibitors.itors.

Inhibition of Microsomal $H^+\;-\;ATPase$ Prepared from Tomato Roots by Various Anions (음이온에 의한 토마토 뿌리조직 마이크로솜 $H^+-ATPase$ 활성 저해)

  • $H^+-ATPase$ located on plasma and vacuolar membranes play major roles in various cellular physiological processes. In order to investigate the physiological roles of $H^+-ATPase$, microsomes were prepared from tomato roots and the effects of various anions were measured on the activities of $H^+-ATPase$. $H^+-ATPase$ was inhibited by various anions. Citrate and phosphate were chosen to investigate detailed inhibitory mechanisms on $H^+-ATPase$ since they showed different levels of inhibition. Inhibitory effect of citrate was observed at the concentrations above 3 mM. When 20 mM citrate was added, the ATPase activity was decreased by 50-60%. However, the inhibitory effect of citrate was decreased by increasing the concentration of$Mg^{2+}$ The citrate-induced inhibited activity was recovered by the addition of $Mg^{2+}$ Addition of 7 mM $Mg^{2+}$ completely removed the inhibitory effect of citrate and the activity recovered to the level of the control experiment. These results imply that citrate chelates $Mg^{2+}$ and thus inhibits $H^+-ATPase$. Meanwhile, the inhibitory effect of phosphate was observed at the concentration above 3 mM and the activity was decreased by 50% in the presence of 30 mM phosphate. Further addition of $Mg^{2+}$ showed no recovery on the activity. These results imply that the inhibitory effect of phosphate is not dependent upon the concentration of $Mg^{2+}$.
