시아노에틸화한 폴리비닐알콜의 표면사이징에 따른 종이의 유전적 성질 향상

  • 종이는 목재섬유와 물의 현탁액을 스크련을 통해 균일하게 분산시킨 판상섬유 제품 으로서 가격이 저렴하고, 열, 전기절연성, 필터기능, 난연성 및 폐기성이 우수하여 다양한 분 야에서 활용되고 있다. 특히 최근에는 종이의 고부가가치화, 차별화를 위하여 내열성, 전기 절연성, 내수성, 기계적 특성, 내약품성, 생체적응성을 지닌 다양한 기능지가 개발되어 식품, 전기, 전자, 건축, 화학, 의료 관련 산업에 있어서 주요 재료로 이용되고 있다. 특히 전기, 전 자산업 발전과 더불어 전기절연성이 우수한 종이의 역할과 수요는 크게 증대되고 있으며 산 업발전의 핵심적인 제품으로서 자리매김을 하고 있다. 하지만 국내의 경우 전기절연지와 관 련된 제품 및 생산기술 부족으로 전량을 수입에 의존하고 있는 실정이며, 그 수입량은 앞으 로 전기 및 전자 산업의 발전으로 인해 더욱 증가할 것으로 예상된다. 이러한 수입의존성을 극복하기 위해서는 전기절연지와 관련된 제품개발 및 생산기술에 관한 연구가 시급히 요청 되고 있다. 이러한 기술의 국산화는 수입 대체 효과 뿐만 아니라 관련 산업의 발전에도 기 여할 수 있을 것으로 예상되며 수출산업으로 성장할 수 있을 것으로 예상된다. 종이의 전기적 성질은 유전상수, 유전손실율과 같은 유전적 특성과 전기저항, 절연 파괴강도의 기계적 특성으로 구분할 수 있다. 종이의 전기적 성질 가운데 유전율은 전기장 에 대한 종이의 물리화학적인 반응으로 일반적으로 종이의 밀도와 종이를 구성하는 성분의 쌍극자 모멘트에 비례하며 온도에 따라서도 변화한다. 일반적으로 온도가 상승하면 열에너 지를 얻게된 쌍극자가 전기장에 배열됨으로써 유전율이 상승하지만 온도가 유리전이점 이상 으로 높아질 경우 열적 교란에 의해서 분극 능력이 감소하게 되어 유전완화 현상이 나타난 다. 전기절연지로 사용될 종이의 절연특성을 이해하기 위해서는 사용환경에 따른 유전적 특 성 및 tan$\delta$에 관한 연구가 펼요하다. 본 연구에서는 종이의 유전특성을 개선하기 위해서 펼름형성능력이 우수한 polyvinyl alcoholCPV A)과 aery lonitrile을 이용하여 시아노에틸화한 P PYA의 표면처리에 따른 종이의 유전적 특성의 변화를 검토하였다. 그 결과 PYA에 도입된 시아노에틸기에 의해 유전율을 증가시킬 수 있음을 확인하였다. 또 본 연구에 적용된 시험 범위에서는 온도상승에 따라 유전율이 상승하였다.

AGB (Ancestral Genome Browser): A Web Interface for Browsing Reconstructed Ancestral Genomes (AGB (Ancestral Genome Browser): 조상유전체 데이터의 시각적 열람을 위한 웹 인터페이스)

  • With the advancement of next-generation sequencing (NGS) technologies, various genome browsers have been introduced. Because existing browsers focus on comparison of the genomic data of extant species, however, there is a need for a genome browser for ancestral genomes and their evolution. In this paper, we introduce a genome browser, AGB (Ancestral Genome Browser), that displays ancestral genome data reconstructed from existing species. With AGB, it is possible to trace genomic variations that occurred during evolution in a simple and intuitive way. We explain the capability of AGB in terms of visualizing ancestral genomic information and evolutionary genomic variations. AGB is now available at http://bioinfo.konkuk.ac.kr/genomebrowser/.

  • 한국인 최초 전체 유전체 서열(KOREF; Koreanindividualgenomesequence) 은 한국인을 위한 참조 서열로써 사용될 수 있다. 2009년 1월에 남성 한국인 유전체를 솔렉사(Solexa)를 통해 전장서열을 결정하였다. 이는 NCBI의 인간게놈프로젝트에서 생산한 게놈의 99.83%를 커버하며, 또한 NCBI게름서열의 약 20배를 커버할 정도의 유전체 서열을 결정하여 매우 높은 정확도를 가진 한국인 고유유전체이다. 한국인 유전체 서열의 분석결과 현재까지 밝혀지지 않았던 한국인 특이적인 3백만 개의 SNP를 밝혀냈다. 먼저 보고된 중국인 게놈은 한국인 게놈과 매우 가까운 민족 그룹임에도 불구하고 38%(3,186,352 SNP중에 1,217,362 SNP) 의 특이적인 차이를 나타내었으며, 또한 미토콘드리아 서열 비교를 통해서도 특이적인 다양성을 보여주는 SNP데이터를 확인 할 수 있었다. 차세대 게놈서열결정의 기술은 적은 노력과 비용으로 인간 유전체 데이터를 얻을 수 있게 되었으며, 이러한 개인유전체 데이터는 개인유전체 의학으로 가는 초석이 될 것이다.

Genetic Diversity and Population Genetic Structure of Exochorda serratifolia in South Korea (가침박달 집단의 유전다양성 및 유전구조 분석)

  • Genetic diversity and population genetic structure were estimated in nine natural populations of Exochorda serratifolia in South Korea using ISSR marker system. Average of polymorphic loci per primer was 5.8 (S.D.=2.32) and percentage of polymorphic loci per population was 78.7% with total 35 loci from 6 ISSR primers. In AMOVA, 27.8% of total genetic variation came from genetic difference among populations and 72.2% was resulted from difference among individual trees within populations. Genetic differentiations by Bayesian inference were 0.249 of ${\theta}^{11}$ and 0.227 of $G_{ST}$. Inbreeding coefficient for total populations was 0.412. There was significant correlation between genetic distance and geographic distance among populations. On the results of Bayesian cluster analysis, nine populations were assigned into three groups. The first group included 5 populations, and the second and the third had two populations per group, respectively. These three regions could explain 10.0% of total genetic variation from hierarchical AMOVA, and the levels of among-population and among-individual were explained 19.7% and 70.3%, respectively. The geographic distribution of populations following the three Bayesian clusters could be explained with mountain range as Baekdudaegan which is the main chain of mountains in Korea. The mountains as the physical barrier might hamper gene flow in the pearlbush. So when protected areas are designated for conservation of this species, we should consider those three regions into considerations and would better to choose at least one population per region.

유전율을 이용한 토양오염 모니터링 기법의 제안

  • 지반오염을 조사하기 위해서는 시추작업을 통하여 시료를 채취하는 방법이 일반적이지만, 실시간으로 원위치에서 다양한 오염물질들의 오염도 변화를 체계적으로 모니터링하는 것은 대단히 어렵다. 본 연구에서는 Frequency Domain Reflectometry(FDR) 장비를 고안하여 지반의 유류오염을 파악하기 위한 유전율 측정법의 실험적 접근을 시도하였다. 구체적으로 포화 및 불포화 매질에 대한 오염도의 측정 및 오염도와 체적함수비의 관계를 측정하여 유전율 반응으로부터 매질의 물성치를 파악할 수 있는 것을 확인하였다.

Genetic diversity and population structure of Atractylodes japonica $K_{OIDZ}.$ in Korea (한국내 삽주의 유전적 다양성과 집단구조)

  • The study of genetic diversity was carried out in Atractylodes japonica $K_{OIDZ}$. Although this species has been regarded as medically important one, there is no report on population structure in Korea. Starch gel electrophoresis was used to investigate the allozyme variation and genetic structure of eight Korean populations of this species. Of the 15 genetic loci surveyed, nine (60.0%) was polymorphic in at least one population. Genetic diversity was high at the species level $(H_{es}=0.144)$, whereas, that of the population level was relatively low $(H_{ep}=0.128)$. Nearly 87% of the total genetic diversity in A. japonica was apportioned within populations. The sexual reproduction, high fecundity, and perennials are proposed as possible factors contributing to high genetic diversity. The indirect estimated of gene flow based on Gst was 1.69.

Determination on the Minimum Area for Conservation of Four Halophyte Species from the Southwestern Coast of Korea Based on AFLP (AFLP에 근거한 서남해안 염생식물 4종 개체군의 최소 보존 면적 결정)

  • To determine the minimum area for conservation of four Halophytic species populations, we evaluate the genetic diversity of four species based on the AFLP method using thirteen primer sets. Four species populations, Phragmites communis Trin, Suaeda japonica Makino, Zoysia sinica Hance, and S. maritima (L.) Dumort, from the southwestern coast of Korea, were selected for this study. The genetic diversity index ($\Psi_{ST}$) of Phragmites communis was 0.3856, Suaeda japonica 0.1445, Suaeda maritima 0.1669, and Zoysia sinica 0.2422. Based on the genetic diversity of population, we could determine the minimum area for conservation of each species as follows. P. communis needs $500{\times}500m^2$, S. japonica, S. maritima, and Z. sinica $100\times100m^2$ for keeping their genetic identity.

Genetic Diversity and Genetic Structure of Phellodendron amurense Populations in South Korea (황벽나무 자연집단의 유전다양성 및 유전구조 분석)

  • Genetic diversity and genetic structures were estimated in seven natural populations of Phellodendron amurense Rupr in South Korea using ISSR markers. The average of polymorphic loci per primer and the proportion of polymorphic loci per population were 4.5 and 78.8% respectively with total 27 polymorphic loci from 6 ISSR primers. The Shannon's diversity index(I) was 0.421 and the expected heterozygosity($H_e$) was 0.285, which was similar to the heterozygosity (hs =0.287) inferred by Bayesian method. In AMOVA, 7.6% of total genetic variation in the populations was resulted from the genetic difference among populations and the other 92.4% was resulted from the difference among individuals within populations. Genetic differentiation(${\theta}^{II}$) and inbreeding coefficient(f) for total population were estimated to be 0.066 and 0.479 by Bayesian method respectively. In Bayesian clustering analysis, seven populations were assigned into three groups. This result was similar to the results of genetic relationships by UPGMA and PCA. The first group included Hwachoen, Gapyeong, Bongpyeong and Yongpyeong population, and the second included two populations in Sancheong region. Muju population was discretely assigned into the third group in spite of the geographically short distance from the Sancheong region. There was no significant correlation between genetic relationship and geographic distribution among populations in Mantel's test. For conservation of the phellodendron trees, it would be effective to consider the findings resulted from this study with ecological traits and life histories of this species.

Genetic Diversity of an Endangered Fish, Iksookimia choii (Cypriniformes), from Korea as Assessed by Amplified Fragment Length Polymorphism (AFLP 분석에 의한 멸종위기어류 미호종개, Iksookimia choii의 유전 다양성)

  • Genetic diversity and population genetic structure within or among three stream populations (Gab, Baekgok and Ji streams) of Korean endangered natural monument fish, Iksookimia choii, were assessed by amplified fragment length polymorphism (AFLP). AFLP analysis using three primer combinations generated 104 to 106 AFLP bands, and percent polymorphic bands were similar in those three populations ranging 21.5 to 24.5%. Heterozygosity and genetic diversity within or among populations were quite low for all of these populations with average values ranging from 0.067 to 0.084 and from 0.076 to 0.087, respectively. Analyses of pairwise distance and genetic similarity among three populations of I. choii also revealed the similar results with very low genetic differentiation one another. Although pairwise Fst values were very low, our data clearly indicated distinct genetic differentiation among the three populations. This is the first report concerning the genetic diversity and differentiation of this species, and provides basic genetic information that should facilitate attempts to conserve this species.

Genetic Differentiation of the Largemouth Bass Micropterus salmoides from the Major Rivers and Reservoirs in Korea Assessed by AFLP (우리나라 주요 강과 호수에 분포하는 외래어종 배스 Micropterus salmoides의 AFLP 분석에 의한 유전적 분화)

  • Genetic diversity and differentiation within or among nine populations of introduced fish, largemouth bass Micropterus salmoides were assessed by AFLP. The AFLP analysis using three primer combinations generated 299.2. AFLP bands and percentage of polymorphic bands were similar in those nine populations, ranging 14.1 to 21%. Heterozygosity and genetic diversity within or among populations were quite low for all of these populations with average values ranging from 0.054 to 0.067 and from 0.069 to 0.085, respectively. Analyses of pairwise distance and genetic similarity among nine populations of Micropterus salmoides also revealed the similar results with low genetic differentiation one another. Although pairwise Fst values were low, they were indicated a clear distinct genetic differentiation among the nine populations. These results indicate that very small population of the largemouth bass was first introduced to Paldang reservoir and they are widely spread at most of aquatic habitats in Korea.
