  • These days sex has become a tool of interest, a constant stream of sex crimes occurs and it brings the age of population cliffs to an age of low birth rates. As an alternative to overcome these problems, we should establish the praying for a child, which has been our ancestors' way of life, as a sacred culture that holds the mystery of life-birth with a new awareness. Therefore, the purpose of this study is not to re-enact the Zhaungam area as a fun and interesting place, but as a sacred place to pray for the nobility, longevity and wealth of life. Based on the spirit of natural love, meeting, healing, and love based on the spirit of traditional culture, Doridori was chosen as the main theme of the project based on the spirit of natural love and life-respect, and the name "Doridori Secret Garden" was given out of 10 medals in Dandong for tourism storytelling. Cultural content becomes a marketing tool for the region and acts as a force for tourists to visit the area. In this context, it was confirmed that the creation of "Doridori Secret Garden" could lead to a high value-added industry that attracts tourists by enhancing the brand value of Andong and showing sufficient significance to become a new attraction. This study is intended to re-create the Zhaungam area as a sacred place to pray for the nobility of life and longevity and wealth of life, noting that people still pray for the birth of their children in Zhaungam and Zhagungam.

Identification of Monoecious and Dioecious Plants of. Schisandra nigra Using the RAPD Markers (RAPD 표지인자를 이용한 흑오미자의 자웅동주 및 자웅이주 식물의 동정)

  • RAPD (Random Amplified Polymorphic DNA) analysis was conducted to Schisandra nigra plants in order to select the specific markers for monoecious and dioecious individuals. RAPD results using eighty random 10-mer primers revealed that S. nigra had a different banding pattern from S. chinensis and Kadsura japonica. When DNA isolated from leaves of monoecious and dioecious plants were used as PCR template, only five primers, OPA-17, OPA-19, OPB-03, OPB-09 and OFB-16, showed polymorphic band patterns. No variation in banding profiles within male or female individuals was observed when these five primers were used whereas three monoecious plants (No 1, No 2 and No 3) showed different banding patterns one another, A 750 bp segment was amplified by primer OPB-3 from male individuals. On the other hand, two segments, 950 bp and 1690 bp, with OPA-19 and 700 bp of segment with OPB-3 were amplified in female individuals. These result indicate that the specific buds of male and female S. nigra could be used as genetic markers for the early discrimination of male and female individuals.

Sex and Sex Reversal of Bivalves (이매패류의 성과 성전환)

  • The sex of bivalves is classified into gonochorism and hermaphroditism, and hermaphroditism is further divided into simultaneous (synchronous or functional), and sequential (asynchronous). The sequential hermaphroditism signifies sex reversal in accordance with seasons. In general, in the case of gonochoristic bivalves, manifestation of sex as female and male, after having identified the morphological sex, is limited to a period of the life cycle in any given individual. In order to accurately identify sex, continuous monitoring of changes in the sex during life cycle is necessary by tagging the sex. In the bivalves, sex reversal and sequential hermaphroditism has been reported in the Ostreidae, Pectinidae, Veneridae and Arcidae etc.. Most of these studies have reached this conclusion based on evidence derived from sex ratio change in accordance with the age within same populations. Sex change in the sequential hermaphroditic bivalves normally occurs when the gonad is undifferentiated between spawning seasons. Two factors are involved in sex determination and sex change in bivalves, namely genetic and environmental. However, more detailed research is needed on the effect of the genetic and environmental factors involved in sex determination and sex change of bivalves.

Variation of RAPD patterns between Male and Female Genomic DNAs in Dioecious Rumex acetosa L. (자웅이주 식물 수영 (Rumex acetosa L.)에서 암.수에 따른 RAPD pattern의 다양성 분석)

  • The genetic variation of random amplified polymorphic DNA (RAPD) patterns of genomic DNAs was investigated in dioecious plant Rumex acetosa L., which carries different sex chromosome complements in female (2n=12A+XX) and male (2n=12A+XY$_1$Y$_2$). One hundred and twenty random primers consisted of 10-mer were used for PCR amplification. Polymorphic bands were found in 24 primers. Specific bands for female and male were 16 and 18, respectively. Especially, a band of 1,440 bp from the OPC-10 primer was male specific. These sex specific RAPD markers are used to understanding the sex determination mechanism in plants.

Morphology and Chlorophyll Contents of Leaf and Wood Anatomical Characteristics of Three Schisandraceae Species in Korea (국내자생 오미자과(科) 3수종의 잎 형태, 엽록소 함량 및 목부해부학적 특성)

  • It was investigated that morphological characteristics of leaf and stomata, chlorophyll contents and wood anatomy characteristics in Schisandra chinensis, Schisandra nigra and Kadsura japonica. Leaf morphological characteristics were significant at 5% levels within the species and within the individuals as a male, a female and monoecious. K. japonica was the longest in the leaf length, followed by S. chinensis and S. nigra. For the leaf length among S. nigra individuals, the female was longer than male and monoecious. As for the Petiole length, S. nigra was twice longer than S. chinensis and K. japonica. Stomata length was ranged from $92.5\;{\mu}m$ to $105.9\;{\mu}m$ for S. nigra, $79.9\;{\mu}m$ for S. chinensis, $76.2\;{\mu}m$ for K. japonica. S. nigra was longer than the others in length and width, while its stomata density was lower than the others. The size of vessel elements and xylem fiber showed a similar tendency to stomata, significant at 5% levels within S. nigra individuals (male, female and monoecious). Chlorophyll contents were also variable in inter and intra species and in general K. japonica showed higher levels of chlorophyll contents than Schisandra spp. plants.

Development of a Female-associated SCAR Marker in Schisandra nigra Max. (Schisandra nigra Max.에서 암그루에 연관된 SCAR 마커의 개발)

  • Schisandra nigra Max., a dioecious plant native to Jeju Island in Korea, is cultivated on a small scale for fruit production. As fruit-producing female individuals are generally considered to be more valuable than male, early identification of plant sex at the seedling stage is important. In this study, a sequence-characterized amplified region (SCAR) marker associated with a female-specific region in the genome of S. nigra was investigated. Of 120 randomly amplified polymorphic DNA (RAPD) primers, one primer (OPB-03) consistently amplified a 749 bp band in female plants. The female-specific PCR product was isolated and cloned, and the nucleotide sequences were then determined. Southern hybridization performed using the female-specific fragment as a probe produced positive signals only in genomic DNA from the female plants. This result revealed that the 749 bp segment of DNA was present in the genome of female plants but absent in the genome of male plants. A SCAR primer pair was designed based on the RAPD marker to amplify a 436 bp fragment in the genomic DNA of female plants. This primer pair amplified the expected size of DNA fragment in female plants and four monoecious individuals collected from a natural population. The SCAR marker identified in this study can be used to distinguish female-flowering individuals at the seedling stage.

  • The sex ratio and gonadal histology were investigated in the Cobitis lutheri Rendahl collected from the Tamjin River of Chollanam-do Province. Korea. The ratio of males declined in proportion to increase in size, while that of females increased in proportion to increase in size. Hermaphrodites were found in the intermediate size fishes. They had degenerating testicular tissue and developing ovarian tissue simultaneously. These phenomena were discussed in relation to skewed sex ratios.

Morphology of Utricles and Maturing Period in Codium fragile(Suringar) Hariot (청각, Codium fragile(Suringar) Hariot의 포낭의 형태 및 성숙시기에 관하여)

  • The present work was carried out to investigate the maturing period and the morphology of utricle Codium fragile collected from April 1984 to March 1985 in the coast of Chumgmu southern of Korea. Ranges of water temperature and salinity during the survey period were $7.5\~27.0^{\circ}C$ and $32.9\~34.5\%_{\circ}$, respectively. The gametangia were produced throughout the year. Mature gametangia were present on plants collected from June to November, whereas most of plants collected from January to May occured as the elongated utricles with both mature and immature gametangia. As for the sexuality, most of plants were dioecious and some monoecious, and there were more male gametophytes rather than female gametophytes. Some plants collected during winter months of December to February were observed as thalli with both brunted and elongated utricles, and male and female gametangia occured together on a single utricle.

  • Sexual maturation and gametogenic cycle of the hard shelled mussel, Mytilus coruscus were investigated by means of histological method. Specimens were collected monthly in Hansan Bay of south coast, Korea from January to December 2002. Sex of the clam was dioecious. The gonads are composed of a number of gametogenic follicles. The gonad index (GI) was reached the maximum in February (3.40), and the minimum in August (1.18). The monthly variation of condition index (CI) coincides with gonad index. The main spawning occurs between February and March. The gametogenic cycle of the clam could be divided into six stages: resting (August to November), multiplicative (November to December), growing (December to January), maturation (January to February), partially spent (February to April) and completely spent/degenerative stage (April to July). Sex ratio (♀/♂) was 1:0.96.

Antiestrogen등 약물이 cytochrome P450와 AEBS에 미치는 효과의 관계

  • Antiestrogen은 에스트로젠 의존성 유방암 치료에 사용되는 약물로써 표적 세포 내에서 에스트로젠 수용체와 작용하여 세포 증식 억제 작용을 나타내고 동시에 에스트로젠 수용체와는 구분되는 소포체 분획의 antiestrogen specific binding site (AEBS) 와도 결합을 하는 것으로 알려져 있다. 그러나, 아직 이 AEBS의 생리적 또는 약리적 기능은 밝혀져 있지 않다. 따라서 본 실험에서는 AEBS의 기능을 조사하기 위하여 cytochrome P45O III 효소군과 AEBS와의 관계를 자옹 백서를 이용하여 면역 화학 반응 실험 및 경쟁적 결합 반응 실험을 하였고, 그 결과는 다음과 같다. 1) AEBS에 대해서 SKF-525A와 metyrapone은 결합 능력을 나타내었다. 2) 자성쥐에서는 주령이 증가함에 따라 cytochrome P450양이 감소하였다. 3) 자옹성쥐 모두에서 phenobarbital 처치에 의해 cytochrome P450 III 효소양이 증가하였고, AEBS도 증가하였다. 4) 웅성쥐에서는 testosterone에 의하여 AEBS가 증가하였다. 5) 자웅성쥐 모두에 tamoxifen 관류시 cytochrome P450 III 효소양이 증가하였고 estradiol과 병용 관류시에는 tamonifen 단독 관류시보다 감소하였다. 이상의 결과에서 tamoxifen이 cytochrome P450 III을 유도할 수 있는 것으로 사료되며 cytochrome P450 III 효소군과 안티에스트로젠 결합부위와 밀접한 관련이 있는 것으로 생각된다.
