Physiological Characteristics of Resistant Starch (HI-MAIZE DIET) Fortified with Other Dietary Fiber Components (식이섬유의 기능이 강화된 저항전분 (HI-MAIZE DIET)의 생리적 특성)

  • This study was performed to investigate the influences of resistant starch (HM: HI-MAIZE) and HM-D (HI-MAIZE DIET) fortified with D-factor (consisted of Psyliium husk, polydextrose and hydrocitric acid) on the glucose and bile acid absorption and production of short chain fatty acids (SCFA). HM-D absorbed more glucose and bile acid than did HM. The glucose transport of HM and HM-D against dialysis membrane showed 77% and 68% for 4h, respectively. After 24h, bile acid transport of HM and HM -D showed 65% and 62.3%, respectively. The HM and HM-D produced 217.8 mM and 264.0mM of SCFA, respectively. The production of butyric acid in HM-D (32.7mM) showed higher than that of HM (26.9mM). The addition of D-factor to HM increased the physiological function of dietary fiber through the glucose and bile acid absorption and production of SCFA.

Effects of Commercial Salts on the Growth of Kimchi-Related Microorganisms (시판소금이 김치발효 미생물의 생육에 미치는 영향)

  • This study was carried out to investigate the effects of various kinds of commercial salts including Chunil, Hanju, Guwoon and Bamboo salts on the growth of microorganisms involved in kimchi fermentation. Among various microorganisms related to the kimchi fermentation, the growth of Leuconostoc mesenteroides, Lactobacillus plantarum, Pichia membranaefaciens and E. coli was examined. Based on the conditions of kimchi fermentation, 3% and 5% concentration of each salt were studied. Also, the temperatures at 1$0^{\circ}C$, 18$^{\circ}C$ and 37$^{\circ}C$ of the cultural condition were examined. The growth of Leuconostoc mesenteroides was inhibited depending on the reduction of cultural temperature and increase of concentration of salts. There was no considerable difference on the growth of Leu. mesenteroides in the different the kind of salts. However, the growth of Lactobacillus plantarum was strongly inhibited by the 5% concentration of Bamboo salt during incubation at 18$^{\circ}C$. When Lactobacillus plantarum was cultured at 1$0^{\circ}C$, its growth was remarkably decreased regardless of the kind and concentration of salts. In the case of Pichia membranaefaciens, Bamboo salt strongly inhibited its growth at all cultural temperatures. Apparent inhibitory effect on the growth of E. coli was observed by the Bamboo salt treatment during the incubation at 18$^{\circ}C$. At the cultural temperature of 1$0^{\circ}C$, similar results obtained.

The Antioxidative Activities of Torreya nucifera Seed Extracts (비자(Torreya nucifera) 추출물의 생리활성)

  • This study was investigated to analyze the contents of flavonoid and polyphenol compounds, and inhibitory activities of tyrosinase and antioxidation to measure physiological effect of reflux water extraction (WE), reflux ethanol extraction (EE) and hot water extract under high pressure (HWE) of Torreya nucifera seed. HWE yields the highest contents of flavonoid compounds (176.34 mg/g) and polyphenol compounds (112.95 mg/g). The tyrosinase inhibitory rates were $5.62{\sim}28.71%$ at 2.0 mg/mL and HWE showed the highest inhibition rate. The nitrite scavenging abilities of all extracts were over 90% at pH 1.2 and 3.0 at the concentration of 2.0 mg/mL. The superoxide dismutase (SOD)-like activities of HWE was the highest value of 33.58%. The electron donating abilities (EDA) were $66.46{\sim}89.72%$ and HWE was the highest when the extracts were tested at 0.1 mg/mL. The EDA of all extracts were decreased with an increment of the extracts concentrations. The xanthine oxidase inhibitory rate of HWE was the highest value of 89.29% at the concentration of 2.0 mg/mL and the WE and HWE were over 75% rate of xanthine oxidase inhibition at 0.5 mg/mL.

Inhibitory Effect of $Zn^{+2}$ on Tolaasin-induced Hemolysis ($Zn^{+2}$에 의한 Tolaasin의 용혈활성 저해효과)

  • Tolaasin, a pore-forming toxin, is a 1,985 Da peptide produced by Pseudomonas tolaasii and causes a brown blotch disease on cultivated mushrooms. Tolaasin forms pores on the plasma membrane of various cells including fungi, bacteria, plant as well as erythrocytes, and destroys cell structure. $Zn^{+2}$ has been known to block the tolaasin activity by an unknown mechanism. Thus, we investigated the inhibitory effects of $Zn^{+2}$ on the tolaasin-induced hemolysis to understand the molecular mechanism of tolaasin-induced pore formation. $Zn^{+2}$ and $Cd^{+2}$ inhibited the tolaasin-induced hemolysis in a dose-dependent manner and their Ki values were 170 ${\mu}M$ and 20 mM, respectively. The effect of $Zn^{+2}$ was reversible since the subsequent addition of EDTA chelates $Zn^{+2}$ and removes the inhibitory effect of $Zn^{+2}$. When an osmotic protectant, PEG 2000, was added, the tolaasin-induced hemolysis was not observed. After the removal of osmotic protectant by centrifugation, resuspended erythrocytes with fresh medium were immediately hemolyzed, while the addition of $Zn^{+2}$ prevented from hemolysis, implying that tolaasin-induced pores on the membrane were already formed in the medium containing osmotic protectant. These results suggest that $Zn^{+2}$ inhibits the activity of tolaasin pores and it has minor effects on the membrane binding of tolaasin and the formation of pore.

Inhibitory activities on biological enzymes of extracts from Oplismenus undulatifolius (주름조개풀(Oplismenus undulatifolius) 추출물의 생리활성 효소 억제 효과)

  • The phenolic contents which were extracted with water and 70% ethanol from O. undulatifolius were 7.7, 10.1 mg/g, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 78, 82% at $50{\mu}g/mL$ phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical cation decolorization activity were 92, 76% at $100{\mu}g/mL$ phenolics. Antioxidant protection factor in water and ethanol extracts at $200{\mu}g/mL$ phenolics were 1.51 and 2.08 PF, respectively. Thiobarbituric acid reactive substance were 84% in water extracts and 99% in ethanol extracts at $50{\mu}g/mL$ phenolics, respectively. The inhibition activity on ${\alpha}-Glucosidase$ was 44% in ethanol extracts at $200{\mu}g/mL$ phenolics. The inhibition activity on ${\alpha}-amylase$ was 37-88% in water extracts at $50-200{\mu}g/mL$ phenolics. The tyrosinase inhibition activity as whitening effect were 82% in ethanol extracts. The elastase inhibition activity were 4, 61% in water and ethanol extracts, respectively. The collagenase inhibition activity of antiwrinkle effect showed an excellent wrinkle improvement effect as 39% in water extracts and 67% in ethanol extracts at $200{\mu}g/mL$ phenolics, respectively. The hyaluronidase inhibition activity as anti-inflammation effect of ethanol extracts was confirmed to 46% of inhibition at $200{\mu}g/mL$ phenolic. The astringent effect of water and ethanol extracts was confirmed to 13, 32% of effect at $200{\mu}g/mL$ phenolic, respectively.

The chemical structure of polyphenols isolated from cacao bean and their inhibitory effect on ACE (Cacao bean으로부터 분리된 polyphenol 성분의 화학구조분석과 ACE 저해효과)

  • Seven kinds of polyphenol compounds having ACE activities were isolated and purified by Sephadex LH-20, MCI-gel CHP-20, ${\um}-Bondapak\;C_{18}$ and Fuji-gel ODS $G_3$ sucessively from cacao bean(Ghana). The chemical structures of each compound were determined and identified using analyzers such as $^1H-NMR$, $^{13}C-NMR$, IR, MS, polarimeter and Elemental Analysis. Inhibition effects of isolated polyphenols on angiotensin converting enzyme (concerned with hypertension) were also observed. The results obtained were as follows,; The compounds isolated and identified were confirmed and determined as compound 1 [(+)-catechin], compound 2 [(-)-epicatechin], compound 3 [procyanidin B-1 : (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(+)catechin], compound 4 [procyanidin B-2 : (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin], compound 5 [procyanidin B-7 : (-)-epicatechin-$(4{\beta}{\rightarrow}6)$-(+)-catechin], campound 6 (procyanidin B-2,3,3'-O -digallate), compound 7 [cinnamtannin A-2 : (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin]. In the inhibition effect on ACE, procyanidin B-2,3,3'-O-digallate (compound 6) showed a higher value of 94.6% for ACE in $100\;{\um}M$ than other compounds such as (+)-catechin (compound 1), (-)-epicatechin (compound 2), procyanidin B-1 (compound 3), procyanidin B-2 (compound 4), procyanidin B-7 (compound 5) and cinnamtannin A-2 (compound 7) showing 67.9%, 61.9%, 88.6%, 82.5%, 72.2% and 82.3% for ACE, respectively. Inhibition of $4{\beta}{\rightarrow}8$ in coupling bond on the ACE enzyme was more effective than that of $4{\beta}{\rightarrow}6$. Procyanidin containing gallate inhibited more effectively than those containing not any. It was also observed that a lot of hydroxy group in the compounds increased the inhibitory effect.

Inhibitory activity against biological activities and antimicrobial activity against pathogenic bacteria of extracts from Hericium erinaceus (노루궁뎅이버섯 추출물의 생리활성 및 부패세균에 대한 항균효과)

  • The aim of this study is to investigate the biological activities of Hericium erinaceus. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of H. erinaceus extract was higher than positive control. The inhibitory activities of xanthin oxidase, ${\alpha}$-glucosidase, and hyaluronidase was measured as functional food activity, and inhibitory activities on collagenase, tyrosinase, and astringent effect as beauty food activity in water and ethanol extracts from H. erinaceus. In functional food activity, xanthin oxidase inhibitory activities at $50-200{\mu}g/mL$ phenolic concentration in ethanol extracts from H. erinaceus showed inhibitory activity in dose dependent manner. ${\alpha}$-Glucosidase inhibitory activities at $50{\mu}g/mL$ phenolic concentration showed high activity of higher than 80%. Inhibitory activities on hyaluronidase as anti-inflammation factor showed inhibition effect in dose dependent manner both in water and ethanol extracts. In beauty food activity, Inhibitory activities on collagenase at $200{\mu}g/mL$ phenolic concentration in water and ethanol extracts showed high activity to 65.09 and 58.38% dose dependently. Tyrosinase inhibitory activity in water extract showed 9.4-58.24%. Astringent activity as pore shrink effect in ethanol extracts also showed a very high activity of 18.94-100%. Antimicrobial activity on pathogenic bacteria was highly effective on Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus and Escherichia coli at 2.5 mg/mL or above. Therefore, the extracts from H. erinaceus can be used as a functional food and beauty food resources and natural antimicrobial agent on pathogenic bacteria in food.

  • 최근 새로운 천연가스 수송/저장 방법으로 가스 하이드레이트 형성법이 주목받고 있다. 본 연구에서는 천연가스의 저장 매체로 다공성 실리카 젤을 사용하였다. 다공성 매질을 사용할 경우 물이 기체와 접촉하는 면적이 극대화되어 하이드레이트로의 전환율이 높아진다. 기공의 직경이 각각 6.0, 15.0, 30.0 nm의 실리카 젤을 사용하여 270 - 285 K의 온도 범위와 0.5 - 3.0 MPa의 압력 범위에서 기공 크기의 분포를 고려하여 에탄 하이드레이트의 3상 평형점 (하이드레이트 (H) - 물 (LW) - 기상 (V))을 측정하였다. 기공의 크기가 작아질수록 벌크 상태의 에탄 하이드레이트에 비해 하이드레이트 평형 조건이 온도는 낮아지고 압력이 높아지는 저해효과가 커짐을 알 수 있었다. 천연가스 수송/저장으로의 응용을 고려할 경우 저해효과가 적은 30.0 nm 이상의 실리카 젤을 사용하는 것이 유용할 것으로 사료된다. 본 연구의 결과는 심해저 천연가스 개발, 이산화탄소 심해저장 등의 가스 하이드레이트 응용 연구에도 유용한 기초자료가 될 것이다.

Inhibitory Effects of Sasa borealis Leaves Extracts on Carbohydrate Digestive Enzymes and Postprandial Hyperglycemia (조릿대잎 추출문의 탄수화물 소화효소활성 저해 및 식후혈당강하효과)

  • This study was designed to investigate whether Sasa borealis leaves extracts (SLE) may inhibit yeast ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities and postprandial hyperglycemia in STZ-induced diabetic mice. Freeze-dried SLE was extracted with 70% methanol and followed by a sequential fractionation with dicholoromethan, ethylacetate, butanol, and water. Both ethylacetate and butanol fractions showed high inhibitory activities against the ${\alpha}-glucosidase$ and ${\alpha}-amylase$ enzymes. The $IC_{50}$ of ethylacetate and butanol fractions against ${\alpha}-glucosidase$ were 0.54 and 0.63 mg/mL, respectively, indicating a greater inhibition effect than acarbose (0.68 mg/mL) (p<0.05). Likewise, the two fractions exhibited a smaller $IC_{50}$ against ${\alpha}-amylase$, compared with acarbose (p<0.05). However, the yield of ethylacetate fraction of SLE was relatively small. Postprandial blood glucose testing of normal mice and STZ-induced diabetic mice by starch soln. loading (2 g/kg B.W.) showed that postprandial blood glucose level at 30, 60, and 120 min were markedly decreased by single oral administration of SLE butanol fraction (200 mg/kg B.W.) in both normal (p<0.0l) and diabetic mice (p<0.0l). Furthermore, the incremental area under the curve (AUC) was significantly lowered via SLE administration (5,745 versus 12,435 $mg{\cdot}mim/dL$) in the diabetic mice (p<0.0l). The incremental AUC in normal mice corroborated the hypoglycemic effect of SLE (p<0.0l) found in the diabetic mice. These results suggest that SLE may delay carbohydrate digestion and thus glucose absorption. In addition, SLE may have the potential to prevent and treat diabetes via its ability on lowering postprandial hyperglycemia.

Studies on the Comparative Analysis of Immunofunction of Agaricus blazei Murill Cultivated with Fermented Media Containing Pueraria thunbergiana (칡혼합 발효배지로 생산된 신령버섯의 면역기능성 비교 분석에 관한 연구)

  • This research was performed to investigate the immnomodulative effects of ploysaccharides extracted from the fruiting body of Agarcus blazei cultivated with the media which are fermented with sugar cane bagasse containing Pueraria thunbergiana in open-air storage. In MTT test, methanol extracts from the fruiting body of A. blazei cultivated with P. thunbergiana media showed in colon carcinoma line(HT29) by 1.5∼3.5 fold and human heptoma cell line (HepG2) by 1.3 ∼2.4 fold antitumor activites compared to two types media (rice straw plus sugar cane bagasse, rice straw only) often used in the fauns. To clarify the antimutagenic principles, three extracts, Ab-l, Ab-2 and Ab-3, were separated by the solvent fractionations such as hot water, cold & hot sodium hydroxide respectively, and their antimutagenic effects was determined against N-methyl-N'-nitro-N-cnitrso-guanidine(MNNG) using Salmonella typhymurium. There was no significant differencies of inhibition levels among the used media, but Ab-3 tractions still showed a high antimutagenicity in the Ames test regardless of cultivating areas or media. To prove the cell immunofunction, nitric oxide (NO) produced from Raw 264.7 matrophage cultured with three fractions (Ab-l, Ab-2, Ab-3) was measured, and showed generally increase about 45 ∼58 percent compared to another two media (rice straw plus sugar cane bagasse, rice straw only), in the fraction of hot alklai extracts of the fruiting body cultivated with P. thunbergiana, which means that the media selection could be very important factors for improving medicinal effects in agaricus blazei fruiting body.
