Inhibitory Effects of Flavonoids Isolated from the Leaves of Stewartia koreana on Nitric-oxide Production in LPS-stimulated RAW 264.7 Cells (노각나무 잎에서 분리된 플라보노이드에 의한 대식세포에서 산화질소 생성 억제효과)

  • Five phenolic compounds were isolated from the ethyl acetate fraction of leaves from Stewartia koreana, and their nitric-oxide (NO) inhibitory activities were measured to identify the major active constituents responsible for the efficacy of the extract against inflammatory reactions. These five compounds were quercetin (1), quercitrin (2), hyperin (3), quercetin-3-O-(6"-O-galloyl)-${\beta}$-D-galactopyranoside (4), and kaempferol 3-O-[2",6"-di-O-(trans-p-coumaroyl)]-${\beta}$-D-glucopyranoside (5). Among the separated compounds in the EtOAc fraction, compounds 4 and 5 were isolated for the first time, and no study has yet reported their anti-inflammatory effects. The compounds were identified by spectroscopic analysis, and the isolated compounds showed significant NO inhibitory effects in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Compound 5 showed the most potent inhibitory effect (63.35% inhibition) against LPS-induced NO production compared to that of compound 1 (17.17%), compound 2 (5.0%), compound 3 (3.92%), and compound 4 (6.32%) at $10{\mu}g/ml$ concentration. NO production was inhibited by suppressing the protein expression of inducible nitric-oxide synthase in LPS-stimulated RAW 264.7 macrophages. These results indicate that kaempferol 3-O-[2",6"-di-O-(trans-p-coumaroyl)]-${\beta}$-D-glucopyranoside might be the major active compound responsible for the anti-inflammatory effects of S. koreana.

Anti-inflammatory and Anti-allergic Effects of Phlorofucofuroeckol A and Dieckol Isolated from Ecklonia cava (감태에서 분리된 플로로탄닌의 염증 및 알러지 억제 효과)

  • The anti-inflammatory effect of brown alga Ecklonia cava is well known, and several phlorotannins have also been reported. In this study, major active components for anti-allergy and anti-inflammation were identified by NMR and MS analysis, and the levels of effectiveness were compared. Six major phlorotannins-phloroglucinol, eckol, eckstolonol, triphlorethol-A, phlorofucofuroeckol A, and dieckol-were isolated from the ethyl acetate fraction of E. cava. In order to analyze the major active substances in E. cava, antioxidant, anti-inflammatory, and anti-allergic effects were evaluated for the six separate substances. Antioxidant capacities of each phlorotannin were evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals, where phlorofucofuroeckol A and triphlorethol-A had the highest radical scavenging capacity in respective radical scavenging assays. Phlorofucofuroeckol A exhibited the highest inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells among phlorotannins tested. Dieckol inhibited the release of ${\beta}-hexosaminidase$, a marker for the release of histamine in mast cells, in a dose-dependent manner in antigen-stimulated RBL-2H3 mast cells. Additionally, no cytotoxicities were observed at 1 and $2{\mu}g/ml$ in both phlorofucofuroeckol A and dieckol. These results suggest that phlorofucofuroeckol A and dieckol may play a key role in allergic inflammatory reactions.

Study on Quantitative Growth Analysis in Yam(Dioscorea spp.) (마의 생장해석(生長解析)에 관(關)한 연구(硏究))

  • This study was conducted to obtain the basic information on the production of high yield and qualitied yam in Andong, major yam-producing district, in Kyongbuk province, using three major Yam cultivars, such as Danma, Jangma and Alata (round type). Alata showed the longest tuber width while Jangma showed the longest tube length. Danma showed the highest tube yield at 2,123 kg/10a. Tubes of Alata had higher dry ratio than those of other cultivars, suggesting that Alata could be used as good processing materials. Characteristics of top parts, such as vine length, leaf number, and lateral vine number showed typical sigmoid curves. Tuber characteristics such as length, width and fresh weight of tuber were linearly increased as the growth period was progressed. Relative growth rate (RGR) for new tuber and stem reached the highest value at 86 DAT (days after transplanting). There were similar tendencies among the cultivars in net assimilation rate (NAR). The highest crop growth rate (CGR) was appeared at 156 DAT regardless of cultivars. In the changes of leaf area ratio (LAR), there were significant differences among three cultivars. LAR were decreased at 86, 106 and 136 DAT for Alata, Danma and Jangma, respectively. However, specific leaf weight (SLW) was greatly increased at 176 DAT, and leaf area index (LAI) was decreased at 156 DAT in all cultivars.

  • The Benangkimi wetland, which serves as a stopover place for migratory birds in the Dadohae Marine National Park in Wando-gun, Jeollanam-do, is a place where various species and a large number of migratory birds are observed. In order to provide an effective and scientific management plan for the habitat environment of migratory birds, we carried out field survey of Benangkimi wetland. The field survey and survey on habitat use of migratory birds were conducted in parallel to obtain the basic data of hydraulics and hydrology. The hydraulic and hydrological survey of Benangkimi wetland was conducted 1-2 times a year during the period of 2015-2016 (two years). It was classified into 4 sectors according to the topography and geology, rainfall, hydraulic characteristics, and wetland conditions. The 3 sectors same as the migratory birds survey sector and the 1 sector as the inflow of the wetland were analyzed. The survey also focused on small migratory birds arriving at Benangkimi wetland during the spring and autumn season. As a result of investigating the population and species in each section, the habitat environment was different according to the water depth, sediment thickness and salinity. Migratory birds prefer hydraulic and hydrological characteristics. This study will be used for the scientific management of Benangkimi wetland, which serves as a stopping point for migratory birds, and it will contribute to basic data of ecology - hydraulics by examining the relationship between habitat environment and hydrological data.

  • This study was carried out to investigate the on the fitness of Polygonatum stenophyllum Maxim. along the coverage of Ambrosia trifida L. We were measured the number of species, death rate of P. stenophyllum, vegetative and reproductive growth of P. stenophyllum and analyzed from June 2015 to October 2016. The coverage gradient of A. trifida was divided none removed control (Control, C), 40 ~ 60% removed treatment (Treatment 1, T1) and 100% removed treatment (Treatment 2, T2) of A. trifida. As a result, When the more than 40% of removal coverage of A. trifida, the number of plant speciecs, the number of P. stenophyllum and vegetative and reproductive growth of P. stenophyllum were higher than none removed the A. trifida during the two years. Therefore, We suggest that the removal of more than 40% coverage of A. trifida from the habitats is effective in preserving the plant species diversity and conserving the populations of P. stenophyllum in this study area.

Optimal Culture Conditions and Food Waste Decomposition Effects of Mixed Strains Separated from Traditional Fermented Food and Soils (전통발효식품과 토양으로부터 분리된 혼합균주의 최적생육조건 및 음식물쓰레기 분해 효과)

  • In this study, for the purpose of decomposing food waste, the strain was screened from traditional fermented food and soils. The enzyme activity (protease, amylase, cellulase, lipase) experiment was carried out using the paper disc method in 212 strains isolated from 5% NaCl media. Among them, only the strains having enzyme activity of more than 2 (soil) or more than 4 (traditional fermented food) with the halozone of enzyme activity of 15 mm or more were selected first, and microorganism identification through 16S rRNA sequencing was performed. Finally, were identified such as Bacillus subtilis, Bacillus amyloliquefaciens, Bacillus siamensis, Bacillus licheniformis, Bacillus aquimaris, Bacillus megaterium, Bacillus koreensis, Bacillus stratoshericus, Bacillus aryabhattai, Bacillus safensis, Marinobacter hydrocarbonoclasticus. 11 species of mixed strains were confirmed that the culture time was 24 hours, the incubation temperature was $30^{\circ}C$ and the optimum pH was 7.0. In order to confirm the degree of decomposition of standard food wastes (100 g) by treating 11 kinds of mixed strains (25%), solid content of more than $2000{\mu}m$ was determined to be 103 g for the sterilized water group and 18 g for the mixed strains group. And the rest was decomposed to a size of less than $2000{\mu}m$.

  • The aim of this study is toxicity assessment using two types of sea urchins (H. pulcherrimus, M. nudus) that can representative primary consumers in potential coastal environments pollutants, Zinc undecylenate (ZU), which is used for various purposes, such as pharmaceutical agents and anti-bacterial and anti-fungi. The Fertilization rate and normal embryogenesis rate of H. pulcherrimus and M. nudus were concentration-dependent decreased. Besides, EC50 of fertilization rate with H. pulcherrimus and M. nudus were 11.27 mgl-1 and 1.48 mgl-1, and EC50 of normal embryogenesis were 0.94 mgl-1 and 3.78 mgl-1. NOEC of normal embryogenesis were 0.20 and 0.78 mgl-1, respectively. In addition, to find the safety criteria of the ZU on the marine environment. PNEC value was 0.0094 mgl-1, calculated using the toxicity values of two species of sea urchin derived from this study and the acute toxicity results of the coastal area through literature research. The above results will be used as basic data for establishing environmental protection strategies for marine environmental pollutants.

Spatial and Temporal Distribution of Fish Communities with Rainfall in Jungrang Stream (강우에 따른 중랑천 어류군집의 시공간적 분포 특성)

  • In this study, we surveyed the fish community at the four sites(St. 1, 3, 5, 7) in Jungrang Stream from 2008 to 2016. We found 37 species grouped into nine families. There were three exotic species; Cyprinus carpio, Carassius cuvieri and Micropterus salmoides after rainfall. Dominant species was Zacco platypus (57.3%) and subdominant species was Carassius auratus(10.4%) in a community. The water quality, surveyed at the six sites(St. 1, 2, 3, 4, 5, 6), based on eight factors(Water temperature, pH, DO, BOD, SS, EC, TN and TP), largely varied depending on each site and period. Minimum values in four factors(BOD, EC, TN and TP) were observed in rainy season, indicating an ionic and nutrient dilution of stream water by precipitation. In contrast, a maximum value in SS was occurred in rainy season at St. 2 and St. 5. The precipitation, Maximum value observed in July ($497.5{\pm}297.2mm$), minimum value in January ($12.9{\pm}8.6mm$). In July and August, the precipitation was divided into before and after the rainfall season in Jungrang stream. Using cluster analysis three fish sites (St. 1, 3, 5) were identified as significantly influence 11 fish species; Hemiculter eigenmanni, Squalidus japonicus coreanus, Hemibarbus labeo, Gnathopogon strigatus, Pungtungia herzi, Rhynchocypris oxycephalus, Pseudogobio esocinus, Pseudorasbora parva, Cyprinus carpio, Carassius auratus and Zacco platypus by rainfall.

The Flowering Characteristics of Non-Spiny Safflowers according to Sowing Time (잇꽃 무가시형 유전자원의 파종시기에 따른 개화특성)

  • 잇꽃(Carthamus tinctorius L.)은 국화과에 속하는 두해살이 초본 식물로 한자명은 홍화(紅花), 영명으로는 Safflower를 사용하고 있다. 우리나라의 잇꽃 재배는 2010년 39ha 수준이 재배되다가 2014년에는 76ha까지 확대되었으나, 점차 감소하는 추세를 보이고 있으며 2020년에는 52ha가 재배되고 있는 것으로 보고되었다. 잇꽃은 종자를 약용으로 이용하거나 꽃잎을 건조하여 천연 염색제 및 향신료로 이용하고 있으나, 일본 산형(山形)현에서는 잇꽃의 경관적 가치를 이용해 홍화 축제를 개최함으로써 소득작목으로 자리매김하고 있다. 본 연구는 지리산을 중심으로 한 준고랭지 지역에서 잇꽃의 경관적 가치의 활용 가능성을 검토하고자, 국립농업유전자원센터로부터 총포에 가시가 없는 무가시형 유전자원(IT323225, IT333473, IT333482)을 분양받아 가시가 있는 재배종과 파종시기별로 개화 특성 등을 비교하였다. 표고 500m 준고랭지의 비가림 하우스에서 3월 하순부터 6월 상순까지 파종시별로 출현율을 검토한 결과, IT 323225 자원은 관행의 3월 하순 파종에서도 40.5%의 낮은 출현율을 보였다. 3월 하순 파종에서 재배종은 6월 22일경에 개화기에 도달하였으나, 무가시형 유전자원들은 7월 2일~5일경에 개화기에 도달하여 만생종의 특성을 보였다. 파종시기를 늦춰 5월 하순에 피종하는 경우 무가시형 자원들은 7월 27일~29일경에 개화기에 도달하였으며, 개화기 전후의 개화 지속기간은 8~10일이 소요되어 3월 하순에 파종하여 7월 상순에 개화하는 경우보다 2~4일이 단축되는 경향이었다. IT333473 자원의 개화기 초장은 5월 하순 파종하는 경우 관행의 3월 하순 파종보다 43.6%가 즐어든 71.1cm를 보였고, 분지수는 41.6% 감소한 8.0개/주 수준을 보였다. 식물체당 착화수 또한 파종시기를 늦춤에 따라 감소하는 경향이었으나 화당 종자수는 IT323225 자원은 5월 중순과 하순 파종시에 증가하는 양상이었고 IT333473 자원과 IT333482 자원은 재배종과 같이 감소하였다. 하계 휴양지로 각광받고 있는 지리산권에서 무가시형 잇꽃 자원의 경관적 가치 활용 가능성을 검토한 결과, 파종시기를 5월 하순으로 늦추는 경우 성수기인 7월 하순과 8월 상순에 개화가 가능하였다. 또한 식물체의 초장이 71.1~83.6cm 수준으로 단축되어 경관 조성에 유리한 특성을 보였다.

Malignant Melanoma (악성 흑색종)

  • Background : The incidence of malignant melanoma is currently increasing at a rate greater than any other cancer occuring in human. At this time, early diagnosis and surgical excision were the mainstay of treatment for patients with malignant melanoma. We reviewed the results of average 4 years of follow-up after surgical excision of total 16 cases of malignant melanoma since 1985. Materials and Methods : There were 16 patients (mean age 58.5 years, 5 men, 11 women). The site of the primary lesion was foot and toe (6), back (3), hand (2), thigh (2), shoulder (1), lower abdomen (1) and lip (1). The lymph node was involved at 9 patients. The histologic diagnosis was made with H-E, S-100 stain, and HMB-45 stain as a special stain. Results : Histologically, there were Clark's stage I for 3 patients, II in 4, III in 2, IV in 3, and stage V in 4 patients. The wide excision only greater than 2cm margin was performed for 4 patients. The wide excision and lymph node dissection were performed for 4 patients. The amputation was only performed for 3 patients, and the amputation and lymph node dissection were performed for 5 patients. After surgical excision, chemotherapy was done with Taxol for each 2 patients of stage IV and V. After long term follow-up for mean 4 years, 4 patients died related with melanoma, 1 patient was recurred, and 11 patients were cured. Conclusion : The incidence of malignant melanoma was rare in Korea, but early involvement of lymph node at initial diagnosis was found in many cases (9/16, 56%). And then, early detection and appropriated excision as well as careful dissection of adjacent lymph nodes will offer the patient the best chance for cure.
