Effect of Water Soluble Extract of Lichens on Weights of Various Organs, Blood Components and Activities of Transaminases and 5-Nucleotidase in Rat (지의류(地衣類)의 수용성 추출물이 흰쥐의 각 장기무게, 혈액성분 및 Transaminases와 5-Nucleotidase 활성에 미치는 영향(I))

  • In order to evaluate the effect of water soluble extract of lichens (Physcia, Parmelia and Clandonia species) on liver damage, activities transaminase(GPT) and 5'-nucleotidase in serum and liver were measured in rats fed lichens extract. DNA and RNA were measured in liver and spleen, as well as various organ weights and blood components. Control group was fed water to compare with the lichen group. Three sets of experiments were conducted: the first set was done with normal rats, the second one with rats with liver damage induced by $CCl_4$ injection was divided into three subgroups. i.e. no treatment group, pre-treatment group and post-treatment group, and the third one was with rats with acute and chronic liver damage. In normal rats, lichens extract feeding reduced serum GOT and GPT activities. In liver damaged rats, both pre-and post-treatment had suppressing effect against increase of serum enzymes. In rats with acute and chronic liver damage, lichens fed group had lower activities of serum GOT, GPT and 5'-nucleotidase but higher activities of liver enzymes than control group. This effect was more pronounced in rats with acute liver damage. Liver weight increased considerably with lichens intake. Hemoglobin and hematocrit levels were also higher in lichens fed group. Nucleic acid contents in spleen but not in liver were increased in lichens fed group. The latter increase was more significant with chronic liver damage. It is suggested from the present study that water soluble lichens extract play protective and therapeutic roles in organs against infection and atrophic disease.

Anti-Obesity Effect of Enterococcus faecalis MD366 isolated from Raw Milk in Diet-Induced Obese Mice (원유에서 분리한 Enterococcus faecalis MD366가 고지방식이를 급여한 마우스의 비만 억제에 미치는 영향)

  • This study aimed to investigate the anti-obesity effects of Enterococcus faecalis MD366, isolated from raw milk, in diet-induced obese mice. To examine the effect, male C57BL/6J mice were fed for six weeks on three different diets, i.e., a normal diet and orally administrated saline solution (ND), a high-fat diet and orally administrated saline solution (HFD), and a high-fat diet and orally administrated E. faecalis MD366 ($10^9CFU/day$) in saline solution (HFD+MD366). After six weeks, the rate of increase in body weight was 18.1% lower in the HFD+MD366 group compared to that in the HFD group. In addition, the weight of epididymal fat pad in the HFD+MD366 group was lower than that in the HFD group. The average levels of triglycerides, total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) were slightly reduced in the HFD+MD366 group compared to those in the HFD group. However, there were no significant differences between the groups. Measuring the adipocytes revealed that the percentage of adipocytes with a size of $2,000{\mu}m^2$ was higher than the percentages of other size classes in ND and HFD+MD366 groups, while the percentage of adipocytes larger than $5,000{\mu}m^2$ was highest in the HFD group. The mean adipocyte size in the HFD+MD366 group was smaller than that in the HFD group.

전력구 내 지중선을 이용한 2W급 상용주파수 무선전력 수신장치 개발

  • 본 논문은 전력구 내 지중선에서 발생하는 상용주파수(60 Hz) 자기장을 이용한 2 W급 무선전력 수신장치를 제안한다. 제안하는 수신장치는 자성체와 구리도선을 포함하는 수신 코일, 장치의 개폐를 위한 손잡이, 직렬공진회로 및 정류기를 포함하는 부가회로로 구성된다. 수신장치는 3상 지중선 중 하나에 설치되어 전력구 내 조명 및 온습도 센서를 구동하는데 사용 가능하다. 수신장치의 자성체로는 높은 기계적 강도 및 포화 자속밀도를 가지는 규소강판 박편이 사용되었으며, 직렬공진회로를 사용하여 최대전력을 수신하였다. 수신장치의 공진회로는 3 이하의 낮은 공진계수를 가지기에 50 Hz 및 60 Hz 지중선에 범용으로 사용이 가능하다. 사용된 규소강판 및 구리선의 무게와 가격의 곱을 성능지수(Figure of Merit)로 선정하여 이를 최소화하는 설계를 수행하였다. 제작된 시작품의 무게는 약 750 g중으로 2.2 W LED부하 구동이 가능하며, 이때 지중선의 전류는 $100A_{rms}$로 가정하였다.

Effects of Dietary Supplementation of Bacteriophage on Growth Performance, Nutrient Digestibility, Blood Profiles, Carcass Characteristics and Fecal Microflora in Broilers (육계 사료 내 Bacteriophage의 첨가가 생산성, 영양소 소화율, 혈액 특성, 도체 특성 및 분내 미생물 조성에 미치는 영향)

  • This experiment was conducted to investigate the effects of bacteriophage SE supplementation on growth performance, nutrient digestibility, blood profiles, visceral organ weight, meat quality and excreta microflora in broilers. A total of 340 1-d-old ROSS 308 broilers (mixed gender) with an initial average body weight (BW) of $41.71{\pm}0.16$ g were randomly allotted to 4 treatments with 5 replicate pens per treatment and 17 broilers per pen for 31 days. Dietary treatments were: 1) CON, control diet, 2) SE05, CON+0.05% bacteriophage, SE 3) SE10, CON+0.10% bacteriophage SE, and 4) SE15, CON+0.15% bacteriophage SE. During d 15 to 31, broilers fed SE15 diet had a higher (P<0.05) body weight gain than broilers fed CON diet. Overall, body weight gain in SE10 and SE15 was greater (P<0.05) than that in CON. Apparent total tract nutrient digestibility and blood characteristics did not differ (P>0.05) among treatments. The water holding capacity was increased (P<0.05) in SE15 compared with CON. Other meat quality in terms of pH value, breast muscle color ($L^*$, $a^*$, $b^*$) and drip loss were unaffected by dietary supplementation with bacteriophage SE. The visceral weight of bursa of Fabricius was increased (P<0.05) in broilers fed the bacteriophage SE incorporated diets compared with those fed the CON diet. No difference (P>0.05) was observed in visceral weight of liver, spleen, breast muscle, abdominal fat, gizzard and excreta concentrations of Lactobacillus, Clostridium perfringens, Escherichia coli, and Salmonella. In conclusion, dietary supplementation with 0.10 and 0.15% bacteriophage SE could improve the growth performance, breast muscle water holding capacity and bursa of Fabricius visceral weight in broilers.

  • The objectives of this study was to determine whether a new physically modified cornstarch by ultra-fine- or nanoscale pulverizer to reduce particle size offers better bioactive function than native cornstarch in weanling Sprague-Dawley rats. Male weaning Sprague-Dawley rats were fed diets containing native cornstarch (NAC), ultra fine pulverized cornstarch (UFC) or nano-scale pulverized cornstarch (NSC) for 4 weeks. In vitro rate of starch hydrolysis, growth performance, organ weight, intestine length intestinal proliferation and the fermentation by Bifidobacterium of rat cecum were evaluated. The diet with reduced particle size (UFC or NSC) significantly increased body weight gain and organ weight. Feed efficiency was increased in NSC fed rats and was not affected in UFC fed rats. Intestinal proliferation was decreased in NSC group. Reduction of particle size also increased cecal short chain fatty acid concentration and the growth and acidifying activity of Bifidobacterium. It is concluded that a reduction of particle size of starch granules by physically modification may increase growing performance and gut function.

Effect of Magnetic Field Exposure on Semen Characteristic and Organ Weight in Mice (자기장이 웅성 생쥐의 정액성상과 장기무게에 미치는 영향)

  • This study were performed to investigate the effect of magnetic field exposure on semen characteristic and the weights of body, reproductive organs and liver, kidney and spleen in mice. In magnetic field exposure for 15 days, sperm concentrations and viability were significantly lower in magnetic field(15.7$\times$10$^{6}$ $m\ell$, 29.3%) than that in control group(25.1$\times$10$^{6}$ $m\ell$, 34.4%)(P<0.05). The proportion of sperm abnormality were significantly increased in magnetic field exposure groups for 15 days than that in control group(P<0.05). The exposure of magnetic field in mice didn't affect the body and reproductive organ weight such as testis, epididymis, vasicular gland and coagulatin gland. The weight in liver and kidney were not affect in magnetic field exposure groups. However, the spleen weight were significantly higher(P<0.05) in group exposed with than without magnetic field. This studies indicate the short or long term magnetic field exposure in mice were noxious effects on the sperm characteristics and spleen weight, but didn't affect body, reproductive organs, and liver and kidney weight.

  • This study were performed to investigate the effect of Bisphenol A(BPA) administration on semen characteristic and body or organ weight in mice. In BPA administration for 15 days or 30 days, sperm concentration and viability were lower in group treated with BPA than in control group. The proportion of sperm abnormality were significantly(P<0.05) increased in groups administrated with BPA for 15 or 30 days than in control group. The administration of BPA in mice didn't affect the body and reproductive organ weight such as testis, epididymis, vasicular gland and coagulatin gland. The spleen weight were significantly affect, but liver and kidney weight were not affect in BPA administration groups for 15 days or 30 days This studies indicate that the short or long term BPA administration in mice were noxious effects on the semen characteristics and organ weight, but didn't affect body weight and reproductive organs.

  • 형질전환 (유전자 결핍; Knockout) Min 마우스를 이용하여 대장암 발생에서 배추, 양배추 주성분인 indole-3-carbinol (I3C)의 대장암 예방효과를 규명하고자 하였다. 실험동물로는 C57BL/6J-Apc$^{min/+}$(Min 마우스)계의 5내지 6주령의 수컷 이형접합체 형질전환 마우스 70마리와 C57BL/6J계의 동일 산자, 동일 주령의 수컷 wildtype 비형질전환 마우스 10kfl를 The Jackson Laboratory 사 (Bar harber, ME, USA)로부터 직접 구입하였다. C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 (Min)마우스 70마리를 각 군 20내지 25마리씩 세군으로 나누었다. Group 1에는 20마리, Group 2에는 25마리, Group 3에는 25마리를 배치하고, I3C 투여 실험군 (Group 1과 2)에는 실험시작시에 AIN-76A 분말사료에 I3C가 각각 100 및 300ppm이 함유되도록 조제하여 공급하였다. 그리고 실험군(Group 3)에는 실험시작부터 종료시까지 AIN-76A 정제고형사료(Teklad사, WI, USA)를 자유로이 급이하였다. 각군간의 체중, 사료 및 음수소비량을 매 2주마다 측정하였고, 10주간 (16주령)의 실험종료시에는 최종체중과 간장, 신장, 비장 등의 장기무게를 측정하여 상대장기 무게비를 산출하였다. 대조군으로서 C57BL/6J계의 동일 산자, 동일 주령의 수컷 wildtype 비형질전환 마우스 10마리는 같은 조건의 사육실에서 AIN-76A 정제고형사료를 33주간 자유로이 급이하였다. 실험동물은 부검전에 하룻밤 동안 절식하고 이산화탄소 흡입 마취하에서 흉대동맥을 절단하여 방혈하고 각 장기(심장, 폐, 위)를 적출하여 생리심염수에 넣어 장기무게를 측정하고 포르말린에 고정하였다. 소장과 대장의 검사를 위하여 위의 식도부위와 직장을 실로 결찰하여 적출하고 생리심염수를 주입하여 팽창시켜, 십이지장, 공장, 및 회장, 그리고 대장으로 나누어 여과지에 펼친 후 포르말린에 고정하였다. 소장과 대장은 육안 및 자동 영상분석길ㄹ 이용한 분석이 끝난 후에 각 부위별로 4-6개의 절편을 작제하여 포르말린에 재고정하고, 통상적인 조직처리과정, 파리핀 포매 및 3-4$\mu$m 두께의 조직절편을 제작하여 H&E 염색을 실시하여 현미경으로 검경하였다. 약 1주일간의 포르말린 고정이 끝난 소장 및 대장을 부위별, 별 종양개수 및 분포를 자동영상분석기(Kontron Co. Ltd., Germany)로 분석하였다. 체의 변화, 장기무게, 사료소비량 및 마리당 종양의 개수에 대한 통계학적 유의성 검증을 위하여 Duncan's t-test로 통계처리 하였고, 종양 발생빈도에 대하여는 Likelihood ration Chi-square test로 유의성을 검증하였다. C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 마우스에 AIN-76A 정제사료만을 투여한 대조군의 대장선종의 발생률은 84%(Group 3; 21/25례)로써 I3C 100ppm 및 300ppm을 투여한 경우에 있어서는 각군 모두 60%(Group 1; 12/20 례, Group 2; 15/25 례)로 감소하는 경향을 나타내었다. 대장선종의 마리당 발생개수에 있어서는 C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 마우스에 AIN-76A 정제사료만을 투여한 대조군은 1.40$\pm$0.24(100%)에 비하여 I3C 저농도 투여 실험군(Group 1; 0.85$\pm$0.23; 61%, P<0.01), 그리고 I3C 고농도 투여 실험군(Group 2 ; 1.32$\pm$0.29 ; 94%)의 순으로 감소하였다. 선종의 크기별 종양의 발생개수의 분포에 있어서 I3C 저농도 투여 실험군에 있어서는 선종의 크기가 3mm이하의 수가 현저하게 감소하였다. C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 마우스에 AIN-76A 정제사료만을 투여한 대조군의 부위별 소장선종의 발생수는 십이지장부위를 제외하고 각 군에서 유의한 변화는 관찰되지 않았다. 십이지장 종양의 발생개수에서만 I3C 저농도 투여 실험군(Group 1 ; 3.11$\pm$0.85)이 대조군 (Group 3: 1.48$\pm$0.35) 및 I3C 고농도 투여 실험군(Group 2: 1.56$\pm$0.47)에 비하여 유의성 있게 증가하였다. (P<0.05). 따라서 I3C은 소장에서는 암예방 효과가 뚜렷하지 않으나, 대장에 대한 암에방 효과가 있을 것으로 생각된다. 소장 및 대장을 제외한 간장, 신장, 비장, 심장, 폐 그리고 위 등의 기타 장기에서의 조직병리학적 변화는 관찰되지 않았다. 소장 및 대장의 종양은 선종(polyps)으로 관찰되었다. 지난 10여년간 형질전환 및 유전자 결핍 실험동물의 종류가 기하 급수적으로 증가하여 이용되고 있다. 가족성 대장 선종성 용종증(FAP)의 대표적인 모델로 이용되고 있는 C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 마우스를 사용하여 배추나 양배추의 주요성분인 Indole-3-carbinol(I3C)의 대장암 예방효과가 있는지를 검색하여 본 결과 AIN-76A정제사료만을 투여한 대조군의 대장선종의 발생률 84%에 비하여 I3C 100 및 300ppm을 투여한 실험군에 있어서 각군 모두 60%로서 감소하는 경향을 나타내었으며, 대장선종의 마리당 발생개수에 있어서는 대조군의 1.40$\pm$1.041를 100%로 환산하였을 경우 I3C 저농도 및 고농도 투여 실험군에서는 각각 약 61%와 94%를 나타내여 감소하였다. 특히 대장선종의 크기별 분포에 있어서 선종의 크기가 3mm이하의 수가 현저하게 감소하였다. 따라서 저농도 I3C의 투여는 실험적 유전성 가족성 대장 선종성 용종증 모델에 있어서 어느정도 암 예방효과가 있는 것으로 생각된다. 그러나 소장 선종의 발생에는 별 영향이 없는 것으로 생각된다. 그러나 본 실험에 사용된 C57BL/6J-Apc$^{min/+}$계 수컷 이형접합체 형질전환 마우스는 실험개시 시점이 7내지 8주령이 경과하여 이미 태생기부터 소장 및 대장의 선종 발생이 진행되어 온 것을 감안하고 특히 비스테로이드계 항염증 소염제(NSAIDS)와 같은 강력한 COX-2억제제가 아님을 고려하면, 상당한 선종의 발생을 억제할 수 있는 가능성이 매우 높다고 생각한다. 또한 이제까지 배추나 양배추 성분의 복합성분들에 대한 실험적 대장암 모델에서의 촉진효과 등에 대한 보고들이 있어 온 점을 고려할 때 위암(Kim 등, 1994) 간암(Kim 등, 1994), 유방암(Grubbs, 등, 1995; Bradlow 등, 1995)에 대한 예방효과가 있을 것으로 생각된다. 앞으로 이러한 종양조직내에서의 COX-2 및 iNOS mRNA와 단백질의 발현정도를 분자병리학적으로 연구중에 있으며, 향후 십자화과식물 성분인 indole-3-carbinol이 마우스뿐 만 아니라 랫드의 화학발암물질에 의한 대장종양에 대한 억제효과 있는지 연구 필요가 있다. Min 마우스와 같은 형질 전환(유전자결핍;knockout) 실험동물을 이용한 새로운 중기 발암성 시험범의 확립을 통한 각종 환경 유해물질의 발암성 유무 및 COX-2 억제작용이 있는 식품인자의 암예방 후보물질을 체계적으로 검색하는데 유용하게 활용될 수 있을 것으로 생각한다.

Effects of Various Sodium Fluoride (NaF) Administration on Blood Glucose, Lipid Composition and A/G Ratio in Rats (NaF 함량을 달리한 투여가 흰쥐의 지질 및 혈당농도와 A/G 비에 미치는 영향)

  • The aim of the study was to investigate the influences of sodium fluoride on the body weight, lipid metabolism functional improvement, blood glucose level, protein component in the sera of rats which fed experimental diets for 5 weeks. Body weight gain of rats was higher in the NF25 group, NF30 group and NF50 group showed a significant difference from the basal diet group (BD group). The changes of the liver and the heart of each argan weights were observed compared to the BD group tends to appear lower in the oral NaF groups (p<0.05). Organ weight of each group of the others showed no significant difference (p<0.05). Serum concentrations of total cholesterol, low-density lipoprotein (LDL)-cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL) and blood glucose were lower in BD group than basal diet plus sodium fluoride groups. Conversely depending on the concentration of sodium fluoride for the high-density lipoprotein (HDL)-cholesterol level, it showed lower results. Serum total protein was observed low showed a significant difference in the NF50 group, there was no significant difference in the other groups (p<0.05). globulin and albumin concentration levels also showed the lowest results in the NF50 group.

Oral Single-dose Toxicity Studies on Germanium-fortified Lettuce, in Mouse (게르마늄을 함유한 상추의 단일 경구투여 독성 검사)

  • Single-dose toxicity test of germanium-fortified lettuce was investigated in mice. Both sexes of C57BL/6 mice were orally administered once at a dose of 2,000 mg/kg. No death, clinical signs and pathological findings related to the treatment were observed. In addition, no significant changes in feed consumption and body weight gain were obtained during the treatment period, in spite of day-to-day fluctuation of water consumption. There were no considerable changes in hematology and serum biochemistry, except a significant decrease in GPT, GOT and LDH. Several alterations were observed in organ weight and blood biochemistry, including thymus, ovaries, heart, kidney and platelet in male or female mice. The ability of spleen cells proliferation was almost same level as shown in control group. However the population of B cells, helper T cells and cytotoxic T cells was not comparably changed in all groups. Taken together, it is suggested that single oral dose of germanium-fortified lettuce to C57BU6 mice did not cause apparent toxicological change at the dose of 2,000 mg/kg body weight.
