Anti-microbial, Anti-inflammatory and Anti-oxidative Effects of Herbal Medicine Extracts as Anti-gingivitis Ingredients (치주 질환 예방 및 치료용 소재로서 수종의 생약성분 추출물에 관한 항균, 항염, 항산화 효능 연구)

  • This study was to estimate the effect of herbal medicines on periodontal disease. To screen effective materials for periodontal disease, we performed a series of test for 15 herbal medicine extracts. MIC test for P. gingivalis, IL-$1{\alpha}$ inhibition test, MMP-1 inhibition test and SOD activation test were carried out for herbal medicine extracts, respectively. The results are as followings : 1) Eunhang, Youkdoogu, Daewhang, Hoobak, Goojulcho, Yongacho, Mokhyang, Sesin, Sancho and Hoehyang extracts were effective for reduce P. gingivalis, 2) Eunhang, Jacyak, Youkdoogu, Hagocho, Hoobak, Guaru and Sanyak extracts showed high IL-$1{\alpha}$ inhibition rates, 3) Eunhang and Youkdoogu extracts have effects on MMP-1 inhibition, 4) Eunhang, Sanyak and Youkdoogu extracts effectively activate SOD. Especially, Eunhang extract has high performance through all estimations and can effectively prevent and treat periodontal disease.

Changes in the Volatile Flavor Components of Nutmeg(Myristica fragrans Houttuyn) during Aging (숙성에 의한 육두구(Myristica fragrans Houttuyn)의 향기성분 변화)

  • The change of volatile flavor components In nutmeg (Kernels of the fruits of Myristica fragrans Houttuyn) during aging at $37^{\circ}C$ were studied by using a fused silica capillary GC & GC/MS. Volatile flavor components having the low boiling point showed a general decrease during aging, but those of the middle and high boiling point showed a reactionary tendency Myristicin and myristic acid among volatile flavor components showing the high boiling point had the amount increased considerably, and those were composed of 24.50% and 18.69% in aging for 6 months, respectively. The amount of whole volatile flavor concentrate showed the increased tendency till the aging period for 4 months, and then subsequently decreased.

The antioxidant ability of nutmeg ethanolic extract in bulk oil and oil-in-water emulsion matrices (식물성 유지 및 수중유적형 유화계에서 육두구 종자 에탄올 추출물의 항산화활성 평가)

  • The antioxidant ability of 80% ethanolic extract of nutmeg seed (NM80) was evaluated using in vitro assays and bulk oil and oil-in-water (O/W) emulsion matrices. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging, and oxygen radical antioxidant capacity (ORAC) in vitro assays were used to evaluate the antioxidant ability of the extract. The DPPH radical scavenging activities of 25, 50, 100, and 200 ㎍/mL NM80 were 12.5, 20.9, 35.1, and 62.8%, respectively, while the ABTS cation radical scavenging activities were 2.7, 6.5, 30.5, and 29.8%, respectively, demonstrating a dose-dependent effect. The ORAC value was significantly higher at an NM80 concentration of 25 ㎍/mL than the positive control (p<0.05). The conjugated dienoic acid (CDA), ρ-anisidine, and tertiary butyl alcohol values in 90-min-heated corn oil containing 200 ppm of NM80 were significantly reduced by 3.26, 16.94, and 17.34%, respectively, compared to those for the sample without NM80 (p<0.05). However, the headspace oxygen content and CDA value in the O/W emulsion containing 200 ppm of NM80 at 60℃ had 6.29 and 82.85% lower values, respectively, than those for the sample without NM80 (p<0.05). The major volatile compounds of NM80 were allyl phenoxyacetate, eugenol acetate, and eugenol. NM80 could be an effective natural antioxidant in lipid-rich foods in bulk oil or O/W emulsion matrix.

Antimicrobial Effects of Ethanol Extracts from Korean and Indonesian Plants (국내 및 인도네시아산 식물의 에탄올 추출물의 항균효과)

  • Antimicrobial effects of 150 kinds of Korean and 82 kinds of Indonesian plants were investigated to develope natural food preservatives. Extracts of the plants with 70% ethanol were tested their antimicrobial effects against several food spoilage microorganisms, Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger. Seventeen kinds of Korean and eighteen kinds of Indonesian plants were found relatively effective, of which Myristica fragrans and Melaleuca leucadendra were the most effective, respectively. The major fractions of the two plant extracts showing antimicrobial activity were further purified by solvent fractionation, silicagel column chromatography and preparative HPLC. The purified substances were identified as limonene and caprylic acid in M. fragrans, and ${\alpha}-terpineol$ in M. leucadendra, respectively.

  • Myristica fragrans Houtt were extracted in 80% aq. MeOH and solvent fractionated sing $CHCl_3$, EtOAc, n-BuOH and water, successively. The n-BuOH fraction gave three phenylpropanoids through application of silica gel column chromatographies. The chemical structures of the phenylpropanoids were determined by the interpretation of several spectral data, including NMR and MS as meso-dihydroguaiaretic acid (1), nectandrin B (2) and syringin methyl ether (3). Compound 1, which was first isolated from this plant by authors, showed inhibitory activities with $60.0{\pm}2.1%\;(100\;{\mu}g/ml),\;42.6{\pm}0.9%\;(140\;{\mu}g/ml)\;and\;12.2{\pm}0.2%\;(200\;{\mu}g/ml)$ on ACAT(acyl-CoA:Cholesterol Acyltransferase), chitin synthase III and HMG-CoA reductase (3-hydroxy-3-methylglutaryl coenzyme A reductase), respectively. Compound 3 showed inhibitory activities with $27.2{\pm}0.9%\;(100\;{\mu}g/ml),\;45.5{\pm}0.8%\;(200\;{\mu}g/ml)$ on ACAT and chitin synthase III.

생약재 추출물의 수율 및 항고혈압 활성

  • 본 실험에 사용한 생약재는 금산 약초시장에서 2003년 6월에 구입하여 추출수율을 조사하였다. 그 결과, 생약재의 물 추출물의 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 갈근, 맥문동, 산수유가 있고, 20~30%에 달하는 생약재는 감초, 구기자, 당귀, 오미자, 천궁, 황기가 이에 속했으며, 중량(%)이 10~20%에 달하는 생약재는 복분자, 오수유, 애엽, 작약, 진피, 지유, 파고지, 황련이 이에 속했다. 마지막으로 10%이하의 추출 수율을 나타낸 생약재들은 가시오가피, 강황, 계피, 석곡, 소목, 영지, 오약, 육두구, 정향, 죽엽, 필발, 초두구, 호장근이 이에 속했다. 생약재의 70% Ethanol 추출물의 동결건조중량을 살펴본 결과, 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 당귀, 산수유가 이에 속하였고, 20~30%에 달하는 생약재는 감초, 맥문동, 구기자, 오미자, 오수유, 천궁, 황기가 속하였으며, 동결건조 중량(%)이 10~20%에 달하는 생약재는 복분자, 육두구, 애엽, 작약, 정향, 지유, 파고지, 호장근, 황련이 이에 속했다. 또한 10% 이하의 수율을 나타내는 생약재는 가시오가피, 강황, 갈근, 계피, 석곡, 소목, 영지, 오약, 익지인, 죽엽, 필발, 초두구가 이에 속하였다. 생약재의 추출 시간 및 온도에 따른 수율을 조사한 결과, 추출 시간이 경과함에 따라서 수율이 점차 증가하는 경향을 보였으며, 가자, 갈근, 황련, 복분자, 지유, 정향, 계피, 소목 순으로 높은 수율을 얻을 수 있었다. 생약재에 여러가지 효소를 이용한 가수분해 결과, 대부분의 생약재에 대해 95$^{\circ}C$에서 2시간동안 추출한 수율과 그다지 큰 차이는 보이지 않았지만 대부분의 생약재가 Termamyl 효소를 사용하여 가수분해하였을 때 가장 높은 수율을 나타내었다. 또한 지유의 경우는 Viscozyme 효소를 처리하였을 경우 가장 좋은 수율을 나타내었다. 또한, 효소의 최적 온도로 조정한 Water bath에서 0, 30, 60, 120, 210분간 효소 반응 시켜, 420nm에서 갈변도를 시간별로 측정해본 결과, Viscozyme 효소를 처리한 생약재에서 시간이 흐름에 따라 흡광도 수치의 변화가 크게 나타났다. 32종의 생약재 물 추출물의 항고혈압 활성을 측정한 결과, 파고지, 소목, 죽엽의 항고혈압 활성이 높은 것으로 나타났으며, 파고지의 효과가 가장 우수한 것으로 나타났다. 10%에탄을 추출물의 경우에는 오미자, 오수유, 소목에서 항고혈압 활성이 높은 것으로 나타났으며, 오미자의 효과가 가장 우수한 것으로 나타났다. 파고지 물 추출물을 칼럼(Sephacryl S-300, High Resolution)으로 분획하여 3개의 Peak를 얻었으며, 2번째 Peak의 항고혈압 활성이 매우 높은 것으로 나타났다.

  • Myristica fragrans has been used for the treatment of stomachache in East Asia. It is very important to determine the amount of myristicin in Myristica fragrans, because excess myristicin causes side effects. In this study, we developed and validated the method for determination contents of myristicin in Myristica fragrans which was purchased from various regions of Korea. The average content of myristicin in Myristica fragrans was 2.10% with the validated HPLC analysis method.

Effects of Some Crude Drug Extracts on the Brain Neurotransmitters in the Ethanol-Treated Rats (수종의 생약 추출물이 에탄올 투여 흰쥐의 뇌 부위별 신경전달물질에 미치는 영향)

  • The concentration of neurotransmitters in rat brain was determined by HPLC-ECD (electrochemical detection) method and the effects of methanol extracts of some crude drugs, such as Polygala Radix, Myristicae Semen, Zizyphi Semen, Acori graminei Rhizoma, Visci Herba, Liriopsis Tuber, Myrrha on the concentration of neurotransmitters in the ethanol-treated rat brain were investigated. By the administration of ethanol, dopamine (DA), 3, 4-dihydroxyphenyl acetic acid (DOPAC) and serotonin (5-HT) levels in frontal cortex and 5-HT level in hippocampus were significantly increased compared with the neurotransmitter levels in the brain of saline-treated rats. The ${\gamma}$-aminobutyric acid (GABA) level in frontal cortex was decreased by the same treatment. There was a tendency that the DA level in frontal cortex and striatum of ethanol-treated rats were increased by the administration of crude drug extracts. Especially, Myrrha and Visci Herba significantly increased the DA level of frontal cortex in ethanol-treated rats, while they significantly decreased the 5-HT level in the same region of the brain. GABA level in striatum of ethanol-treated rats was significantly decreased by Myristicae Semen, Visci Herba and Myrrha. These results suggest that the tested crude drug extracts have selective interaction with neurotransmitters in specified region of central nervous system.

  • Objectives: The aim of this experimental study is to investigate the effects of MYRISTICAE SEMEN, GRANATI PERICARPIUM and ALPINIAE KATSUMADAI SEMEN on the anticatharsis by using mice and Guinea pigs. Methods : We evaluated the effect of herbs on the extracted organs contraction, the barium sulfate transport, and the anticathartic action induced by acetylcholine chloride, barium chloride, histamine chloride, neostigmine, castor oil, pilocarpine in the ileums, large intestines, small intestines of animals. Results : In the extracted organs, all herbs showed concentration-dependent inhibitory effect on the constriction of extracted ileum. The transport of barium sulfate in the large and small intestine was increased in comparing with the control group. And every three herbs showed the anticathartic action. Conclusions : These results demonstrate that MYRISTICAE SEMEN, GRANATI PERICARPIUM and ALPINIAE KATSUMADAI SEMEN extracts have an advantageous effects on diarrhea induced by drugs and will contribute to the development of diarrhea treatment through further related studies.
