포자생성 유산균의 종양세포 증식 억제 활성과 지방산 산화 억제 활성 (Tumor Cell Proliferation Inhibitory and Antioxidative Activities of Spore Forming Lactic Acid Bacteria)

  • Tumor cell proliferation inhibitory, antioxidative activities and glutathione content were analyzed in a variety of spore forming lactic acid bacteria. Tumor cell proliferation inhibitory activity varied widely depending upon the strains of spore forming lactic acid bacteria and the types of carcinoma cell lines(0${\simn}$56.7%), Bacillus coagulans KTCC3625 has shown a marked antipro-liferative effect against the carcinoma cells and NCL-H1299 human lymphoma cell line tended to be least affected by the spore forming lactic acid bacterial cell extracts. Antioxidative activity analyzed in the lipid peroxidation occurred in all the test strains varied on the strains(5.0 to 52.0%) an extensively high degree of antioxidative activity was demonstrated by three strains of Bacillus coagulans KTCC3625, Bacillus coagulans KTCC1015 and Lactobacillus sporogens CU 815. Concentrations of glutathione were highest in a strain of Lactobacillus sporogenes CU 815 followed by Sporo-lactobacillus inulinus ATCC13538 (5.34 to 8.19 mol/g). Spearmans' rank correlation quotient between cellular GSH levels and linoleic acid peroxidation inhibitory effects of the spore forming lactic acid bacteria revealed highly significant correlation quotient of 0.78. Spearmans' rank correlation quotient between the Caski human cervix carcinoma cell proliferation inhibitory activity and the linoleic acid peroxidation inhibitory effects of the spore forming lactic acid bacteria and that between Caski carcinoma cell proliferation inhibitory activity and the cellular GSH levels were shown to be 0.29 and 0.32,respectively, which means an insignificant positive correlation however.

Effect of Mixed Microbes Addition on Chemical Change and Silage Storage of Spent Mushroom Substrates (복합생균제 첨가가 버섯부산물의 화학적 성분 변화와 발효 저장성에 미치는 영향)

  • This study was conducted to evaluate effects of mixed microbes addition on physico-chemical, fermentative and microbial parameters of sawdust-based spent mushroom substrates(SMS). The SMS was inoculated with mixed microbes(Enterobacter ludwigii, Bacillus cereus, 2 strains of Bacillus subtillis, Saccharomyces cerevisiae and Lactobacillus plantarum) at 1% level(wet basis) and anaerobically fermented during the different periods(up to 8th week). Compared with the SMS before ensiling, the ensiled one had higher CP, NDF and ADF percentages and lower DM and NFC percentages. However, levels of change were very low. The in situ ruminal disappearance of SMS DM and NDF decreased with the ensiling period prolonged. For fermentative parameters, pH reduced and lactic acid contents increased after ensiling, compared with those after ensiling. At 8th week of ensiling, pH increased and lactic acid contents reduced again, compared with those at 4th week of ensiling; however, the silage still showed favorable fermentation status. Lactic acid bacteria counts did not change throughout 8 weeks of ensiling. Counts of total microbes and yeast reduced after 4th week of ensiling period. Counts of lactic acid bacteria and yeast at 8th week of ensiling were in the levels of 108cfu/g. These results indicate that anaerobic fermentation with microbial addition could be an effective way for the long term(8 weeks) storage of the SMS.

Lactic Acid Fermentation and Biological Activities of Rubus coreanus (복분자의 유산발효와 생리활성 평가)

  • The puree of Rubus coreanus was fermented using lactic acid bacteria and its biological activities were examined. Lactobacillus acidophilus KCCM 32820, L. casei KCCM 12452, Lactococcus lactis subsp. lactis KCCM 40104, and Streptococcus thermophilus KCCM 40430 were used as a single or mixed starter for the lactic acid fermentation, and their cultures at the late logarithmic growth phase were inoculated to final concentration of 2% (v/v). L. casei fermented the puree of Rubus coreanus best when used as a single starter, and the culture of L. casei and L. lactis with the inoculation ratio of one to one showed the highest fermentation activity when used as a mixed starter. However, the fermented broth of the puree of Rubus coreanus using L. acidophilus and S. thermophilus showed the best results in the sensory evaluation. The optimal lactic acid fermentation conditions were as follows; the concentration of oligosaccharide added was 1% (w/v), pH of puree and fermentation temperature were 4.0 and $37^{\circ}C$, respectively, and fermentation time was $72{\sim}96$ hours. Glucose and fructose were major free sugars, and the content of lactic acid was 698.2 mg/100 g in the fermented broth. The fermented broth of the puree of Rubus coreanus showed the electron donating ability and nitrite scavenging ability with the value of 69% and 38.3% at pH 1.2, respectively. SOD-like activity and inhibitory activity on xanthine oxidase were also found in the fermented broth with the value of 60.3% and 41.8%, respectively. When the antimicrobial activities of the fermented broth were examined, it showed the highest growth inhibitory activity against Escherichia coli O-157:H7, and also contained antimicrobial activities against Salmonella typhimurium, Bacillus cereus, and Staphylococcus aureus.

Basic Physiological Activities of Bifidobacterium infantis Maeil-K9 and Lactobacillus plantarum KCTC3099 Selected by Anticarcinogenic Activities. (항암 활성능이 우수한 Bifidobacterium infantis Mneil-K9과 Lactobacillus plantarum KCTC3099의 기초 생리활성)

  • This study was conducted to confirm the availability of lactic acid bacteria as probiotics haying inhibitory effects to cancer cell line. Five lactic acid bacteria showing anti-cancer activity were compared by acid tolerance, bile tolerance, antibiotics resistance, milk fermentation, stability, and cell adherence activity to colon epithelial cell. The results obtained are as follows : In acid tolerance, all strains did not have a resistance below pH 3.0 and 3.5 except Lactobacillus plantarum KCTC3099. In antibiotics resistance, Lactococcus lactis and L. plantarum KCTC3099 were resistant to cotrimoxazol (128 mg/1), and Bifidobacterium adolescentis Maeil-K8 and B. infantis Maeil-K9 were resistant to doxycylin and gentamycin (4 mg/1). In case of cell adherence ability to Caco-2 cell, B. infantis Maeil-K9 was found to be superior to others as 3.1%, while the others were less than 0.5%. When the strains were cultured to milk base, viable counts of the strains tested increased more 1 log cycle than inoculation, but acid production was very low except L. plantarum KCTC3099. Also, L. plantarum KCTC3099, B. adolescentis Maeil-K8, and B. infantis Maeil-K9 were stable in fermented milk base during storage. In conclusion, L. plantarum KCTC3099 and B. infantis Maeil-K9 were confirmed to be superior for the availability as probiotics.

Studies on the Utilization of Wastes from Fish Processing I - Characteristics of Lactic Acid Bacteria for Preparing Skipjack Tuna Viscera Silage (수산물 가공부산물의 이용에 관한 연구 I -가다랭이 내장 발효 silage 제조를 위한 유산균주의 배양특성)

  • In order to utilize fish by-products from the skipjack tuna (Katsuwonus pelamis) canning manufactures Lactobacillus buigaricus KCTC 3188 and L. piantarum KCTC 1048 were used as a starter culture for the preparation of fermented fish silage with skipjark tuna viscera. The optimum temperature and pH on barterial growth and lactic acid production of L. bulgaricus and L. plantarum in MRS broth were $35^{\circ}C$ and around pH 6.0, respectively. And the optimum concentrations of the carbohydrate sources added to the broths were $7\%$ for dextrose and $10\%$ for molasses on the basis of total weights of skipjack tuna viscera. The pH of acid treated skipjack tuna viscera silage (ASS) slightly increased from 4.0 to 4.5, while that of fermented skipjack tuna viscera silages by the use of lactic acid bacterias (FSS) was significantly declined from 5.9 to about 40 after 42 days of storage at $35^{\circ}C$. Though the content of volatile basie nitrogen (VBN) in ASS was lower than those of FSS after 42 days of storage at $35^{\circ}C$, VBN content in silages slightly increased from an initial value of $62\~65{\cdot}mg/100g$ to final value of $113\~155\;mg/100g$ over 42 days. The fermented silage by L. piantarum reached a maximum concentration of amino nitrogen and showed $81\%$ of hydrolysis degree after 4 days of storage at $35^{\circ}C$.

Isolation and Characterization of Kimchi Lactic Acid Bacteria Showing Anti-Helicobacter pylori Activity (Helicobacter pylori 억제능 김치 유산균의 분리와 특성 규명)

  • One bacterium, which showed strong antagonistic activity against H. pylori KCCM 41756, was isolated from kimchi. The strain NO1 was designated as Lactobacillus plantarum NO1 based on Gram staining, biochemical properties, and 16S rRNA gene sequencing. The culture medium $(2{\sim}4{\mu}g/ml)$ of Lb. plantarum NO1 reduced $(40{\sim}60%)$ the urease activity of H. pylori KCCM 41756. Lb. plantarum NO1 inhibited the binding of H. pylori to human gastric cancer cell line, AGS cells, by more than 33%. Lb. plantarum NO1 exhibited high viability (maintained initial viable cell count of $10^9CFU/ml$) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastricjuice for 2 h and in 0.3%, 0.5% oxgall for 24 h. Hemolysis phenomena did not observed when Lb. plantarum NO1 was incubated in the blood agar media. We concluded that Lb. plantarum NO1 can be a good candidate as a probiotic, harboring anti-H. pylori activity.

Analysis the Impact of Topographic Factors on the Structure of Forest Vegetation in Deogyusan National Park (덕유산 국립공원 산림식생구조의 지형적 영향 분석)

  • The purpose of this study was to analyze the topographic effect of the LAI (Leaf Area Index), which has been widely used as an index that quantifies the structure of forest vegetation in Deogyusan National Park. With this aim, the study was conducted through a regression analysis which took as explanation the following variables: the elevation, slope, aspect, and soil moisture conditions. The LAI was taken as the response variable. Overall, the correlation between the Field-LAI and topographic factors was less than 0.5, which was relatively low. Except for topographic altitude, there was no statistical significance regarding the correlation with other factors. Meanwhile, regarding the orientation of the correlation, the higher the attitude, the steeper slope, the lower the soil moist, the lower the LAI value. The topographic altitude was found as a statistically significant explanation variable. The TWI (Topographic Wetness Index), which was used in this study to explain the soil moisture conditions, was not significantly related to the LAI distribution. The results of this study are expected to be utilized as basic data in more accurate forecasting the LAI distribution using remote sensing data.

Effect of Aerobic Exercise and Gym-ball Exercise on the Liver Function Test Index, Adipokines, and Cardiovascular Risk Factors in Obese Children with Nonalcoholic Fatty Liver Disease (유산소 운동과 짐볼 운동이 비알코올성 지방간 비만아의 간 기능평가 지수와 Adipokine 및 심혈관 질환 위험인자에 미치는 영향)

  • The aim of the present study was to investigate effects of a 12-week aerobic exercise training program and a gym-ball exercise training program on body composition, aspartate aminotransferase (AST), alanine aminotransferase (ALT), adipokines, and cardiovascular risk factors in obese children with nonalcoholic fatty liver disease. The subjects were separated into two groups, an aerobic exercise group (n=10), which practiced moderate aerobic exercise training for 12 weeks, and a gym-ball exercise group (n=13), which practiced resistance exercise training for 12 weeks. The results of the analyses are as follows: Weight, body mass index, and body fat were significantly lower (p<0.01, respectively), whereas the $VO_2$ max was higher in both groups (p<0.01). Fasting glucose, insulin and HOMA-IR levels were significantly decreased in the gym-ball exercise group (p<0.05), whereas adiponectin, AST, and ALT levels were significantly increased (p<0.05, p<0.001, p<0.001, respectively) in both groups after the 12-week exercise training program. In addition, our results showed that HOMA-IR, insulin, and concentrations of C-reactive protein (CRP) were significantly lower in both groups. They demonstrate that a 12-week program of regular aerobic exercise or gym-ball exercise yields beneficial effects such as an amelioration of cardiovascular risk factors, body indices, and liver function in obese children with nonalcoholic fatty liver disease.

Probiotic Properties of Lactic Acid Bacteria isolated from Feces and Kimchi (베트남인 분변 및 김치로부터 분리된 유산균의 프로바이오틱스 기능성 연구)

  • The purpose of this study was to investigate the probiotic properties and antioxidant capacity of lactic acid bacteria isolated from Vietnamese feces and the Korean traditional food kimchi. Six isolated strains were identified as Lactobacillus sp. by 16S rRNA sequencing. All strains showed good resistance to low pH (1.5, 2.0, and 3.0) and 0.3% oxgall bile acids. Culture filtrates from the six strains showed various antioxidant effects, including DPPH, ABTS, reducing power, and metal chelating ($Fe^{2+}$) activities. Two of the six Lactobacillus strains showed potential probiotic activity. Heat resistance and adhesion assays were conducted by mixing the selected strains, Lactobacillus acidophilus V4, Lactobacillus plantarum V7, and Lactobacillus paracasei DK121 isolated from kimchi. The results showed that the heat resistance of these strains was similar to that of a commercial strain, L. plantarum LP. In addition, a mucin attachment assay using the mixture of selected strains (V4, V7, and DK121) showed high binding activity to the mucous layer. In conclusion, a mixture of V4, V7, and DK121 shows promising probiotic activity and may be useful for the development of health-related products.

Antimicrobial effect of Enterococcus faecalis BMSE-HMP005 isolated from human breast milk against multidrug-resistant bacteria (모유 유래 유산균 Enterococcus faecalis BMSE-HMP005의 다제내성 균에 대한 항균효과)

  • In this study, Enterococcus faecalis BMSE-HMP005 isolated from human breast milk demonstrated antimicrobial effects against multidrug-resistant (MDR) bacterial strains. The bacteriocin produced by E. faecalis BMSE-HMP005 was fractionated using reverse-phase high-performance liquid chromatography. This fraction showed antimicrobial effects against both gram-positive and gram-negative MDR bacteria. No hemolytic reactions were observed. E. faecalis BMSEHMP005 was resistant to vancomycin; however, kanamycin, ampicillin, and erythromycin showed minimum inhibitory concentrations that were lower than the acceptable range provided by the European Food Safety Authority. For artificial gastric juice and bile acid, the survival rates were 98.67% and 95.70%, respectively. These results show the potential utility of E. faecalis BMSE-HMP005 as a probiotic with remarkable antimicrobial effects against MDR bacteria.
