Compositions of Nitrogen Compound and Amino Acid in Soybean Paste(Doenjang) Prepared with Different Microbial Sources (미생물 급원을 달리한 숙성 된장의 질소성분과 아미노산 조성)

  • Fermented traditional soybean paste(doenjang), koji soybean paste by Aspergillus oryzae, natto soybean paste by Bacillus natto and koji & natto soybean paste by Aspergillus oryzae and Bacillus natto were analyzed for compositions of amino acids and contents of nitrogens. Amino type nitrogen was the highest in fermenting for 90 days as $271{\sim}868\;mg/100g$, and its contents were in the order of koji soybean paste>traditional soybean paste>koji & natto soybean paste>natto soybean paste in all samples tested. In compositions of total amino acids, glutamic and aspartic acids were rich in koji soybean paste but big differences were not observed in all samples. But some differences were observed in free amino acid compositions in all samples, that is, glutamic acid, tyrosine, lysine and aspartic acid were detected more abundantly. Sum of free amino acids for 90 days were in order of koji soybean paste>traditional soybean paste>koji & natto soybean paste>natto soybean paste. The ratios of free to total amino acids were $3.28{\sim}19.81%$ for 45 days, but increased to $10.88{\sim}25.10%$ for 90 days, and in order of traditional doenjang>koji doenjang>koji & natto doenjang>natto doenjang. Methionine and histidine showed higher ratios of free to total amino acid but lower in glutamic acid and aspartic acid. These results suggest that koji and traditional soybean paste of having high ratios of free amino acids to free and total amino acids may be more favorable in soybean paste fermentation.

Changes of Components Affecting Organoleptic Quality during the Ripening of Traditional Korean Soybean Paste -Amino Nitrogen, Amino Acids, and Color- (한국 전통 된장의 숙성중 관능적 품질에 미치는 성분의 변화 -아미노산성질소, 아미노산 및 색도를 중심으로-)

  • This study was performed to investigate the changes of amino nitrogen, total amino acids, free amino acids, and cole. of traditional Korean soybean paste (doen-jang) during the ripening and storage for 12 months. All of the preparation methods for soybean paste followed the recommendations of the Korea Food Research Institute. The components of soybean paste were analyzed at 0, 6, and 12 months. The content of amino nitrogen of soybean paste was significantly higher than that of soybeans or meju (soybean cakes) at the initial stage of storage (p〈0.05), and decreased during the storage. The composition of total and free amino acids and their ratios of soybean paste were changed during the storage. The ratios of free to total amino acids of soybeans, meju, and soybean paste were 0.8%, 17.3%, and 20.4∼32.9%, respectively. Glutamic acid, which represents the savory taste, was detected the most abundantly in soybean paste during the storage. The ratios of free to total amino acids of glutamic acid were 21.1∼41.5% in soybean paste. Lightness, redness and yellowness of Hunter color of soybean paste decreased over time (p〈0.05). The results of this study indicate that the ratios of fee to total amino acids of soybean paste were much higher than those of soybeans, although its contents of total amino acids were much lower than those of soybeans. The results also indicate that this comes from the preparation and fermentation of meju. It was suspected that the organoleptic quality of soybean paste derived from these three components might be inferior over 1 year of storage time.

  • Coral damselfish (demoiselle), Chromis notatus, has been one of the most tasty sea foods in Jeju-Do, Korea, The free amino acids in tile extracts of Chromis notatus were analysed by amino acid autoanalyser. In the extracts, taurine, lysine, glycine and alanine were abundant, holding $30.3\%,\;22.7\%,\;16.0\%\;,\;11.5\%$ of total amino acid content, respectively and followed threonine, glutamic acid and serine, in order. While methionine, histidine, leucine, isoleucine, valine were poor and aspartic acid and proline were trace in content.

  • Changes of free amino acids and free sugars in steamed and roasted green tea were determined after heat treatment at $110^{\circ}C$. Sixteen kinds of free amino acids and four kinds of free sugar were analyzed by HPLC. Free amino acids isolated were aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine and arginine. Free sugars were sucrose, glucose, fructose, and raffinose. After the heat treatment, free amino acids and free sugars were decreased considerably.

The Taste Compounds in Commercial Toha-jeot (시판 토화젓의 정미성분에 관한 연구)

  • 전남지방의 전통 수산발효식품인 토하젓의 인지도 향상 및 국가경쟁력을 갖춘 지역특산품으로 발전시키기 위한 일련의 연구로써, 식품학적 기초자료를 얻기위하여 숙성기간이 다른 시판 토하젓을 구입하여 정미성분을 분석.검토하였다. 저온(5$\pm$2$^{\circ}C$)에서 90일간숙성시킨 토하젓의 전체 유리아미노산 함량은 건물당 1873.0mg/100g)보다 2배이상 많았다. 한편 숙성된 토하젓의 주요 유리아미노산(719.4mg/100g) 보다 2배 이상 많았다. 한편 숙성된 토하젓의 주요 유리아미노산은 glutamic acid, alanine, leucine, valine 및 phenylalanine으로 전체 유리아미노산에 대해 58.6%를 차지하였다. 핵산 관련물질은 담금 직후 및 숙성된 토하젓 모두 hypoxanthine의 함량이 월등히 많았으며, 그 다음으로 AMP 및 IMP순이였다. 그리고 숙성된 토하젓은 담금 직후의 토하젓과 비교해서 AMP 및 IMP는 적었으나, hypoxanthine의 함량은 많았다. 불휘발성 유기산을 GC분석한 결과, 담금 직후 및 숙성된 제품 모두에 lactic acid가 가장 많아 각각 건물당 389.3mg/100g 및 752.3mg/100g이였고, succinic acid는 미량 함유되어 있었다. 90일간 숙성된 TMAO, betaine 및 total creatinine 함량은 건물량 기준으로 각각 63.6mg/100g, 179.1mg/100g 및 123.7mg/100g이었다. Omission test를 이용하여 각정미성분을 관능검사한 결과, 유리아미노산 및 불휘발성 유기산이 토하젓의 맛에 중요한 역할을 하는 것으로 밝혀졌으며, 핵산 관련물질은 그다지 맛에 대한 기여도가 높지 않았다. 따라서 시판 토하젓의 맛에는 유리 아미노산 및 불휘발성 유기산이 중요한 역할을 하며, 핵산 관련물질이나 TMAO, betaine 및 total creatinine은 보조적인 역할을 할 것으로 생각된다.

Changes in Free Amino Acids of Yellow Corvenia, Pseudosciaena manchurica, during Gulbi Processing (굴비 가공(加工)중의 유리아미노산의 변화)

  • Gulbi, salted and dried yellow corvenia, is one of widely consuming fish foods in this country. This study was attempted to establish the basic data for evaluating the taste compounds of Gulbi. The free amino acids were analysed by amino acid autoanalyser. In the free amino acid composition of raw yellow corvenia, abundant amino acids were glutamic acid, alanine and lysine and then leucine, glycine, serine and threonine in order. Such amino acids like proline, valine, methionine, tyrosine and phenylalanine were poor in content. The changes of free amino acid composition in the extract of yellow corvenia during Gulbi processing was not observed. In the extract of Gulbi product, glutamic acid, lysine, leucine, alanine and valine were dominant holding 20.0%, 12.9%, 11.1%, 10.1% and 8.5% of total amino acids respectively. The amounts of total free amino acids of yellow corvenia increased more than 3.6 times as compared with that of raw sample during Gulbi processing. It is convinced that the characteristic flavor of Gulbi was attributed to such amino acids like lysine and alanine known as kweet compounds, as glutamic acid with meaty taste, and as leucine known as bitter taste.

  • 볶음조건에 따라 치커리의 유리당 함량은 무처리구에서는 sucrose 4.78%, fructose 0.81% 및 glucose 0.53%이었는데 볶음온도가 증가하고 볶음시간이 길어질수록 감소하여 16$0^{\circ}C$ 40분 볶음처리구에서는 sucrose 2.21%, fructose 0.35% 및 glucose 0.20%로 전체 유리당 함량으로는 약 45.10% 잔존하였다 아미노산은 18종이 분리되었고, 그 중 유리아미노산 함량은 arginine이 504.49 mg%, asparagine이 500.78 mg%, glutamic acid가 97.82 mg%이었고, 충아미노산 함량은 arginine이 676.7 mg%, glutamic acid가 583.9 mg%, aspartic acid가 319.2 mg%, prolineol 297.4 mg%로 다른 것에 비하여 높게 나타났다 볶음온도가 증가하고 볶음시간이 길어질수록 아미노산의 함량은 감소하였고, 그 정도도 다르게 나타났다. 전체함량으로 보면 유리 아미노산의 경우 무처리구에서는 1728.6 mg%이었던 것이 16$0^{\circ}C$ 40분 처리구에서 4.37 mg%로 약 0.25%만 잔존하였고, 총아미노산의 경우 무처리구에서 3159.4 mg%이었던 것이 16$0^{\circ}C$ 40분 볶음처리구에서는 1732.5 mg%이었다.

  • Yellow sea bream, Branchiostegus japonicus, has been one of the widely consumed food fish in Jeju-Do, Korea. This study was attempted to establish the basic data for evaluating changes of taste compounds of the yellow sea bream during dehydration. The free amino acids were analysed by amino acid autoanalyser. The free amino acid composition of the raw yellow sea bream, abundant amino acids were lysine, alanine, threonine and arginine and then taurine, serine, proline, glycine, glutamic acid, leucine in order. Such amino acids as valine, methionine, tyrosine, isoleucine, phenylalanine were poor in content. The changes of free anino acid composition in the extract of the yellow sea bream during dehydration was not found. In the extract of dried product, lysine, alanine, glutamic acid and arginine were dominant holding $18.4\%,\;14.7\%\;8.0\%\;and\;7.3\%$ and $7.3\%$ of total amino acids respectively. The amount of total free amino acids of the yellow sea bream increased to more than 1.4 times during dehydration process, that of raw sample especially aspartic and glutamic acids increased to more than 2.9 times and 2. 1 times whereas taurine and histidine decreased ranging $40-50\%$ of the original content. It is presumed that the characteristic flavor of dried yellow sea bream was attributed to such amino acids like lysine and alanine known as sweet compounds, glutamic acid as meaty taste and TMAO known as plain sweet taste.

Free Amino Acids of Xylem-Pith in Panax ginseng Root. (인삼근동체중심부의 유리아미노산)

  • Composition of free amino acids (FAA) in the central part (xylem plus piths,of tap root in P. ginseng was investigated in velation to stem status at harvest. The sum of FAA tended to be higher with dead stem than with healthy one but both were not significantly different. The sum of FAA (3.6-4.9% dried weight) was much less than total FAA, suggesting that water soluble nonprotein fraction contained large quantity of ninhydrih positive components except FAA. Pattern of amino acid composition between both stem status was not different. Ten of all 17 amino acids showed increasing tendency with dead stem and two, glutamic acid and cysteine, decreasing. Major FAA were arginine (relative content 58%), glycine (8.2), lysine (5.9), serine (5.7), glutamic acid (4.2) and aspartic acid (3.5). Above facts strongly suggest that the inside white of red ginseng did not closely related with FAA and that early defoliation or stem death did not decrease FAA. The content of arginine was heighest in all cases reported indicating the important role of nitrogen metabolism. Pattern of PAA composition except arginine was not different in present samples but greatly different with other cases reported mainly due to alanine, phenylalanine, glycine and proline.

Free Sugar, Free Amino Acid, Non-Volatile Organic Acid and Volatile Compounds of Dongchimi added with Jasoja(Perillae semen) (자소자 첨가 동치미의 유리당, 유리아미노산, 비휘발성 유기산 및 휘발성 향기성분)

  • All optional ingredient, Jasoja(Perillae semen) was adopted to improve Dongchimi in qualify during fermentation. Free sugar, free amino acid, non-volatile organic acid and volatile compounds were determined during fermentation at 10$^{\circ}C$ for 45 days. Free sugar content was slightly higher in 0.5%-Jasoja-treated samples than that of control. The contents of free amino acids in control Dongchimi (without jasoja) increased slowly during fermentation while those in 0.5 %-treated samples began to decrease after reaching their maximum value on the day 11 when Dongchimi became most acceptable. There were 6 non-volatile organic acids, such as lactic, fumaric, succinic, malic, tartaric, and citric acid. Among these, only lactic and succinic acid increased consistently with fermentation while others decreased. Volatile components in Dongchimi were mostly identified as sulfur-containing compounds by gas chromatography. Their numbers and % peak areas in the gas chromatogram decreased slightly with the increase in organic acids and alcohols during fermentation period. On the other hand, Dongchimi prepared with Jasoja maintained its contents of total acids as well as the level of sulfur-containing compounds.
