Generic Characters of the Fishes of the Family Tetraodontidae (Teleostei: Tetraodontiformes) (참복과(복어목) 어류 속의 외부형태적 특징)

  • Twenty seven species of pufferfishes (family Tetraodontidae) have been examined to find their generic characters based on olfactory organ, lateral line system, prickles, body form and color characters of specimens collected in 20 places along the coast of Korea from September, 1989 to July, 1994. Based on nostril shape, the genera of Takifugu, Lagocephalus, Plurancanthus, Spheoides and Feroxodon have two nostrils, and those of Ephippion and Canthigaster have a single nostril or nasal tentacle, while Boesernanichthys, Arothron and Chelonodon have no nostril. In the genera of Lagocephalus, Spheoides, Boesemanichthys, Arothron and Feroxodon have only a single well-developed lateral line on the body, in those of Takifugu and Plurancanthus usually have two lateral lines, and those of Chelonodon and Ephippion have two lateral lines merged on the caudal peduncle. Based on the lateral line system and olfactory organ, genus Canthigaster has different characters from the other genera of the family Tetraodontidae. In Ephippion guttifer, the prickles on the dorsal and lateral of the body are enormously enlarged into plates forming a partial carapace. Of the 22 species which have prickly spines, the genera, Lagocephalus, Takifugu stictonotus and Arothron Stellatus have the larger ventral pricklyy spines than the dorsal ones. In genera Lagocephalus most of the species have prickly spines that coverd both the back and belly, while L. inermis has no prickly spines.

  • Since the 1970s, manufacturing has been one of the key driving forces that has led to Korea's economic growth. However, this growth rate has been reduced significantly since the 2000s, and shows that revenues and employment are steadily decreasing. In addition, while manufacturing investment in Korea has dropped sharply, the United States, Germany, Japan, and other major countries have increased investment in manufacturing. These countries have promoted manufacturing innovation strategies that include the convergence of information and communications technologies (ICT) and manufacturing. For manufacturing innovation, it is important for time and cost savings required for product development to be achieved by changes in the production process, especially product design. Modeling and simulation (M&S) is a process that replaces physical product design, mockup making, and testing, with virtual product creation (modeling) and engineering analysis (simulation). In this paper, we analyze the economic ripple effect of supercomputing M&S using an input-output model technique based on the input-output tables published by the Bank of Korea. When we set the M&S budget (about US$16 million for the last 10 years) of the Korea Institute of Science and Technology Information (KISTI) as input coefficients, the effect on production inducement, value-added inducement, and employment inducement was analyzed to be US$24 million, US$13.4 million, and 267, respectively.

Effect of Chungpaesagan-tang on ischemic damage induced by MCAO in spontaneously hypertensive rats

  • Chungpaesagan-tang (CPSGT) is most frequently used to treat ischemic brain injury in tradition Korean medicine. Clinically, cerebral ischemia is likely to be accompanied by preexisting or complicating disease. However, animal models used to examine the effects of herbal medicines on cerebral ischemia have not given this issue sufficient consideration. The present study was undertaken to determine the effects of CPSGT on focal cerebral ischemia in normal and SHR rats subjected to transient middle cerebral artery occlusion (MCAO). Animals were divided into four groups: Normal (Sprague-Dawley) rats subjected to MACO (the NC+MCAO group), normal rats subjected to MCAO and then administered CPSGT (NC + MCAO + CP), SHR rats subjected to MCAO (SHR + MCAO), and SHR rats subjected to MCAO and then administered CPSGT (SHR + MCAO + CP). MCAO was performed using the intraluminal method. CPSGT was administrated orally twice (1 and 4 h) after MCAO. All animals were sacrificed at 24 h postoperatively. Brain tissues were stained with hematoxylin & eosin, to examine the effect of CPSGT on ischemic brain tissues. In addition, changes in TNF-$\alpha$ expression in ischemic areas were examined by immunostaining. CPSGT was found to significantly reduce infarction areas in normal and SHR rats and infarction volumes in SHR rats. Similarly, CPGST markedly increased neuron numbers and sizes in all treated groups, except cell sizes in SHRs. Furthermore, CPSGT reduced TNF-$\alpha$ expression in MCAO administered SHR rats. The findings of the present study suggest that CPSGT effectively ameliorates neuron damage caused by MACO-induced cerebral ischemia, and that it has a significant neuroprotective effect after cerebral ischemia in SHR.

  • An undisturbed healing process without micromotion at the implant-bone interface is essential for achievement of osseointegration of dental implant. Therefore, initial stability was advocated as prerequisite for successful clinical outcome. Adequate bone quality and quantity were important to achieve initial stability and to prevent early failures. However there were few published data available regarding the effect of design change in implant geometry on initial stability of the implants. The purpose of the current study was to assess the initial stability of various designs of implants when placed into artificial bone materials of varying qualities and shapes of insertion holes. Within the scope of this study, the following results were drawn. Bone quality was major importance to achieve initial stability. Initial stability was higher on GS II which had additional design feature of double thread. With a tapered design of implant such as GS III showed a higher initial stability than straight one. An insertion hole with the similar shape of implant would lead to reduce a compression force on cortical bone and enhance a bone anchorage on cancellous bone.

Production of a Monoclonal Antibody to Human $\alpha$-Fetopotein and Development of Monoclonal Antibody-Based Enzyme-Linked Immunosorbent Assays for Human $\alpha$-Fetoprotein (인간 $\alpha$-fetoprotein에 대한 모노클로날 항체의 제조 및 모노클로날 항체를 이용한 효소면역분석법의 개발)

  • This study was attempted to generate a monoclonal antibody against human $\alpha$-fetoprotein (AFP) and to produce an immunoassay, recognizing AFP in plasma and amniotic fluid. AFP was purified from human amniotic fluid and used to immunize mice. Spleens were taken from the mice and the cells were fused with mouse myeloma cells (Sp2/0-Ag-14) for the production of monoclonal antibodies by employing the hybridoma technology. As a result, a hybridoma cell line producing anti-AFP monoclonal antibody was cloned out and designated as MabF22. From isotyping analysis, it was found that monoclonal antibody MabF22 was IgG type with IgG1 heavy chain and k light chain. The binding specificity of MabF22 was analyzed by immunoblotting as well as by ELISA. MabF22 was highly specific, reacting with only AFP-containing samples. The binding affinity was determined by ELISA (free-capture mode) and Scatchard analysis. As a result, the value of Kd was 0.8$\times$10$^{-10}$M. The validity of the MabF22 for AFP assay was examined by two kinds of ELISAs, i.e., non-competitive and competitive ELISA. Both assays revealed that MabF22 reacted well with AFP in sample in a concentration-dependent manner. Standard curve and antibody titration curve were obtained by using purified AFP and MabF22. These results indicate that the monoclonal antibody produced in this study would be useful not only for research purposes but also for further development of immune-diagnostic kit for the measurement of AEP concentration.

Effects of $\alpha$-Tocopherol Acetate on Puromycin Aminonucleoside-Induced Glomerular Injury in Spontaneously Hypertensive Rats (선천성 고혈압 흰쥐에 Puromycin Aminonucleoside로 유도된 사구체 손상에 대한 $\alpha$-tocopherol Acetate의 효과)

  • In order to study the effects of $\alpha$-tocopherol acetate in glomerular injury, the minimal change nephrosis disease was induced by puromycin aminonucleoside (PAN) in spontaneously hypertensive rats, and we examined biochemical analysis in serum and morphological changes. The experimental animals were divided to control, PAN-treated (30 mg/kg, I.p.), vitamin E-treated (200 mg/kg, P.O.), and PAN+vitamin E-treated groups. After PAN injection, the rate of increase of body weight was lower than the other treatments. In addition, at 8 days after PAN injection, total protein content in serum was the lowest, whereas both blood urea nitrogen and serum creatinine contents were the highest in all experimental groups, which their changes of serum parameters were statistically significant. In morphological changes, the glomerular tissue at 8 days after PAN injection clearly showed obstruction of urinary space and proliferation of mesangial cells, and that loss and fusion of pedicles, vacuolization and edema of endothelial cells, and thickness of basal lamina were ultrastructurally showed in the glomerulus. Glomerular injury was significantly prevented by administration of vitamin E having an antioxidant effect. It suggested that the glomerular injury induced by PAN was accelerated by hypertension, and renal dysfunction might be induced by oxidative injury.

Effects of Cadmium Exposure on Tissues of Carassius auratus (카드뮴 노출이 붕어(Carassius auratus) 조직에 미치는 영향)

  • The present study aimed to investigate the level of accumulated heavy metal in various tissues of Carassius auratus after exposure to Cadmium (Cd), histologically and physiologically. After treating C. auratus with Cd, the accumulated Cd in gill tissues was detected to be of the highest content, and showed the lowest content in integument tissues. Also, Cd content increased in a time dependent manner and showed the highest accumulation in the tissues exposed for 20 days. Antioxidant enzyme activities showedhigher activity in the gill and integument than in the kidney and liver tissues. In the case of SOD, antioxidation activity of SOD in all Cd exposed tissues was higher than in unexposed tissues. The activities of SOD and CAT also became higher after Cd exposure. Gill tissues exposed to Cd showed an increased number of mucous cells between lamella in a time dependent manner. In addition, the gills showed morphological changes such as edema, exfoliation of epithelial cells, and fusion of the secondary lamellae. Also, exposure to Cd for 20 days had an effect on gill tissues, causing membrane damage in the mitochondria and nucleus. In kidney tissues, atrophied glomerulus was observed, and the empty space in Bowman's capsule was wider.

Regulation of Mitochondrial Homeostasis in Response to Endurance Exercise Training in Skeletal Muscle (지구성 훈련에 반응한 골격근의 미토콘드리아 항상성 조절)

  • Mitochondrial homeostasis is tightly regulated by two major processes: mitochondrial biogenesis and mitochondrial degradation by autophagy (mitophagy). Research in mitochondrial biogenesis in skeletal muscle in response to endurance exercise training has been well established, while the mechanisms regulating mitophagy and the relationship between mitochondrial biogenesis and degradation following endurance exercise training are not yet well defined. Studies have demonstrated that endurance exercise training increases the expression levels of mitochondrial biogenesis-, dynamics-, mitophagy-related genes in skeletal muscle. However, the increased levels of mitochondrial biogenesis marker proteins such as Cox IV and citrate synthase, by endurance exercise training were abolished when autophagy/mitophagy was inhibited in skeletal muscle. This suggests that both autophagy/mitophagy plays an important role in mitochondrial biogenesis/homeostasis and the coordination between the opposing processes may be important for skeletal muscle adaptation to endurance exercise training to improve metabolic function and endurance exercise performance. It is considered that endurance exercise training regulates each of these processes, mitochondrial biogenesis, fusion and fission events and autophagy/mitophagy, ensuring a relatively constant mitochondrial population. Exercise training may also have contributed to mitochondrial quality control which replaces old and/or unhealthy mitochondria with new and/or healthy ones in skeletal muscle. In this review paper, the molecular mechanisms regulating mitochondrial biogenesis and mitophagy and the coordination between the opposing processes is involved in the cellular adaptation to endurance exercise training in skeletal muscle will be discussed.

Clinical and molecular biological aspect of the hyaluronidases: basis and clinical overview for oriental medical application

  • Components of extracellular matrix and the matrix-degrading enzymes are some of the key regulators of tumor metastasis and angiogenesis. Hyaluronic acid (HA), a matrix glycosaminoglycan, is known to promote tumor adhesion and migration, and its small fragments are angiogenic. Until now, we have compared levels of hyaluronidase, an enzyme that degrade HA, in normal adult prostate, benign prostate hyperplasia and prostate cancer tissues and in conditioned media from epithelial explant cultures, using a substrate (HA)-gel assay and ELISA-like assay (Kim et al., unpublished results). The present review described an overall characterization of hyaluronidases and its application to human diseases. The hyaluronidases are a family of enzymes that have, until recently, deed thorough explication. The substrate for these enzymes, hyaluronan, is becoming increasingly important, recognized now as a major participant in basic processes such as cell motility, wound healing, embryogenesis, and implicated in cancer progression. And in those lower life forms that torment human beings, hyaluronidase is associated with mechanisms of entry and spread, e.g. as a virulence factor for bacteria, for tissue dissection in gas gangrene, as a means of treponema spread in syphilis, and for penetration of skin and gut by nematode parasites. Hyaluronidase also comprises a component of the venom of a wide variety of organisms, including bees, wasps, hornets, spiders, scorpions, sh, snakes and lizards. Of particular interest is the homology between some of these venom hyaluronidases and the enzyme found in the plasma membrane of mammalian spermatozoa, attesting to the ancient nature of the conserved sequence, a 36% identity in a 300 amino acid stretch of the enzyme protein. Clearly, hyaluronidase is of biological interest, being involved in the pathophysiology of so many important' human disorders. Greater effort should be made in studying this family of enzymes that have, until recently, been overlooked. Also, oriental medical application of the hyaluronidase will be discussed with respect to inhibition and suppression of inflammation and malignacy.

Effects of polysaccharide fractions from phellodendron chinese SCHNEID on tumor progression and immunopontentiation

  • In the previous paper (Kim et al., Glycoconjugate Journal 16, 247-252, 1999), heteropolysaccharides from Korean medicinal plant, Phellodendri cortex (Hwangbek) showed a poten B-Iymphocyte-stimulating activity in a system using polyclonal antibody forming cells in C57BL/6XC3H mice at dosages of 2-10 mg. In a series of biolgical active polysaccharides from natural medicinal plants, the polysaccharide fractions were isolated and purified from Phellodendron chinese SCHNEID, and antitumor activities were examined at dosages of 2, 5 and 10 mg/100 g. F-7 and F-8 showed the highest tumor inhibitory activities (inhibition ratio 96.4 and 98.2% in 2 mg/100 g), and in dose of 5 mg/100 g, the inhibitory ratios were 95.3 and 97.5%, respectively. Furthermore, 10 mg/100 g of intraperitoneal (i.p.) injection gave 97.3 and 98.7% of inhibition. In oral administration, the inhibitory activities were not markedly observed, indicating that the polysaccharides are directly acting to immune system. When the effects on TS and TK activities were determined, TS activities in the F-2 and F-7-treated mice were markedly suppressed to 73.7% and 79.5% of that in the control (p<0.01), while there was little difference in TK activity with a slight decrease in F-2 only. However, in i.p. injection, TS activities in the F-2, F-5, F-7 and F-8-treated mice were markedly suppressed to 83% to 85% of that in the control (p<0.01). Furthermore, there was also significant differences in TK activities in F-2, F-5, F-7 and F-8-treated mice (p<0.05). Therefore, polysaccharide fraction F-8 was further purified to active fractions of F-9 and F-11 by gel permeation chromatography using TSK Gel HW50S. The purified polysaccharides of F-9 and F-11 were composed of GlcNAc (47.3%), Gal (24.7%) and Man (28.0%). These results clearly indicated that the i.p. injection is much effective to suppress tumor growth than oral administration.
