Genetic Analysis of Three Overwintering Diamondback Moth, Plutella xylostella (Linne), Populations in Korea (국내 세 지역의 배추좀나방(Plutella xylostella (Linne)) 월동집단에서 나타나는 유전변이 분석)

  • Four polymorphic allozymes were used for genetic analysis of overwintering populations of field diamondback moth, Plutella xylostella (Linne). Different allele frequencies were found among three local populations of Andong, Youngchon, and Yangsan in all loci. Two allozyme loci (acid phosphatase and phosphoglucomutase) showed significant deviation from Hardy-Weinberg equilibrium in allele frequencies among these populations. Estimated Nei's genetic distance varied from 0.0151 between Yangsan and Youngchon to 0.0877 between Andong and Youngchon. Compared with the previous genetic distances in this moth, a little higher genetic differentiation among these overwintering populations suggests that there would be a specific genetic bottleneck in each local population during overwintering period.

  • In this paper, determination of the scalability of the cluster composed common personal computer was performed when optimization of reinforced concrete structure using genetic algorithm. The goal of this research is watching the potential of multi-core-PC cluster for optimization of seismic design of reinforced-concrete structures. By increasing the number of core-processer of cluster, decreasing of computation time per each generation of genetic algorithm was observed. After classifying the components in singular personal computer, the estimation of the expected bottle-neck phenomenon and comparison with wall-clock time and Amdahl's law equation was performed. So we could obseved the scalability of the cluster appear complex tendency. For separating the bottle-neck phenomenon of physical and algorithm, the different size of population was selected for genetic algorithm cases. When using 64 core-processor, the efficiency of cluster is low as 31.2% compared with Amdahl's law efficiency.

Genetic Differentiation of Overwintering Populations of Oriental Fruit Moth, Grapholita molesta, and Their Movement (월동세대 복숭아순나방(Grapholita molesta)의 지역적 분화 및 이동)

  • Spring phonology of the oriental fruit moth, Grapholita molesta, was monitored using sex pheromone traps in apple cultivating areas. Their occurrence was earlier in southern areas and their population sizes were significantly different among orchards even in a local cultivating zone. The overwintering populations appeared to move between local orchards, based on the fact that monitoring data obtained at the sites between orchards were similar to those of nearby orchards. However, within orchards, these adult movements appeared to decrease and showed skewed occurrences at the side of upwind direction or close to neighboring orchards. At initial occurrence peak(April 20-25), the ovenvintering populations of the different localities were collected and analyzed in their genetic distances. PCR-RAPD analysis indicated that there were significant genetic differences among the overwintering populations of G. molesta. This genetic differentiation of overwinterin populations may be due to genetic bottleneck following differential selection pressures against the subpopulations of G. molesta during winter on the basis of the RAPD analysis that each early spring population was significantly different to its previous fall population in the same locality.

  • Enzyme electrophoresis was used to estimate genetic diversity and population genetic structure of Pseudobagrus fulvidraco in Korea. Nine of the 14 loci (64.3%) showed detectable polymorphism. Genetic diversity at the population and species levels were 0.286 and 0.277, respectively. Analysis of fixation indices, calculated for all polymorphic loci in each population, showed a substantial deficit of hetero-zygotes relative to Hardy-Weinberg expectations. This deficit is expected that it is due to a limited effective number of individuals per population. The average $G_{ST}$ for polymorphic loci was 0.064, indicating that most (93.6%) of the genetic diversity occurred within populations. The indirect estimate of gene flow based on mean $G_{ST}$ was 3.67. Given limited gene flow is expected to diverge genetically due to drift and reduced populations. Most populations in our study experience annual, severe demo-graphic bottlenecks due to drought and floods.

Mitochondrial DNA Swquence Variation of the Firefly, Pyrocoelia rufa(Coleoptera: Lampyridae), in Korea (늦반딧불이 Pyrocoelis rufa(딱정벌레목: 반딧불이과)의 미토콘드리아 DNA 염기서열 변이)

  • We have sequenced a portion of mitochondrial CO! gene (403 bp) of the firefly, Pyrocoelia rufa, to investigate genetic diversity within population, geographic variation, and phylogenetic relationships among haplotypes. A total of seven mtDNA haplotypes ranging in sequence divergence from 0.2% to 1.2% were obtained from 26 fireflies collected at four localities in Korea: Namhae, Pusan, Muju, and Yongin. The samples collected at the urban area, Pusan, were all fixed with one haplotype, differently those collected at the forest and/or agricultural areas. This appears to suggest that habitat fragmentation and population bottleneck caused by urbanization might have been severe in Pusan. On the other hand, from Muju known as the largest habitat and sanctuary for the firefly, four haplotypes with the maximum sequence divergence of 1.0% were obtained, and this estimate was the highest among the areas studied. The fireflies collected at the isolated islet, Namhae, revealed relatively low haplotype diversity(H=0.25), but one haplotype (PR7) was phylogenetically differentiated from others. This phenomenon was explained in terms of biogeographic history of the island and gene flow in the recent past. Grouping of Muju- Y ongin and Pusan-Namhae, respectively, in the hierarchical genetic analysis suggests the presence of historically occurred, biogeographic barrier against gene flow between them.

  • The travel speed between two locations within the downtown differs according to time horizon and district. Also, There is delay time on numerous traffic signals and bottle neck areas. It has an influence on planning the vehicle routing. However, there are almost no studies focusing on delay time for distance and travel time between two locations among the existing researches for vehicle routing problem (VRP). In this paper, we approach the real VRP by designing the model which estimates the delay time for traffic signal and bottle neck areas. The results of computation experiment demonstrate that proposed method performs well and have better solution than other method not considering the delay time.

  • Salix koriyanagi is a deciduous shrub and native to Korea. The spatial distribution of multilocus allelic frequencies and geographical distances of the natural population in upper swampy area at the Namgang River in Korea were studied. The species showed a significant positive and negative spatial autocorrelation according to geographical distances as measured by Moran's I. Genetic similarity of individuals was found among subpopulations at up to a scale of a 12 m distance, and this was partly due to a combination of allelic frequencies, and therefore, a significant spatial autocorrelation was composed of a scale of 12 m intervals. Within S. koriyanagi in swampy area at the Namgang River, a strong spatial structure was observed for allozyme markers, indicating a migration within subpopulations.

감압증류법에 의한 $^{32}$P 제조

  • $^{32}$P는 순수한 $\beta$$^{-}$ 방출핵종(방출에너지 = 1.71 MeV, 반감기 = 14.3일)이며 의료용, 표지화합물 합성용, 유전공학 실험용 등으로 널리 사용되므로 고품질의 $^{32}$P의 수요에 부응하기 위해 감압증류법을 개발하였는 바 그 방법과 결과는 다음과 같다. 연구로 2호에서 중성자 조사된 황 표적을 감압 증류용기내에서 5~10 mmHg의 감압하에 200~30$0^{\circ}C$로 가열하여 황을 증류해 낸 다음 묽은 염산을 역류시켜 넣고 $^{32}$P 를 울궈냈다. 이 용액을 이온교환 수지로 정제하여 약 60 mCi/batch의 정제 $^{32}$P를 얻었다. 이온교환수지에 흡착되는 $^{32}$P의 방사능은 전체의 3% 미만이었고 여기에 흡착되는 불순 핵종은 $^{131}$ Ba, $^{85}$ Sr, $^{59}$ Fe, $^{65}$ Zn, $^{60}$Co이었다. 이 방법으로 얻은 $^{32}$P 최종제품은 핵종순도 >99%, 방사화학적 순도 >98%, 고형성분 함량 <1.2 mg/mL 이어서 그 품질이 우수함을 알 수 있었다.

Low Genetic Diversity and Shallow Population Structure of the Japanese Halfbeak Hyporhamphus sajori Revealed from Mitochondrial DNA in the Northeast Asia (Mitochondrial DNA를 이용한 동북아시아 학꽁치 Hyporhamphus sajori의 유전적 다양성과 집단 구조)

  • This study was conducted to know the genetic diversity and population structure of Japanese halfbeak (Hyporhamphus sajori) in the Northeast Asia, using mitochondrial DNA control region. In the present study, a total of 70 individuals were collected from three locations of China (Liaoning), Korea (Tongyeong) and Japan (Wakasa Bay), and 47 individuals sequences from three locations of Japan (Wakasa Bay, Toyama Bay and Mikawa Bay) were downloaded from genbank. A total of 7 haplotypes were identified with 7 polymorphic sites from 358 bp length sequences. Haplotype and nucleotide diversity were very low and ranged from 0 to 0.295±0.156 and 0 to 0.0009±0.0011, respectively. Ancestral haplotype was shared by 94% individuals. An extremely low haplotype and nucleotide diversity, and starlike minimum spanning tree indicated that the species have undergone a recent population expansion after bottleneck. Pairwise FST values were low and there was no significant differences among populations suggesting a gene flow among the populations. Dispersal of the eggs with the aid of drifting seaweed and currents might be the major responsible factor for the genetic homogeneity.
