To establish the mass proliferation system of Ardisia pusilla DC, the shoot tips of Ardisia pusilla DC were cultured on the MS and half-strength MS medium supplemented with $0{\sim}5.0$ mg/L BA or $0{\sim}0.5$ mg/L thidiazuron(TDZ), respectively. A few multiple shoot formation observed when the shoots were cultured on MS medium containing TDZ. However, the frequency of multiple shoot formation was reached up to 82.4%, when the shoots were cultured on half-strength MS medium supplemented with 0.5 mg/L BA. Also the number of shoot per explant was 7.1. To promote rooting from multiple shoot, newly formed shoots were transferred to half-strength MS medium containing 0.5 mg/L IBA or 0.5 mg/L NAA, respectively. Regenerated plantlets were grown to normal mature plants in soil.
To investigate the function of the chloroplast small heat shock protein (small HSP), transgenic tobacco plants (Nicotiana tabacum L., cv. SRI) that show constitutive expression of the chloroplast small HSP were generated. Effects of constitutive expression of the introduced gene on thermotolerance were first probed with the chlorophyll fluorescence. After a 5-min incubation of leaf discs at high temperatures, an increase in the Fo level and a decrease in the Fv level, indications of separation of LHCII from PSII and inactivation of electron transport reactions in PSII, were mitigated by constitutive expression of the small HSP. When tobacco plantlets grown in Petri dishes were incubated at $52^{\circ}C$ for 45 min and subsequently incubated at $25^{\circ}C$, leaf color of nontransformants was gradually became white and all plantlets finally were died. Under conditions in which all nontransformants were dying, more than 80% of the transformants remained green and survived. These results suggest that the chloroplast small HSP plays an important role in protecting photosynthetic machinery during heat stress.

최근 호두 기내 소비량은 급증하나 대부분 실생묘로 조림되어 결실까지 장기간 소요되며, 무성 번식률이 낮아 보급확대가 어렵다. 본 연구는 호두나무 관행 묘목 번식법 개선이 가능한 우량 국산묘 대량증식 기술 개발로 최적 배양조건을 구명하고자 수행하였디. 호두나무 등 목본류는 식물 조직배양 기본배지에서 산화방지제를 첨가하여 사용하는 경우, 배양체에서 생기는 페놀 화합물(Phenolic compound)의 작용을 억제하여 배양 식물체의 갈변(Browning) 또는 흑변(Blackening) 현상을 감소시키는 효과가 있다. 페놀 추출물은 배지로 빠져나와 갈변시키며 생존율을 저하시키는 원인이 되고 이를 제거하는 산화방지제는 배양 식물체의 생산효율을 높이는 효과를 나타내는 강력한 항산화제(Antioxidant) 역할을 한다. 호두나무의 액아를 배양하게 되면 조직 및 배지의 갈변화가 심하게 일어나는데, 휴면 정도가 깊을 수록 정단에서 추출되는 페놀성 물질의 함량이 증가하며, 식물 조직 내 페놀성 물질의 함량 정도와 배양의 고사율과는 상관관계가 있다. 이러한 식물 배양체에서 생기는 페놀 화합물에 의해 발생되는 배양상의 문제점을 해결하기 위하여 계대배양을 자주해 주거나 활성탄(Activated charcoal), Ascorbic Acid 등의 첨가물을 배지에 추가하는 방법이 있다. 이에 따라 호두 '신령' 품종의 액아배양의 페놀집적을 제거하기 위해 배지 내에 산화방지제 첨가물질 등을 농도별로 처리하였다. 호두나무 액아 배양 후 8주차 산화화방지제 첨가물질의 농도별 처리 효과는 생존율은 85.3%를 보였으며, 신초 형성율 76.4%, 신초수 1.9개/주, 신초장 27mm/주의 양호한 결과를 나타냈으며, 페놀 제거 및 유식물체 고사 억제 효과를 보였다.
Experiments were conducted to find the optimal acclimatization conditions for Eleutherococcus senticosus plantlets regenerated from bioreactor cultured somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. Among the various temperature and artificial soil tested, the highest survival rate (88%) was observed when plantlets were acclimatized in Klasmann bed soil at $10^{\circ}C$ When in vitro plantlets directly transplanted to field environment, shading treatment was necessary and 50% shading was more effective than 30% shading. Transplanting season were also important for successful acclimatization of in vitro cultured plantlets, transplanted on March 20 with 50% shading exhibited the best survival rate and further growth.
To measure the time required for ginseng explants to become determined to form flower buds, we cultured zygotic embryos, seedlings, and cotyledonary nodes on MS medium supplemented with BA and GA$_3$of 5 ${\mu}$M each (flower inducing medium, FIM) for various periods and transferred to the basal medium. The explants required a minimum of 10 days on FIM to be determined. Histological observations revealed that the axillary meristem to be fated to develop into flower bud remained in a state of shoot meristem during the first 10 days of culture and differentiated into flower bud after 15 days of culture. We suggest that the in-vitro flowering system described in this study is useful in investigating (a) regulatory element(s) to cause the phase change from the vegetative to reproductive state by comparing predetermined explants with determined ones at the molecular level.
To develop herbicide-resistant cucumber plants (Cucumis sativus L. cv Green Angle) embryogenic suspension cultures were co-cultured with Agrobacterium tumefaciens strain LBA4404 carrying a disarmed binary vector pGA-bar. The T-DNA region of this binary vector contains the nopalin synthase/neomycin phosphotransferase Ⅱ (npt Ⅱ) chimeric gene for kanamycin resistance and the cauliflower 35S/phosphinothricin acetyltransferase (bar) chimeric gene for phosphinothricin (PPT) resistance, After co-cultivation for 48 h, embryogenic calli were placed on maturation media containing 20 mg/L PPT. Approximately 200 putatively transgenic plantlets were obtained in hormone free media containing 40 mg/L PPT. Northern blot hybridization analysis confirmed the expression of the bar gene that was integrated into the genome of five transgenic plants. Transgenic cucumber plants were grown to maturity. Mature plants in soil showed tolerance to the commercial herbicide (Basta) of PPT at the manufacturer's suggested level (3 mL/L).
This experiment was conducted out to investigate the effect of plant growth regulators on callus and shoot formation. The callus formation was effective on 1/2 MS medium containing 2,4-D, while shoot formation was suppressed. Shoot formation and differentiation were the highest in combination 0.1 mg/L of IAA and 0.1 mg/L of BA. The polarity of explants was investigated from cotyledon, which excised 20% of each basal and terminal parts. Formation of shoot was induced from excised ends of the basal part. In excised ends of the basal part, callus was induced vigorously and shoots were produced lately. Root induction was easily achieved in 1/3 MS medium from the adventitious shoot and more than 90% of regenerated plantlets acclimatized successfully and flowered normally.

To observe the stimulating effects of low dose ${\gamma}-radiation$ on the seed germination, early growth and physiological activity in the gourd, seeds of three cultivars (Partner, Support, FR Yongja) were irradiated at the dose of $0.5{\sim}30\;Gy$. The hormetic effects of the low dose ${\gamma}-radiation$ on the growth were different from each cultivar. Low dose ${\gamma}-radiation$ had promoting effects on the germination at the optimum dose of 2 Gy and 8 Gy and on the early growth at the optimum dose of 4 Gy, 16 Gy and 20 Gy in Partner and Support cultivar and 1 Gy in FR Yongja cultivar. Irradiation didn't have significant effects on the chlorophyll contents of gourd plantlet. The antioxidant enzyme activity of gourd plantlet and early growth increased in low dose irradiation group.
This study was investigated to establish a regeneration system of plant via adventitious bud formation from leaf explant cultures of strawberry (Fragaria ananassa Duch). Effects of plant growth regulators (2,4-D, BAP), agar sucrose and myo-inositol on adventitious bud formation were investigated. When the leaf explants were cultured on MS medium supplemented with 0.1 mg/L 2,4-D and 3 mg/L BAP, the adventitious bud formation was most promoted. The adventitious bud formation was not induced from leaf explants cultured on MS medium containing 2,4-D alone. Adventitious bud formation was enhanced to almost 3 times on medium with low level of agar concentration (0.4%) in comparison with those on the medium with high level of agar (1%), but almost of shoot was vitrificated on the medium. Therefore, the normal adventitious bud formation from leaf explants was most effective on the medium containing 0.05 mg/L 2,4-D,1 mg/L BAP, 0.8% agar, 30 g/L sucrose and 100 mg/L myo-inositol. Thus, the mass propagation of healthy strawberry could be established using leaf explants.
This study was conducted to establish the optimal condition for plant regeneration and acclimatization from somatic embryos of Panax ginseng. Cotyledon segments of Panax ginseng produced primary and secondary somatic embryos when cultured on MS and WPM media supplemented with 7% sucrose. To induce plantlet conversion, cotyledonary somatic embryos were cultured on WPM solid medium with $GA_3$ at various concentrations (1~30 mg/L) for 4 weeks. Plantlets were transferred to 1/2 WPM solid medium with $GA_3$ at various concentrations (0~5 mg/L) and 0.5% activated charcoal for shoot and root elongations. Elongated plantlets further developed into well-developed leaf and root system on 1/3 SH medium with 0.5% activated charcoal under ventilation condition for 5 months. The highest survival rate to soil was 75% when plantlets were regenerated on 1/3 SH medium without sucrose under ventilation condition.
