Effect of Copper on Lethal Dose and Pupation of Aedes albopictus (Diptera: Culicidae) (흰줄숲모기, Aedes albopictus (Diptera: Culicidae)의 치사농도 및 용화에 대한 구리의 영향)

  • The mosquito, Aedes albopictus is important vector of humen phatogen such as dengue fever. And Aedes albopictus has widly distribution in oriental regions. This study was conducted to investigate the biological effects of cooper on the development of the Asian tiger mosquitoes, Aedes albopictus (Skuse). Aedes albopictus was affected with pupation ratios, mortalities, and wing lengths of the 3th and 4th instars by exposure to different concentration $CuCl_2$ (0.0, 2.5, 25.0 and 50.0 ppm) for 24 h and 48 h depending on dose and exposure period. The lowest $LC_{50}$ (median lethal concentration) value of $CuCl_2$ was 18.1 ppm at the 3rd instars for 48 h exposure. The pupation ratio of the larvae exposed at 50.0 ppm for 48 h was 14% which was much lower than those exposed to other treatment groups Changes of adult wing length showed significant decreased by Cu treatment. In conclusion, the most prominent effects of high concentration and longer exposure period were to reduce in the survival and pupation rates of larvae and wing length of Ae. albopictus.

Variation in Development and DNA Methylation of Spodoptera exigua Fed with Different Diets (먹이에 따른 파밤나방 발육과 DNA 메틸화 변이)

  • Physiological plasticity of insects can be closely related with their epigenetic change. This hypothesis was tested using a polyphagous lepidopteran insect, Spodoptera exigua, by assessing the effects of different diets on development and DNA methylation. Three different diets (Welsh onion (WO), Chinese cabbage (CC), artificial diet (AD)) were assessed by feeding a cohort of larvae from neonate to last instar. There were significant differences in larval developmental rate, pupal weight and adult emergence according to diet treatments. AD-fed larvae exhibited the fastest developmental rate along with the highest pupal weight and adult emergence. Among natural hosts, WO was more favorable for development of S. exigua than CC. Total hemolymph proteins and sugars in the last instar larvae were varied among different diets. Gene expression of an insulin-like peptide (SeILP1) presumably associated with development was also varied among diets. Cytosine methylation of genomic DNA was assessed using a monoclonal antibody. Genomic DNA of S. exigua larvae was methylated. DNA methylation was apparently varied among different diet-fed larvae. The facts that a cohort of S. exigua was differentiated in developmental rate and cytosine methylation by different diets suggest that epigenetic factor(s) may play a crucial role in the physiological plasticity.

Developmental Ecology of Persimmon Fruit Moth, Stathmopoda masinissa Meyrick (Lepidoptera: Stathmopodidae) (감꼭지나방 (나비목: 감꼭지나방과)의 발육생태)

  • Persimmon fruit moth, Stathmopoda masinissa, collected from roadside persimmon trees planted as shade trees in yeongdong area was used to investigate their developmental ecology under laboratory conditions; $25\pm$ $1^{\circ}C$, 16L : 8D and 85% RH. Developmental periods for eggs, larvae, and pupae were 7.4, 34.8 and 15.5 days, respectively. The duration for each larval instar from the 1st to the 5th was 3.5, 4.2, 5.2, 6.5 and 15.4 days, respectively, and the head capsule width of corresponding stage was 0.20, 0.40, 0.65, 0.87 and 1.07 mm. Rates of pupation and emergence were 68.0 and 59.9%, respectively. Longevity of adult males was 6.2 days while that of females was 10.1 days. Average fecundity was 25.4 eggs.

Breeding of Major Characteristics of the Wild Silkmoth, Antheraea yamamai indoor-and outdoor-reared (실내외 사육에 의한 천잠 우량계통 육성)

  • We investigated fertilization rate, hatchability rate, pupation rate and cocoon quality etc to selecting good characteristics of the wild silkmoth, Antheraea yamamai using method of indoor-and outdoor-rearing. As results of indoor rearing, the healthiness of A. yamamai was better in light green colour strain than that of dark green colour strain, but cocoon quality was better in dark green colour strain. There was a little effect to selecting major characteristics in grade of A, B and C strain classified with cocoon weight. $F_1$ hybrid was lower in fertilization rate and hatchability rate than parent, but higher in pupation rate and cocoon quality. There was no effect of heterosis in $F_2$ hybrid. In case of outdoor rearing, mortality rate was lower than that of indoor rearing but missing larva was higher. In addition, outdoor rearing is higher that indoor rearing in cocoon weight, cocoon shell weight and cocoon shell rate.

  • This paper reports a simple rearing system for Cnaphalocrocis medinalis based on corn seedlings diet. C. medinalis population under this system has been maintained by four stages (egg, young larva-1st to 3rd instar, old larva-4th to prepupa, 3nd adult) at $25{\pm}2^{\circ}C,\;40{\pm}10%$ RH, 3nd 16L:8D photoperiod in a laboratory. We have elaborated a new egg collection method using a polystyrene container $(top\;{\phi}11.3\;{\times}\;bottom\;{\phi}\;{\times}\;H8cm)$ which has made a great contribution in easy collection of eggs and storage of them for long period. Under this system, pupation and adult emergence rate, and pupal weight of C. medinalis population were close to the other reports by com seedling diet, and superior to those by artificial diets. Therefore, this rearing system would be useful in obtaining C. medinalsis population suitable to each specific needs.

Effect of Popcorn, Wolbachia Variant, on Development and Reproduction in Drosophila melanogaster (Wolbachia 변이종 Popcorn이 초파리의 발육과 생식에 미치는 영향)

  • Wolbachia, vertically transmitted bacterial endosymbionts, is known to induce cytoplasmic incompatibility (CI), parthenogenesis, or feminization in some insect species. The Wolbachia found to exist symbiotically as a non-virulent form in Drosophila melanogaster, however popcorn is a lethal strain of Wolbachia, which causes tissue degeneration and early death of its adult host. Popcorn-infected flies showed the delayed duration of an egg and larvae, and their pupal period and life span reduced. The oviposition and egg-hatching rate of popcorn-infected flies were decreased 15% and 80%, respectiv~ly, compared to those of the normal Wolbachia-infected flies. The pupation and emergence rates of popcorn-infected flies were 67% and 65%, respectively. When popcorn-infected flies were crossed with Wolba-chia-infected flies, and vice versa, both crosses resulted in a significant reduction in egg production, egg¬hatching, pupation, and emergence rate, and their progeny revealed the popcorn syndrome. When popcorn-infected male flies were crossed with uninfected females, popcorn was not detected in their progeny. Popcorn also maternally transmitted in flies, but it did not induce CI and affected on its host life cycle as a virulence.

Mass-rearing Method of the Fungus Gnat, Bradysia difformis (Sciaridae, Diptera) in Laboratory (실내에서 작은뿌리파리(Bradysia difformis, Sciaridae, Diptera)의 대량사육시스템)

  • The fungus gnat, Bradysia difformis, has been recognized as an important pest of greenhouse crops. There is a need for research on the control of the fungus gnat. However, it is difficult to obtain many generations of the fungus gnat for several kinds of research. Indoor propagation is a very useful method for obtaining enough individuals in cases where the need is for larvae in soil. This study was conducted to determine the optimum growing media and temperature conditions for rearing the fungus gnat in the laboratory. Under experimental temperature conditions, hatching, pupation, and eclosion rates were the highest at $20^{\circ}C$. The developmental period of the fungus gnat was shortened with higher temperatures. The greatest number of eggs was an average of 144 at $20^{\circ}C$. Using different types of larvae growth media, the highest hatching rates were 84.7 and 84.4% in water agar and potato disks, respectively. The larval period was the shortest, at 14.7 days, when grown on potato disks. The highest pupation and eclosion rates were 85.2 and 82.6% on potato disks, respectively. The highest number of eggs was an average of 125.6 on potato disks. Regarding the effects of different growth media on the eclosion rate of B. difformis, the highest eclosion rate was 88.4% on the soil mix, and was 50% on oatmeal, 25% on shredded potato. The results of four different inoculation levels of larvae on eclosion rate of B. difformis showed that the highest eclosion rate was 84.7% for 1,000 larvae. The eclosion rate was shortened with a higher number of larvae inoculated/cage. In the growth medium used, 3,000 eggs were better for the initial level of inoculation, showing a relatively high emergence rate and short developmental period. Mass rearing procedures were explained in detail.

Insecticidal Activities of Prunus mume Extract Against Rice Leaf Folder (Cnaphalocrocis medinalis Guenee) (혹명나방에 대한 매실나무 추출물의 살충활성)

  • Insecticidal activities of methanol extracts of the different parts of Prunus mume were investigated against the larvae of Cnaphalocrocis medinalis (C. medinalis) by topical application. The mortalities of the larvae of C. medinalis were 34% at the concentration of 4,000 ppm, 36% of 8,000 ppm, 40% of 16,000 ppm and 58% of 32,000 ppm of the extracts from the fruits, and 38% of 4,000 ppm, 45% of 8,000 ppm, 58% of 16,000 ppm and 75% of 32,000 ppm from the stem + flower of Prunus mume (P. mume), respectively. The rates of pupation of C. medinalis treated with either fruit or stem+flower were gradually low according to the concentration of extracts increased. The weight of pupae of C. medinalis has no significantly different in between the control and the treated groups of the extracts from both of fruit and stem+flower. However, the differences of the pupal weight and the rate of pupation of C. medinalis treated with the extracts from the stem+flower and fruit of P. mume were significant as the level of 56% ($R^2$=0.56363, P=0.4364). Both of stem+flower and fruit extracts of P. mume showed good effects of anti-feeding against C. medinalis. The extracts of the stem+flower of P. mume could be advocated for developing as birational agent for the control of C. medinalis.

Indoor Rearing Method of Diving Beetles: Cybister japonicus, Cybister tripunctatus orientalis, Cybister brevis (물방개류 실내 사육법)

  • The aim of this study is to develop indoor-rearing methods of the diving beetles. In nature, both the adult diving beetle and its larvae are voracious aquatic predators. The larvae beetles hunt relatively bigger size of tadpoles and small fish for food source. However, due to difficulties of the food supplement for rearing diving beetles at indoor-condition further motivated us to develop new artificial food. Three separate experiments were performed. In the first experiment, adult beetles were provided with one of the several food choice treatments to self-compose their preferred foods that are affordable on the market at lower price. The second experiment was also to develop artificial diet that is possible for rearing larvae beetle under indoor condition. The larvae beetles were restricted to raw squid, artificial food source and mosquito larvae as a control at the first stadium and small fish and raw squid during second to third stadium duration. According to our result, adult beetles selected a food that made of boiled squid and dead small fish while, the young larvae consumed small fish, mosquito larvae and raw squid. Although, the larval food restriction on law squid caused noticeable decrease in survival, the result still supported the possible survival rate of keeping larvae at indoor condition. Moreover, pupation rate experiments, in which groups of larvae were placed at different mats, natural soil and fermented sawdust, showed that 80% of diving beetles pupated on the sawdust. This result indicates that female beetle preferentially selected to oviposit along soft and moist area.

Rearing and Innoculation Condition of Nd hybrid Silkworms for Paecilomyces tenuipes Production (Paecilomyces tenuipes 생산을 위한 나용교잡종 누에 사육 및 균 접종 조건)

  • To shorten the labor cost for the production of the fungus, Paecilomyces tenuipies, uncocooning mutant hybrid silkworms as a host was used. The yield of P. tenuipies was reduced when Nd (uncocooning mutant) was used as a host because Nd was smaller than the hybrid Baegokjam that has been used as a typical host. To increase the host weight, Nd was crossed with Jam l23 or Jam 124, and resulted in not only greater weighter but also higher yield of P. tenuipies. The optimum temperature for rearing Nd hybrid silkworms was $3^{\circ}C$ higher than standard temperature of rearing silkworms to increase the naked pupation rate. The optimum time for the infection of P. tenuipies was 12 h at $28.5^{\circ}C$ and 95% R.H., when $1{\times}10^7spores/ml$ of P. tenuipies were sprayed on the surface of the silkworm. The conditions were applicable to both spring-rearing and autumn-rearing.
