Transformation of Populus alba × P. glandulosa by Agrobacterium rhizogenes (Agrobacterium rhizogenes 에 의한 현사시나무의 형질전환(形質轉換))

  • The widely cultivated hybrid poplar Populus alba ${\times}$ P. glandulosa in Korea was transformed with Agrobacterium rhizogenes agropine type strain A4. Genetic transformation was confirmed by the presence of agropine. Alteration in growth rate of hairy roots was seen following changes in the dilution rate of medium and concentration of sucrose, suggesting that improved growth might be achieved by more precise manipulation of the nutrient medium. Plant regeneration occurred from transformed hairy roots on MS medium supplemented with 0.5 mg/l BAP. Transformed plantlets grown in vitro exhibited a more developed root system characterized by fast growth behavior in comparison to normal plantlets. This work demonstrates that the root transformation would be useful in improving plantlet establishment and growth through the effective root system.

  • Six flue- cured cultivars of Nicotiana tabacum L., 15 possible $F_1$ hybrids and $F_2$ Populations among them, and 15 haploid Populations from Fl hybrids and haploids from Parents, were evaluated. Comparisons of the $F_1$, hybrids and $F_2$ Populations with the Parents indicated that heterosis values were small but significant for yield, plant height, days to flower, leaf length and width and total alkaloids from-6.0% to 5.4% in $F_1$ hybrids, and from -3.4% to 3.6% for Plant height days to flower leaves per plant in $F_2$ populations, respectively. There were positive correlations for yield, plant height, days to flower, leaves per plant and total alkaloids between diploid and haploid populations. Increase or decrease ratio of haploids to diploids of total alkaloids was appeared to 30.3%. Those for yield, leaf length and width, value arid reducing sugar were ranged from -4.1% to -27.6%.

Analysis of Phase Noise of High Stable Microwave Phased Locked Oscillator with Gate Voltage Tunning (게이트 전압 제어에 의한 마이크로파 고안정 위상동기발진기의 위상잡음 특성 분석)

  • In this paper, we design a high stable Ku-band phase-locked dielectric resonant microwave oscillator with the gate voltage controls of p-HEMT. By adapting the nonlinear equivalent elements which affects phase noise of microwave oscillator, we optimize the nonlinear elements of p-HEMT to have low phase noise operation. Using the scattering parameters according to bias voltages, we designed the gate voltage control microwave dielectric resonant oscillator and phase-locked loop circuits is applied to have the high stable operations. Designed microwave oscillator as a local oscillator of digital microwave communication shows that output power is 9.17dBm at 10.75GHz and it's phase noise is -88dBc/Hz at 10KHz offset frequency.

  • 최근 미관을 위하여 소형 네온관 등이 사용되고 있다. 따라서 네온관 등 구동에 사용되는 인버터는 안정기 역할을 하면서 소형화와 경량화가 중요한 요소이다. 본 연구는 압전 특성이 우수한 PZT계 세라믹스 조성인 $(Pb_{0.94}Sr_{0.06})[(Ni_{1/2}W_{1/2})_{0.02}(Mn_{1/3}Nb_{2/3})_{0.07}(Zr_{0.51}Ti_{0.49})_{0.91}]O_3$에 특성 향상을 위해 PbO, $Fe_2O_3$, $CeO_2$, Xwt% $Nb_2O_5$ 조성별로 첨가하였으며 그에 따른 유전, 압전 특성을 조사하고 또한 Rosen type의 압전 트랜스포머를 제작하여 각 조성별 변환효율을 측정하였다. 실험방법은 일반적인 세라믹스 제조공정으로 파우더 혼합 후 24시간 ball milling하고 $850^{\circ}C$ 에서 2시간 하소 후 $1230^{\circ}C$에서 2시간 소결하였다. 또한 최종 소결 시편을 이용하여 Rosen type의 압전 트랜스포머를 제작하였다. 상 분석을 위해 XRD를 이용하여 perovskite구조를 확인하고 미세구조확인을 위해 SEM으로 관찰하였다. 압전 특성을 평가하기 위해 압전 $d_{33}$ Meter를 사용하였으며, Impedence analyzer HP 4194A를 이용하여 전기적 특성을 측정하였다.

Effects of Variety and Acetosyringone Influencing Transformation of Rice Mediated by Agrobacterium tumefaciens (Agrobacterium tumefaciens에 의한 벼 형질전환에 미치는 품종과 Acetosyringone의 영향)

  • The cytosolic glutathione reductase (GR) gene of Brassica campestris L. was introduced into several Japonica cultivars of rice by Agrobacterium tumefaciens and a large number of transgenic plants were produced. Three-week old calli were co-cultivated with A. tumefaciens strain EHA101 carrying the plasmid pIGR1. The efficiency of transformation was differed from rice cultivars. A Japonica cultivar, 'Daeribbyeo' appeared the highest efficiency (42.5%) of transformation among the four cultivars tested. The addition of acetosyringone (50 $\mu$M) during co-cultivation was a key to successful transformation. Transgene fragments were identified by PCR amplification and further confirmed by Southern blot analysis. Mendelian inheritance of the transgenes was confirmed in T$_1$ progeny.

  • This experiment was conducted to determine the optimum dosage of gamma rays for inducing artificial mutation of several mutant characters in the flue-cured tobacco. 1) In Hicks and BY 104, the gammarays irradiation has no significantly different effect on seed germination from the control. However, the average dosage for 50% growth inhibition was 25-30kr for all the varieties tested, which inhibition 46-52% and 43-57% of the seedling growths for Hicks and BY 104, respectively. 2) A mutant line 83H-5 was selected from Hicks by irradiation gamma ray at the level of 30kr. It has white flower, more resistance to bacterial wilt, Pssudomonas solanacearum, lower plant and stalk height, narrower leaf width, larger leaf shape index(lento width) and later days to flower when compared with the original variety Hicks. 3) White flower was recessive to pink flower in F, and Br (F1 X Hicks) progenies. F2 population of the cross gave segregation ratio of 3 pink flower:1 white flower, and B, (F1 X 83H-5) Population gave 1:1 ratio. Results showed that the white flower character is governed by a single recessive gene.

The complete genome sequence of a white spot syndrome virus isolated from Litopenaeus vannamei (흰다리새우(Litopenaeus vannamei )에서 분리된 WSSV의 전장유전체 분석)

  • The full genome sequence of a Korean white spot syndrome virus (WSSV, isolate: WSSV-GoC18) is presented here. We obtained a total of 12,320,554 reads with 291,172 bases, 170 gene, and 170 coding DNA sequence, which were assembled in 1 contig. Phylogenetic analysis revealed that the WSSV-GoC18 was closely related to Chinese isolate (WSSV-PC) and distinctly different with previously reported a Korean isolate (WSSV K-LV1). The complete genome sequence of WSSV isolates will be of great help in molecular epidemiological studies, contributing to molecular diagnosis and disease prevention in shrimp aquaculture.

  • The upper limit of solid solution of $Al_2O_3$ in $LiTaO_3$ was investigated using X-ray diffraction and Raman spectroscopy. By substituting cations in $LiTaO_3$ with $Al^{3+}$, the melting temperature was lowed and the ferroelectric properties can be improved. It is easier at lower temperature to fabricate the single crystal used for SAW filters and IR sensors. From the measured lattice constants and Raman band broadening, the solubility limit was X=0.25mol in $Li_{1-X}Al_{2X}Ta{1-X}O_3$, above which $Al_2O_3$ was obsered as a second phase. The Raman band of sintered $LiTaO_3$ was compared with that of the single crystal to see the effect of grain size on the band broadening.

Gene Expression Profiles and Antioxidant Effects of Houttuynia cordata Thunb Extract in Human Keratinocyte HaCaT Cells (인간 피부각질세포 HaCaT에서 어성초 추출물의 유전체 발현 분석 및 항산화 효과)

  • Based on the antioxidative effects in organic solvent fractions obtained from the main methanolic extract of Houttuynia cordata Thunb, the cytoprotective effects by oxidative-stress were here analyzed. Regarding the antioxidant activity of organic solvent fractions, the electron-donating ability of DPPH increased in a dose-dependent manner, and $ED_{50}$ exhibited the highest concentration at $175{\mu}g/ml$ in the Hc-EtOAc fraction. The cell viability of Hc-EtOAc fractions on $H_2O_2$-induced HaCaT cell death ($IC_{50}$) increased in a concentration-dependent manner and a visible cell survival rate of 74% was observed at a concentration of $100{\mu}g/ml$. Meanwhile, the gene expression patterns in HaCaT cells treated with $100{\mu}g/ml$ of the Hc-EtOAc fraction for 6 and 24 hr were identified with microarray analysis. The genes involved in signal transduction, cell division, antioxidant activity, and epithelial cell proliferation were found to be 2-fold up-regulated genes in HaCaT cells following the Hc-EtOAc fraction treatment. Especially, proinflammatory cytokines (IL1B, TNF, and IL6) were identified as involved in antioxidant activity based on the expression patterns of the HaCaT cells, and pathway analysis indicated that TLR4 might be considered an upstream regulator of these genes. In order to verify the activity of IL1B, TNF, and IL6, qRT-PCR showed that the expression increased more than 2 times in HaCaT cells treated with at least $100{\mu}g/ml$ of the Hc-EtOAc fraction. The activity of the upstream regulator TLR4 protein was also increased by the Hc-EtOAc fraction. As a result, the antioxidative activity of the Hc-EtOAc fraction is predicted to pass from TLR4 through cytokines such as IL1B, TNF, and IL6.

Analysis of right border flanking sequence in transgenic chinese cabbage harboring integrated T-DNA (Agrobacterium을 이용하여 형질전환시킨 배추에서 T-DNA Right Border 인접염기서열 분석)

  • We developed 14 transgenic lines of Chinese cabbage (Brassica rapa) harboring the T-DNA border sequences and CryIAc1 transgene of the binary vector 416 using Agrobacterium tumefaciens-mediated DNA transfer. Six lines had single copy cryIAc1 gene and four of them contained no vector backbone DNA. Of the left border (LB) flanking sequences six nucleotides were deleted in transgenic lines 416-2 and 416-3, eleven nucleotides in line 416-9, and 65 nucleotides including the whole LB sequences in line 416-17, respectively. And we defined 499 bp of genomic DNA (gDNA) of transformed Chinese cabbage, and blast results showed 96% homology with Brassica oleracea sequences. PCR with specific primer for the right border (RB) franking sequence revealed 834 bp of PCR product sequence, and it was consisted of 3' end of cryIAc1, nosterminal region and 52 bp of Chinese cabbage genomic DNA near RB. RB sequences were not found and the 58 nucleotides including 21 bp of nos-terminator 3' end were deleted. Also, there were deletion of 10 bp of the known genomic sequences and insertion of 65 bp undefined genomic sequences of Chinese cabbage in the integration site. These results demonstrate that the integration of T-DNA can be accompanied by unusual deletions and insertions both in transgenic and genomic sequences.
