In an outbreak of acute porcine diarrhea in newborn piglets, an etiological study was carried out using piglets submitted in Cheju Province Institute for Livestock Promotion(Cheju Veterinary Service for the disease diagnosis). Sixteen piglets(2-7 days old) were collected from 4 farms during outbreaks of diarrhea disease(from January to April 2000). Specimens were taken after necropsy and examined by immunohistochemistry using of monoclonal antibodies for porcine epidemic diarrhea(PED) virus, transmissible gastroenteritis(TGE) virus, and porcine rotavirus. Immunohistochemistry showed that PED virus antigens, but both TGE virus and rota virus antigens not, were localized in the some epithelial cells of the intestines of 14 animals among 16 piglets examined. PEB virus antigens were mainly detected in the cytoplasm of enterocytes. Infected cells, which were most abundant in the villous epithelial cells of the jejunum and ileum, were uncommon in the crypt, epithelial cells, the lamina propria and Peyer's patches of piglets examined. The results suggest that PED virus is one of the most prevailing agents in an outbreak of fatal diarrhea in newborn piglets on Cheju island and PED virus was need to further study to prevent this disease.

The innate immune response is fundamental defense response of vertebrates and invertebrates. Especially, the innate immune response important for larvae that lack of resistance to infectious diseases in the early stages. Galectin is one of the kinds of lectin and presents in the fish mucous that involves innate immune response. Galectin have been studied from various fishing species, but expression analysis of galectin is still unclear during early developmental stage in olive flounder. In this study, we investigated gene expression of galectin-1 from various developmental stage and tissues. We excised several tissues including the muscle, fin, eye, gill, brain, stomach, intestine, kidney, spleen and liver from adult olive flounder and confirmed gene expression of galectin-1 using RT-PCR and quantitative real-time PCR. Expression of galectin-1 was significantly higher in muscle, stomach and intestinal tissue than other tissue in adult fish (5 and 29 months). Also, galectin-1 gene was detected from 0 DAH and gradually increased to 35 DAH and since then decreased after stomach development period. Induction of galectin-1 during the early developmental stage suggest that muscle, fin and eye tissue is formed and begins the secretion of galectin this period. In addition, increased expression levels at 35 DAH suggest that due to complete formation of stomach and intestine, increase of secretion and activation of enzyme. This study shows that expression of galectin-1 during early developmental stages and adult period in olive flounder and can be expect that galectin-1 play essental role in the innate immune system throughout the whole life time. Galectin-1 is primary barrier such as skin and digestive tissue against pathogen infection, also digestive tract developmental period is important for pathogen invasion can be expected that it will serve. Mass mortality due to the disease in seed production is continuing damage, therefore these result will be meaningful about infectious disease during early developmental stages as a basic data for the study.
In the study of investigate the expression and effects of adrenomedullin (AM) in the tissue injured by ethanol in our laboratory, there were lymphosarcomas involving mesenteric lymph node and intestine discovered in experimental mice. Lymphosarcoma is probably the second most common spontaneous neoplastic disease, the incidence of which is dependent on the strain of mouse. The diagnosis of lymphosarcoma was based on gross and light microscopic studies. Crossly, the excised intestine and mesentery showed soft-to-firm, cream-colored, displayed protuberance into the lumen. The neoplastic cells had mainly infiltrated into the mucosal and mesenteric lymph node, and have round hyperchromatic nuclei and a small amount of cytoplasm. The origin of these tumor cells was determined by immunohistochemistry positive of CD20, CD3 and Vimentin.
This study was conducted to determine the effects of probiotics supplementation on growth performance, nutrient digestibility, occurrence of diarrhea and immune response in weaning pigs. Treatments were 1) NC (basal diet), 2) PC (basal diet + 0.12% avilamycin) and 3) A (basal diet + 0.2% Aspergillus oryzae), 4) B (basal diet + 0.2% Lactobacillus casei), 5) C (basal diet + 0.2% Bacillus subtilis), 6) D (basal diet + 0.2% Lactobacillus crispatus). A total of 120 pigs ($7.16\pm0.01$ kg average body weight, weaned at $23\pm3$days of age) were assigned to 6 treatments, 5 replicates and 4 pigs per pen in a randomized complete block (RCB) design. During the whole experimental period, body weight (P<0.01), average daily gain (ADG; P<0.01), and average daily feed intake (ADFI; P<0.05) of treatment PC were higher than other treatments. However, the probiotics treatments tended to increase ADG and G:F ratio compared to treatment NC. The G:F ratio in treatment A (Aspergillus oryzae) was similar to treatment PC during the whole experimental period (P<0.05). The supplementation of probiotics in the diet of weaning pig did not change nutrient digestibility (dry matter, crude protein, crude fat and ash) and nitrogen retention of weaning pigs. In blood urea nitrogen (BUN) concentration, treatment B had lower value than other treatments at 2 and 4 weeks (P<0.05). Treatments PC and C tended to decrease diarrhea score than other treatments (P=0.18). At 3h after lipopolysaccharide (LPS) injection, treatments NC and PC had higher count of $CD^{4+}$ T-cells than probiotics treatments, and treatment C showed the lowest value (P<0.01). There were no differences on count of $CD^{8+}$ T-cells and $CD^{4+}:CD^{8+}$ ratio among all treatments (P>0.10). These results suggest that the dietary probiotics are likely able to improve the growth performance, occurrence of diarrhea and immune response although they do not have similar effects like antibiotics in weaning pigs.
Background: In this study, we investigated the early time course of expression of the major histocompatibility(MHC) antigens, intercellular adhesion molecule-1(ICAM-1), vascular cell adhesion molecule-1(VCAM-1), interleukin-6 and the histopathological changes in the coronary arteries of cardiac allografts exchanged between inbred mice strains that differ in one loci of class I major histocompatibility antigen (B10.BR to B10.A). Material and Method: No immunosuppressive therapy was used. Both allografts and the hearts of the recipients were harvested at 7(group 1, n=6), 15(group 2, n=6), 21(group 3, n=6), and 30(group 4, n=6) days after transplantation. They were examined by immunohistochemistry, microscopy and morphometry. All allografts had contractions at the time of harvest. Result: A strong MHC class I antigen expression was present on the endothelial and medial cells of the coronary arteries in group 1 and remained unchanged in the rest of the groups. However, MHC class II reactivity was none or very little at any time. Mild to moderate ICAM-1 expression was observed on the endothelial cells, but not on the medial cells at any time by 30 days. VCAM-1 expression was strong both on the endothelial and medial cells at any time. Moderate degree expression of interleukin-6 was observed from 7 to 30 day specimens. Histopathologically, percentage of affected vessels(vessels with intimal thickening) was less than 10 % in 7 day group and increased up to 50 % at 30 days. Mean percent narrowing of the lumen of the affected vessels revealed less than 20 % at 7 days and 40 % at 30 days. The area occupied by tropomyosin positive cells in the intimal lesion, graded from 0 to 3, showed gradual increase but remained between grade 0 to 1 by 30 days. Medial integrity was also well preserved at any time. Moderate perivascular mononuclear cell infiltration was observed at 7 days and it was progressively increased upto 30 days. Recipients' heart revealed no positive immunopathologic findings. Conclusion: In this study, the early time course of progression of the transplantation vasculopathy was demonstrated in the murine heterotopic heart transplant model.
In order to determine the antigenic localization in the tissues of the adult Metagonimus yokegawai, immunogoldlabeling method was applied using serum immunoglobulins (IgG) of cats which were infected with isolated metacercariae from Plecoglossus altivelis. The sectioned worm tissue was embedded in Lowicryl HM 20 medium and stained with infected serum IgG and protein A gold complex (particle size: 12 nm) , It was observed by electron microscopy at each tissue of the worm. The gold particles were observed on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum. The gold particles were not observed on the basal lamina of the tegument, interstitial matrix of the parenchyma, the muscle tissue and mitochondria of the tegument. The gold particles were specifically labeled in the secretory granules in the vitelline cells. They were also labeled on the lumen of bladder and egg shell. The above findings showed that antigenic materials in the tissue of adult worms were specifically concentrated on the tegumental syncytium as well as cytoplasm of tegumental cells and epithelial lamella of the caecum.
The aim of this study was to develop an activator, 3-10 kDa fraction from radish water extracts, that will improve the intestinal function and bowel movement in the colons. Radish water extracts were investigated for their intestinal function effects according to the charcoal meal transit method, employing Balb/c mice: also, their anti-constipation activities were compared utilizing the loperamide-induced constipation method, employing SD rats. The result suggested that the effects of the charcoal meal transit increased remarkably in radish water extract administrated rats in comparison to loperamide administrated rats. Futhermore, the effects of various solvent extracts of radish on charcoal meal transit in Balb/c mice increased remarkably in radish water fraction administrated rats than in different solvent fraction administrated rats. Radish extraction was tested and isolated into 4 groups: below 3 kDa, 3-10 kDa, 10-300 kDa, and over 300 kDa. 3-10 kDa was the most effective on the intestinal function and bowel movement in the colons; also, 3-10 kDa fraction of radish water extraction was found to be the most effective charcoal meal transit. The dry weight and moisture content of feces remarkedly increased in the 3-10 kDa administrated rats group than in the loperamide only group. Experimental results revealed that 3-10 kDa fraction of radish water extract was the most effective on the intestinal function and bowel movement was the crypt epithelial cells that contained more MUC2 in the 3-10 kDa administrated group than the loperamide only group: in addition, the thickness of mucus layer stained with alcian blue was significantly thicker in 3-10 kDa administrated rats than in loperamide administrated rats. Crypt epithelial cells secreted more MUC2 in the 3-10 kDa administrated group than the loperamide only group and the stained cells clearly showed the MUC2 with antibody Biogenex AM358.
Suppression subtractive hybridization (SSH) was performed to construct an cDNA library of olive flounder Paralichthys olivaceus peripheral blood leukocytes (PBL) stimulated with lipopolysaccharide (LPS). Total 470 clones were randomly selected and sequenced, 214 out of 470 sequences showed identities with known genes and 252 sequences were unknown genes. Among the 218 known genes, 34 sequences were found to be homologous to IL-1RII gene, and 14 sequences were identified as IL-$1{\beta}$. RT-PCR analysis showed that IL-$1{\beta}$ mRNA was induced 1h after LPS-stimulation, and IL-1RII was increased from 3h after stimulation, indicating that most of SSH clones are associated with inflammatory responses in fish, and this SSH cDNA library would be useful to identify bio-defense and immuno-related genes in fish.

A study investigated the effects of a mixture of Lactobacillus plantarum (L. plantarum) and Bacillus subtilis (B. subtilis) on diarrhea incidence, immune response, and fecal microflora counts in post-weaning piglets. One hundred 28-day-old piglets were randomly assigned to five treatment groups: negative control (NC), free of antibiotics; positive control (PC), 0.03% chlortetracycline; LB 1, a mixture of L. plantarum and B. subtilis (LB) 0.5 kg/ton feed; LB 2, LB 1.0 kg/ton feed; and LB 3, LB 2.0 kg/ton feed. Diarrhea scores for LB 2 and LB 3 from the 2nd week were significantly reduced compared to NC (P<0.05). In terms of the level of IgG and IFN-${\gamma}$, all treatment groups were significantly higher than NC (P<0.05), and the IgG level of LB 3 was significantly higher than that of PC (P<0.05). Furthermore, fecal lactic acid bacteria (LAB) counts for LB 2 and LB 3 were significantly higher than those of NC and PC (P<0.05). In addition, fecal Enterobacteriaceae (ENT) counts for PC, LB 2 and LB 3 were significantly lower than those of NC (P<0.05). Based on the results from this study, it was concluded that a combination of L. plantarum and B. subtilis strains could be used as potential alternatives to antibiotics to prevent diarrhea incidence in piglets.
'Tight junctions (TJ)' have recently been identified in the granular cell layer of the human epidermis, where they contribute to the normal adhesion between keratinocytes and to the physiologic barrier function of the epidermis. Among the TJ proteins in the epidermis, occludin is an important transmembrane protein, which is considered as a major component. The purpose of this study is to investigate whether regional variation exists in the expression of the tight junction protein occludin in normal human epidermis. Indirect immunofluorescence staining for occludin was performed with specimens taken from different areas of normal skin (4 from each of 7 different anatomical sites, including the scalp, face, posterior neck, upper arm, abdomen, lower back, and inner thigh). The degrees of the expression-intensity in each specimen were estimated with the reciprocals of positive end-point titer of occludin in an indirect immunofluorescence study. The highest degree expression-intensity of the TJ protein occludin among the different areas of normal epidermis was observed on the face and abdomen with a titer of 600 (p=0.001). The lowest intensity of expression of occludin was seen in the epidermis from the upper arm. Skin specimens from the scalp, neck, back, and leg demonstrated intermediate degrees of the expression in intensity. The expression of occludin in the skin samples obtained from different locations of the body showed a statistically significant variation. This suggests that there is a certain degree of regional variation in the expression-intensity of TJ protein 'occludin' in the human epidermis.
