온청음(溫淸飮)과 삼황세제가미방(三黃洗劑加味方) 병용이 NC/Nga 아토피 생쥐에 미치는 영향 (Effects of Onchung-eum Administration along with Samhwangseze-gamibang on Treatment Atopic Dermatitis Development in NC/Nga Mice)

  • The purpose of this study is to examine closely effect that Onchung-eum(OC) and Samhwangseze-gamibang(SG) used to atopic dermatitis disease patient get in atopy eruption control experimentally. Atopic dermatitis(AD) of molecular mechanism underlying it's effectiveness is unknown. We analyzed the expression the clinical severities in 13 and 16 weeks old NC/Nga mice, and the spleen weight of OC with SG treated NC/Nga mice, and mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of OC with SG treated NC/Nga mice, and IL-1${\beta}$, TNF-${\alpha}$, IL-6 express of gene, and Histological observation of the ear and skin tissues, and than IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of OC with SG treated NC/Nga mouse group compared to the untreated control mouse group. Also, We examined cell toxicity that of OC is safety the strength of 10, 50, 100ppm and inflammatory RAW 264.7 in the serum of OC. Thus in these present study diverse immune responses in terms of chemical mediators related to AD were investigated using an atopic mouse model NC/Nga after OC along with 5G. At the result that OC along with SG treat is can effective use for the treatment of atopic dermatitis(AD).

온청음 물 추출물의 세포독성, 피부재생, 주름개선, 미백 및 보습 효과 (In Vitro Cytotoxicity, Skin Regeneration, Anti-wrinkle, Whitening and In Vivo Skin Moisturizing Effects of Oncheongeum)

  • 목 적: 본 연구에서는 한의학에서 다양한 피부질환과 대사질환에 빈번히 사용되고 있는 온청음 물 추출 동결건조물(수율=13.82%)의 피부 노화 개선 효과 평가의 일환으로 세포독성, 피부재생, 주름개선, 미백 및 보습 효과를 각각 평가하였다.방 법: 본 연구에서는 human normal fibroblast(CRL-2076) 및 B16/F10 murine melanoma(CRL-6475) 세포에 대한 온청음의 세포독성을 MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium Bromide) 방법으로 평가하였으며, 피부 재생 및 주름 개선 효과를 transforming growth factor(TGF)-β1와 비교한 fibroblast의 collagen type I 합성능, phosphoramidon disodium salt(PP)와 비교한 elastase 활성 억제, oleanolic acid(OA)와 비교한 hyaluronidase, collagenase 및 matrix metalloproteinase (MMP)-1 활성 억제를 통해 각각 평가하였고, 미백효과를 B16/F10 murine melanoma cells의 melanin 생성 억제 정도 및 tyrosinase 활성 억제를 통해 arbutin과 비교 평가하였으며, 모든 실험은 OCE의 농도별로 군을 나누어 농도에 따른 효과의 변화를 함께 분석하였다. 보습효과는 흰쥐의 피부 수분함량 변화를 통해 평가하였다.결 과: 본 실험의 결과, 온청음은 human normal fibroblast 및 B16/F10 murine melanoma 세포에 대해 의미 있는 세포독성을 나타내지 않았으며, fibroblast의 collagen type I 합성을 증가시켰고, 세포외 기질의 파괴에 관여한다고 알려진 hyaluronidase, elastase, collagenase 및 MMP-1 활성을 억제하였으며, 피부의 색을 결정하는 melanin 의 생성에 관여하는 tyrosinase의 활성 및 B16/F10 murine melanoma cells의 melanin 생성을 억제하는 것으로 관찰되었다. 이 반응의 효과들은 모두 농도에 비례하여 증가하였고, 이와 함께 정상 매체 대조군에 비해 흰쥐의 피부 수분 함량이 세 용량의 온청음 경구 투여군 모두에서 투여용량 의존적으로 의미 있는 증가를 보였다.결 론: 이상의 결과에서, 온청음은 세포 독성 없이 비교적 우수한 피부 재생, 주름개선, 미백 및 보습 효과를 나타내는 것으로 관찰되어, 차후 피부 노화 억제 개선제 또는 기능성 화장품의 주요 소재로서 그 가치가 매우 높을 것으로 판단되나, 금후 개별 구성 약재 각각에 대한 효능 및 생리활성을 나타내는 화학성분의 검색과 더불어 다양한 방면으로 기전적인 연구와 피부 보호 효과에 대한 in vivo 평가를 체계적으로 수행해야 할 것으로 판단된다.

약진(藥疹)으로 심한 소양감을 보인 환자에 대한 온청음(溫淸飮) 투여 1례 (A case report of Onchungeum(溫淸飮) for drug eruption with severe itching)

  • Drug eruption is a relatively commin dermatological disorder and may cause significant problems. The treatment is to stop using a drug that may cuase a drug eruption. In some cases, steroids and antihistamines can be used supportively. According to some reports, Onchungeum(溫淸飮) has steroid-like effects and antihistaminic effects. We administered Onchungeum(溫淸飮) a patient with severe itching because of a drug eruption. After administration of Onchungeum(溫淸飮), itching decreased to much degrees. It can be suggested that Onchungeum(溫淸飮) can be used to treat symptoms of allergic skin disease like a drug eruption.

소아의 온청음(溫淸飮) 신제형 선호도 조사 (Survey on the Preference for the New Dosage Form of Onchung-eum in Children)

  • Objectives : Medication is important in treatment for children, but prescribing traditional herbal medicine for them is very difficult. The aim of this study was to evaluate the preference for new and traditional dosage form of Onchung-eum. Methods : A total of 24 children who visited Oriental Medical Center of Daegu Hanny University during one month since March 2008 were included in this study. They compared new dosage form of Onchung-eum with traditional thing and evaluated items such as taste, perfume, color, sensation of chewing and texture. Results : As a whole they prefered new dosage form more than traditional thing in a sensory test. And 13(54%) children choose new dosage form as the better one. Conclusions : Considering the above results, the new dosage form might be efficacious to be taken by children. Further studies in other methods and new dosage forms are needed to make prescribing it for children easily.

아토피성 피부염에 활용되는 온청음(溫淸飮)에 대한 한중일의 논문 비교 (Comparative Analysis of Korean, Chinese and Japanese Articles about Oncheongeum Used for Curing Atopic Dermatitis)

  • Background and Purpose : Oncheongeum(溫淸飮) composed of Samultang(四物湯) and Hwangryunhaedoktang(黃連解毒湯) was mainly used for healing metrorrhagia in the Qing dynasty of China. At present, Oncheongeum is used for treating a broad spectrum of diseases such as skin disease, stomatitis, behcets disease, diabetes mellitus and, especially in Japan, atopic dermatitis. To our knowledge, however, neither the precise constituents and their effects of Oncheongeum nor the criteria for the prescription of Oncheongeum were defined. To address this issue, we searched and performed analysis of Korean, Chinese and Japanese articles reporting the clinical and experimental studies of Oncheongeum. Methods : We searched articles in the national assembly library of Korea by using keyword 'Oncheongeum' korean. Similarly, we did chinese articles in the CNKI and japanese ones in the CiNii, respectively. Results : We found 13 korean articles about Oncheongeum in the national assembly library, 34 chinese articles in the CNKI, and 23 japanese in the CiNii. The papers were divided into clinical and experimental articles. The clinical articles were mainly published from China and Japan, and their subjects were predominantly on skin diseases. Conclusions : There were a lot of case reports about Oncheongeum used in the clinical studies. In order to better understand the effects of Oncheongeum, systematic review of the studies seems essential. The efficacy of Oncheongeum reported in the clinical studies should be supported by experimental data. Not much were clinical reports in Korea, although we are expecting more to come. It is possible to not only analyse but compare Oncheongeum with other prescriptions used for atopic dermatitis. More comprehensive and comparative analysis of three countries' prescriptions might provide a way of how to standardize prescriptions, which leads oriental medicine to an evidence based medicine.

온청음(溫淸飮)이 인체 간암세포의 세포주기 G1 Arrest에 미치는 영향 (G1 Arrest of the Cell Cycle by Onchungeum in Human Hepatocarcinoma Cells)

  • Onchungeum, a herbal formula, which has been used for treatment of anemia due to bleeding, discharging blood and skin disease. In the present study, it was examined the effects of extract of Onchungeum (OCE) on the growth of human hepatocarcinoma cell lines Hep3B (p53 null type) and HepG2 (p53 wild type) in order to investigate the anti-proliferative mechanism by OCE. Treatment of Hep3B and HepG2 cells to OCE resulted in the growth inhibition in a dose-dependent manner, however Hep3B cell line exhibited a relatively strong anti-proliferative activity to OEC. Flow cytometric analysis revealed that OCE treatment in Hep3B cells caused G1 phase arrest of the cell cycle, which was associated with various morphological changes in a dose-dependent fashion. RT-PCR and immunoblotting data revealed that treatment of OCE caused the down-regulation of cyclin D1 expression, however the levels of cyclin E expression were not changed by OCE. The G1 arrest of the cell cycle was also associated with the induction of Cdk inhibitor p27 by OCE. Because the p53 gene is null in Hep3B cells, it is most likely that the induction of p21 is mediated through a p53-independent pathway. Moreover, p27 detected in anti-Cdk4 and anti-Cdk2 immunoprecipitates from the OCE-treated cells, suggesting that OCE-induced p27 protein blocks Cdk kinase activities by directing binding to the cyclin/Cdk complexes. Furthermore, OCE treatment potently suppresses the phosphorylation of retinoblastoma proteins and the levels of the transcription factor E2F-1 expression. Taken together, these results indicated that the growth inhibitory effect of OCE in Hep3B hepatoma cells was associated with the induction of G1 arrest of the cell cycle through regulation of several major growth regulatory gene products.

온청음(溫淸飮)의 조성 용량변화가 Hep3B 세포의 G1 arrest 기전에 미치는 영향 (Change of Ratio of Onchungeum Composition Induces Different G1 Arrest Mechanisms in Hep3B Cells)

  • Onchungeum(OCE), a herbal formula, has been used for treatment of anemia, discharging blood and skin diseases. In the previous study, we investigated the anti-cancer effect of OCE by G1 arrest of the cell cycle in human hepatocarcinoma cells, Hep3B cells. In this study, it was examined that the difference of anti-proliferative mechanisms by change in the ratio of OCE composition (OCE I) in Hep3B cells. Treatment of OCE I exhibited a relatively strong anti-proliferative activity and caused various morphological changes such as membrane shrinkage and cell floating. In addition, OCE-I arrests the cell cycle at G1 phase, which was associated with the down-regulation of cyclin D1 and Cdk6 expressions. The G1 arrest was also associated with the induction of Cdk inhibitors p27 and p21. Moreover, both p21 and p27 were detected by immunoprecipitation with anti-Cdk4 and anti-Cdk2 antibodies in OCE I-treated cells but in case of OCE, p21 did not make any complexes with Cdk4 and Cdk2. These results suggest that the change in the ratio of OCE composition might induce different mechanisms in anti-cancer efficacy of OCE, which may confer characteristic principles in oriental medical formula.

흰쥐의 전층피부손상에서 온청음(溫淸飮)이 신생혈관형성에 미치는 영향 (Angiogenetic Effect of Onchung-Eum on Full-thickness Skin Wound in Rats)

  • The wound healing process can be categorized as follows : inflammation, fibroplasia, neovascularization, collagen deposition, epithelialization, and wound contraction. During the healing process, various growth factors are secreted to accelerate wound healing. Previous studies have demonstrated that endogenous growth factors, such as vascular endothelial growth factor(VEGF) are the important regulatory polypeptides for coordinating the healing process. They are released from macrophages, fibroblasts, and keratinocytes at the site of injury and participate in the regulation of reepithelization, granulation tissue formation, collagen synthesis and neovascularization. Onchung-Um has been used clinically to treat various skin diseases. In addition, Onchung-Um has been also used for congestive inflammations. In the present study, we evaluated the effects of Onchung-Um on wound healing process and wound size reduction in rats. Full-thickness skin wounds ($15mm\;{\times}\;15mm$) were created on the back of rats. Rats were then divided into 2 groups : The Onchung-Um treated group that was orally administered with a dose of 193.9mg/100g of Onchung-Um extract per day for 15 days and Control group without Onchung-Um administration. Moreover, the histological changes and VEGF immunoexpressions of two groups were estimated. In results, wound closures were significantly accelerated by oral administration of Onchung-Um extract. Furthermore, in Onchung-Um treated group, there were significant increases in fibroblast migration, epithelialization compared with the Control group. VEGF expressions were also increased in Onchung-Um treated group. This study has therefore demonstrated the Onchung-Um can significantly improve the quality of wound healing and scar formation and the oral administration of Onchung-Um extract may increase early tissue angiogenesis in the incisional wound of an experimental animal model.

황연해독탕(黃連解毒湯)과 온청음(溫淸飮)이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響) (A Experimental Study on the Effect of Hwangryeonheadock-Tang and Onchung-Eum on Hyperlipidemia & Hypertension)

  • The aim of the present study was to investigate the effect of Hwangryeonheadock-Tang and Onchung-Eum on essential hypertension and hyperlipidemia. Rats were orally administered for 30days with Hwangryeonheadock-Tang and Onchung-Eum and the blood was withdrawn at 10, 20 and 30days after an oral administration. The heart rate, tail blood pressure, plasma renin activity, plasma level of aldosterone. catecholamine, sodium and angiotensin II were measured after an oral administration of Hwangryeonheadock-Tang and Onchung-Eum in spontaneously hypertensive rat(SHR). In addition, serum levels of total cholesterol, triglyceride, HDL-cholesterol. LDL-cholesterol and total lipid were measured cholesterol-fed rats. The results were summarized as follows ; 1. A significant decrease of tail blood pressure was shown at 10, 20 and 30days after Hwangryeonheadock-Tang and Onchung-Eum treatment in SHRs. compared with saline. 2. Heart rate was significantly decreased at 30days in SHRs after Hwangryeonheadock-Tang treatment and at 20, 30days after Onchung-Eum treatment in SHRs. compared with the effects of saline group. 3, A significant decrease of plasma aldosterone level was elicited at 10, 20days after Hwangryeonheadock-Tang treatment in SHRs, compared with the effects of saline group, 4. Plasma renin activity was significantly decreased at 10days after Onchung-Eum treatment compared with the effects of saline group in SHRs. 5. Plasma norepinephrine level was significantly decreased at 20 and 30clays after Onchung-Eum treatment in SHRs, compared with the effects of saline group. 6. A significant decrease of plasma epinephrine level was induced at 30days after Hwangryeonheadock-Tang treatment and at 10, 20 and 30days after Onchung-Eum treatment, compared with the effects of saline group in SHRs. 7. Plasma sodium level was. significantly decreased at 20days after Hwangryeonheadock-Tang and Onchung-Eum treatment, compared with the effects of saline group in SHRs. 8. Plasma angiotensin II level was significantly decreased at 30days after Onchung-Eum treatment, compared with the effects of saline group in SHRs. 9. A significant decrease of body weight was observed at 20 and 30days after Hwangryeonheadock-Tang treatment and at 10, 20 and 30days after Onchung-Eum treatment. compared with the effects of saline group in hyperlipidemia rats. 10. Hwangryeonheadock-Tang and Onchung-Eum showed a significantly decreasing effect at 30days on serum total cholesterol level in hyperlipidemia rats, compared with the saline treatment. 11. Hwangryeonheadock-Tang and Onchung-Eum saw 20 and 30days respectively on serum triglyceride level in the saline treatment. 12. Hwangryeonheadock-Tang and Onchung-Eum decreased on serum HDL-cholesterol level significantly, compared with the saline treatment in hyperlipidemia rats. 13. A significant decrease of serum LDL-cholesterol was observed at 10 and 30days after Hwangryeonheadock-Tang treatment and at 30days after Onchung-Eum treatment, compared with the effects of saline group in hyperlipidemia rats. 14. Hwangryeonheadock-Tang had a significantly decreasing effect at 10, 20 and 30days on serum total lipid level, compared with the saline treatment in hyperlipidemia rats. 15. Hwangryeonheadock-Tang elicited a significantly decreasing effect on weight of kidneys, spleen and testes respectively and Onchung-Eum induced on weight of liver and spleen respectively in hyperlipidemia rats, compared with saline treatment. These Findings suggest a possible anti-hypertensive and anti-hyperlipidemic effect of Hwangryeonheadock-Tang and Onchung-Eum.

온청음 가미방 3개월간 장기복용의 유효성과 안전성 보고 (Case Report of Efficacy and Safety of Long Term Medication of Oncheongeum-gamibang)

  • This study is designed as pilot study to report the efficacy and safety of long term medication of Oncheongeum-gamibang on atopic dermatitis. The patient of atopic dermatitis was asked to take Oncheongeum-gamibang for 3 months. Every end of month, he had gotten hematological test and SCORAD Index examination. After the clinical trial was done, he had additional examination to see the changes and duration of medicinal effect. During the period of taking Onchung-um, estimation of symptoms by SCORAD Index examination showed little bit of improvement while taking medicine and after one month, the improvement was maintained. There were no significant changes in hematologic test and liver function test during and at the end of the clinical trial. We were able to find out that it is effective and safe to take long term medication of Oncheongeum-gamibang for an individual. However, due to deficiency of the cases and immunological values, there will be additional study to make up for better result.
