Anti-oxidative Activities of Extracts from Chionanthus retusus leaves, Fruits and FLower (이팝나무 잎과 열매, 꽃 (Chionanthus retusus) 추출물의 항산화 효과)

  • The aim of this research was to investigate the anti-oxidantive activities of extracts from Chionanthus retusus leaves, fruits and flower. The content of phenolics were 20.8 mg/g in water extracts and 32.2 mg/g in 90% ethanol extracts from flower. The DPPH free radical scavenging activity of extracts from Chionanthus retusus was above 70% at phenolic concentration $100{\mu}g/mL$. The ABTS radical decolorization activities of water and ethanol extracts were both above 80% at $100{\mu}g/mL$ phenolic concentration, respectively. The antioxidant protection factor(PF) of water and 80% ethanol extracts from leaves was the highest as 2.27 PF and 1.70 PF at $50{\mu}g/mL$ phenolic concentration. The TBARs inhibition rate of the Chionanthus retusus extracts, was above 70% at $200{\mu}g/mL$ phenolic concentration. The anti-oxidantive activities of extracts from leaves were shown more active than BHT as a positive control except TBARs. These results confirmed that the extracts from Chionanthus retusus leaves, fruits and flower was shown the high anti-oxidant activity. The results can be expected isolated phenolic compounds from Chionanthus retusus to use as functional beauty food resource.

Assessment of physical condition of old large Chionanthus retusus(Chinese Fringe Tree) using structural stability analysis (천연기념물 이팝나무 노거수 구조안정성 진단을 통한 물리적 생육상태 평가)

  • Decay or large cavities inside trees are the main causes of trees overturning and broken branches, and structurally weakened trees are more vulnerable to strong winds and heavy snowfall. Recently, as strong winds and typhoons increase due to climate change, the damage to human life and property due to trees overturning continues to increase, and cultural assets are in a similar situation. In particular, old big trees are structurally vulnerable to external shocks such as strong winds and heavy snowfall. This study was aimed at providing a scientific basis for preventive protection measures by conducting a structural stability diagnosis of seven retusa fringe trees designated as natural monuments. For the structural stability diagnosis, tree risk assessment and internal tree defect measurements were performed. As a result of the tree risk assessment, the Retusa Fringe Trees in Sinjeon-ri, Yangsan and Gwangyangeupsu had the highest risk of broken branches due to weak branch attachment strength. As a result of the diagnosis of internal defects of cross sections of measured trees, there were suspected cavities or severe decay in all except two trees of the population of Retusa Fringe Trees in Pyeongji-ri. Natural disasters due to climate change are increasing, and the scale is getting larger, so it is very important to preemptively manage large old trees through scientific structural safety diagnosis to manage trees that are vulnerable to environmental changes.

First Report of Rust Disease on Fringe Tree by Puccinia sp. and Its Alternative Host (Puccinia sp.에 의한 이팝나무 잎녹병 발생 및 중간기주 보고)

  • In July 2018, a serious rust symptom was found throughout the fringe trees planted in Gangjin-gun, Korea. Yellow and brown spots were observed on the adaxial (topside) surface of the collected fringe tree leaves, and yellow color aecia were observed on the abaxial (underside) surface leaves. The size of aeciospore and urediniospores of JCK-KCFR1 strain were measured to 41.2 ㎛ (Φ) and 28.84 ㎛ (Φ) with a light microscope. Phylogenetic analysis of the small subunit rRNA, internal transcribed spacer, and large subunit rRNA region indicated that JCK-KCFR1 strain is novel species of the genus Puccinia and closely related to Puccinia kusanoi, which has been reported a rust pathogen on bamboo. In May 2019, rust symptoms were also discovered on the bamboo leaves planted around the fringe tree on Muwisa-ro, and their telia and teliospores were observed on the abaxial leaf surfaces of the bamboo with 100% sequence homology with the rust of the fringe tree. This is the first report that Puccinia sp. JCK-KCFR1 is a new species that requires both primary (fringe tree) and alternative (bamboo) host plants to complete its life cycle in Korea.

이팝나무의 항산화 및 항균활성

  • 이팝나무(Chionanthus retusus)는 물푸레나무과에 속하는 낙엽교목으로 육도목, 유소수라고도하며 중국이나 일본에서는 잎을 차대용으로 사용한다고 하여 다엽수라고도 부른다. 지사, 건위, 중풍 등의 치료에 효과적이고, 강장제, 해열제, 이뇨제 등으로 사용되었다고 알려져 있는 이팝나무 잎의 생리활성을 검색하여 유용자원으로 활용하기 위한 기초 연구로 메탄을 추출물을 각종 용매를 이용하여 분획한 후 DPPH를 이용한 항산화 활성, 아질산염 소거 및 항균 활성 등의 생리활성에 대한 실험 결과는 다음과 같다. DPPH에 의한 항산화 활성을 측정한 결과 에틸 아세테이트 분획물과 부탄올 분획물에서 가장 높은 활성을 나타내었으며, reducing power의 경우 에틸 아세테이트 분획물에서 그리고 농도가 증가함에 따라 환원력이 증가하는 것으로 나타났다. 아질산염 소거능은 pH가 낮을수록, 농도 의존적으로 그 활성이 높게 나타났다. 항균활성은 에틸 아세테이트 분획물에 대하여 특히, Escherichia coli와 Bacillus cereus 균주에 대해 각각 16 및 18mm 로 높은 항균력을 나타내었다.

  • The morphological characteristics of Chionanthus retusus Lindl. & Paxton in Korea are reviewed in this study through the comparison with its previous records and a distribution map of this species is created using previous flora and voucher specimens. The leaf epidermal micromorphology and the petiole anatomy are also briefly described.

Isolation and Identification of Triterpenoids and Sterols from the Flowers of Chionanthus retusus Lindl. & Paxton (이팝나무(Chionanthus retusus Lindl. & Paxton) 꽃으로부터 Triterpenoid 및 Sterol 화합물의 분리 및 동정)

  • The flowers of Chionanthus retusus Lindl. & Paxton were extracted with 80% aqueous MeOH at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAc fraction, two triterpenoids and two sterols were isolated using the repeated silica gel ($SiO_2$) and octadecyl $SiO_2$ (ODS) column chromatographies. According to the results of physico-chemical and spectroscopic data including nuclear magnetic resonance, mass spectrometry, and infrared. The chemical structures of the compounds were respectively determined as ursolic acid (1), corosolic acid (2), ${\beta}$-sitosterol (3), and daucosterol (4). All the compounds were isolated for the first time from the flowers of Chionanthus retusus Lindl. & Paxton.

Analysis on the Growth Environment of Chionanthus retusus Community at the Wansanchielbong in Jeonju (전주 완산칠봉 이팝나무 자생지의 생육환경으로 본 자연유산 가치 분석)

  • This study analyzed the distribution, structure and environmental condition of the vegetation of the Chionanthus retusus Lindly et Paxton community at the Wansanchielbong in the Jeonju city to offer basic data for sustainable conservation and ecological management system. And the results are as follows; 1. The average pH of soil at the community was pH 5.69 and it was slightly higher than the average of forest soil pH of Korea. But if the degree of pH will be down, it will be needed some more fertilization of Calcium. 2. The total average for contents of organism was 4.98%. And the nitrate - nitrogen content(mg/kg) of A, B, C, D quadrat was 20.29%, 28.87%, 7.65%, and 23.3% respectively. And there were good condition except quadrat C which was contaminated by amount of earth and sand. 3. The flora of the Chionanthus retusus Lindly et Paxton community was listed as 60 taxa; 37 families, 50 genera, 47 species, 10 varieties and 3 forms. The average appearance species of each Quadrat were A sector 30, B sector 26, C sector 19 and D 19 taxa respectively. 4. Surveyed woody plants in the community were as follows : Chionanthus retusus, Zelkova serrata, Quercus variabilis, Cornus walteri, Robinia pseudo-acacia and those were mixed status. And Chionanthus retusus, Zelkova serrata, Robinia pseudo-acacia, Albizzia julibrisin, Cudrania tricuspidata, Symplocos chinensis for. pilosa were mixed in mid layer trees. Herbaceous plants were founded such as Chionanthus retusus, Zelkova serrata, Robinia pseudo-acacia, Grewia parviflora, Rosa multiflora, Trachelospermum asiaticum was dominant with 35~64% in the ground cover, and Commelina communis, Calamagrostis arundinacea, Dryopteris bissetiana, Lilium lancifolium were founded also. 5. The importance values of Chionanthus retusus was 40.2% in the quadrat A1, 50.2% at quadrat A, 50.0% B1, 45.2% B2, 22.4% C1, 73.6% C2, 33.2% D1 and the total average of I.V. was 44.9%. 6. The average height of surveyed Chionanthus retusus was 5.7m and the average DBH was 12.4cm. The number of trees higher than 2m were 107 and the number of trees lower than 2m were 63. The total numbers of Chionanthus retusus were 170. 7. The age of surveyed Chionanthus retusus were analyzed 42 thru 87 years old and that of Zelkova serrata were 42, Quercus variabilis were 60, Quercus aliena were 48, Robinia pseudo-acacia were 40. 8. The number of trees with DBH 40 through 50cm were 6, and that of 30~39cm were 3, and that of 20~29cm were 16, so the total number that was over 20cm was 25. And there were 70 trees under 10cm of DBH and 63 seedlings. It will be very important data to conserve the habitat that the structure and environmental condition of the Chionanthus retusus Lindly et Paxton community at the Wansanchielbong was stable, and sustainable monitoring will be needed. Now that community is nurse forest of Jeonju City but more positive preservation plan will be needed and assigning monument of city or province also be necessary.

Antioxidant and anti-aging activities of water extracts from Chionanthus retusus flesh according to different extraction temperatures (추출온도에 따른 이팝나무 과육 물 추출물의 항산화 및 항노화 활성)

  • The fringe tree, Chionanthus retusus (Oleaceae) has been used as a natural remedy that helps a paralytic stroke, dementia, phlegm and malaria. This study was conducted to investigate the antioxidant and anti-aging activities of water extracts obtained from the flesh of C. retusus by reflux extraction at room temperature and $80^{\circ}C$, and pressure heating extraction at $110^{\circ}C$, for an increment of their practical use. The highest extraction yield from the flesh of C. retusus was 67.94% extract by pressure heating extraction at $110^{\circ}C$. The content of total polyphenol compounds was the highest by 72.71 mg/g in $80^{\circ}C$ extract by reflux extraction. The highest content of total flavonoid compounds was 7.60 mg/g at $110^{\circ}C$ extract. The results of soluble protein and reducing sugar contents showed the highest content of 4.93 mg/g and 46.77 mg/g in $110^{\circ}C$ extract, respectively. In the analysis of DPPH radical scavenging activity, all extracts showed an excellent effect of 92% in 1.0 mg/mL concentration. The highest effect of ABTS radical scavenging activity was 88.67% at $110^{\circ}C$ extraction in 1.0 mg/mL concentration. Nitrite scavenging ability was the highest as 66.16% at $110^{\circ}C$ extract at pH 1.2 and 1.0 mg/mL concentration. The results of SOD-like activity and xanthine oxidase inhibition activities showed the highest effect of 14.95% and 59.45% at $110^{\circ}C$ extract in 1.0 mg/mL, respectively. Tyrosinase inhibition activity for whitening effect was the highest of 35.31% at $85^{\circ}C$ in 1.0 mg/mL. In the analysis of anti-aging effect, collagenase and elastase inhibition activity showed the highest effects of 37.78% at $110^{\circ}C$ and 20.39% at $80^{\circ}C$, respectively. All results of antioxidant activities and anti-aging effects increased along with the concentration increases. These results indicated that the extracts extracted from the flesh of C. retusus at $80^{\circ}C$ and $110^{\circ}C$ have a large amount of useful ingredients, an excellent antioxidant activity, like as DPPH and ABTS radical scavenging activity, and anti-aging effects to develop functional products than those of $25^{\circ}C$.
