Optimization of Submerged (Ginseng Root Culture Conditions for the Production of Saponin (사포닌 생산을 위한 인삼 root 액체배양조건의 최적화)

  • This study was carried out to determine the optimal liquid medium composition of ginseng (Panax ginsengC.A. Meyer) root induced by growth regulators in order to improve the yield of saponin production. Submerged culture conditions were optimized using the fractional factorial design with 4 factors and 3 levels by a RSM computer program The effects of various pH values of medium, sucrose, nitrogen and phosphate concentration on the saponin content of the ginseng root were investigated. The optimum phosphate concentration determined by a partial differentiation of the model equation, pH of medium, sucrose and nitrogen concentration were phosphate 93 mg/L, pH 5.5, sucrose 5% and nitrogen 50 mg/L, respectively. Under these conditions, the predicted saponin content of ginseng root was estimated at 0.308%.

The Effect of Ginseng Saponin Fraction on Antioxidant Activity of $\alpha$-Tocopherol ($\alpha$-tocopherol의 항산화작용에 미치는 인삼사포닌분획의 영향)

  • The effect of the saponin fraction extracted from Panax ginseng C.A. Meyer on the antioxidant activity of $\alpha$-tocopherol was investigated in vitro as well as in vivo. Microsomal preparation of rat(Winter, 180-200g) liver was incubated in the mixture containing NADPH, $Fe^{3+}$, ATP, $\alpha$-tocopherol with and/or without ginseng saponin fraction for 30 minutes and the malondialdehyde formed was assayed and found that the saponin fraction stimulated the antioxidant activity of $\alpha$-tocopherol cooperatively. It was also realized that the cooperative stimulation of the antioxidant activity of $\alpha$-tocopherol was most eminent when the concentration of the saponin fraction was around $10^{-5}$% in the reaction mixture. Alcoholic suspension of $\alpha$-tocopherol with and f or without ginseng saponin fraction was administered orally to rats in which the lipid preoccupation was induced by ethanol administration and the lipid peroxide contents of the liver were assayed at certain periods of time after $\alpha$-tocopherol administration in this animal. From the previous work and present experimental results, it seemed that the saponin fraction accelerated the absorption of $\alpha$-tocopherol and therefore stimulated the antioxidant activity of $\alpha$-tocopherol more effectively in the animal body.

  • Effect of saponin fraction of ginseng C.A. Meyer on blood serum lipoprotein distribution of high cholesterol fed rabbits for two to four weeks was investigated. A. significant increase of very low density lipoprotein(VLDL) and low density lipoprotein(LDL) occured while high density lipoprotein(HDL) were decreased in the blood of both groups which were fed high cholesterol diet with (test group) and/or without ginseng saponin(control group) for 2-4 weeks. However the degrees of VLDL and LDL increase and HDL decrease were relatively small in ginseng administered group compared with control group. This suggests that the hypocholesterolemic action of the saponin might be brought about by decreasing the raised VLDL and LDL level and increasing the lowered HDL level.

  • The present study was undertaken to determine the inhibitory effects of orally administered ginseng saponins(SP), protopanaxadiol saponins (PD), and protopanaxatriol saponins(PT) on the development of morphine-induced tolerance and physical dependence in mice. The study also sought to determine the hepatic glutathione contents. which are closely related to the degree of detoxification of mine the effects of GS on morphine 6-dehydrogenase, which catalyzes the production of morphinone from morphine, and the roles of spinal descendign inhibitory systems in the production of antagonism. The results of the present study showed that GS, PD and PT administered orally inhibited the development of morphine induced tolerance and dependence. GS. PD and PT inhibited the reduction of hepatic glutathione concentration in mice treated chronically with morphine and the activity of morphine 6-dehydrogenase, and the activation of spinal descending inhibitory systems was inhibited by GS. So we hypothesized that the results were partially due to the dual action of the test drugs, the inhibition of morphinone production and the activated formation of morphinone-glutathinone conjugation, and the inhibition of the activatin of apinal descending inhibitory systems and the others.

Changes of Saponin and β-Glucan Content on the Cultured Ginseng with Mushroom Mycelia (버섯균사체로 배양된 인삼 Saponin과 β-Glucan 함량 변화)

  • This study investigated the changes of saponin and $\beta$-glucan content on the cultured ginseng with mushroom mycelia of Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE). Cultured ginsengs with mushroom mycelia were extracted with 80% ethanol, fractionated with n-buthanol, and analysed for ginsenosides by high performance liquid chromatography (HPLC). Crude saponin content of raw ginseng was 4.11% (d.b) but cultured ginseng with mushroom mycelia of PL, GL, and HE were increased to 6.74, 6.77 and 6.23% (d.b), respectively. Ginsenoside-Rd, among the 12 ginsenosides which were available for analysis, was remarkably increased to 13.61, 24.26, and 32.69 mg/g, respectively (raw ginseng: 0.80 mg/g). The $\beta$-glucan content of cultured ginseng with mushroom mycelia of PL, GL, and HE were decreased to 8.85, 5.51 and 5.46% rather than mushroom mycelia of 29.14, 19.44, and 23.39% (d.b), respectively.

Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples. Inhibition Effect of Ginseng Saponin on the Growth of Citrobacterer sp. Isolated from Contaminated Ginseng (오염된 인삼으로부터 분리된 Citrobacter sp.에 대한 인삼사포닌의 생육억제 효과)

  • A bacterium isolated from contaminated white ginseng was identified using API kit and electron microscope. This isolate was determined as rod shaped bacterium having about 1.0 ${\mu}m$ in diameter and 2.0 to 6.0 ${\mu}m$ in length. It had motility by peritrichous flagellum. The isolate had ${\beta}-galactosidase$, arginine dihydrolase and ornithin decarboxylase. It did not have ability not only to use citrate as sole carbon source and but also to produce $H_2S$. However, it could ferment glucose, manitol, sorbitol, rhamnose, arabinose and amygdalin. From these obserbations, the isolate was identified as Citrobacter sp. Ginseng saponin was added to culture of Citrobacter sp. in order to investigate saponin's influence on its growth. The strain was incubated at $38^{\circ}C$ for 3 days after addition of 0.05, 0.5, 2.0 and 4.0% (w/v) of saponin, respectively and the growth rates was investigated. The relative bacterial growth inhibition rates showed 28.6, 66.7, 92.4 and 97.7%, respectively, when compared with saponin non-treated group. These results suggest that the growth of Citrobacter sp. is inhibited by saponin in a concentration-dependent manner.

Saponin and Ginsenoside Content in Korean Red Ginseng Products (고려홍삼 제품류의 사포닌 및 진세노사이드 함량)

  • Red ginseng products manufactured by the Korea Ginseng and Tobacco Corporation were analyzed to determine the crude saponin , total saponin and ginsenoside contentents by gravimetry, spertrometry and HPLC, respectively, to see if effective quality control of the components in the products can be achieved. Medicinal powders, powders, tablets and capsules which were made from ginseng powder showed similarity in saponin content, the ratio of PD to PT saponin, and the ginsenoside content and composition, while extract powder, extract, extract tea, extract pills and tea, which were made of ginseng extract, showed difference in saponin content, ratio of PD to PT saponin, and the content and composition of ginsengside. It is, accordingly, believed that ginseng products which are uniform in contents and saponin composition can be produced by carrying out strict quality control throughout the processes of making raw red ginseng into final products.
