기능성 탄수화물소재를 이용한 당뇨치료식 개발

  • 본 연구는 저열량 기능성 탄수화물 소재(저항전분, 식이섬유)를 이용하여 당뇨병 환자의 혈당과 지방대사를 조절하는데 도움을 줄 수 있는 치료식을 개발하기 위해 동물실험과 임상실험을 수행하여 제품의 혈당 및 지방대사 조절, 혈당조절호르몬, 장내기능조절 효과를 검증하였다. 또한 동물실험을 통해 대장에서 염증을 일으키는 것으로 알려진 deoxycholate (DOC)를 식이무게의 2%로 첨가하고, HAS는 일반전분의 50%로 대체시켰을 때 HAS가 DOC의 염증작용에 어떠한 영향을 미치는지 살펴보아 HAS의 장기능 개선에 대한 기능성을 환인해보았고 저항전분이 혈당 및 혈액내 지질조성에 미치는 영향을 살펴보았다. 인체실험 연구결과는 저항 전분을 섭취한 군에서 체중이 유의적으로 감소하였고 비만도를 나타내는 BMI도 감소하여 개발된 선식제품의 체중감소 효과가 입증되었다. 당대사 결과는 동물실험의 경우는 저항전분을 섭취한 군에서 혈당감소를 나타내고 있으나 인체실험의 경우 선식제품 섭취 후 혈당과 인슐린의 농도가 영향을 받지 않았으며 혈액 내 지방성분은 동물실험에서는 혈액내 총지방과 중성지방이 저항전분 섭취시 감소하였고, 인체실험의 경우 저항전분 시제품 섭취 후에 혈액내 중성지방량은 약간 증가하였으나 총지방량과 총콜레스테롤량, LDL-콜레스테롤량은 감소하는 효과를 보여주었다. 또한 면역능력에 대한 지표는 저항전분 시제품 섭취 후 혈액 내 면역글로부린 G가 유의적으로 증가 하였다. 동물실험을 통한 저항전분의 장기능에 관한 연구결과는 저항전분의 한 형태인 HAS의 섭취가 대장길이와 맹장무게를 증가시키며, 맹장에서의 비피더스균의 증식을 가져와 장내 SCFA의 생산을 증가시키고, 맹장내 pH 감소를 가져오며 대장 점막에서의 PGE2의 생산을 감소시켜 장 기능개선의 효과를 나타내고 있다. 그리고 이러한 효과는 DOC섭취에 의한 부작용을 완화시키는 작용을 하였다. 또한 저항전분의 동물실험 겨로가는 혈당을 감소시키며, 혈액지방 수준을 감소시키는 효과를 나타내므로 저항전분의 섭취는 장긴\ulcorner 개선, 혈당유지, 혈액지방수준 개선의 효과가 있는 것으로 보인다.

  • Structures of Civil Engineering have been designed with a safety as their main purpose. However recently, the greenness side of the technology of civil engineering regarded as most Important and also utility values and stability of the structures. Also developments in the technology make materials to be higher strength and they shortened scales and stiffness of the structural members. This means that it brings an immoderate deflection and vibration of bridge structures simultaneously. Accordingly, this study ai ed to find and estimate the most idealized structure models on the effects of the deflection and vibration of bridges from the traffic lane load and human vibration - the main pass of bridge structures.

  • This paper attempts to evaluate driving stability by the vibration of human body. The purpose of this research is to establish an evaluation methodology for controling the quality of road surface. The study was conducted to investigate the relationship between road surface profiles (IRI, International Roughness Index) and the fatigue caused by the vibration of human body. Furthermore, 3-axis acceleration in driving vehicles was examined based on sampling sections under various road conditions. The acceleration value of frequency bands were analyzed by the characteristics of road surface, and realized the range of human influence by conditions and type of road surface with ISO-2631 standards. In general, more human fatigue by vehicle vibration was appeared in concrete pavements with high IRIs based on the analysis from given test data. Whereas, The SMA asphalt pavement and the diamond grinded concrete pavement reduce the human fatigue.

  • Intra-Body Communication (IBC) is a communication method using the human body as a communication medium. The fact that our human body consists of water and electrolyte allow such communication method could work and have strength in low-power. However, because the IBC directly affects to human body by using it as a medium, there was a lack of research in communication protocols of each communication layer. In this paper, we suggests MAC parameters which affects the performance of communication in human body channel, and propose new MAC protocol. Our results shows that our MAC is suitable for supporting high data rate applications with comparable radio duty cycle performance.

Effect of Epigallocatechin Gallate on Apoptosis in MDA-MB-231 Human Breast Cancer Cells (Epigallocatechin Gallate가 인체 유방암 세포인 MDA-MB-231의 세포사멸에 미치는 영향)

  • Among the numerous polyphenols isolated from green tea, epigallocatechin gallate (EGCG) is a predominate and is considered to be a major therapeutic agent. To elucidate the mechanical insights of anti-tumor effect, EGCG was applied to human breast cancer MDA-MB-231 cells. We investigated the effect of EGCG on protein and mRNA expression of proteins related to cell apoptosis in MDA-MB-231 human breast cancer cell lines. We also identified caspase-3 activity. We cultured MDA-MB-231 cells in the presence of 0, 5, 10, and $20\;{\mu}M$ of EGCG. Protein and mRNA expression of bcl-2 were decreased dose-dependently in cells treated with EGCG. However, protein and mRNA expression of bax were increased (p<0.05). Caspase-3 activities were increased dose-dependently in cells treated with EGCG. These results suggest that EGCG induces cell apoptosis by increase of caspase activity through decreasing of protein and mRNA expression of bcl-2 and increasing of protein and mRNA expression of bax.

  • Hydrofluoric acid is a less acidic substance than hydrochloric acid, nitric acid, and sulfuric acid, but it is one of the most dangerous substances for humans. In recent years, it has become an indispensable substance in industries such as chemical plants and the semiconductor industry, and although it is a threat to the human body, its use is increasing for various purposes, and the amount of use is constantly increasing due to the expansion and development of the industry. The dangers of hydrogen fluoride have been highlighted since the 2012 accident, which led to a more than fivefold increase in management standards for handling facilities. Hydrogen fluoride converts to hydrofluoric acid when exposed to the air, which can be fatal to humans. This study simulates the effects of a release of a toxic substance in the workplace, even though a protection layer has been provided to minimize the damage caused by the released toxic substance, and recommend ways to control the risk to workers in the event of a release in the workplace.

Effect of Cyanidin on Cell Motility and Invasion in MDA-MB-231 Human Breast Cancer Cells (Anthocyanin계 성분인 Cyanidin이 인체 유방암세포 MDA-MB-231의 이동성과 침윤성에 미치는 영향)

  • Anthocyanidins, the aglycones of anthocyanins, are natural colorants belonging to the flavonoid family. Cyanidin is one of the anthocyanidins, used for their antioxidant properties. Furthermore, previous studies have shown anthocyanidin-rich material extracts or aglycone form inhibit growth and induce apoptosis of cancer cells. But, Tumor metastasis is the most important cause of cancer death, and various treatment strategies have targeted on preventing the occurrence of metastasis. This study investigated the effects of cyanidin on metastasis processes, including motility, invasion and activity of MMP-2 and MMP-9 in MDA-MB-231 human breast cancer cell lines. We cultured MDA-MB-231 cells in presence of various concentrations 0, 5, 10 and 20 ${\mu}M$ of cyanidin. The cell motility was significantly decreased dosedependently in cells treated with cyanidin (p < 0.05) and cyanidin treatment caused the significant suppression of the invasion (p < 0.05). MMP-2 and MMP-9 activities, and MMP-9 mRNA express were not affected by anthocyanin treatment. In conclusion, cyanidin inhibits cell motility, invasion in MDA-MB-231 human breast cancer cell lines.

  • 산업화가 급속히 진행됨에 따라 삶의 질은 향상된 반면, 대기오염으로 인한 피해는 증가하고 있다. 대기 중 존재하는 휘발성 유기화합물 (VOCs)은 인체에 독성을 가지며 암을 유발하고, 오존의 전구물질(precursor), 광화학 스모그의 원인물질, 성층권의 오존층 파괴 및 지구온난화에도 영향을 미칠 뿐 아니라 (Derwent et al., 1996) 악취를 유발하기도 한다. 악취는 황화수소(hydrogen sulfide), 메르캅탄류(mercaptans), 아민류(amines) 등 기타 자극성 있는 기체상 물질이 사람의 후각을 자극하여 불쾌감과 혐오감을 주는 냄새로 정의되며, 주로 인체의 위해성보다는 정신적ㆍ심리적인 피해를 일으키는 감각공해라고 할 수 있다 (환경부, 2002).   • The purpose of this study is to show distribution pattern of comfortable Indoor-Temperature in South Korea based on 10-days normal data(June 3rd, July 2nd, August 1st). Comfortable Indoor-Temperature values and Heat flow equation values for the 68 stations are constructed to show how man tend to feel in various areas. In the early summer, comfortable Indoor-Temperature is very low in Taegwallong regions and the Southern interior regions due to the influence of the orographic effects or the altitude. In the hot summer, All regions except Taegwallong and Central interior regions have been the high value. These are largely derived from the low Clo-values due to the stable weather with the highest insolation, humidity and strong radiation heat by the North Pacific Anticyclone appears in Eastern Asia.

  • 배양한 치은섬유아세포와 3T3 세포에서, glycyrrhetinic acid가 cyclosporin A를 처리 세포의 활성에 미치는 영향을 알아보기 위해서 MTT 방법을 이용하여 측정하였다. Cyclosporin A는 $1{\sim}10ng/ml$의 농도에서 인체 치은 섬유아세포와 마우스 3T3 세포 활성을 증가시켰으며, glycyrrhetinic acid는 농도와 비례하게 마우스 3T3 세포의 성장을 억제시켰다. 특히 $25{\mu}g/ml$ 이상의 농도에서는 현저하게 억제시킴을 관찰할 수 있었다. 반면, 인체 치은 섬유아세포에서는 $50{\mu}g/ml$ 이상의 농도에서 조차도 성장을 억제 시키지 않았다. 또한, 마우스 3T3와 인체 치은 섬유아세포에서 일정한 cyclosporin A 농도에 $1-100{\mu}g/ml$의 다양한 농도로 glycyrrhetinic acid를 첨가하였을 때, cyclosporin A 단독으로 처리한 세포에 비하여 유의성 있게 세포의 활성을 억제시켰으며, 특히, $100{\mu}g/ml$의 glycyrrhetinic acid를 첨가하였을 경우 세포활성을 현저하게 억제시켰다. 3T3 세포에 cyclosporin A와 $50{\mu}g/ml$의 glycyrrhetinic acid를 함께 처리한 경우 glycyrrhetinic acid단독처리한 군에 비하여 세포활성에 첨가 효과를 보였으며, 인체 치은 섬유아세포 에서는 같이 처리한 경우 glycyrrhetinic acid단독 처리에 비하여 뚜렷한 억제 효과를 나타내었다. 이러한 상승효과는 1ng/ml 의 cyclosporin A와 $50{\mu}g/ml$ glycyrrhetinic acid를 같이 처리한 군에서 가장 뚜렷하게 관찰되었다. 이러한 결과로, 인체 치은 섬유아세포에 cyclosporin A와 glycyrrhetinic acid를 동에 처리하였을 경우 세포의 활성에 상승 효과가 있음을 알 수 있다.
