Studies on the Associative N2-fixing Microbes in Crop Root Systems -I. Associative Nitrogen Fixation by Azospirillum Isolated from Weed Roots grown Rice Paddy Field and its Distribution (작물근계(作物根系) 협생질소(協生窒素) 고정미생물(固定微生物)에 관(關)한 연구(硏究) -I보(報) : 논 토양(土壤) 잡초근계(雜草根系) 협생질소(協生窒素) 고정미생물(固定微生物)의 분포(分布)와 질소고정(窒素固定))

  • This experiment was carried out to investigate the distribution of associative nitrogen fixers, Azospirillum spp. and their nitrogenase activities measured by ARA in the rhizosphers of rice, soybean, and weed grown in the rice paddy field at ear formation stage of rice. Nitrogenase activities produced by Azospirillum spp. enriched from histophere ranged from 16 to 53 n mole/tube/hr.. High nitrogen fixing activities more than 30 n mole/tube/hr. were observed in the histophere of the Echinochloa crus-galli L., Finbristylis miliacea L., and Monochoria vaginalis var.. Nitrogen fixing activities of Azospirillum spp. obtained from single colonies which originated from the rhizoplane of rice (pot-kwang var.), Finbris tylis miliacea L., Monochoria vaginaliz var., Glycine max L. were higher over 100 n mole/tube/hr. than those histophere. Genus of Azopsirillum isoltated from roots of the Graminease (Oriza sativa L., $C_3$-plant, Echinochloa crus=galli L., $C_4$-plant, Cyperus difforuis L.. $C_4$-plant), and Aeschynomene indica L. (Leguminosae, $C_3$-plant) was identified as A. brasilense. However, both strains, A. lipoferum and A. brasilense ($nir^-$ or $nir^+$ strain) were isolated from other plant roots, Both $nir^-$ and $nir^+$ strains of A. brasilense were associated with the same host plant.

Identification of a Carduus spp. Showing Anti-Mycobacterial Activity by DNA Sequence Analysis of Its ITS1, 5.8S rRNA and ITS2 (Mycobacteria에 대해 항균력을 나타내는 엉겅퀴의 분류를 위한 ITS1, 5.8S rRNA, ITS2의 염기서열 분석)

  • It has been reported that extracts of globe thistle (Echinops spp.) and thistle (Circium spp., Carduus spp. and Onopordum spp.) have anti-bacterial and anti-fungal activities. Methanol extracts of Echinops setifer and Carduus spp. were used to test and see if the extracts of these plants could suppress growth of Mycobacterium smegmatis and Mycobacterium fortuitum. Although extract of Echinops setifer showed no anti-mycobacterial activities, extract of Carduus spp. showed inhibition zones when tested with filter discs. Genomic DNA was isolated from Carduus spp. and PCR was performed to clone a DNA fragment containing ITS1, 5.8S rRNA gene and ITS2. A 733-bp PCR product was obtained and its DNA sequence was reported to the GenBank (accession number GU188570). BLAST search of the obtained DNA sequence did not show a match with any DNA sequences in the Genbank. Carduus crispus and Carduus defloratus had the closest phylogenetic relationships to this plant.

Studies on Distributions and Morphological Characteristics of Calanthe discolor, C. sieboldii, and C. bicolor Native to Cheju Province (제주 자생 새우란, 금새우란, 왕새우란의 분포와 외형적 특성에 관한 연구)

  • Distribution analysis on natural habitats for the genus Calanthe in Mt. Halla of Cheju province revealed that C. discolor and C. sieboldii were found mostly in the range of 120~400 m and 300~600 m above the sea level, respectively. C. bicolor was found in 300~400 m where both C. discolor and C. sieboldii were overlapped. The present morphological assessments showed that C. discolor had the smallest sizes of leaves and floral parts, C. had the largest, while C. bicolor had the intermediate between the above mentioned two species. Flower colors of C. discolor were brownish red and simple white, and that of C. sieboldii was merely golden yellow, whereas C. bicolor had a diverse mixed colors originated from the other two species. The specific distribution of natural habitats and morphological characteristics of three Calanthe species led the suggestion that C. bicolor can be regarded as new species derived originally from natural hybridization between C. discolor and C. sieboldii.

Experiments on the Resistant Force of the Trees in Rivers (하천 내 수목의 내력 시험)

  • This study was focused on the investigation of the distribution of trees which is an important factor for the improvement of the river environment, and the experimental examination of resistant force of trees to the external forces. The investigation of plant distribution performed in 11 major rivers in Korea showed that the willow family grows spontaneously over the whole country. The field experiments on the resistant force of trees were carried out on the 78 trees of 8 species in 3 different sites to estimate whether pulled out trees damage downstream hydraulic structure. The experiments were performed by the method that a backhoe drew trees and the forces were measured when the trees were overturned and pulled out. The analysis of the experimental results showed that there is a linear correlation between the resistant force and DHB (diameter at breast height).

  • Innovation clusters have different factors depending on different stages. Innovation clusters in Gyeonggi now are early stages, which are necessary to investigate factors so that they go into upstages. Silicon Valley is known as one of the best innovation clusters, which are now represented as the forms of related variety. The purpose of the paper is to investigate current situation of innovation clusters in Silicon Valley and suggest appropriate policies for the development of innovation clusters in Gyeonggi. In order to do this, we visit several representative institutions regarding infra, human-training, industry-academic-institution cooperation, start-up assistance and local government. Throughout the case study, we propose implications, and produce policy tasks for the development of cluster policy in Gyeonggi.

Effect of Herbs on the Growth-Inhibition of Lactic Acid Bacteria and Quality Characteristics of Dongchimi (허브가 젖산균의 생육억제와 동치미의 품질특성에 미치는 효과)

  • Effects of antimicrobial herbs on growth of tactic acid bacteria were investigated to extend shelf-life of Dongchimi, kimchi made with radishes and spices. Marta rosemary, among eight foreign and domestic herbs, showed strongest antimicrobial activity against seven kinds of lactic acid bacteria. Spices such as spring onion, garlic, and ginger showed no antimicrobial activity against lactic acid bacteria. During fermentation of Dongchimi for 6 days at $35^{\circ}C$, addition of marta rosemary inhibited acid formation, maintaining optimal pH of 3.8-4.2 throughout fermentation period, while no inhibitory effect was observed from Sambaekho (Saurus chinensis), pepper mint, and stevia. Results show shelf-life of Dongchimi was extended by addition of marta rosemary, which controlled over-production of tactic acid by inhibiting bacterial growth.

Comparison of Biological Activities on Extracts and Fractions in Compositae Plants (국화류 추출물 및 분획물의 생리활성 비교연구)

  • This study was conducted to investigate the anti-oxidative, anti-oxidative and tyrosinase inhibitory effects of MeOH 80% extract and hexane, chloroform, ethyl acetate, butanol and aqueous fraction on three kinds of compositae plants in Korea. In the antimicrobial effect, the extract and chloroform fraction of Eclipta prostrata and hexane fraction of Carpesium abrotanoides L. and chloroform fraction of Siegesbeckia glabrescens exhibited significant inhibition. The antioxidant activity of ethyl acetate and butanol fractions was more than 90% in all three plants. In case of tyrosinase activity, showed a potent inhibition ethlyacetate fraction of Siegesbeckia glabrescens and Carpesium abrotanoides L, which were higher than control group. In MeOH 80% extracts, there was not found to have antimicrobial, anti-oxidant and tyrosinase inhibitory activity, however there was ethylacetate fraction of Siegesbeckia glabrescens to show effectss commonly in the three assay system.

Analysis of antibacterial, anti-inflammatory, and skin-whitening effect of Centella asiatica (L.) Urban (병풀의 추출용매에 따른 항균, 항염증 활성 및 피부 미백효능 등의 분석)

  • The imports of Centella asiatica L. Urban are increasing year-by-year due to the fact that its extract is a raw material used for skin wounds and in cosmetics. However, studies on the cultivation and identification of native C. asiatica species in Korea have been extremely rare. Therefore, this study was conducted in order to investigate the physiological and functional activity of Korean native C. asiatica plant cultivated in Hapcheon, Gyeongsangnam-do, Korea. As a result, the highest antibacterial and anti-inflammatory activities were examined with methanol extract while skin-whitening and wrinkle improvement were examined with water extract. Seven bacterium and one fungus were treated with 50% methanol extracts of C. asiatica and most of the bacterium showed similar or low levels of antibacterial activity compared to the control group of Omiza (Schisandra chinensis) extract, except for Streptococcus pyogenes, which showed higher antimicrobial activity than that of Omiza extract. However, neither C. asiatica and Omiza extracts showed antimicrobial activity against the fungus, C. albicans. The results of anti-inflammatory effect analyses with Raw 264.7 cells confirmed that the treatment of methanol extract reduced the level of NO by 50% or more compared to the control group. In addition, the water extract showed the highest reduction of melanin content of up to 20% more than the control group when examined with B16F10 cell line, indicating a significant skin-whitening effect. Furthermore, we were able to show the significant skin wrinkle improvement caused by C. asiatica extract with NHDF cell as an indicator, but strong cytotoxicity was also observed, suggesting that further studies are necessary.

Potential of Antifungal Lactic Acid Bacteria Isolated from Kimchi as Cheese Starters (김치 분리 항진균 유산균의 치즈 스타터로서 이용 가능성)

  • This study was performed to identify the cheese starter potential of antifungal lactic acid bacteria isolated from Kimchi. Eight fungi were isolated from cheese or the cheese ripening room, and identified as Penicillium and Cladosporium by ITS-5.8S rDNA analysis. Twenty-two lactic acid bacteria species with antifungal activity were isolated from Kimchi, and identified as Lactobacillus and Pediococcus by 16S rRNA sequence analysis. Six lactic acid bacteria species were selected (L. sakei subsp. ALJ011, L. sakei subsp. ALI033, L. sakei subsp. ALGy039, P. pentosaceus ALJ015, P. pentosaceus ALJ024, and P. pentosaceus ALJ026) based on higher antifungal activity from the initial 22 species. Out of the six identified species, L. sakei subsp. ALI033 had the highest antifungal activity. For growth of the six lactic acid bacteria, optimal temperature and pH were $30{\sim}37^{\circ}C$ and 7.0, respectively. Proteolytic activities of the six lactic acid bacteria were almost as strong as the commercial strain Str. thermophilus Body-1. Coagulative activities of L. sakei subsp. ALI033, P. pentosaceus ALJ015, and P. pentosaceus ALJ024 were higher than those of L. sakei subsp. ALJ011, L. sakei subsp. ALGy039, and P. pentosaceus ALJ026. The acid resistance of L. sakei subsp. was higher than that of P. pentosaceus. The major organic acid component of the lactic acid bacteria culture medium was lactic acid.

Perception of Visitors on Traditional Marketplaces becoming Tourist Attractions and Supporting Policies - Focused on Tongin Market, Seoul - (전통시장의 관광지화 및 지원정책에 대한 이용객들의 인식 - 서울 통인시장을 중심으로 -)

  • Tourism is being used as a tool to regenerate declining spaces. Therefore, the government is carrying out various projects to create tourist attractions to revitalize the traditional marketplace, which is declining nationwide. Because of these government-led projects, traditional marketplaces have been transformed into tourist attractions, and some markets have received enough positive feedback that they have been successfully revitalized economically. However, as marketplaces have become tourist destinations, negative views about government support projects are also increasing. This also leads to conflicts due to differences in perception and interests of stakeholders. This paper examines the process of the marketplace becoming a tourist destination focused on the Tongin Market, in Seoul which has been regenerated as a tourist attraction through a government support project. The purpose of this study is, first, to examine the background of tourist markets becoming tourist attractions and the issues surrounding them. The second was to explore in depth the types of conflict perceptions of visitors, residents and tourists, and the perception of their responses. As a research method, the process of the revitalization of traditional markets through news and existing literature, issues and the difference of perceptions about tourist-centric traditional marketplaces were examined. In addition, the Q methodology was used, which is a qualitative research methodology that can measure the difference of viewpoints and perceptions of specific events in order to analyze the difference of perception among visitors in traditional marketplaces. The results of the study show that there are three main perspectives of difference among visitors' perceptions about government support projects in traditional marketplaces and the marketplace becoming a tourist attraction. The 'policy dependent perspective' emphasizes the necessity of government regulations, a 'critical perspective' criticizes the government's traditional marketplace support policy, and the 'neutral perspective' acknowledges the need for government-led projects and accentuating the internal competitiveness of the marketplace. The study provided the environment for academic discussion about traditional marketplaces, which are becoming tourist attractions nationwide. In addition, this paper can contribute to providing implications for future government projects by analyzing the perception of marketplace visitors.
