  • The rate constants for the solvolysis of anthraquinone-2-carbonyl chloride in binary solvent mixtures, methanol-acetonitrile, methanol-acetone, ethanol-acetonitrile and ethanol-acetone, have been studied by means of conductometry. Maximum rate phenomena were observed at the methanol mole fraction, $X_{MeOH}$ = 0.73~0.81 for methanol-acetonitrile and at $X_{MeOH}$ = 0.83 for methanol-acetone mixtures. The Kivinen and Grunwald-Winstein plots indicated that the reaction proceeds via $S_N2$ type mechanism. Application of Taft's solvatochromic correlation on the solvolysis rate revealed that both $\alpha$ and ${\pi}^*$ are important for reactions in methanol-acetonitrile and methanol-acetone mixtures, while only ${\alpha}$ influences significantly on the rate in ethanol-acetonitrile and ethanol-acetone mixtures.

  • Kinetic studies on the solvolysis of para-methyl benzyl nitrate and benzyl nitrate were carried out in aqueous methanol, ethanol, acetonitrile, acetone, tetrahydrofuran and dioxane mixtures at 60$^{\circ}$C. The rates were faster in protic solvent mixtures than in aprotic solvent mixtures. This was considered in the light of transition state stabilization by hydrogen bonding solvation of protic solvent mixtures. Grunwald-Winstein equation, extended Grunwald-Winstein equation and correlation between E$_T$(30) and rate constant were applied in order to discuss the transition state variations caused by changing benzyl substituents and solvents. The results showed that strong electrophilic assistance of solvent is operative in the the water-rich solvents.

  • The extraction of taxol and baccatin III from the ground needles of Taxus cuspidata were carried out by using supercritical carbon dioxide with and without cosolvents such as ethylacetate and methanol at 300 bar and 313K. Taxol is a promising anticancer agent and baccatin III is a precursor of semisysthesis of taxol. The taxol and baccatin III contents in the extracts were determined by a HPLC. The highest yields of taxol and baccatin III could be obtained by the supercritical extraction with 3wt% methanol and ethylacetate, respectively, as cosolvents. It was also found that the selectivity of taxol and baccatin III were 0.117 and 1.245wt%, respectively, with 0.7wt% ethylacetate, which demonstrated that the selectivity of taxol and baccatin III were increased about 1.8 and 19 times than those of conventional organic solvent extraction.

  • This study was carried out to examine some factors affecting the supercritical carbon dioxide extraction of genistein from soybean. The factors investigated in this study were pressure, temperature, flow rate of carbon dioxide, and concentration of modifier. It was foumd out that genistein is not extracted in the absence of modifier. Ethanol was found to be more effective modifier than methanol. 70% of genistein was extracted at 35$^{\circ}C$, 300bar and ethanol 15% (w/v) as compared with the performances of organic solvent extraction.

  • Effects of modifier and soaking on extraction of triterpenoid saponin (glycyrrhizin) from liquorice were examined using supercritical $CO_2(SC-CO_2)$ at 50 MPa, $60^{\circ}C$, and flow rate of 3 mL/min, and glycyrrhizin content was analyzed by HPLC. Additon of undiluted methanol, ethanol or isopropanol as modifier to $SC-CO_2$ had little influence on extraction yield of glycyrrhizin. Soaking process using water increased the extraction yield as the sample to solvent ratio was increased. Addition of 70% methanol, ethanol or isopropanol to $SC-CO_2$ significantly increased the extraction yields, with 70% methanol resulting in the highest yield. When water at 90% (w/w) of sample weight was used for soaking, the extraction yield and rate increased, 70% ethanol-modified $SC-CO_2$ was almost equal to that obtained using 70% methanol.

  • Coprinus cinereus peroxidase (CiP) was often used as a catalyst for oxidative polymerization of a variety of phenol derivatives to produce a new class of polyphenols. Economical point of view, to know the mechanism of enzyme deactivation is significantly important because cost of enzyme is critically high. Hydrogen peroxide being used as oxidizing agent induced deactivation of peroxidase by destruction of heme structure. In the presence of hydrogen peroxide the stability of peroxidase was unexpectedly improved by adding organic solvent. Especially 2-propanol significantly improved enzyme stability among tested solvents. Radical scavenging by organic solvents may play a major role in protecting peroxidase from the oxidation of oxidizing radicals.

  • The effect of organic solvents on the stability and catalytic activity of the microbial lipase from Rhizopus arrhizus for interesterification of fats and oils has been examined. The reaction system used was nonaqueous solvent system (two phase system). The solvents examined were 5 hydrocarbons (n-hexane, n-heptane, n-octane, isooctane, and cyclohexane) and 3 ethers (diethylether, diisopropylether, di-n-butylether). The results revealed that diisopropylether and isooctane are superior to the other solvents examined for interesterification of fats and oils in two phase systems.

  • Hydrogen bonding, dielectric effects and steric effects are all factors which determine the carbonyl stretching frequency, νC=O of ketones in solution. The νC=O frequency of ketones is affected by change in concentration of ketones in various solvents. The νC=O frequency shifts to lower frequency in nonpolar solvents and shifts to higher frequency in polar solvents with the increasing volume% of ketones. In acetonitrle, the νC=O frequency shifts to higher frequency as the volume% of ketones is increased except dimethyl ketone. The νC=O frequency shifts to lower frequency as the solvent system becomes increasingly polar or with the increasing extent of intermolecular hydrogen bonding, as in the case of increasing volume% chloroform in $CHCl_3$/$CCl_4$ solvent system.

  • An extractive fermentation process was developed to produce citric acid from g1ucose. Citric acid is a strong inhibitor to this fermentation. A mixture of tertiary amine and oleyl alcohol was used to selectively extract citric acid from the fermentation broth, hence enhancing the productivity by over 200%. Although the toxicity of the solvent was significant in the range of higher than 30% of amine, immobilization in polyurethane foam was useful to protect the cells from the toxicity of the solvent.

  • To elucidate whether or not solvent-tolerant bacteria use anti-oxidative defense systems to defend themselves against toxic solvents, oxidative enzyme activity and total anti-oxidative capacity (T-AOC) were investigated in two tolerant strains of Pseudomonas sp. under toluene stress. The superoxide dismutase (SOD) activities of solvent tolerant BCNU 106 exhibited relatively increased levels at a toluene concentration of 100 mg/l, where those of solvent tolerant BCNU 171 increased at 200 mg/l. A greater than three-fold increase in catalase (CAT) levels was observed at concentrations of 200 and 300 mg/l in BCNU 106, and a two-fold increase was monitored at the same concentrations in BCNU 171. High glutathione S-transferase (GST) levels were also observed in the solvent tolerant bacteria. Higher levels of T-AOC was expressed in the solvent tolerant strains than in the ordinary non-tolerant KACC 10266. The highest plateau of SOD in BCNU 171 was observed at 1 hr of toluene exposure. CAT levels plateaued at 1 hr and 14 hr in BCNU 106 and reached the highest plateau at 3 hr in BCNU 171. The highest peak of T-AOC occurred at 9 hr in BCNU 106, and two high peaks occurred in BCNU 171, at 1 hr and at 9 hr of toluene exposure. The solvent-tolerant bacteria showed active antioxidant responses and could survive under harsh environments, including the presence of solvents, through means of antioxidant defense systems.
