Effects of Environmental Factors on the Bacterial Community in Eutrophic Masan Reservoir (이화학적 수질인자가 부영양화된 마산저수지의 세균분포에 미치는 영향)

  • The total bacterial numbers, Eubacterial community structures and environmental factors which affect bacterial community were estimated monthly using DAPI and fluorescent in situ hybridization monthly, from June to November 2000 to evaluate the correlation between the bacterial community and environmental factors in eutrophic agricultural Masan reservoir in Asan. Average water temperatures varied from 12.3 to $27.5^{\circ}C$, pH 7.5 to 9.0, DO 7. I~12.8 mg/L, COD 6.4~13.0 mg/L, chlorophyll a 30.5~99.0 mg/㎥, SS 7.S~25.7 mg/L, TN 1.748~3.543 mg/L., and TP 0.104~0.581 mg/L, respectively. Total bacterial numbers showed high ranges from 0.4 to 9.6$\times$ $10^{6}$ cells/ml, and these indicated the mesotrophic or eutrophic state. The ratio of Eubacteria to total bacteria was 67.6-88.0%, which was higher than that in other reservoir. The relationships of total bacteria and Eubacteria community were more significant with organic nitrogen (Org-N), and organic phosphorus (Org-P) than with water temperature. Proteobacteria groups showed strongly significant relationships with Org-P and Org-N and significant relationships with water temperature, conductivity, COD, and inorganic nitrogen. C-F group was the most significant with Org-N, and HGC group with water temperature. However, relationships of Chl-a, pH, DO and SS showed no significance with any bacterial community. These results were different from other studies, because of the specific characteristics of Masan reservoir such as old, shallow and eutrophic states. The seasonal variation of bacterial community in Masan reservoir does not seem to depend on phytoplankton dynamics but on storm event and organic materials from watershed and the sediment of reservoir.

Analysis of Attached Bacterial Communities of Biological Activated Carbon Process Using DGGE Method (DGGE 기법을 이용한 생물활성탄 공정의 부착 박테리아 군집분석)

  • The concentration of organic compounds was analyzed at each step of BAC (biological activated carbon) process though BDOC (biodegradable dissolved organic carbon) total/rapid/slow. Further, bacteria communities and biomass concentrations measured DGGE (denaturing gradirnt gel electrophoresis) and ATP (adenosine triphosphate) methods were analyzed. The bed volume of steady state is different based on assessment of organic compounds removal. Bed volumes at steady state in DOC, $BDOC_{rapid}$ and $BDOC_{total/slow}$ removal were around 27,500, 15,000 and 32,000, respectively. A biomass didn't change after the bed volume reached 22,500 according to analyzing HPC (heterotrophic plate count) and ATP concentration of bacteria. The concentration of HPC and ATP were $3.3{\times}10^8$ cells/g and $2.14{\mu}g/g$, respectively. The number of the DGGE band were only 5 at the bed volume 8,916, but increased up to 11 at the bed volume 49,632. As operation time increase, bacterial group were more diversity. Four bacteria species including Pseudomonas fluorescens, the uncultured bacterium similar to Acinetobacteria, uncultured Novosphingobium sp. and Flavobacterium frigidarium have detected from the early stages and Proteobacteria group were dominantly detected.

The Pretreatment Effects on Methane Fermentation of Microalgal Biomass (미세조류의 전처리에 따른 메탄발효 특성)

  • This study was conducted to identify the characteristics of methane fermentation and lysis effects of pre-treated microalgae. Chemical compositions of microalgae showed that the VS(volatile solid) was 86.1% of TS(total solid), and the protein was 63.5% of VS. These values were higher than those of activated sludge. The cell lysis test of raw microalgae biomass was conducted by many physicochemical methods. presenting that the degree of cell lysis was affected by following order: ultrasonic(100min.), alkali(pH 13), ultrasonic(10min), thermal($120^{\circ}C$), thermal($50^{\circ}C$), and acidic(pH 3) treatment. Methane fermentation with many pre-treated samples was performed, showing that the concentration of acetic acid was the highest. followed by propionic acid, butylic acid and valerie acid among all VFA(volatile fattic acid). In methane production. ultrasonic samples were only more effective than untreated one in total gas and methane productivity. but other samples were less effective. Especially. the alkalic sample had an inhibitation effect on methanogens.

Sorption of Dissolved Inorganic Phosphorus to Zero Valent Iron and Black Shale as Reactive Materials (반응매질로서의 영가철 및 블랙셰일에 용존무기 인산염 흡착)

  • In order to reduce the availability of dissolved inorganic phosphorus in surface water, lakes, and estuaries, black shale and zero valent iron can be used as reacitve materials. Sorption of phosphate to sampled sediment, black shale, and zero valent iron was quantitatively evaluated in this research. Effect of coexistence of calcium was also tested, since coexisting ions can enhance the precipitation of phosphate. An empirical kinetic model with fast sorption(k$_t$), slow sorption(k$_s$), and precipitation(k$_p$) was well fitted to experiment data from this research. Langmuir and Freundlich sorption isotherms were also used to evaluated phosphate maximum sorption capacity. Calcium ions at 0, 1 and 5 mM affected the precipitation kinetic coefficient in empirical kinetic model but did not have impact on the maximum sorbed concentration.

Effect of Inorganic Coagulants on the Performance of Electro-Chemical Treatment Process Treating Hospital Wastewater (병원폐수의 전기화학적 처리시 무기응집제 주입 효과에 관한 연구)

  • Effect of inorganic coagulants dosing on the performance of electro-chemical process was studied when treating hospital wastewater having low electrolyte concentration. It is thought that adding inorganic coagulants caused increase in concentration of electrolyte and this caused increase in free chloride concentration and consequently, caused increase in indirect oxidation effect. Thus, COD removal efficiencies more than doubled in percentage terms at the 2 hrs of reaction time and current density of $1.76A/dm^2$ compared with the results obtained from the parallel experiments without adding inorganic coagulants. T-N removal efficiencies approximately doubled in percentage terms at the 2 hrs of reaction time and 700 ppm of coagulants addition and applied current density of $1.76A/dm^2$ due to the increase of free residual chlorine such as HOCl caused by increase of electrolyte concentration through the addition of inorganic coagulants. Under the same experimental condition, more than 90% of T-P removal efficiencies was obtained. The reason can be explained that increase of chemical adsorption rate between phosphate and insoluble metal compounds caused by dissolved oxygen generated from anode by the increased electrolyte concentration through inorganic coagulants addition make a major role in improving T-P removal efficiencies. It can be concluded that inorganic coagulants addition as the supplemental agent of electrolyte is effective way in improving organic and nutrient salt removal efficiency when treating hospital wastewater having low electrolyte concentration.

Effect of Advanced Treatment Process for Residual Chlorine Decay and THM Formation in Water Distribution System (고도처리공정이 관로 내 잔류염소 감소 및 THM 생성에 미치는 영향)

  • According to increase of consumer's desire for clean tap water, advanced treatment processes include with membrane, ozone, and granular activated carbon(GAC) were introduced. In order to evaluate the effect of advanced treatment processes for residual chlorine decay and trihalomethane(THM) formation in water distribution system, dissolved organic matter(DOC) removal of each advanced treatment process was investigated. The residual chlorine decay and THM formation using bottle tests were also evaluated. $UV_{254}$ removal in all advanced treatment was better than DOC removal. Especially, DOC by ozone treated was removed as 4% in contrast with sand filtered water, but $UV_{254}$ was removed about 17%. This result might be due to convert from hydrophobic DOC to hydrophilic DOC by ozonation. Ozone/GAC process was most effective process for DOC removal. The residual chlorine decay constants in treated water by sand filtration, ozonation, GAC adsorption, and ozone/GAC processes were 0.0230, 0.0307, 0.0117 and 0.0098 $hr^{-1}$, respectively. The sand filtered water was produced 81.8 ${\mu}g/L$ of THM after 190 hours of reaction time, as the treated water by ozone, GAC, and Ozone/GAC was less produced 6.0, 26.2, 30.3% in contrast with sand filtered water, respectively. Consequently, the durability of residual chlorine and reduction of THM formation were improved by advanced treatment processes.

Effects of Sewage Effluent on Organic Matters of Nakdong River: Comparison of Daily Loading (낙동강 수계 내 하수처리 방류수가 하류 하천 유기물에 미치는 영향: 부하량 비교)

  • This study investigated the water quality of effluents from the wastewater treatment plants, located at the Gumi Complex 4, Gumi, and Wonpyong, in Gumi. DOC accounted for higher than 70% of TOC, and oxidation efficiencies, calculated from carbon, were 13~43% for BOD and 37~73% for CODMn, respectively. Based on the biological decomposition experiments, R-DOC account for higher than 70% of DOC, mostly being occupied by refractory organic matters. This indicated that the biodegradable organics occupied more proportions of organic loadings than the refractory organics. The effect of the organics from a discharge of a sewage treatment plant on rivers, Gumi industrial Complex 4, Gumi, and Wonpyong on lower streams of the Nakdong River were found to be 15%, 6% and 16% respectively. The ratio of 15% suggests that comparatively, no large portions of TOC loadings are occupied, but the problem is that the biodegradable organic matters occupy a lot more proportions than that of the refractory organic matters. Thus, it is highly estimated that the refractory organics can gradually increase the pollution level of organics and precursors of disinfection by-products to the down-stream water treatment plants.

Application of Newly PAC Selection Method Based on Economic Efficiency (경제성을 고려한 새로운 PAC 선정방법의 적용)

  • In order to applicate a newly method for powdered activated carbon(PAC) selection based on economic efficiency, PAC adsorption tests were performed for removal of MIB and dissolved organic carbon(DOC) in drinking water supplies. The removal rate of MIB increased when the PAC dose increased. The Coal-based PACs were superior for adsorption of MIB compared to wood-based PACs. PAC adsorption of DOC and $UV_{254}$ were a little different for different PACs and types of raw water, but both were lower than adsorption of MIB. Among the tested PACs, the one called P-1000 was most effective for removal of MIB, DOC and $UV_{254}$. Most of the organics in the tested samples were proven by excitation emission matrix(EEM) results to be fulvic-like materials. Especially, fulvic-like materials, humic-like materials, and soluble microbial byproduct(SMP)-like materials decreased after contact with PAC. P-1000 which had the lowest MIB cost index(MCI) was selected as the optimum PAC for the target water. PAC efficiency and treatability, particle size and distribution, and the cost associated with PAC dosing for MIB removal according to DOC concentration should all be considered before making the final selection of the best PAC for the target water.

Removal of organic Carbon, Nitrogen and Phosphorus in Wastewater based on tapered Aeration with Bacillus sp. (점감포기에 의한 바실러스 특성을 이용한 폐수의 유기물질 및 질소, 인 처리에 관한 연구)

  • This study was conducted to investigate an aeration tank with RBC process attached Bacillus sp. known as a suitable microorganism for the removing of organic carbon, nitrogen and phosphorus. An aeration tank was based on tapered aeration because Bacillus sp. was well grown in this like environment conditions. The biofilm process with Bacillus sp. as an advanced treatment process could be a best technology for the prominent removal of organic carbon, nitrogen and phosphorus if the mechanism in the process is verified. The operation conditions of DO in the tapered aeration tank were maintained as $1.2{\sim}1.5mg/L$ in aeration tank1, as $0.3{\sim}0.5mg/L$ in aeration tank 2 and less than 0.2 mg/L in aeration tank 3, respectively. Lab-scale experiments were conducted, at room temperature, internal recycle rate was from 200% to 50% and returned sludge rate was from 100% to 50%. As a result, concentration of organic carbons, nitrogen and phosphorus in Period 1 (the time of Bacillus sp. adapted to environment) were decreased gradually. Ultimately, each removal rate in this biological experiment were TCODCr 94%, BOD 87%, T-N 85%, T-P 89% in Period 2. Hence, this process showed an excellent performance of the removal of organic carbon, nitrogen and phosphorus and this is an effective system fur treating of wastewater.

Remediation of Soils Contaminated with Arsenic and heavy Metals by Soil Washing (토양세척에 의한 비소 및 중금속 오염토양의 복원)

  • In order to remediate soils contaminated with oxyanionic As and cationic Zn and Ni through the pilot-scale acid washing, the effectiveness of acid washing and the properties of contaminated soils, fine soil particle and dissolved contaminants were evaluated. $H_{2}SO_4\;and\;H_{3}PO_4$ washing at pH $2{\sim}3$ enhanced the removal of As by the presence of competitive oxyanions and HCl washing effectively removed simultaneously As, Zn and Ni. The effectiveness of soil washing was little enhanced above the critical reaction time, and the carbonate, Fe/Mn oxide and organic/sulfides associated fraction were dominantly removed. The washing of coarse soil particles was highly efficient, but that of fine soil particles($<74{\mu}m$) was recalcitrant due to the enrichment with contaminants. Moreover, the physical separation of fine particles($<149{\mu}m$) enhanced the overall efficiency of soil washing. Therefore, both chemical extraction and separation of fine soil particles showed the high effectiveness of soil washing in the intersection point to minimize the amount of fine soil particles and to maximize the chemical extraction of contaminants.
