The effects of low temperature storage and aging of Jeot-kal on the microbial counts and microflora (젓갈의 숙성 및 저온 저장이 미생물 균수 및 균총에 미치는 영향)

  • The addition of 5% NaCI to standard plate count (SPC) and bromcresol purple (BCP) agar showed the highest viable cell counts for Jeot-kal samples. The use of 15% glycerol as cryoprotectant showed the highest microbial survival rate at both temperatures, $-20^{\circ}C$ and $-170^{\circ}C$, and on both colony count media, SPC and BCP. During the aging, the pH of Bajirak Jogae-Jeot (fermented clam) decreased from 6.8 to 5.0. Crude protein content was 10% for Bajirak Jogae-Jeot and $6{\sim}7%$ for Myeolchi-Jeot (fermented anchovy). Microbial population of Bajirak Jogae-Jeot was $10^9\;CFU/g$ after 4 weeks of aging, but was only $10^{3-5}\;CFU/g$ in the case of Myeolchi-Jeot. The proportion of Gram positive and catalase negative bacteria in Bajirak Jogae-Jeot increased drastically during the 4 weeks of aging, which showed typical lactic bacterial fermentation. After 2 years' storage of Jeot-kal in liquid nitrogen tank, the cell counts of total aerobic or lactic bacteria were decreased, resulting in about 10% survival rate. Microbial floral change of Jeot-kal was also investigated. In the case of Bajirak Jogae-jeot, the ratio of rod to cocci and that of Gram negative to positive increased after liquid nitrogen storage. But, rod to cocci ratio of Myeolchi-jeot decreased after liquid nitrogen storage. The ratio of yeasts decreased in both cases after storage.

Effect of Root Zone Temperature on the Induction of Inflorescence of Phalaenopsis in Summer (하절기 근권 온도가 팔레놉시스의 화경 발생에 미치는 영향)

  • The influence of root zone temperature to the induction of inflorescence and growth of Phalaenopsis was investigated. Root zone temperatures were 15, 20, 25, and $30^{\circ}C$, while the air temperature was kept over $28^{\circ}C$ during three months. $CO_2$ uptake, fresh weight, dry weight and branched root number of Phalaenopsis were highest at $25^{\circ}C$ and lowest at $15^{\circ}C$. But, the anthocyanin content was highest at $15^{\circ}C$ and lowest at $25^{\circ}C$. Inflorescence was not induced by root zone cooling temperature below $25^{\circ}C$ for three months. The concentrations of K, Ca and Mg in leaves were changed according to the root zone temperature, but those of N and P were not changed. K content was high at $20^{\circ}C$, whereas Ca and Mg contents were high at $25^{\circ}C$ root zone temperature. This study indicates that Phalaenopsis perceives temperature by shoot and the optimum root-zone temperature for the vegetative growth is $25^{\circ}C$.

Radiological Safety Assessment for KAERI Incineration Plant on the Basis of Trial Burn Results (시험소각결과에 기준한 한국원자력연구소 소각시설의 방사학적 안전성 평가)

  • Radiological safety for the conventional operation of Demonstration-Scale Incineration Plant (DSIP) was assessed on the basis of the results of trial burns using the simulated and real radioactive wastes. Radiation dose assessments for routine releases on an annual basis as well a several severe accidental releases on a short-term basis (2h) revealed that there would be no significant environmental impact when low-level waste Is incinerated in DSIP. For semivolatile radioactive cesium species, expected emission concentrations slightly exceeded 10% of maximum permissible concentration. Removal characteristics of the bag filter for condensed-phase cesium species was investigated by the trial burns of simulated waste with inactive cesium tracer. In the off-gas before passing through bag filter, distributions of condensed cesium species in the transition size ranging between the diffusional and inertial region are less than 5%. The overall collection efficiency of the bag filter for cesium species was higher than 99.9%, showing enough decontamination capability as a primary filter for the low-temperature dry off-gas system in radwaste incineration plant.

  • The temperature-corrected values of blood gas analysis were compared to uncorrected values in 40 cases of open heart surgery under moderate hypothermic cardiopulmonary bypass. The results were as follows. 1. The corrected value of pH was significantly higher than uncorrected value, and it's relationship was ${\Delta}pH=-0.015$ ${\Delta}Temp+0.005$(r=0.81, P<(0.01). 2. The corrected value of $PCO_2$ was lower than uncorrected value, and it's relationship was ${\Delta}PCO_2=1.11$ ${\Delta}Temp+1.81$(r=0.50, P<0.01). 3. The corrected value of $PO_2$ was lower than uncorrected value, and it' s relationship was ${\Delta}PO_2=5.21$ ${\Delta}Temp-1.45$(r=0.32, P<0.01). But there was no clinical significance. 4. The corrected values of $HCO_{3^-}$, base excess, $CO_2$ content and oxygen saturation were similar with uncorrected values. In summery, the values of pH and $PCO_2$were significantly changed by temperature-correction. Because of the neutral point of water (pH=pOH) rises as temperature falls and it change in parallel with the changes in blood pH, a corrected pH of 7.4, $PaCO_2$ of 40mmHg during deep hypothermia would reflect a profound respiratory acidosis. Therefore, the use of the uncorrected value at $37^{\circ}C$ is more preferable and valid means of assessing acid-base management regardless of actual patient temperature.

  • This study was carried out to develop the normal temperature-circulatable retort sterilized imitation crab leg (RSIC) with long shelf-life by retort sterilization. We have examined the optimum retort sterilization conditions and quality characteristics of the RSIC. Central composite design for response surface methodology (RSM) was adopted for optimization of retort sterilization, and experimental central composite design consisted of 11 samples such as 4 factorial points; 4 star points; 3 central points. Sterilization temperature and $F_0-value$ for retort sterilization were chosen for independent variables, and shearing force, whiteness and sensory score about texture were selected as a dependent variable. As a results of RSM analysis, multiple response optimization for the RSIC by Minitap statistical programing was $F_0-value$ 3.3 min. at $117.5{^{\circ}C}$. Also RSM analysis indicated such as sterilization temperature during retort sterilization was the most influential factor, while $F_0-value$ little affected on quality of the RSIC.

  • To illustrate the characteristics of marine environment in coastal waters of stationary fisheries, field observations were carried out in August and November, 2001. Water temperatures in Jindong Bay compared with those in Masan Bay were about $2.0^{\circ}C$ higher in August and about $0.7^{\circ}C$ in November. Salinities were lowest in the western part of Jindong Bay. It was found that salinity increased gradually from the western part of Jindong Bay to Masan Bay. Sea water in Masan Bay can be characterised as lower temperature and higher salinity, compared with those in Jindong Bay. Concentrations of suspended solids in the inner parts of Jindong Bay were higher than 15mg/L in August. Suspended solids in both regions of Jindong Bay and Masan Bay showed contrastive distributions, especially in November. That is, higher concentrations of suspended solids were found in the inner parts of Jindong Bay and off Masan Bay. Concentration of chemical oxygen demand in Masan Bay was highest, 7mg/L.

Study on the Storage Stability of Horse Fat in Jeju (제주산 말지방(Horse Fat)의 저장 안정성 향상에 관한 연구)

  • Horse fat is known to be an effective ingredient in Asia, and the horse fat itself, which is mixed with other ingredients at the additive level, is often sold as a finished product. In this case, physical properties of the horse fat raw material are important. Many horse fats produced in Korea (Jeju) have low temperature stability, so if not stored at low temperatures, segregation may occur. In the case of Japanese horse fat, it is partially hydrogenated or is used the solid phase as the horse fat by separating the liquid phase and the solid phase that is harder and more stable than the horse fat of Jeju. In this study, the physical properties were tested to improve the temperature stability even without the partial hydrogenation process of Jeju horse fat. Various oil gelling agents were used in the study. Results confirmed that the physical properties of the hydroxystearic acid added Jeju horse fat were improved. In addition, stability evaluations at temperatures of 25 ℃, 40 ℃, 45 ℃ and flow behavior evaluations at temperatures of 25 ℃, 30 ℃, 40 ℃ were performed for Jeju horse fat with hydroxystearic acid, 100% Jeju horse fat, and 100% Japanese horse fat. Results showed that the Jeju horse fat improved in flow behavior by adding hydroxystearic acid similar to that of Japanese horse fat. In addition, when the crystal state was observed under a microscope, the thermal stability was improved by decreasing the size of the needle-type crystals with the addition of hydroxystearic acid. Jeju horse fat containing hydroxystearic acid was found to have no physical problems even when stored at room temperature for a long time.

Effects of Calcium and Indole-3-butyric acid Treatments on Calcium Concentration and Stem-End Browning in 'Fuyu' Sweet Persimmons (칼슘제 및 IBA 처리가 '부유' 단감과실의 칼슘함량 및 과정부 갈변현상에 미치는 영향)

  • 'Fuyu' (Diospyros kaki L.) is an important sweet persimmon cultivar, and the fruits are often stored in a modified atmosphere after harvesting in South Korea. However, blossom-end browning and darkening of fruit often occur after harvest or during storage, which decreases fruit quality in the fresh fruit market. High fruit calcium concentration would reduce oxidation of phenolic compounds in the cytoplasm such oxidation is responsible for fruit browning. This study investigated the effects of soluble calcium fertilization and foliar application, and indole-3-butyric acid (IBA) fertilization on fruit quality and browning. Trees received one of the following five treatments: 1) control (no calcium or IBA); 2) calcium fertilization (Ca FG, 2 mL per tree); 3) calcium foliar application (Ca FA, 2 mL); 4) calcium and IBA fertilization (Ca+IBA) 5) IBA fertilization (IBA, 2 mL. Fruit calcium concentration was highest in trees treated by Ca FA, and lowest in control trees. Generally, fruit calcium concentration was high in the stem end but low in the blossom end, which usually first develops fruit-browning symptoms. There were no apparent differences in fruit qualities such as firmness, soluble solid content, and weight among treatments. Fruit browning occurred at frequencies of about 14%, 20%, and 50% on Ca FA, Ca FG, and control trees, respectively. Therefore, the improved fruit calcium level seen when trees received Ca or IBA application tended to prevent fruit browning, which increased fruit quality and storage properties.

Effect of split achene pericarp, Giberellin, and germination temperature on the germination of Rosa davurica Pall (생열귀나무 종자 발아에 있어서 개갑처리, 지배렐린, 발아온도의 효과)

  • This study was conducted to investigate the suitable conditions for seed germination of Rosa davurica PALL. In the characteristics of the fruit and seed of Rosa davurica, fruit length and width was 1.3cm and 0.9cm , respectively, and seed number was eighty-nine. Artificial and low temperature storage at 4$^{\circ}C$ increased the rate of split achene pericarp until 46.6% and storage at 15$^{\circ}C$ incubator decreased the rate of split achene pericarp (10.5%). The rate of seed germination of split achene pericarp in control at 15$^{\circ}C$ was 90% and of non-split achene in GA$_3$100ppm at 15$^{\circ}C$ was 36.8%. Average germination day of split achene pericarp seeds in GA$_3$150ppm at 2$0^{\circ}C$ was 4.2 days and non-split achene pericarp in GA$_3$100ppm at 2$0^{\circ}C$ was 7.3 days. Seed germination was not different between various concentrations of GA$_3$ treatments in split achene pericarp seeds but the rate of germination was more reduced in high concentration of 200ppm. Only the treatments of GA$_3$ was increased germination rate at 4$^{\circ}C$ and immature seed of Rosa davurica.

Effect of Immersion Temperatures and Times on Moisture Absorption and Germination of Cryptomeria japonica Seeds (침지 온도와 시간이 삼나무 종자의 수분흡수 및 발아에 미치는 영향)

  • This study was carried out to enhance germination of the seed by different immersion temperatures(15, 20, 25, 30 and $35^{\circ}C$) and times(1, 6, 12, 18, 24, 36 and 48 hours) in Cryptomeria japonica that is a major plantation species in the southern Korea. Optimum germination temperature was between $20^{\circ}C$ and $25^{\circ}C$, and the lowest germination was observed at $35^{\circ}C$ Moisture absorption increased with increasing immersion time as a whole and was high at $30^{\circ}C$ and $35^{\circ}C$. Initial moisture absorption rate was highest at $30^{\circ}C$ whereas was lowest at $15^{\circ}C$ Moisture absorption rate constant was also high at $30^{\circ}C$ and $35^{\circ}C$, however, it was relatively low at $15^{\circ}C$ and $20^{\circ}C$ Germination was the highest when the seeds were immersed for 48 hours at $30^{\circ}C$. In mean germination time(MGT) the seeds immersed for 48 hours at $30^{\circ}C$ did not differ from non-treatment, while the others showed higher values than non-treatment. Consequently, germination could be enhanced and MGT was kept by optimum immersion temperature and time.
