암 관련 유전자를 후성유전학적으로 제어하는 방법에는 miRNA, DNA 메틸화, 그리고 히스톤 단백질의 변형에 의해서 가능하다. 그러나 후성유전학적 방법을 통해서 암 관련 유전자를 제어하기 위해서는 첫째, 한 유전자에 여러 miRNA들에 의해서 조절되기 때문에 선택의 문제가 있으며, 둘째, 암 관련 유전자를 제어하는 DNA 메틸화 패턴이 다양하며, 셋째, 히스톤 단백질의 변형 자체가 다양하며 각 유전자에 대한 히스톤 변형의 특이성이 있다. 따라서 후성유전학 기반 하에서 암 관련 유전자를 제어하기 위해서는 시스템 수준에서의 접근이 바람직하다. 본 연구에서는 암 관련 유전자의 네트워크를 구축하고, 이 네트워크를 기반으로 암 유전자를 제어하는 miRNA에 최우선 순위를 부여하는 방법, 암 유전자의 DNA 메틸화 모티프 패턴을 분석하는 방법, 히스톤 변형과 암 관련유전자의 상관관계를 분석하는 방법을 제시하고자 한다.
In order to evaluate the availability of lacZ as a reporter gene for producing transgenic mud loach, foreign DNA, bacterial \beta-galactosidase$ gene (lacZ) was microinjected into mud loach eggs and its insertion and expression were examined. X-gal based histochemical assay, fluorimetric analysis of \beta-galactosidase$ with 4-methylumbelliferyl-$\beta$-D-galactoside (MUG) and molecular biological examination using polymerase chain reaction (PCR), dot blot, southern blot and sequence analysis of PCR products were carried out to analyze both microinjected group and non-injected controls. The results are disccussed in this paper.
$\textrm{T}_{5}$ progeny of one transgenic tomato line (To9) carrying antisense polygalacturonase (PG) cDNA was generated by selfing. Five $\textrm{T}_{5}$ plants were used to analyse in detail. The PG antisense gene was stably inherited through fifth generations. In all five $\textrm{T}_{5}$ plants, expression of the antisense transcripts were detected. In consequence, it led to a reduction of the PG enzyme activity in ripe fruit to between 37% and 65% that of normal. In two plants the expression of endogenous PG gene was inhibited in ripe fruit.
The Human gene testing act (GenDG) in Germany starts from the characteristic features of gene testing, i.e. dualisting structure consisted of anlaysis on the one side and the interpretation on the other side. The linguistic distincion of 'testing', 'anlaysis' and 'judgment' in the act is a fine example. Another important basis of the regulation is the ideological purpose of the law, that is information autonomy. The normative texts as such and the founding principle are the basis of the classification of testing types. Especially in the case of gene testing for medical purpose is classified into testing for diagnostic purpose and predictive purpose. However, those two types are not always clearly differentiated because the predictive value of testing is common in both types. In the legal regulation of gene testing it is therefore important to manage the uncertainty and subjectivity which are inherent in the gene-analysis and the judgment. In GenDG the system ensuring the quality of analysis is set up and GEKO(Commity for gene tisting) based on the section 23 of GenDG concretes the criterium of validity through guidelines. It is also very important in the case of gene testing for medical purpose to set up the system for ensurement of procedural rationality of the interpretation. The interpretation of the results of analysis has a wide spectrum because of the consistent development of technology on the one side and different understandings of different subjects who performs gene testings. Therefore the process should include the communication process for patients in oder that he or she could understand the meaning of gene testing and make plans of life. In GenDG the process of genetic counselling and GEKO concretes the regulation very precisely. The regulation as such in GenDG seems to be very suggestive to Korean legal polic concerning the gene testing.

마이크로어레이는 생명과학 분야에서 사용되는 대규모의 유전자 발현정도를 동시에 측정할 수 있는 도구이다. 마이크로어레이 실험은 많은 양의 데이터를 생성하기 때문에, 자동화된 효과적인 분석기법이 필요하다. 이 논문에서는 약물의 영향 분석을 위해 약물의 투여량 및 투여후의 시간대별로 샘플을 추출하여, 마이크로어레이를 이용하여 유전자의 발현량을 분석하는 경우에, 약물에 대해서 반응하는 유전자를 추출하는 데이터 마이닝 기법을 제안한다. 제안한 방법에서는 유전자의 발현정도값을 이전 시간의 값을 기준값으로 하여 증가, 감소, 답보에 해당하는 기호로 매핑하여, 분석자가 원하는 패턴을 보이는 유전자를 추천한다. 한편, 유전자의 상호간에 많은 영향을 주고 받기 때문에 특정 약물을 투여할 때, 이에 직접적인 영향을 받는 것도 있지만, 이와는 전혀 상관없이 동작하는 것도 있기 때문에, 제안한 방법에서는 이러한 약물 투여와 유의성이 있을 가능성이 있는 유전자만을 전처리과정을 통해서 필터링하는 기법을 활용한다. 제안한 방법은 실제 약물 투여 실험 샘플에 대한 마이크로어레이 데이터에 적용하여 활용가능성을 확인하였다.
Genome sequencing and microarray technology produce ever-increasing amounts of complex data that needs statistical analysis. Visualization is an effective analytic technique that exploits the ability of the human brain to process large amounts of data. In this study, biplot approach applied to microarray data to see the relationship between genes and samples. The supplementary data method to classify new sample to known category is suggested. The methods are validated by applying it to well known microarray data such as Golub et al.(1999), Alizadeh et al.(2000), Ross et al.(2000). The results are compared to the results of several clustering methods. Modified graph which combine partitioning method and biplot is also suggested.

마이크로 어레이 기술로 인해서 유전자의 발현 데이터를 대량으로 얻을 수 있게 되었다. 따라서 실험조건에 따른 유전자 발현 양상을 한눈에 볼 수 있게 되었고. 이를 기반으로 유전자간의 조절 관계를 예측할 수 있게 되었다. 또한 실험 이미지와 분석 파일들이 많아짐에 따라서 이러한 데이터를 효율적으로 관리하고, 저장하는 시스템이 필요하게 되었다. 이 두 가지 시스템을 통합함으로써 유전자 조절 네트워크 분석에 필요한 발현 데이터를 체계적으로 관리하고 손쉽게 얻을 수 있을 뿐만 아니라 분석 결과 또한 효율적으로 관리할 수 있다. 본 논문에서는 유전자 네트워크 분석 시스템과 마이크로 이미지 및 분석 데이터 관리 시스템을 통합한 시스템을 소개하고 각 시스템에서 제공하는 기능과 통합 시스템의 특징에 대해서 소개한다.

바이오 칩 분석 시스템은 유전자와 실험의 두 축으로 이루어진 바이오 칩에서 자료를 추출하고 필요한 정보를 얻기 위해 데이터를 분석하는 시스템이다. 유전자 데이터를 효율적으로 분석할 수 있는 방법으로 바이오 칩 분석 시스템이 각광받으면서 데이터의 양과 종류가 방대해지고 메모리의 효율적인 사용과 이에 따른 속도 개선을 위해 임베디드 시스템이 필요해지고 있다. 이에 따라 본 연구에서는 임베디드 시스템을 위한 PSO 기반의 군집화 알고리즘을 구현하였다. 방대한 양의 유전자 데이터를 분석하기 위해 생태계 모방 알고리즘인 Particle Swarm Optimization 알고리즘과 비슷한 유전자의 분류를 위한 기법으로 군집화를 사용하여 유전자 데이터의 통합 분석 시스템을 구현, 사용자에게 더욱 효율적으로 정보를 제공한다. 본 논문에서는 방대한 양의 데이터의 최적화에 효율적인 생태계 모방 알고리즘 Particle Swarm Optimization 을 이용하여 데이터들을 군집화하는 알고리즘을 임베디드 시스템을 위해 구현한 방법을 기술하고 있다.
The prenatal period in livestock animals is crucial for meat production because net increase in the number of muscle fibers is finished before birth. However, there is no study on the growth and development mechanism of muscles in Hanwoo during this period. Therefore, to find candidate genes involved in muscle growth and development during this period in Hanwoo, mRNA expression data of longissimus in Hanwoo at 6 and 9 months post-conceptional age (MPA) were analyzed. We independently identified differentially expressed genes (DEGs) using DESeq2 and edgeR which are R software packages, and considered the overlaps of the results as final-DEGs to use in downstream analysis. The DEGs were classified into several modules using WGCNA then the modules' functions were analyzed to identify modules which involved in myogenesis and adipogenesis. Finally, the hub genes which had the highest WGCNA module membership among the top 10% genes of the STRING network maximal clique centrality were identified. 913(6 MPA specific DEGs) and 233(9 MPA specific DEGs) DEGs were figured out, and these were classified into five and two modules, respectively. Two of the identified modules'(one was in 6, and another was in 9 MPA specific modules) functions was found to be related to myogenesis and adipogenesis. One of the hub genes belonging to the 6 MPA specific module was axin1 (AXIN1) which is known as an inhibitor of Wnt signaling pathway, another was succinate-CoA ligase ADP-forming beta subunit (SUCLA2) which is known as a crucial component of citrate cycle.
We identify the correct chromosome and locate the corresponding markers close to the QTL in the linkage analysis of a quantitative trait by using the SSVS method. We consider several markers linked to the QTL, as well as to each oyher and thus the i.b.d. values at these loci generate collinear predictors to be evaluated when using the SSVS approach. The results on considering only closely linked markers to two QTL simultaneously showed clear evidence in favor of the closest marker to the QTL considered over other markers. The results of the analysis of collinear markers with SSVS showeed high concordance to those obtained using traditional multiple regression. We conclude based on this simulation study that the SSVS is quite useful to identify linkage with multiple linked markers simultaneously for a complex quantitative trait.
