지방구가 제거된 돼지 미성숙 난포란의 유리화 동결에서 Straw법과 EM Grid법의 비교

  • 많은 연구에서 돼지의 난포란 혹은 배아가 낮은 온도에 대해 민감한 것은 세포질 내에 함유된 지방구와 관련이 있다고 하였으며, 이런 세포질내의 지방구를 제거함으로 동결능력이 향상된다고 하였다. 이렇게 지방구가 제거된 돼지의 난포란과 배아를 효과적으로 동결보존하기 위해서는 새로운 개념의 동결보존 기술이 필요하다고 하겠다. 따라서, 본 연구는 미성숙 단계에서 지방이 제거된 돼지 난포란의 유리화 동결에 적합한 방법을 찾기 위하여 straw를 이용한 유리화 동결법과 electron microscopic grid(EM grid)를 사용한 유리화 동결법을 실시하여 효율적인 동결방법을 검토하였다.

Free Amino Acid and Free Sugar Contents of Liquid Koji Kochujang (액체국에 의한 숙성고추장의 유리아미노산과 유리당의 함량)

  • The three kinds of Kochujang were prepared with liquid koji equivalent to 30%, 50% and 70% of brewing water and the contents of free amino acids and free sugars during aging at $25^{\circ}C$ for 3 months were compared with those of the control group which was made from solid koji. All tested Kochujang were higher in glutamic acid, lysine and aspartic acid while lower in methionine, histidine and tryptophan. The content of total free amino acids was high in the order of control group, 70%, 50%, and 30% liquid koji group. Fructose and rhamnose were found in all groups, but glucose in only the control group and 70% liquid koji group. Fructose content was the highest of the free sugars and total free sugar content was higher in the control group and 70% liquid koji group than other groups.

Thermal Insulation Property of UV Cure Coatings Using Hollow Micro-Spheres (마이크로 중공구를 이용한 자외선 경화 코팅 박막의 단열 특성)

  • In this study, the composite coating materials with improved thermal insulation property were prepared by incorporating the hollow micro-spheres with high heat transfer resistance. The UV curable resin system consisting of hexa aliphatic urethane acrylate (UP118), trimethylolpropane triacrylate (TMPTA), 1,6-hexanediol diacrylate (HDDA), and photoinitiator (Irgacure184) was employed as an organic binder. The glass substrates were coated by the prepared composites via bar coating method and cured under UV radiation. The optical transparency, thermal insulation property, adhesion, and surface hardness of the glass coated with composites containing different type of micro-spheres were investigated. The incorporation of micro-spheres with only 20 vol% of content resulted in remarkable improvement in the thermal insulation property of the coated glass. In addition, the transparent coated glass with light transmittance of about 80% could be obtained when silica micro-sphere (SP) was used as a thermal barrier.

Low Dilution Glass Bead Digestion Technique for the Trace Element Analysis of Rock Samples (저희석 유리구 용해법에 의한 암석시료 미량원소 분석법)

  • Open beaker digestion method is routinely used as the sample preparation technique for trace element determination of rock samples by inductively coupled plasma mass spectrometry, With this method, however, dissolution of Zr and Hf is not always guaranteed especially when the samples contain refractory minerals. In this study, glass bead digestion technique was compared with conventional open beaker digestion technique for the sample preparation of three USGS rock standards such as AGV-2, BHVO-2, and G-3. Thirty trace elements including rare earth elements were analysed by ICP-MS and ICP-AES. There were no clear differences in analytical results for the AGV-2 and BHVO-2 standards between the two techniques, but Zr, Hf, Y, and middle- to heavy- rare earth element concentrations of the G-3 standard prepared by open beaker digestion technique were significantly lower than the recommended values. This can be attributed to the presence of refractory mineral zircon. On the contrary, all the analytical results of the G-3 standard prepared by glass bead digestion technique were in good agreement with the recommended values, indicating complete dissolution of zircon. The analytical results show that the volatile elements such as Pb and Zn were not lost during the preparation of glass bead. Low dilution glass bead digestion technique described here will be very helpful to enhance precision and accuracy of trace element analysis for geological samples containing refractory minerals.

Effects of Exposure to Vitrification Solution on Maturation, Fertilization and Development of Immature Porcine Oocytes In Vitro (유리화 동결액 노출이 돼지 미성숙 난포란의 성숙율, 수정율 및 배발달율에 미치는 영향)

  • This study was conducted to investigate the toxi-cological effects of different vitrification solution on development of immature porcine oocytes in vitro. Oocytes were exposed to EFS solution [40% ethylene glycol (EG) + 18% Ficoll + 0.3M sucrose], ES solution (5.5M EG + 1.0M sucrose) or GE solution [10% glycol (G) + 20% EG], and these oocytes were transferred to sucrose solution directly. Maturation rates were significantly (P<0.05) higher in the ES solution (44.5%) and control (57.6%) than in the EFS solution (38.8%) and GE solution (22.4%). No differences among three solution were found in fertilization rates. Cleavage rates was significantly (P<0.05) higher in the ES solution (47.1%) and control (65.9%) than in the EFS solution (21.9%) and GE solution (19.0%), but no difference among three solutions was found in the blastocyst formation rates. These results indicate that combination of EG and sucrose solutions had effects on development of immature porcine oocytes.

Improvement of Canopy Light Distribution, Photosynthesis, and Growth of Lettuce (Lactuca Sativa L.) in Plant Factory Conditions by Using Filters to Diffuse Light from LEDs (LED 식물공장에서 산란 유리 이용에 의한 상추(Lactuca Sativa L.)의 군락 광분포, 광합성 및 생장 향상)

  • Plant factories with artificial lights require a large amount of electrical energy for lighting; therefore, enhancement of light use efficiency will decrease the cost of plant production. The objective of this study was to enhance the light use efficiency by using filters to diffuse the light from LED sources in plant factory conditions. The two treatments used diffuse glasses with haze factors of 40% and 80%, and a control without the filter. For each treatment, canopy light distribution was evaluated by a 3-D ray tracing method and canopy photosynthesis was measured with a sealed acrylic chamber. Sixteen lettuces for each treatment were cultivated hydroponically in a plant factory for 28 days after transplanting and their growth was compared. Simulation results showed that the light absorption was concentrated on the upper part of the lettuce canopy in treatments and control. The control showed particularly poor canopy light distribution with hotspots of light intensity; thus the light use efficiency decreased compared to the treatments. Total light absorption was the highest in the control; however, the amount of effective light absorption was higher in treatments than the control, and was highest in treatment using filters with a haze factor of 80%. Canopy photosynthesis and plant growth were significantly higher in all the treatments. In conclusion, application of the diffuse glass filters enhanced the canopy light distribution, photosynthesis, and growth of the plants under LED lighting, resulting in enhanced the light use efficiency in plant factory conditions.

찹쌀과 찹쌀가루가 고추장의 품질에 미치는 영향

  • 찹쌀 및 가루를 사용하여 담금한 고추장(시험구 A: 찹쌀가루, 시험구 B: 찹쌀, 시험구 C: 찹쌀 및 가루의 혼용)의 화학성분, 효소력 및 texture를 측정한 결과는 다음과 같다. 수분, 조단백, 조지방, 적정산도, 무기성분, alcohol의 함량 및 PH는 시험구간에 차이가 없었다. 이미노태 질소함량과 산성 pretense 및 당화 amylase 활성은 A구가 높았고 환원당은 Brn가 경시적으로 높았다. Ca, Cu, K, Mn, Mg, P, Fe의 무기성분이 각 시험구에서 검출되었고 이중 K의 함량이 가장 높았으며 Cu, Mn, Fe는 극히 미량이었다. Aspartic acid, glutamic acid, cystine, arginia proline 등의 유리아미노산이 60일 숙성 고추장에서 양적으로 많이 검출되었고 histidine, valine의 함량은 적었으며, 유리아미노산의 총량은 A구 16.49, B구 13.68, C 구 15.84mg/g으로서 A 구가 가장 많았고 B 구의 고추장이 가장 적었다. texture를 측정한 결과 견고성과 부착성은 담금 직후에는 A구가, 숙성 후기에는 B 구가 높았으며 응집성, 탄력성은 A 구가 높았고 B 구가 낮았다.

Free Sugar, Free Amino Acid, Non-Volatile Organic Acid and Volatile Compounds of Dongchimi added with Jasoja(Perillae semen) (자소자 첨가 동치미의 유리당, 유리아미노산, 비휘발성 유기산 및 휘발성 향기성분)

  • All optional ingredient, Jasoja(Perillae semen) was adopted to improve Dongchimi in qualify during fermentation. Free sugar, free amino acid, non-volatile organic acid and volatile compounds were determined during fermentation at 10$^{\circ}C$ for 45 days. Free sugar content was slightly higher in 0.5%-Jasoja-treated samples than that of control. The contents of free amino acids in control Dongchimi (without jasoja) increased slowly during fermentation while those in 0.5 %-treated samples began to decrease after reaching their maximum value on the day 11 when Dongchimi became most acceptable. There were 6 non-volatile organic acids, such as lactic, fumaric, succinic, malic, tartaric, and citric acid. Among these, only lactic and succinic acid increased consistently with fermentation while others decreased. Volatile components in Dongchimi were mostly identified as sulfur-containing compounds by gas chromatography. Their numbers and % peak areas in the gas chromatogram decreased slightly with the increase in organic acids and alcohols during fermentation period. On the other hand, Dongchimi prepared with Jasoja maintained its contents of total acids as well as the level of sulfur-containing compounds.

감잎 품종별 성분분석과 항산화활성 비교

  • 품종별 감잎의 주요성분과 항산화 활성을 비교하여 고기능성 감잎차의 우수성을 알리는 기초자료로 활용하고자 이 실험을 수행하였다. 상주둥시, 갑주백목, 월하시, 청도반시, 녹차(대조구)의 생엽을 7월 초순에 채엽 후 건조시켜 시료로 사용하였고, 비타민C, 카페인, 카테친, Gallic acid, 향기성분, 유리당, 유리아미노산, 칼슘, 항산화 활성을 조사하였다. 비타민C는 월하시 0.089%, 청도반시 0.079%, 갑주배목 0.032%, 상주둥시 0.015%, 녹차 0.01%로 비타민C 함량이 가장 많은 월하시가 녹차에 비해 8배정도 높았고, 카페인은 감잎 4품종 모두 발견되지 않았고, 녹차는 6.63%이었다. 카테친은 청도반시 0.35%, 갑주백목 0.34%, 월하시 0.24%, 상주둥시 0.18%, 녹차(대조구) 0.07%이였고, Gallic acid는 상주 둥시 0.32%, 갑주백옥 0.2%, 월하시 0.05%, 청도반시 0.03%, 녹차(대조구) 1.41%이었다. 칼슘은 청도반시 9516.1PPM, 영동월하시 6863.5PPM, 봉옥 6563.5PPM, 상주둥시 5420.1PPM, 녹차(대조구) 2349.7PPM이였고, 유리당은 감품종간에는 큰 차이는 없었으나 녹차에 비해 Xylose, fructose, Glucose, Sucrose의 함량이 높았고, Maltose의 함량에 일어서는 녹차가 높았다. 유리아미노산은 31항목을 조사하였고, 전체 함량은 상주둥시 60.40, 봉옥 53.21, 월하시 52.29, 청도반시 47.58, 녹차(대조구) 114.72nmo1/${\mu}\ell$이었다. 시료의 향기성분은 생엽을 건조시켜 전자코를 이용하여 분석하였고, 감잎 품종간에는 향기 패턴이 비슷한 경향이였으나, 녹차의 패턴과는 차이가 있었다. 감잎의 DPPH radical 소거활성은 상주둥시 RC$_{50}$($\mu\textrm{g}$)=64.5 청도반시 64.0, 월하시 42.0, 갑주백목 47.0, 녹차(대조구) 19.0으로 김잎품종중 월하시의 항산화 활성이 높았고, 녹차의 항산화 활성이 감잎에 비해 높았다.

Changes in Free Sugars and Organic Acids of Banana Fruit at Various Storage Temperatures (저장온도에 따른 바나나의 유리당과 유기산)

  • Banana purchased from local market was stored at 5, 15 and $25^{\circ}C$ for 10 days after which free sugars and organic acids were determined. The amount of glucose, fructose and sucrose was $0.30{\sim}0.36%$ at the initial time of storage. The changes of free sugar was influenced by storage temperature. At $5^{\circ}C$, little change in the content of each free sugar was measured and sucrose had a slightly higher value. Glucose and fructose showed a predominant increase while sucrose content decreased at 15 and $25^{\circ}C$. The values of total sugar, reducing sugar, glucose, fructose and free sugar contents at $25^{\circ}C$ were higher than at any other storage temperature. Citrate, malate, lactate and acetate in Banana were identified as organic acids. The total amount of organic acid decreased from $305{\sim}373.3$ mg% at the initial time to $45.9{\sim}67.9$ mg% after 9 days of storage. The reduction of organic acids contents was relatively high at $25^{\circ}C$.
