  • Anatomy of Pinus koraiensis needles and Ginkgo biloba leaves was investigated after exposure to simulated acid rain, drought and salt solution. All of the stresses applied to the needle of Pinus koraiensis caused collapse of mesophyll tissues; epidermis was collapsed due to acid rain, severe compress and transformation of epidermis, mesophyll, and endodermis to drought, and collapse of phloem and transfusion tissue to salt solution. Spongy mesophyll tissues, however, collapsed in the leaves of Ginkgo biloba; epidermis and palisade tissues collapsed due to acid rain, compress and transformation of spongy mesophyll tissues to drought, and hypertrophy and collapse of endodermis and transfusion tissues to salt solution.

Feeding Quantity of the Black-tipped Sawfly, Acantholyda posticalis posticalis(Hymenopera : Pamphiliidae), infesting the Korean Pine (잣나무넓적잎벌의 섭식량(攝食量)에 관(關)한 연구(硏究))

  • The damage by the black-tipped sawfly, Acantholyda posticalis posticalis, has been increasing all over the area of Korean pine(Pinus koraiensis)stands in Korean peninsula. This study was conducted to provide basic information for the management and control of this pest by investigating the feeding quantity of pine needles during the larval stage. The results were summarized as follows : The total needles damaged in the larval stage of the black-tipped sawfly were 9,584mm in length on an average and those of female and male were 11,774mm and 7,394mm, respectively. The total length ingested actually was 7,797mm and it was equivalent to 81.4% of the total length damaged. Early larval stage showed a little feeding and it was equivalent to 17% of total quantity. On the contrary, the feeding quantity in late larval stages of fourth and fifth instars amounted to 83%. The number of the frass excreted per larva was about 1,160, and the difference between male and female was not showed, whereas it showed remarkable difference among larval instars.

Identification of Lophodermium Species Associated with Needle-cast Disease of Pines in Korea (한국(韓國) 소나무류(類)에 잎떨림병을 일으키는 Lophodermium 속(屬) 균(菌)의 동정(同定))

  • Fallen and living pine needles bearing ascocarps were collected throughout the country from February, 1987 to October, 1988 to identify and describe Lophodermium species associated with Lophodermium needle cast disease of pines in Korea. All of the observed characteristics of given species were compared with published descriptions of the Lophodermium species. As a result of this study, six Lophodermium species were identified and described. L. pinastri was previously recorded while the other five species, i.e., L. australe, L. durzlabrum, L. nitens, L. pini-excelsae and L. pini-pumilae, were unrecorded in Korea. One Lophodermium species has not been fully identified in this study, and tentatively named as L. sp-1. L. australe was observed on the needles of Pinus rigida, P. rigida ${\times}$ taeda and P. taeda, L. pini-excelsae on P. parviflora and P. strobus, L. nitens, L. durilabrum and L. pini-pumilae on P. koraiensis, L. sp-1 on P. thunbergii and L. pinastri on P. densiflora and P. taeda. L. sp-1 was collected from the diseased regions of 2-year-old needles of P. thunbergii and L. durilabrum from P. koraiensis, suggesting pathogenic nature of these fungi.

  • Gas exchange rates and concentrations of ascorbate and glutathlone were measured in needles of eastern white pine(Pinus strohltr) trees differing in foliar sensitivity to ambient oxidant pollution during a ten month period beginning in mid-June, 1988. Current-year needle dry mass and length was 60 to 75% and 45 to 60% less, respectively, in sens~tive trees than in a tolerant tree. Net photosynthesis ($P_n$) and needle conductance ($g_n$) were greatest in the tolerant individual through late September when the rates begin to decline In trees. Needle transpiration rates showed a trend similar to $P_n$ and $g_n$. Ascorbate and total glutathione concentrations in current-year needles increased through the summer and fall, reached a maximum in mid-winter, and then decreased in the spring. Consistently throughout the year, ascorbate concentration was highest in the tolerant tree until the initial springtime decline began in April. The difference In needle ascorbate between the tolerant and sensitive individuals was greater in the summer months (25 to 30%) than in the winter months (8 to 19%). Glutathione content was similar, as was the ratio or oxidized /reduced glutathione, in both tolerant and sensitive trees.

Injury Responses of Woody Landscape Plants to Air Pollutants(I) -Sulfur and Heavy Metal Content- (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 피해반응(被害反應)(I) -엽내유황(葉內硫黃) 및 중금속(重金屬) 함량(含量)을 중심으로-)

  • This study was conducted to investigate the absorbing and/or filtering capacity of woody landscape plants growing in urban and industrial regions of Seoul and Taejon. Korea. Total sulfur content and heavy metal content were analyzed in tree foliage of Pinus densiflora, Pinus koraiensis, Ginkgo biloba. Liriodendron tulipifera, Platanus occidentalis. The total sulfur content in the leaves of woody landscape plants in Seoul and Taejon were remarkably higher than those of the tree leaves in Kwangreung. The total sulfur content of deciduous trees had 2.6 times more than that of coniferous trees. the former had better absorbing capacity of $SO_2$ gas. Especially Ginkgo biloba and Platanus occidentalis were recommanded as absorption trees of $SO_2$ because sulfur content levels of them were the highest among all species and increased until tall coloration. While coniferous trees showed no changes in total sulfur content between four seasons. deciduous trees increased in sulfur during the growing season. Pb content of all species in polluted area were higher than those in unpolluted area. In polluted area, while coniferous trees had higher absorption and accumulation of Pb, deciduous trees had higher absorption of Zn. The content of Pb, Zn and Cu in needle of Pinus koraiensis were more than those of Pinus densiflora, In Pinus koraiensis and Pinus densiflora, heavy metal content in the 2nd year needle were higher than thous in the current year needle. In Pinus densiflora, Pinus koraiensis Liriodendron tulipifera and Platanus occidentalis there was a positive correlation between total sulfur and heavy metals [(Pb+Zn+Cu)/3] content.

Four Species of Endophytic Fungi Isolated from Leaves of Woody Plants in Mt. Hambaek (강원도 함백산에 서식하는 목본식물의 잎에서 분리한 4종의 내생균)

  • In this study, endophytic fungi were isolated from leaves of four species of woody plants, Acer tagmentosum, Larix kaempferi, Abies holophylla, and Pinus koraiensis, on Mt. Hambaek, Gangwondo, Korea. The endophytic fungi were identified using morphological and sequences analysis of ITS regions. The fungal endophytes were identified as Talaromyces radicus, Myceliophthora verrucosa, Cryptosporiopsis diversispora, and Sphaerulina berberidis, which are the first record in Korea. The morphological and molecular phylogenetic characteristics of each strain were described.

Changes in Chlorophyll Contents of Leaves and pH of the Extracted Solutions from the Leaves of 7 Tree Species by pH Level (7개(個) 수종(樹種)의 잎이 pH수준별(水準別) 처리(處理)에 따른 엽록소(葉綠素) 함량(含量) 및 침출액(浸出液) pH변화(變化))

  • We conducted this study as a fundamental study on the response of various tree species against acid rain. The tree species used for this study were Zelkova serrata, Robinia pseudoacacia, Quercus acutissima, Prunus serrulata, Ginkgo biloba, Pinus koraiensis and Pinus densiflora. The leaves were examined for the pH changes by treatment time and the chlorophyll content into various pH solution in vitro. The results obtained were as follows ; 1. When the leaves were immersed in the solution of various pH(pH 3.0-pH 6.0) levels, the pH were changed to species specific pH ranges as pH 5.0~pH 5.5 of Z. serrata, pH 5.5~pH 6.0 of R. pseudoacacia, pH 4.5~pH 5.0 of Q. acutissima, pH 5.5~pH 6.0 of P. serrulata, pH 3.5~pH 4.5 of G. biloba, pH 3.5~pH 4.5 of P. koraiensis until 48 hours. However, in case of P. densiflora, it was difficult to find specific pH range of the species. Z. serrata, R. pseudoacacia and P. serrulata showed a little pH increasing by pH 2.0 solution treatment, while other species showed no change by the solution. 2. The amount of chlorophyll contents in Z. serrata, R. pseudoacacia and P. serrulata were decreased after immersing in the pH 2.0 solution. Chlorophyll content was almost constant in other pH levels. Other species showed almost constant chlorophyll contents in various pH levels and treatment time.

Fatty Acid Composition of Total Lipids from Needles and Pollen of Korean Pinus densiflora and Pinus koraiensis (소나무 및 잣나무의 잎과 꽃가루의 지방산 조성)

  • The needles and pollen of Pinus densiflora and Pinus koraiensis were studied for their lipid contents and fatty acid composition. The total lipid contents in needles of Pinus densiflora and Pinus koraiensis were 5.0 and 4.5%, whereas in pollen of Pinus densiflora and Pinus koraiensis 3.5 and 5.6%, respectively. Twenty-four fatty acids ranged from lauric acid to docosahexaenoic acid(22:6${\omega}$3) were identified in the needle lipids. In needles, linolenic acid and palmitic acid were the major fatty acids. The needles of Pinus densiflora showed higher proportions of docosahexaenoic acid and 5-olefinic nonmethylene-interrupted polyenoic acids than those in the Pinus koraiensis. Twenty fatty acids ranged from myristic acid to lignoceric acid were identified in the pollen lipids. Linoleic acid was the major fatty acid in the pollen followed by oleic and palmitic acid. The fatty acid profile of pollen of Pinus densiflora was similar to those of the Pinus koraiensis pollen lipids.

Effect of Simulated Acid Rain on Water Status of Pinus densiflora and P. koraiensis (인공 산성비가 소나무와 잣나무 잎의 수분상태에 미치는 영향)

  • The water status of Pinus densiflora and P. koraiensis was observed after the treatment of simulated acid rain for every month August 1997 to March 1998. Both of the species showed increase of dry weight, water saturation deficit and epidermis transpiration after treatment of simulated acid rain (pH 3.5) for first three months. On the contrary, water content was decreased in both the species after three month of simulated acid rain treatment. The effect of simulated acid rain, however, became weaker after that and no big difference was observed after seven month of treatment. Water potential of the leaf of both species became lower than that of the control(pH 5.6) after seven month treatment of simulated acid rain. The cuticle layers of the epidermis and florins ring were damaged seriously and the size of the gap between stomata became bigger after the treatment of simulated acid rain.

Cytotoxic Effect of the Pine needle extracts (솔잎 추출물의 in vitro계 암세포 성장억제효과)

  • This study was performed to observe cytotoxic effect of the pine needle extracts against cancer cell lines including human gastric carcinoma (KATOIII), human lung carcinoma (A549), human hepatocellular carcinoma (Hep3B) and human breast adenocarcinoma (MCF-7) using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) and SRB (sulforhodamine B) method. The extracts were prepared by step-wise fractionation of ethanol extract of pine needles using diethylether, chloroform, ethylacetate, butanol and water. The growth of the cancer cells in medium containing pine needle extracts were significantly inhibited degree in proportion to the increase of the extract concentration. A significant shrinkage of Hep3B cells was observed when the cells were exposed into 0.5, 1 mg/mL of pinus rigida extract.
