Studies on the Processing of Rapid- and Low Salt-Fermented Liquefaction of Sardine(Sardinops melanoslicta)(II) -Changes in Quility during Preheating and Fermentation Chopped Whole Sardine- (저식염 속성 정어리 발효 액화물 가공에 관한 연구(II) -마쇄육의 예열처리 및 숙성중의 품질변화-)

  • As a part of investigation to use sardine(Sardinops melanoslicta) more effectively as a food source, this study was undertaken the processing condition of rapid- and low salt-fermented liquefaction of sardine. To prepare rapid fermented products, the chopped whole sardine was added 8% NaCl and then preheating treatment at $40^{\circ}C,\;45^{\circ}C$ and $50^{\circ}C$ in the manufactured fermenter(180L) for 9 hrs, and then fermentation at $33^{\circ}C$ for 90 days. The chemical changes such as amino nitrogen(amino-N), volatile basic nitrogen(VBN), and histamine in the hydrolysates of fermented sardine were analyzed as well as viable cell count and organoleptic evaluation during fermentation to compare the quality between control and preheating samples. During fermenting, the amino-N in the hydrolysates increased rapidly during the first 30 days and slowly thereafter. The highest content of amino-N appeared at 75 days in control sample and $60{\sim}75$ days in preheating samples. The changes of VBN in the hydrolysates increased rapidly during first 15 days in control samples and 30 days in preheating samples. However they were generally low level in preheating samples. Histamine content in the hydrolysates of the control samples increased markedly after 15 days, but preheating samples were generally low level, and then $75{\sim}90$ days of fermentation reached to the maximum which was about $2.0{\sim}3.0$ times lower than that of control samples. As for the organoleptic flavor evaluation, the control and preheating at $40^{\circ}C$ samples were unpleasant odor after 15 and 60 days, respectively. But preheating at $45^{\circ}\;and\;50^{\circ}$ samples were fresh odor after 90 days fermentation.

  • The KASINICS (KASI Near Infrared Camera System) is a ground-based Near-Infrared (NIR) imaging instrument developed by the Korea Astronomy and Space Science Institute (KASI). In this paper, we report the test results of the KASINICS camera optics system which is comprised of a 1-1 Offner relay. We measure that the surface RMS fluctuations of the Offner mirrors are at the level of $10^{-1}-10^{-2}$ of the target wavelengths, showing that the mirrors are sufficiently smooth for NIR observations. The alignment of the Offner optics system has been checked too. Our ray-tracing simulations find that the image quality should not degrade more than the pixel size of the KASINICS ($40{\mu}m$), if a de-centering or a tilt of the Offner mirrors are within 5mm, or $2.5^{\circ}$. Our measurement shows that the de-centering or the tilt of the Offner mirrors are less than 1 mm or $0.5^{\circ}$, assuring that the KASINICS image quality are not affected by the alignment errors. We have also measured that the optics resolution is $20{\mu}m$ and it does not degrade more than 10% over the detector surface area of 14.3 mm ${/times}$ 14.3mm. Overall, we conclude that the KASINICS optics system satisfies the design requirements for NIR imaging observations.

  • From the stupa of Sujongsa-Temple, located in Yangpyeong, Gyeonggi-do province, was excavated a celadon jar with lid, a gilt-bronze nine-storied miniature pagoda and a silver-plate hexagonal miniature shrine, etc. Among them, the gilt-bronze miniature pagoda has been known as a gilt-bronze product since most of its surface has copper tint and the base part has bronze corrosion. Its formal title registered on the Cultural Properties Administration also begins with "a gilt-bronze". However, it was supposed to be a gold product in many aspects: the color and status of the surface, degree of the bronze corrosion creation and the metal thread joining the wind bell. So the necessity to prove its material by a scientific analysis was raised. We examined it with nondestructive method. The results were traces of open-working on many parts of the surface, but no traces of gilding. Moreover, as we perform an XRF analysis, the main component were gold and silver(16%) and some impurities such as copper and iron were found. Therefore, it could be inferred that this miniature pagoda is a gold product.

Production of protein hydrolysate and plastein from alaska-pollack (명태단백 가수분해물 제조 및 plastein의 합성)

  • In order to enhance the processing quality and utility of alaska-pollack meat, the optimum conditions for the preparation of pronase hydrolysate and the synthesis of plastein were investigated. The optimum temperature and pH for the hydrolysis of alaska-pollack by pronase were $40^{\circ}C$ and pH 7.0. The reaction time and enzyme concentration were 4 hr and 1,000 units per g of substrate. Under the above optimum conditions alaska-pollack was hydrolyzed by pronase yielding a hydrolytic degree of about 89%. Pronase hydrolysate was employed as substrate for plastein synthesis. The 30% pronase hydrolysates were adjusted to pH 7 for fruit-bromelain and pH 5 for stem-bromelain, and then plastein were synthesized by 1% bromelain at $40^{\circ}C$ for 24 hr. The plasteins synthesized by fruit- and stem-bromelain were consisted of peptides having average peptide length of 22.6 and 20.8 under the optimum synthetic conditions. The plastein synthesis reaction reduced considerably the bitterness of pronase hydrolysate.

  • This study was investigated the changes of gel properties about pH, color and water holding capacity at various hydration time and temperature of food-grade additives (BPP: bovine plasma protein, DEW: dried egg white, SPI: soy protein isolate). The changes of rheological properties were checked about hydration time and temperature. Hydration time and temperature affected pH value, hydration decreased pH of SPI and DEW. The BPP was not influenced at hydration time and temperature. Some Hydration condition increased jelly strength of food-grade additive, but SPI did not form a gel at all hydration condition. Hydration increased lightness of food-protein.

  • This study was attempt to improve the quality of rapid- and low salt-fermented liquefaction of sardine (Sardinops melanoslicta). Effect of pretreatment methods such as water adding, heating, and intermittent NaCl adding on fermented liquefaction of chopped whole sardine were investigated. The divisions of the experimental samples by pretreatment methods were as follows; Sample A (water adding and heating): chopped whole sardine adding 20% water and then adding 3 and 5% NaCl consecutively at the intervals of 3 and 6 hrs during heating for 9 hrs at $50^{\circ}C$ and then fermented at $33^{\circ}C$ for 90 days. Sample B (preheating): chopped whole sardine with 8% NaCl and heating at $50^{\circ}C$ for 9 hrs and then fermented at $33^{\circ}C$ for 90 days. Sample C (control): neither pretreatment methods of water adding nor preheating on chopped whole sardine with 13% NaCl and then fermented at $33^{\circ}C$ for 90 days. Comparison of the appropriate fermentation period, yield of hydrolysate, chemical composition of fermented liquefied products were carried out. The highest content of amino nitrogen appeared at 60 days in the sample A, 75 days in the sample B, and 90 days in the sample C during the fermentation period. The appropriate fermentation period of the sample A was shorten 15 days than the sample B and 30 days than the sample C in the processing of sardine. The product A was lower NaCl (8.5%) and lower histamine content (25mg/100g) than the sample B and C. Possibly, three kinds of pretreatment methods such as water adding, heating, and intermittent NaCl adding, might be recommend as the processing of rapid- and low salt-fermented liquefaction product of chopped whole sardine.

  • 다국적 제약회사에서 생산되는 제약화합물은 인간의 질병치료 뿐만 아니라 가축의 생산성을 향상시키는 물질으로도 활발하게 생산되는데 육우의 성장을 촉진하는 기능성 화합물들도 많이 있다. 북미에서 생산되는 소고기의 약 80%가 1번 이상의 성장촉진기술들이 활용되고 있다고 보고된다. 고기소로 생산되는 비육우에 적용되는 방법은 크게 피하 이식에 의해 혈류를 타고 성장을 촉진하는 17β-Estradiol과 합성 남성호르몬제인 Trenbolone acetate가 주로 활용되고, 비육후기 사료에 섞어서 급여하는 사료첨가제 형태인 β2-adrenergic agonist 같은 형태로 적용하게 된다. 근육을 성장하는 기술의 작용기전은 많은 선행연구에 의해 밝혀져 있는 반면 천연 알칼로이드 성분들의 기전은 밝혀진 것이 많지 않다. 한방제재들에서 많이 발견되는 알칼로이드 성분들은 생리활성 기능들을 가진 것으로 알려져 있지만, 생산, 수거, 가공, 추출 등의 공정에서 많은 비용이 발생하므로 비육우의 사료화 가능성은 아주 희박하다. 따라서 비용을 최소화 할 수 있는 후보재료를 검색중에 감자부산물을 확보하였고 기능성 물질의 추출과 사료첨가제 화 하여 비육우에 급여시험을 실시하였다. α-solanine과 α-chaconine은 감자의 잎, 과일 및 괴경에서 발견되는 글리코알칼로이드 화합물로, 쥐, 토끼, 닭과 같은 다양한 동물 모델에서 중독성을 가진 물질로 보고 되고 있다. 최근 연구에서는 비육우의 성능을 유도하는 데 사용되는 것으로 나타났다. 한우 송아지 3마리의 사태(Semimembranosus)와 등심(Longissimus Dorci)근육에서 추출된 근육위성 세포(BSC)에 다양한 수준의 α-solanine(control, 0.001, 0.01, 0.1, 1, 10μM)으로 처리해본 결과 근육관련 지표인 MHC2X과 β2-AR의 발현이 높게 나타난 것을 확인했다. 사료급여실험에서는 대조군에 비해 급여군의 등심단면적과 도체중이 향상되는 것을 확인하였다. 결과적으로 감자유래 농산부산물은 한우 비육우의 근육의 성장을 증가시키고, 그 작용기전은 β2-수용체에 작용하여 단백질 합성을 촉진시켜 근육을 축적시키는 것으로 확인하였다. 농산부산물을 이용한 기능성 사료개발은 최근 이슈가 되고 있는 축산분야 탄소저감을 개선할 수 있는 기술로 축산의 업사이클링 기술로 활용 가능하다.

  • A lot of water soluble proteins and lipids are released from minced mackerel meat and lost into the washing waste during the leaching process of Kamaboko or surimi manufacture. The removed proteins and lipids are not only an edible things but also a big burden for treating the wastewater. In order to recover the proteins from the effluent and to use as food stuff, the "pH-shifting" treatment, a modified isoelectric point precipitation method, was tried. This method is based on a myogen-aggregation phenomenon, which occurs when a solution of sarcoplasmic proteins is acidified or alkalified beyond the critical pH zone of 2∼3 or 12∼13 respectively and then neutralized. The maximum amount of precipitation was obtained by shifting the pH of the wastewater from original pH to isoelectric point (pH 4) or alkali pH 12 and then changing to neutral pH. The precipitates were easily collected by filteration or centrifuging at 10,000rpm. The oils which were only floating in the washing wastewater are easily recovered by seperating with oil separator after pouring. The recovered proteins were slightly denaturated during this pH shifting precipitation process, while the composition of amino acids was good balance as a food.

Sex Identification of Bovine Meat Using Male Specific SRY and ZFY Genes (웅성 특이적 SRY 및 ZFY 유전자를 이용한 쇠고기 성(性) 판별)

  • The objective of this study was to develop a rapid and reliable method for the sex determination of beef using the PCR(polymerase chain reaction) technique. We have used two bovine sex determining genes, SRY and ZFY, on the Y-chromosome to identify the sex of Hanwoo and Holstein beet. We attempted to amplify 1,348 bp and 979 bp fragments from male and female genomic DNA corresponding to the SRY and ZFY genes, respectively, using male specific primers. The amplified PCR products were separated by electrophoresis in a 1.5% agarose gel to detect a male specific DNA band. When DNA from male beef was amplified with primers specific for the SRY gene, a DNA band of 1,348 bp was present in all of the male samples, but absent from all of the female samples. Also, when DNA from male beef was amplified with primers specific for the ZFY gene, a DNA band of 979 bp was observed in all of the male samples, but absent from all female samples. In conclusion, the bovine SRY and ZFY genes are typically found only in male beef. For the practical application of this method for the sexing of commercial beef at the processing and marketing stages after slaughter. a total of 350 beef samples collected randomly from local markets were analyzed for sex determination. The proportions of male and female samples were 252 (72%) and 98 (28%), respectively. Therefore. the SRY and ZFY genes. which are specific for the Y-chromosome, may be useful sex-diagnostic DNA markers to distinguish male meat from female meat.
