Effect of Cold Acclimation and Deacclimation on the Freezing Tolerance, Total RNA, Soluble Protein and Soluble Sugar in Chinese Cabbage (저온순화 및 탈순화가 배추의 내동성 및 total RNA, soluble protein, soluble sugar 함량에 미치는 영향)

  • This study was conducted to determine the change in freezing tolerance of Chinese cabbage (Brassica campestris L. ssp. penkinensis). Four-week old seedlings grown in a greenhouse at $25^{\circ}C$$\pm$0.5 were transferred to 5, 10 or 15$^{\circ}C$ condition for acclimation for one day, and then transferred to a lethal temperature of -3$^{\circ}C$ condition. Optimal acclimation temperatures were 5$^{\circ}C$ and 1$0^{\circ}C$. Freezing tolerance of leaf tissues was investigated during a week-long cold stress. Development of freezing tolerance was shown by survival ratio, and amounts of total RNA, soluble protein and soluble sugar. LT$_{50}$ value increased from -3 to -4$^{\circ}C$ after 5 days acclimation at 5 or 1$0^{\circ}C$, and this was considered to be the first indication of enhanced freezing tolerance. Plants, cold acclimated for five days, reached to a survival ratio of 60%. This increase in freezing tolerance was found to be associated with the increased levels of total RNA, soluble sugar and soluble protein. These metabolic changes imply the association of adjustment of growth and cell metabolism with low temperatures at the beginning of cold acclimation in chinese cabbage.e.

  • This study was aimed for determining the biochemical mechanism of cold tolerance in crops and for searching the biochemical genetic marker related with cold tolerance by the analysis of isozyme pattern. We investigated various biochemical changes induced by the long-term cold acclimation in cold sensitive rape (B. napus) and in cold tolerant 'Sandongchae'(B. campestris) seedlings. The cold shock after long-term cold acclimation to B. napus and B. campestris greatly increased the activities of peroxidase 157% and 50% in root fraction and, 201% and 205% in hypocotyl, respectively. Simultaneously, the activity of superoxide dismutase was largely increased in hypocotyl fraction, too. Protein contents of hypocotyl fractions in B. napus and B. campestris were also increased by 11.4% and 57.8%, respectively. The band of pl 6.4 among peroxidase isozymes newly biosynthesized during long-term cold acclimation was emerged in the hypocotyl fraction of cold tolerant B. campestris as well as in the root of both species. From above and previous results, we presented a model of interconversions of molecular oxygen species due to the cold injury and biochemically inferred the mechanism of cold tolerance in crops.

Studies on the Cold Attenuation and Protective Effects of a Thermostable Newscastle Disease Virus Isolated from Korean Pheasants (한국산 꿩으로부터 분리한 열안정성 뉴캣슬병 바이러스의 저온순화와 방어효과)

  • Newcastle disease virus, CBP-1 strain isolated from Korean pheasants was passaged for 173 times by 9-day-old specific pathogenic free (SPF) embryonated eggs at $37^{\circ}C$ (parent strain) and subsequently passaged for 15 (cold attenuation (CA) -15) and 30(cold attenuation (CA) -30) times by 10-day-old of commercial broiler chicks embryonated eggs at $29^{\circ}C$, respectively, The Physical and chemical properties (sensitivity to lipid solvents, low pH and thermostability), pathogenicity (mean death time, intracerebral pathogenic index and intravenous patho-genic index), safety, booster or protective effect and characterization of temperature sensitivity were measured in cold attenuated CA-15 or 30 strain and compared to those of parent CBP-1 strain. NDV, CBP-1 CA-30 strain acquired cold attenuation and decreased infectivity at $41^{\circ}C$ compared to those of parent strain grown at $37^{\circ}C$. It lost hemagglutination activity (HA) and cell infectivity at $56^{\circ}C$ for 30, 60, and 120 Min. CA-30 strain treated with ethyl ether also lost its HA and cell infectivity. Both CA-30 and parent strains exhibited a little resistant to HA at pH 3.0 glycine HCI buffer. Intracerebral pathogenic index (ICPI) and intravenous pathogenic index (IVPI) of parent strain were 1.12 and 1.45, but decreased to 0.75 and 0.00 in CA-30 strain, respectively. The safety was evaluated by mortality in chicks inoculated with 10$^{4.0}$ EID$_{50}$ /0.1 ml. The mortalities of parent, CA-30 and commercial Bl strains were 17.5, 12.0 and 0.0%, respectively. The safety of CA-30 strain was higher than that of parent strain. The booster effects of CA-30 strain and parent strain performed in 4-week-old chicks after being vaccinated with primary commercial Bl strain.

Growth and Green Pod Yield by Sowing and Acclimation Dates in Autumn Green Pea (추파 풋완두 파종과 순화시기에 따른 생육 및 수량)

  • When sowing green peas in the autumn, proper seedling stands and growth quantity should be secured before winter begins. Also, for proper acclimatization, injuries caused by low temperatures, frost or high temperatures in the P.E. film during mulching, should be avoided during the regeneration period; that being early spring. The days required for growth in each stage in Yeosu are shorter than those in Naju because Yeosu has high temperatures during the growth period. Furthermore, in Yoesu, it was observed that there were more effective branches as well as effective and attached node positions on the branches. The first pods on the main stems and effective branches were observed to be higher than those in Naju. The number of pods per plant and the number of seeds per pod in Yoesu was greater than for those in Naju and the pod length was longer as well. Considering the missing plant rate, growth, and green pod yield, the optimum sowing date for the green pea was mid-November in both location. The stable acclimatizing date for the green pea was early March when the highest yield can be acquired due to a lot of effective branches and pods per plant and with the lowest missing plant rate and rate of injury in acclimatization.

Condition of Storage and Growth Characteristics on Bulb Propagation of Nerine by Tissue Culture (네리네 대량증식을 위한 자구 저장 및 순화재배)

  • Nerine was originated from South Africa and around 30 kinds of species are distributed in worldwide. In present study we have characterized the condition of storage and growing conditions on the bulb propagation of Nerine by the tissue culture. Result indicated that the percentage of decrement, decay and sprouting of bulblets in the tissue culture was high at the higher temperature compare to lower temperature. The optimum conditions for storage and time were 12 weeks at $15^{\circ}C$ which were having 86.4% sprouting rate. The sprouting rate at different temperature and acclimation time, using bulblets in the tissue culture, were optimum at early June when temperature is relatively higher($22^{\circ}C$) that facilitate to breaking the dormancy. Growth characteristics and capacity of photosynthesis of Nerine were better at red and blue mixed lightness.

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks. Comparative Transcriptome Analysis of the Response of Two Lines of Rapeseed (Brassica napus L.) to Cold Stress (유채 두 계통에서 저온 스트레스에 반응하는 전사체 발현 비교 분석)

  • Rapeseed is a typical winter crop, and its freezing stress tolerance is a major feature for winter survival. Therefore, it is important to comprehend clearly the physical and molecular mechanisms of rapeseed under freezing stress conditions. This study investigates the physical and transcriptome changes of two rapeseed lines, 'J8634-B-30' and 'EMS26', under cold acclimation and freezing temperature treatments. The proline content of 'J8634-B-30' at 5 ℃ increased 8.7-fold compared to that before treatment, and there was no significant change in that of 'EMS26' RNA-sequencing analysis revealed 5,083 differentially expressed genes (DEGs) of 'J8634-B-30' under cold acclimation condition. Among the genes, 2,784 (54.8%) were up-regulated and 2,299 (45.2%) were down-regulated. The DEGs of 'EMS26' under cold acclimation condition were 5,831 genes, and contained 2,199 up-regulated genes (37.7%) and 3,632 down-regulated genes (62.3%). Among them, only DEGs annotated in the cold response-related signaling pathways were selected, and their expression in the two rapeseed lines was compared. Comparative DEGs analysis indicated that cold response related signaling pathways are proline metabolism and ABA (Abscisic acid) signaling. And ICE (Inducer of CBF expression) - CBF (C-repeat-binding factor) - COR (Cold-regulated) signaling were the significantly differentially expressed transcripts in the two rapeseed lines. The major induced transcripts of 'J8634-B-30' induced P5CS (Δ'-pyrroline-5-carboxylate synthetase), which is related to proline biosynthesis, PYL (pyrabactin resistance-like protein, ABA receptor) and COR413 (cold-regulated 413 plasma membrane 1). In conclusion, these result provide a foundation for understanding the mechanisms of freezing stress tolerance in rapeseeds. Further functional studies should be performed on the freezing stress-related genes identified in this study, which can contribute to the transgenic and molecular breeding for freezing stress tolerance in rapeseed.

Analysis of the Proteins Accumulated During Cold Treatment in Intermolecular Space of Barley (저온에서 세포밖 공간에 축적되는 보리 단백질)

  • In order to identify an antifreeze proteins responsible for freeze-tolerance in barley the proteins accumulated in extracellular space during cold acclimination were extracted and analyzed. After 42 days cold treatment there were several proteins sized of 70, 21, 16, 14 KDa increased in their amount accumulated in extracellular space. In addition, continuously sized polypeptides smaller than 10 KDa were found to be increased in their amount as cold treatment prolongs. Since these proteins were not detectable in total leaf protein extract it appears that the procedure used to isolate extracellular extract was valid. A similarity in profile of the extracellular proteins isolated from barley and rye may indicate a possibility for these proteins to be an antifreeze proteins since the same extract from rye was reported to show an antifreezing activity.

Growth and Flowering of Campanula Species as Affected by Duration, Temperature, and Light Condition during Chilling Treatment (저온처리 기간, 온도 및 광 조건이 자생초롱꽃의 생육과 개화에 미치는 영향)

  • The experiment investigated effect of duration, temperature, and light condition during chilling treatment on growth and flowering of four Campanula species in a factorial experiment. Two parent species, Campanula punctata Lam. var. rubriflora Mak. and C. Punctata Lam., and their two $F_1$ hybrids, C. punctata Lam. ${\times}$ C. punctata Lam. var. rubriflora Mak. ('Jiknyeo') and C. punctata Lam. var. rubriflora Mak. ${\times}$ C. punctata Lam. ('Gyeonu'), were used. Plants were cultured in vitro for five weeks at $25^{\circ}C$ under about $75{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD before being chilled at 4 or $25^{\circ}C$ for 3, 6, or 9 weeks under a darkened or lighted (about $10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD) condition. After chilling treatment, plants were transplanted to 10 cm pots filled with a commercial growing medium and were transferred to environment-controlled growth chambers and subsequently to a greenhouse to observe their reproductive growth. Growth of all species and flowering of a $F_1$ hybrid 'Jiknyeo' were affected by duration, temperature, and light condition during chilling treatment. The greatest growth and survival percentage were observed in C. punctata Lam. var. rubriflora Mak. The survival percentage was greater when plants were chilled in a lighted than darkened condition, whereas it decreased when plants were chilled more than six weeks in vitro. Among the four species tested, flowering was observed only in a $F_1$ hybrid 'Jiknyeo' with 62.5% flowering plants when it was chilled at $25^{\circ}C$ for three weeks under a lighted condition. Percent flowering plant was affected by duration, temperature, and light condition during chilling treatment. Three-week chilling at $4^{\circ}C$ under a darkened condition significantly reduced days to flowering. These results suggest that the low temperature requirement for flowering is not qualitative but quantitative in Campanula species. Further experiment with more number of plants is necessary to ascertain this conclusion.

Sunlight Causes Skin Blackening in 'Fuyu' Persimmon Fruits during Storage (일광에 의해 야기되는 '부유' 단감의 저장 중 과피흑변)

  • Skin blackening is a serious postharvest disorder occurring in 'Fuyu' persimmon fruits (Diospyros kaki, 'Fuyu'). Phenolic content, polyphenol oxidase (PPO) and peroxidase (POD) activity, and lipid saturation were investigated in skin tissue of 'Fuyu' fruits. The phenolic compounds were accumulated unevenly on fruit skin depending on sunlight exposure, i.e., the most accumulation on sunny side and the least on the opposite, shaded side. The fruits harvested from shaded branches inside the canopy showed relatively even distribution of phenolic compounds in the skin. The activity distribution of PPO and POD were also uneven but only in fruits suffering f rom skin b lackening disorder, w ith the h ighest and the lowest a ctivity of PPO and P OD, respectively, in the blackened area of the fruit. When the sunny, southern sides of fruits were marked at harvesting time and fruits were stored at low temperature, a round black spot, as a symptom of skin blackening disorder, developed during storage exactly on the marked region. In addition, the sunny side of fruit showed higher lipid saturation, thus indicating adaptation to high temperature caused by the sunlight. These results suggest that the skin blackening disorder of 'Fuyu' fruits can be considered as a kind of chilling injury symptom that develops on the high-temperature-acclimated skin tissue of sunlight-exposed fruits when they are later exposed to low temperature.
