Application of a Modified Sublimation Method to Screen for PAH-Degrading Microorganisms (다환 방향족 탄화수소(PAH) 분해 미생물 탐색을 위한 승화법의 개발)

  • Recent studies have described various microorganisms that can degrade PAH, however, there are currently limited methods available to screen for PAH-degrading microorganisms. To screen for PAH-degrading microorganisms, a sublimation method (Alley, Jeremy F. and Lewis R. Brown. 2000. Appl. Environ. Microbiol. 66, 439-442) was modified to produce a simple screening system. In our results, there were several bacterial species capable of pyrene degradation including genera, Coryenbacterium, Gordonia, Rhodococcus, and Streptomyces, which have been screened from 350 bacterial isolates of commercial gasoline and oil-spilled sediment by the sublimation method. The main advantage of this method is that it (i) safely deposits an even, thin and visible layer of PAH onto the agar surface without the use of solvents and (ii) the quantity of PAH sublimed onto the agar can be easily controlled. Overall, this sublimation method may be an effective and simple technique to screen for PAH-degrading microorganisms.

Synthesis of $TiO_2$ Powders by the Hydrolysis of Titanium n-Butoxide and Reaction Mechanism (Titanium n-Butoxide의 가수분해에 의한 $TiO_2$ 분말 합성과 반응 메커니즘)

  • $TiO_2$ powders were prepared via hydrolysis of titanium n-butoxide in n-butanol and hydrolysis mechanism of titanium n-butoxide was studied using UV-Vis spectrometer. Hydrolysis reactions were controlled to proceed to pseudo-first order reaction in the presence of excess water. The phases of $TiO_2$ powders, prepared under the these conditions, were identified by XRD and reaction rates were calculated by Gugggenheim method. Prepared powders were noncrystalline states in their initial stage of formation but transformed to crystalline rutile structure by heating. Reaction mechanism of titanium n-butoxide was proposed as Interchange-Associative(Ia) mechanism, based on the data of n-value and termodynamic parameters which were determined from the rate constants.

  • Hemicellulosic hydrolysates contain not only sugars but also several kinds of ethanol fermentation inhibitory substances such as carboxylic acids, furans and phenolic compounds. In this work, emulsion liquid membrane (ELM) was chosen as a separation technology to remove the inhibitors. A basic simulated hemicellulosic hydrolysate was composed of xylose as sugar, dilute sulfuric acid solution as solvent, and acetic acid as carboxylic acid, and furfural as furan derivative or p-hydroxybenzoic acid(HBA) as phenolic compound was added to the hydrolysate when necessary. Acetic acid and HBA as weak acid could be selectively removed from the hydrolysates in all the ELM systems considered here, but furfural as aldehyde was quite hard to remove. Also, when HBA was added to the basic simulated hemicellulosic hydrolysate, both of acetic acid and HBA in the feed phase could be selectively removed up to 99% in an ELM system with tributyl phosphate as extractant.

Depolymerization of Kraft Lignin over a Ru-Mg-Al-oxide Catalyst (Ru-Mg-Al-oxide 촉매 상에서 크라프트 리그닌의 저분자화 연구)

  • Kraft lignin is a by-product of the pulp and paper industry, obtained as a black liquor after the extraction of cellulose from wood through the Kraft pulping process. Right now, kraft lignin is utilized as a low-grade boiler fuel to provide heat and power but can be converted into high-calorific biofuels or high-value chemicals once the efficient catalytic depolymerization process is developed. In this work, the multi-functional catalyst of Ru-Mg-Al-oxide, which contains hydrogenation metals, acid, and base sites for the effective depolymerization of kraft lignin are prepared, and its lignin depolymerization efficiency is evaluated. In order to understand the role of different active sites in the lignin depolymerization, the three different catalysts of MgO, Mg-Al-oxide, and Ru-Mg-Al-oxide were synthesized, and their lignin depolymerization activity was compared in terms of the yield and the average molecular weight of bio-oil, as well as the yield of phenolic monomers contained in the bio-oil. Among the catalysts tested, the Ru-Mg-Al-oxide catalyst exhibited the highest yield of bio-oil and phenolic monomers due to the synergy between active sites. Furthermore, in order to maximize the extent of lignin depolymerization over the Ru-Mg-Al-oxide, the effects of reaction conditions (i.e., temperature, time, and catalyst loading amount) on the lignin depolymerization were investigated. Overall, the highest bio-oil yield of 72% and the 3.5 times higher yield of phenolic monomers than that without a catalyst were successfully achieved at 350 ℃ and 10% catalyst loading after 4 h reaction time.

  • Separation of deasphalted oil from atmospheric residue using n-pentane as a solvent was carried out to obtain the valuable heavy hydrocarbon products that can be used lube base oil. After separation experiments, it is shown that the separation yield of oil from atmospheric residue was dependent on n-pentane density and increased in the critical region of n-pentane. Also, the metal content of separated oil was much lowered when compared with that of atmospheric residue. However, there's slight differences in sulfur removal between above two cases.

  • The Polarographic behavior of 1-(2-thiazolylazo)-2-naphthol (TAN) in acetonitrile solution was studied. From the DC polarograms of TAN in acetonitrile solution, the type of reduction current and the effect of proton donor such as water have been investigated. In order to explain the reduction mechanism, the number of the electrons for each reduction step was measured by controlled potential coulometric technique and the electrolysis products were identified by UV-Vis spectroscopy and IR spectroscopy. The results were shown that the reduction of TAN in acetonitrile solution occurred with four-one electron steps. In addition, each reduction step was considerably reversible and the reduction current was diffusion controlled.

  • 높은 광흡수 계수를 갖는 Cu(In,Ga)Se2(CIGS) 화합물 박막 소재는 고효율 태양전지 양산을 위해 가장 전도유망한 재료이나 상대적으로 매장량이 적은 In 및 Ga을 사용한다는 소재적 한계가 있다. Cu2ZnSnSe4(CZTSe) 혹은 Cu2ZnSnS4(CZTS)와 같은 Cu-Zn-Sn-Se계 화합물 반도체는 CIGS 내 희소원소인 In과 Ga이 범용원소인 Zn 및 Sn으로 대체된 소재로써 미래형 저가 태양전지 개발을 위해 활발히 연구되고 있는데, 그 화합물 조합에 따라 0.8eV부터 1.5eV까지의 에너지 밴드갭을 갖는 것으로 알려져 있다. 본 연구에서는 열분해법으로 CZTS 나노 입자를 합성하였다. 용매로 Oleylamine을 사용하였는데, $220^{\circ}C{\sim}340^{\circ}C$의 온도 범위에서 3시간 30분 동안 CZTS 나노입자를 합성하였고, $240^{\circ}C$에서 3시간~5시간까지 합성하였다. 헥산을 이용하여 원심분리기와 초음파세척기로 용매인 Oleylamine을 제거하였고, 진공오븐에서 건조된 CZTS 분말의 FE-SEM(Field Emission Scanning Electron Microscope), XRD(X-Ray Diffraction), EDS(Energy Dispersive Spectroscopy) 분석 등을 통해 합성온도에 따른 구조적, 화학적 조성 변화를 조사하였다.

Isolation and Identification of Bioactive Organosulfur Phytochemicals from Solvent Extract of Broccoli (브로콜리 용매추출물의 Bioactive Organosulfur 화합물의 분리 및 동정)

  • Bioactive organosulfur phytochemicals were isolated from fresh broccoli using methlylene chloride as an extract solvent and identified by GC/MSD analyses. Major organosulfur phytochemicals of broccoli extract were found to be isothiocyanates, which constitute 40.42% of total phytochemicals. The isothiocyanates from broccoli extract were identified to be 3-butenyl, 4-methyl thiobutyl, 4-methylthio-3-butenyl, 5-methylthiopentyl, 2-phenylethyl, 3-methyl sulfinyl propyl, and 4-methylsulfinylbutyl isothiocyanates, of which major isothio cyanates were 3 butenyl isothiocyanate and 4-methylsulfinylbutyl isothiocyanate, constituting about 38.55% of total isothiocyanates present in the solvent extract. Also, nitrites, corresponding to products from enzymatic hydrolysis of glucosinolates were identified as 4-methylthiobutyl, 5-methyl thiopentyl, 2-phenylethyl and 4-methylsulfinylbutyl nitrile. In addition, three sulfides were identified as dimethyl disulfide, dimethyl trisulfide and dimethyl tetrasulfide.

Preparation of Nano-size BaTiO3 Powders Using Glycothermal Method (Glycothermal법을 이용한 나노 사이즈 BaTiO3분말의 제조)

  • Barium Titanate(BaTiO$_3$) nanoparticles were prepared at 22$0^{\circ}C$ through glycothermal process by using barium hydroxide and amorphous titanium hydrous gel as precursor and 1,4-butanediol and distilled water as solvent. It is demonstrated that the size of BaTiO$_3$ particles can be controlled by reaction conditions such as various content of 1,4-butanediol/distilled water volume ratio. This processing method can fabricate BaTiO$_3$ powders, which have a narrow distribution and exhibit good dispersion. The particle size of BaTiO$_3$ powders obtained by glycothermal process were about 50 nm to 200 nm on the condition that reaction temperature was 22$0^{\circ}C$ and holding time was 24 h.

  • Nanomaterial $TiO_2-SiO_2$ was synthesized by hydrolysis and condensation process using 2-propanol(2-PrOH) and was characterized by FT-IR, DSC, XRD and FE-SEM. FT-IR spectra were measured to investigate Ti-0-Si absorption peak. DSC thermal analysis results appllied to Ozawa equation were used to calculate to activation energy of crystallization. It was found that the changes of X-ray diffraction patterns and FWHM obtained XRD measurement depended on calcination temperature. In FE-SEM analysis, particle size changed by quantity changes of Ti-alkokide.
