Parasitism of Liriomyza trifolii (Diptera: Agromyzidae) by Hemiptarsenus zilahisebessi(Hymenoptera: Eulophidae) on Tomato (토마토에서 Hemiptarsenus zilahisebessi에 의한 아메리카잎굴파리 기생율)

  • This study was conducted to investigate percentage of parasitism and control effect of Liriomyza trifolii by Hemiptarsenus zilahisebessi on tomato. Percentages of parasitism on L. trifolii larva by ectopatasitoids war e 26∼45% Among them the parasitism by H. zilahisebessi was highest as 47∼75% in tomato. The parasitoids preferred 1 st to 3rd instar of host larvae. In laboratory test, the parasitoids showed high parasitism on 3rd instar larvae of host by 89.8∼93.1% when the female parasitoids were introduced by the ratios of 1 : 10, 1 : 20, and 1 : 30. In field test, 3 or 5 female parasitoids were introduced per plant. In the case, the parasitism increased to 80% 4 weeks after introduction of the parasitoids. This increased parasitism was resulted from density reduction of the host larvae There were no significant differences in parasitism, density of alive host, and percentage of damaged leaf between inoculation density.

  • An in vitro immune effector mechanism against the target encysted metacercariae of Paragonimus westermani was demonstrated in the rat system. Peritoneal exudate cells, mainly macrophages from normal rats, showed adherence to and killing of encysted metacercariae of p. westermani in the presence of complement-independent serum from rats infected with Paragonimus metacercariae. These reactions were specific for the excysted metacercariae, as tissue-migrating juvenile worms were not affected. Damage of encysted metacercariae of p. westermani due to antibody and macrophages was assessed by morphological observation, by cell adherence reaction and by the use of vital dyes. frypan blue dye exclusion proved to be a reliable indicator of judging metacercarial viability. Electron microscopic studies demonstrated that macrophages reacted with fusty material on the tegumental surface and fine structures in the syncytium of the parasites. The tubular tunnels formed between the basement membrane and muscle layers of the damaged parasites were also noticeable. The relevance of these findings to cellular immunity in the early paragonimiasis was discussed.

  • Predation efficiency and preference of the hydrophilid water beetle, Hydrochara affinis (Coleoptera: Hydrophilidae), larvae on two mosquito species, Culex pipiens molestus and Ochlerotatus togoi, were tested under laboratory conditions. Experiments were conducted in a transparent cylindrical plastic container (diameter, 100 mm; height, 40 mm; water level, 15 mm) for 24 hours (16 h, light; 8 h, dark) at $25^{\circ}C$. The predation efficiency of H. affinis on mosquito larvae was investigated under various prey conditions (1, 2, 4, 8, 16, 32, 64, and 128 larvae) with five replicates. Further, in order to investigate the prey preference on two mosquito species, each 32 larvae of C. pipiens molestus and O. togoi were provided simultaneously for the first instar larva of H. affinis with 5 replicates. As a result, the predation curves of H. affinis on two mosquito species were logarithmic; and the number of consumed prey increased as the number of predator instars were increased. As predicted from the predation curves, the estimated maximum number of C. pipiens molestus and O. togoi consumed by a third instar larva of H. affinis per a day was 926 and 304 larvae, respectively. As predicted from the Holling's disk equation, the differences in consumed prey number between predator instars and prey species were caused by handling time rather than attack rate. Handling time decreased rapidly as the predator larvae grew, and that of O. togoi was twice longer than that of C. pipiens molestus.

  • The diapause induction season in Ostrinia furnacalis (Lepidoptera: Crambidae) was estimated in Suwon. Three batches of adult generations were observed, the first one from early May to early July, the second from early or mid-July to early or mid-August, and the third from mid-August to October. In outdoor larval rearing, colony rearing occurring from mid-July to mid-August produced both non-overwintering and overwintering larvae, whereas late-reared colonies produced only overwintering larvae. Larvae collected during July and August in maize fields produced both non-overwintering and overwintering larvae, whereas late-collected larvae produced only overwintering larvae. The results indicated that O. furnacalis has a bi- or trivoltine complex life cycle in this area. In the laboratory, when larvae of all instars within 9 h after molting were first treated to a diapause induction condition (11:13 h = light:dark photoperiod and 20℃), almost all larvae were induced to diapause. However, when similar treatments were conducted age-specifically for the 5th instar larvae, diapause induction rates in 3- and 4-day-old larvae of the 5th instar decreased. In contrast, when larvae were subjected to the diapause induction treatment only during the periods from the hatching stage to the 2nd, 3rd, and 4th instar, almost all larvae were not induced to diapause. The results suggest that the early age of the 5th larval instar is the last stage for sensitivity to diapause induction stimuli. In the diapause-induced larvae, hemolymph trehalose content increased and body supercooling points dropped, compared with those in non-diapause larvae.

Optimal Larval Density and Low Temperature Storage Conditions for Rearing of Protaetia brevitarsis (Coleoptera: Cetoniidae) using a Fermented Mulberry Sawdust-base Diet (뽕나무발효톱밥을 이용한 흰점박이꽃무지(딱정벌레목: 꽃무지과) 유충 사육에서 적정 사육밀도 및 저온처리 조건)

  • We raised the white-spotted flower chafer, Protaetia brevitarsis seulensis, from the larvae stage using a fermented mulberry sawdust-base diet at 25℃ and 16:8 h light:dark photoperiod. First, we determined the optimal density of the neonate larvae to be introduced into a rearing box (543 × 363 × 188 mm). The survival rates of the larvae were higher than 80% at 90 days after rearing at 100-175 larval densities but reduced by more than 10% at 200 larval density. The larval weights at 100 to 150 larval densities were similar; however, the weights at 175 and 200 larval densities were lower than those at 100 larval densities, indicating that the rate of weight gain increased under lower density. Based on these results, we inferred that 100-150 larvae was the optimal density. Second, we investigated the storage conditions of the last instar larvae under low temperatures. Four weight groups (1.8-2.0, 2.0-2.3, 2.3-2.5, and 2.5 g) of larvae were stored at 4, 8, and 10℃, respectively. All the larvae at 4℃ died 70 days after storage, whereas 80% of the larvae survived at 8℃ 70 days after storage, indicating that high larval weight was associated with high survival rates. The adults that emerged from larvae with more than 2.3 g and stored for up to 50 days at 4℃ laid few eggs. However, the adults that emerged from all larval weight groups stored for 70 days at 8℃ and 10℃ laid several eggs successfully. Based on these results, we inferred that the last instar larvae with more than 2.3 g could be stored for 30-50 days at 8℃.

Infection status of the sea eel (Astyoconger muyinster) purchased from the Noryangjin fish market with anisakid larvae (노량진 수산시장에서 구입한 붕장어(Astroconger myriasser)의 아니사키스 유충 감염 상황)

  • Although the sea eel (Astroconger myriaster) is suspected as one of the most important fish host for human anisakiasis in Korea, no report has been made on the infection status of the sea eel with anisakid larvae. In the present study, 26 sea eels (Astroconger myriaster) were purchased from the Noryangjin 6sh market in Seoul, and anisakid larvae were collected from their viscera, muscle, head and skin. The collected larvae were classified by their morphological types. A total of 1,351 anisakid larvae were collected from 15 of 26 fish examined. Among them, 1,269 were recovered from the viscera, 66 from the muscle, and 16 from the head and skin. Morphologically, most of the anisakids were classified into 6 known larval types, Anisakis type I (564 larvae) of Berland(1961) , Contracaecum type A(409) and type D(5) of Koyama et at. (1969), Contracaecum type C'(83) and type D'(117) of Chai et at. (1986), and Contracaecum type V(1) of Yamaguti (1935). Remaining 172 specimens were new in the available literature, hence, designated as Centracaecum type A'(new type). The present results revealed that the sea eels caught in the Korean waters are heavily infected with anisakid larvae, not only in their viscera but also in the muscle, and Anisakis type I was the most common among the 7 larval types.

An Improved Collecting Method of the Infective Juveniles of the Entomopathogenic Nematode, Steinernema carpocapsae Weiser (감염태 곤충병원선충(Steinernema carpocapsae Weiser)의 효과적 회수법)

  • We report here an improved collecting method of the infective juveniles multiplied in the host insect with the entomopathogenic nematodes, Steinernema carpocapsae Weiser. The specific characteristics of this method involved the opening of the host insect hemocoel after the population of their infective juveniles reached at the maximum (6 days at 25$\^{C}$ after nematode treatment to nonimmunized host insects) to facilitate the escape of the multiplied nematodes. It also used 'Baermann funnel'method to select the infective juveniles effectively. This improved 'Baermann funnel'method was compared with a traditional collecting method, which was characterized with a combination of untreated host insects and 'White trap'collecting method, in both yield and pathogenicity of the collected infective juveniles to the fifth instar larvae of beet armyworm, Spodoptera exigua (H bner). More than 95% of the nematode populations collected by the two methods represented the morphological infective juveniles. To prove the nematodes to be infective juveniles functionally, pathogenicity and infective activity were compared in the nematodes collected by the two methods. They were not different in both pathogenicities and infective activities which were measured by the numbers of nematodes penetrated into the hemocoel of the insect hosts after exposure for the specific times to the same dote of infective juveniles. Significant difference between two collecting methods was found in the total yields of the infective juveniles per host insect About 50,000 infective juveniles per infected fifth instar larva of S. exigua after 6 day incubation at 25$\^{C}$ were collected only for 2 days by the improved 'Baermann funnel'method, while about 20,000 infective juveniles per host were collected for 10 days by the classical 'White trap'method.

Characteristics of the Aggregation Pattern of the Striped Rice Borer(Chilo suppressalis(Walker) during the Larval Stage (이화명나방유충(幼蟲)의 공간분포특성(空間分布特性))

  • The data on the striped rice, borer populations, collected in the four years from 1973 to 1976, was reanalyzed to study the characteristics in the aggregation pattern of the insect larvae in the paddy fields. The distribution pattern of the larvae was well fitted to the negative binomial model in both the first and the second generation. With reference to the Green's coefficient of dispersion estimated, the aggregation of the larvae was categorized into three phases: initial high aggregation, changing, and dispersed stable phase. Except the changing phase, each phase in each generation could be defined by a common k. The phase-change was initiated by the larval dispersal between hills of the rice plants: the larvae of the third stage and those of the fifth stage, for the first and second generation, respectively. The characteristics of the aggregation pattern were stable in the second generation. In the first genaration, the pattern was more or less variable, indicating that the life system of the insect in the first generation was more susceptible to the variations of the external physical factors than that in the second generation.

A comparison of the hydrolase activities of excretory-secretory products and somatic extracts from fish parasitic nematodes, Anisakis simplex sensu stricto and Anisakis pegreffii larvae (어류 기생성 선충 Anisakis simplex sensu stricto와 Anisakis pegreffii 유충의 excretory-secretory products 및 somatic extracts의 가수분해효소 활성 비교)

  • Hydrolase activities of excretory-secretory products (ESP) and somatic extracts (SE) from Anisakis simplex sensu stricto (s.s.) and Anisakis pegreffii larvae were investigated by using API ZYM kit. In esterase group, acid phosphatase showed high activity from both of A. simplex (s.s.) and A. pegreffii. Esterase (C4) showed activity only from SE and A. simplex (s.s.) showed higher activity than A. pegreffii. Alkaline phosphatase, acid phosphatase and naphthol-AS-BI-phosphohydrolase showed higher activity in 3rd stage larvae than in 4th stage larvae of both species. In aminopeptidase group, only leucine arylamidase showed remarkable activity in SE of both anisakid species, and A. simplex (s.s.) SE showed higher activity than A. pegreffii SE. In glycosidase group, N-acetyl-${\beta}$-glucosaminidase, ${\alpha}$-mannosidase, ${\alpha}$-fucosidase showed higher activity in A. simplex (s.s.) than A. pegreffii, and 4th larvae showed higher activity than 3rd larvae. These differences in hydrolase activity of anisakid nematodes larvae are thought to be due to different metabolism such as growth, moulting, digestion and feeding.

Life history of Echinopowphium recurvatum (Trematoda: Echinostomatidae) in Korea (한국에서의 오리극구흡충 (Echinoparyphium recurvatum)의 생활사)

  • The present study was performed to observe characteristics of the lifE history of Echinoparvphium recunpatum under both natural and laboratory conditions in Korea. A batch of Rcnix aunculnria corecna was collected from Sunamchon, one of the stream of West Naktonggang (River), in Kangso-gu, Pusan during August and September 1992. Out of 106 snails examined by crushing,52 (49.0%) were infected with larval E. recurvaum i.e. rediae, cercariae and metacercariae. Cercariae naturally shed from snails encysted in the snails of same species and loaches, but not in mud-snails. Adult worms were detected from chicks and ducks experimentally infected with metacercariae, but not from rats and mice. The average recovery rate of adults from chicks was 13.1%. Rediae were sac-like, $2.437{\;}{\times}{\;}0.317{\;}mm$ in average size, with a muscular pharynx and a brownish cecum which reached the anterior half of the body. Cercariae consisted of a spindle-shaped body ($0.262{\;}{\times}{\;}0.129{\;}mm$ in average) and a rod-like tail ($0.528{\;}{\times}{\;}0.056{\;}mm$ in average). In the cercarial body, 45 collar spines were observed on the head crown, and double rows of excretory ducts with fine granules were laterally arranged between the pharynnx and the ventral sucker. Metacercariae were spherical, $0.144{\;}{\times}{\;}0.142{\;}mm in average size, with thick hyaline outer and thin elastic inner walls, and many excretory granules. Adults were slender and more attenuated in the anterior end, $0.2760{\;}{\times}{\;}0.550{\;}mm$ in average size, and had 45 collar spines including four end group spines on both ventral corners. From the above results, it was confirmed that R. auriculnna corennc plays a pivotal role in the life cycle of 5. recuwatum as the first and/or second intermediate hosts in Korea.
