The Preventive and Therapeutic Effects of Probiotics in Allergic Diseases Via Immune Modulation (프로바이오틱스의 면역조절을 통한 알레르기 예방 및 치료효과)

  • The demand to develop more safe and efficient methods for treating allergic patient is now continuously growing due to the increasing prevalence of allergic diseases. Probiotics are endogenous microbial flora that gives health benefits within hosts. Probiotics are now considered as one of solutions to treat allergic patients since recent evidence shows that some of probiotics have immunomodulatory function. Also, the treatment of probiotics to patients is relatively safer than other anti-inflammatory agents. In this review, we summarized on immunomodulatory function of some probiotics which show preventive or therapeutic effects on major allergic diseases such as atopic dermatitis, allergic rhinitis, asthma, or food allergy. Based on previous literature, the treatment of probiotics can alleviate the symptoms of allergic diseases via balancing $Th_1/Th_2$ response or increasing the number of regulatory T ($T_{reg}$) cells.

Neuroprotective Effects of Cheongnoemyeongsin-hwan against Hydrogen Peroxide-induced DNA Damage and Apoptosis in Human Neuronal-Derived SH-SY5Y Cells (인체 신경세포에서 청뇌명신환(淸腦明神丸)의 산화적 스트레스에 대한 세포보호 효과)

  • Objectives : Oxidative stress due to excessive accumulation of reactive oxygen species (ROS) is one of the risk factors for the development of several chronic diseases, including neurodegenerative diseases. Methods : In the present study, we investigated the protective effects of cheongnoemyeongsin-hwan (CNMSH) against oxidative stress‑induced cellular damage and elucidated the underlying mechanisms in neuronal-derived SH-SY5Y cells. Results : Our results revealed that treatment with CNMSH prior to hydrogen peroxide (H2O2) exposure significantly increased the SH-SY5Y cell viability, indicating that the exposure of the SH-SY5Y cells to CNMSH conferred a protective effect against oxidative stress. CNMSH also effectively attenuated H2O2‑induced comet tail formation, and decreased the phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V‑positive cells. In addition, CNMSH exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential (MMP) loss that were induced by H2O2, suggesting that CNMSH prevents H2O2‑induced DNA damage and cell apoptosis. Moreover, H2O2 enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase, a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with CNMSH. Furthermore, CNMSH increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, CNMSH is able to protect SH-SY5Y cells from H2O2-induced apoptosis throughout blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating Nrf2/HO-1 signaling pathway. Conclusions : Therefore, we believed that CNMSH may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.

Protective Effect of Flavonoids on Lymphocyte DNA Damage Using Comet Assay (Comet Assay를 이용한 Flavonoids와 항산화 비타민의 인체임파구 세포 DNA 손상 보호 효과)

  • The present study was attempted to investigate and compare the antioxidant potency of several well-know flavonoids, antioxidant vitamin and commercially available popular beverages. The antioxidant potency was assessed by the effect on reducing oxidative DNA damage of human lymphocytes. Cellular oxidative DNA damage was measured by SCGE (single-cell gel electrophoresis), also known as comet assay. Lymphocytes were pre-treated for 30 minutes with wide ranges of doses of apigenin, kaempferol, luteolin, myricetin, rutin, quercetin, $\alpha$-tocopherol (10,25,50,100,200,500,1000 $\mu$M) ,green tea extract or grape juice (10,50,100,250,500,1000 $\mu$g/mL) followed by a $H_2O$$_2$(100 $\mu$M) treatment for 5 min as an oxidative stimulus. The physiological function of each antioxidant substance on oxidative DNA damage was analyzed as tail moment (tail length $\times$ percentage migrated DNA in tail) and expressed as relative DNA damage score after adjusting by the level of control treatment. Cells treated with $H_2O$$_2$alone (positive control) had an extensive DNA damage compared with cells treated with phosphate buffered saline (PBS, negative control) or pre-treated with all the tested samples. Of all the six flavonoids, quercetin was the most potent antioxidant showing the lowest $ED_{50}$/ of 8.5 $\mu$g/mL (concentration to produce 50% protection of relative DNA damage). The antoxidant potency of individual flavonoids were ranked as follows in a decreasing order; luteolin (18.4 $\mu$g/mL), myricetin (19.0 $\mu$g/mL) , rutin (22.2 $\mu$g/mL) , apigenin (24,3 $\mu$g/mL) , kaempferol (25.5 $\mu$g/mL). The protective effect of $\alpha$-tocopherol was substantially lower (highest $ED_{50}$value of 55.0 $\mu$g/mL) than all the other flavonoids, while the protective effect was highest in green tea and grape juice with low ED5O value of 7.6 and 5.3, respectively. These results suggest that flavonoids, especially quercetin, and natural compounds from food product, green tea and grape juice, produced powerful anti-oxidative activities, even stronger than $\alpha$-tocopherol. Taken together, supplementation of antioxidants to lymphocytes followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species.

The Study on Wound Healing in Rabbit Skins by Low-intensity Laser Irradiation (저강도 레이저 조사에 의한 가토 피부의 상처 치유에 관한 연구)

  • The skin is an organ that has many important roles, including protection against infection, regulation of temperature and fluid loss, and sensory function. Injury to the skin, wound repair normally involves: (1) balanced activity of inflammation, (2) the re-epithelial phase and (3) the matrix formation of remodeling phase. Thus, skin wound healing is a finely controlled biological process involving a series of complex cellular interactions. Laser therapy is being implemented with increasing frequency in medicine. Low intensity laser is one that is capable of producing an energy density so low that any biologic alterations are the result of direct irradiation effect, not thermal events. This study was designed to evaluate the efficacy of low intensity laser therapy on skin wound healing in rabbits. A total of 10 male rabbits (New Zealand White Rabbit), age 8 weeks were used. Skin wound were surgically created dorso-lateral on the flank of 10 rabbits (2$\times$2 cm/damage areas). The experimental animals were treated with 5Hz (830 nm wave length) low-intensity laser (MILTA-01 Model) daily for 10 min (1.6 J/$cm^2$) for 12 days. Control animals were sham treated with the laser head. Laser irradiation animals showed a complete remodeling of the epithelial layer, a positive repair of connective tissues, and enhanced the wound closure rate over time as compared to the control animals. Especially, laser irradiation groups improved fibroblast activity, cellular content, granulation tissue formation, and collagen deposition which is resulted in improving the tensile strength of the wound. These findings suggest that laser photostimulation could accelerate healing of open wound in rabbits, and may be benefit in the treatment of open wound, including decubitis ulcers.

  • A grounding system in a large-scale apartment complex consists of a TT grounding system when viewed in each building unit, but the characteristics of TN grounding system appears when viewed in the total ground system function. Therefore, this study examined the characteristics of a grounding system in a large-scale apartment complex using design draft, field measurements and PSCAD/EMTDC modeling. This paper proposes an algorithm for fault analysis depending on the grounding system structure in large-scale apartment complexes considering the connection of the underground culvert as well as a design algorithm for each grounding system considering the contact voltage and step voltage. The simulation results based on the proposed modeling and algorithm confirmed that it is useful for analyzing the characteristics of a grounding system in a large-scale apartment complex.

  • Firefighters performing disaster response activities such as Fire Suppression Rescue First Aid in fire are being exposed in various adverse factors, heat, smoke, toxic gases, emotional stress, biological toxic factors and physical overload by unnatural ergonomic posture required for firefighters. But so far, there is the study for this problem only separately. There is no study about comprehesive analysis of exposed adverse factors in fire-related disaster response activities and countermeasures. The purpose of this study is to contiribute to solving the health problems and prevention of accidents of firefighters by extracting hazardous agents in disaster such as fire and by proposing countermeasures. After analyzing circumstances such as fire-suppression, rescue first aid and life-environment, exposure factors of fire are derived and exposure status is suggested according to physical chemical biological psychological aspects. The countermeasure against the noise of the physical exposure factors are proposed. The countermeasures such as protective equipment and clean room in chemical factors, infection prevention education, vaccination and periodic check system in biological factors, PTSD alleviation booth and mentoring in psychological factors are proposed.

Effects of carbendazim on DNA, gene and chromosome (살균제 carbendazim이 DNA, 유전자 및 염색체에 미치는 영향)

  • Benzimidazole pesticide carbendazim that is effective against a wide range of fungal plant pathogens is a protective, eradicant, and systemic fungicide. For genetic toxicity evaluation of carbendazim on DNA, genes and chromosome, were investigated with chromosome aberration, bacterial reverse mutation, micronucleus test in mouse born marrow and DNA damage assay by single cell microgel electrophoresis. Substitution and frameshift mutation were not induce at variable concentration of carbendazim on Ames test with or without rat liver microsomal activation. For the result of chromosome aberration test, numerical changes of chromosome were detected at the concentrations higher than $4.0{\mu}g/m{\ell}$, but structural aberration was not induced. Positive control, Mitomycin-C and captafol made a structural aberration, but numerical change of chromosome did not appear. In the micronucleus test for mouse born marrow, carbendazim was negative, but was weak positive in DNA damage assay by single cell microgel electrophoresis because of increased DNA moving length of 20% to control.

The Effect of Judo Training on Body Composition, Blood lipids and Immunity of Security Majoring Obese University Students (유도훈련이 경호전공 비만대학생들의 신체구성, 혈중지질 및 면역력에 미치는 영향)

  • College students majoring in bodyguards learn and acquire bodyguard martial arts to protect the life and property of the subjects. Also, the bodyguard martial arts are helping to improve and maintain the body and spirit of the bodyguards. However, more and more students majoring in security at universities are becoming obese due to lack of time to participate in bodyguard training and excessive food intake, which could have a negative impact on their future security work as well as their health condition. In this study, the effect of 10 weeks of Judo training on body composition, blood lipid and immune function on Security Majoring obese male college students was determined to be as follows. 1. Weight(p<.05), ; body fat ratio(p<.05), BMI(p<.01) etc. of body composition after Judo training were significantly reduced, and in the case of BMI(F=4.488, p<05), there were significant differences in interaction between groups and timing. 2. After judo training, TC (p<.05) and LDL-C(p<.01) of the blood lipid showed a significant decrease. 3. After judo training, lymphocytes of leukocytes were shown to increase significantly(p<.05), lymphocytes(F=11.772, p<.01) showed significant differences among groups. 4. After judo training, IgA and IgG of immunoglobulin were shown to increase significantly (p<.05), IgA(F=7.646, p<.05) and IgG (F=14.267, p<.01) showed significant differences among groups. Considering the above results, it is thought that judo training can prevent obesity due to reduced body fat and reduce neutral fat and cholesterol, thus preventing arteriosclerotic diseases. In addition, it is expected to play a positive role in defense against human bacterial infections, inflammatory reactions, and external antigen in immune function. It is thought that this will not only prevent the bodyguards' obesity but also maintain a healthy life and improve the security work efficiency. However, some studies have shown results that are somewhat different from the results of this study, so studies on blood lipids, immune function, and exercise will require more detailed research on various methods of exercise, such as exercise time and intensity.

Induction of Apoptosis by Citri Pericarpium Methanol Extract through Reactive Oxygen Species Generation in U937 Human Leukemia Cells (진피 메탄올 추출물의 활성산소종 생성을 통한 인체 백혈병 세포의 apoptosis 유발)

  • Citri Pericarpium is one of the most commonly used traditional herbal medicines in Korea, China, and Japan. Its extracts have many properties including the treatment of indigestion and inflammatory respiratory syndromes such as bronchitis and asthma. However, the underlying molecular mechanisms of anti-cancer activity and molecular targets are not fully understood. In this work, we investigated the anti-proliferative activity of Citri Pericapium (EMCP) methanol extract on reactive oxygen species (ROS) production and the association of these effects with apoptotic cell death using U937 human leukemia cells in vitro. EMCP treatment decreased cell proliferation in a dose-dependent manner following an increase of the sub-G1 phase, the down-regulation of Bax proteins, the activation of caspases, the degradation of poly (ADP-ribose) polymerase proteins (PARP), and the induction of ROS generation. However, the quenching of ROS generation by N-acetyl-L-cysteine administration, a scavenger of ROS, reversed the EMCP-induced apoptosis effects. In addition, heme oxygenase-1 expression also recovered by inhibiting the nuclear translocation of phosphorylated NF-E2-related factor 2. Taken together, our data indicate that ROS are involved as key mediators in the early molecular events in the EMCP-induced apoptotic pathway.

The Application of Nanoliposome Composed of Ceramide as an Anti-irritant in Cosmetics (세라마이드를 구성성분으로 하는 나노리포좀의 응용 - 화장품에서의 자극완화제)

  • The objective of this study is to suggest the potentialities of nanoliposome composed of ceramide as an anti-irritant against various irritants used in cosmetics. Ceramides are major structural components of the epidermal permeability barrier, which is known to play an essential part in human physiology by not only preventing the loss of water from the body but also protecting the body from external physical, chemical, and microbial insults. According to the results, better effects on reinforcement of skin barrier function and anti-irritation were obtained with nanoliposome composed of ceramide than with dispersed ceramide. And, we performed in vitro skin penetration test using horizontal Franz diffusion cells with skin membrane prepared from hairless mouse to evaluate the influence of nanoliposome composed of ceramide on the skin penetration of lactic acid in formulations. From the results, we found that the anti-irritation effects of nanoliposome containing ceramide were due to reduced penetration rate of irritants. Conclusively, we could develop a new anti-irritation system and apply this nanoliposome composed of ceramide to the final cosmetic products successfully.
