Major Characteristics Related on Eating Quality and Classification of Inbred Lines of Waxy Corn (찰옥수수 자식계통 식미관련 특성 및 계통 분류)

  • Objectives of this study were to select inbred lines which have a good eating quality and desirable segregates during inbreeding of waxy corn. The 64 inbred lines showed a large variance in their kernel shape and weight. 100-kernel weight, pericarp thickness, kernel length, kernel width, and kernel thickness ranged $11.7\~37.3g,\;11\~77{\mu}m,\;5.8\~9.6mm,\;6.5\~10.0mm$, and $4.1\~6.8mm$, respectively. The physicochemical analysis of 64 waxy corn inbred lines showed crude protein, crude fat, free sugar, and amylopectin content ranging $8.7\~15.8\%,\;2.3\~5.8\%,\;1.1\~11.0\%,\;and\;78.5\~93.8\%$, respectively. The texture property analysis of 64 inbred lines by texture analyzer showed a big difference. Gumminess, hardness, and chewiness of 64 inbred lines ranged $91\~383,\;181\~394,\;and\;73\~370$, respectively. The principal component analysis for 14 characteristics related to kernel quality showed that $73.1\%$ of the total variation could be attributed to the first five principal components. Biological meaning of the principal component was explained clearly by the correlation coefficient between principal components and characters. The first principal component appeared to correspond to small kernel and bad eating quality, The second principal component appeared to correspond to large kernel and good eating quality. The 64 inbred lines were classified into 8 groups by the cluster analysis using the first and second principal component. Among the groups, group VII and VIII included inbred lines with good eating quality that had thin pericarp thickness, low protein content, large kernel, and soft tenderness.

Identification of the Transmissive Fractures in the Vicinity of waterway Tunnel (도수로터널 주변 지역의 지하수 유동성 단열 규명)

  • A field technique for assessing the transmissive fractures in an aquifer was applied to a fractured rock formation in Youngchun area Korea. Geological mapping and detailed acoustic borehole teleview(BHTV) logging were performed to obtain information about the fractures. The study area consists predominantly of two types of fractures. The fracture sets of low angle partings such as bedding and sheeting plains have strike N70-80$^{\circ}$W, 25$^{\circ}$-30$^{\circ}$SW and N3S$^{\circ}$W, 12$^{\circ}$NE, respectively. In areas of high fractures, on the other hand, the major fracture sets show strike N80$^{\circ}$W and dip 70$^{\circ}$-85$^{\circ}$SW, N10$^{\circ}$E.85$^{\circ}$SE in sedimentry rocks, N40-50$^{\circ}$E.85$^{\circ}$SE/85$^{\circ}$NE, N70$^{\circ}$E.80$^{\circ}$SE, and N7$^{\circ}$-75$^{\circ}$W.80$^{\circ}$SW in granites and volcanic rocks. Injection tests have been performed to identify discrete production zones and quantify the vertical distribution of hydraulic conductivity. The calculated hydraulic conductivities range from 3.363E-10 to 2.731E-6, showing that the difference between maximum and minimum value is four order of magnitude. Dominant section in hydraulic conductivity is extensively fractured. Geophysical logging was carried out to clarify characterization of the distribution of fracture zones. Transmissive fractures were evaluated through the comparison of the results obtained by each method. The temperature logs appeared to be a good indicator that can distinguish a high transmissive fractures from a common fractures in hydraulic conductivity. In numerous cases, evidence of fluid movement was amplified in the temperature gradient log. The fracture sets of N70-80$^{\circ}$W.60-85$^{\circ}$NE/SW N75-80$^{\circ}$W.25-30$^{\circ}$SW, N50-64$^{\circ}$W.60-85$^{\circ}$NE, N35-45$^{\circ}$E.65-75$^{\circ}$SE, and N65-72$^{\circ}$E.80$^{\circ}$SE/60$^{\circ}$NW were idenfied as a distinct transmissive fractures through the results of each tests.

Characteristics and breeding of a new cultivar Pleurotus eryngii, Song-A (큰느타리버섯 신품종 '송아'의 육성 및 그 특성)

  • To develop a new cultivar of King oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strains derived from ASI 2824(Keunneutari No.2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-51($G09-21-10{\times}2844-11$) was shown the best cultural characteristics, selected to be a new cultivar and named as 'Song-A'. The 'Song-A' was formed incompatibility line distinctly in the confrontation growth of parental strains Keunneutari No.2, Aeryni 3 and ASI 2844. The optimum temperature for mycelial growth, fruiting body development and pH arrange were $25{\sim}30^{\circ}C$, $14{\sim}16^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about $94.7{\pm}29.5$ g which is almost 106% quantity compared to that of other cultuvar Keunneutari No.2. And also the stip is thick and long but the number of available stipe is few. Analysis of the genetic characteristics of the new cultivar 'Song-A' showed a different DNA profile as that of the control strains, Keunneutari No.2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primers URP4 and 7 were used. This new cultivar 'Song-A' of Pleurotus eryngii is characterized by a small number of primordia formation and the stip is thick and long. Therefore, we expect that this new strain will save of labor and cost by without culling work.

Optimal Desiccation Condition and Moisture Content of Dehisced Seeds of Ginseng (Panax ginseng C.A. Meyer) for Cryopreservation (인삼 개갑종자의 초저온동결보존을 위한 최적 건조조건과 수분함량)

  • This study was conducted to establish the efficient protocols of the germination and cryopreservation of dehisced Korean ginseng seeds for long-term germ­plasm conservation. $GA_3$ and BA treated on seed for 24 hr facilitated germination at 5 and $10^{\circ}C$. Germination percentage of desiccated seeds was decreased under moisture content (MC) of below $7.2\%$. Dehisced ginseng seeds were dried under airflow of laminar floor cabinet and seed drying room. The high levels (more than $90\%$) of germination after cryogenic exposure were obtained after drying under vertical airflow of laminar floor for 12-30 hours (MC $10.6{\~}7.2\%$). Decrease in germination percentage of ginseng seeds due to desiccation damage and freezing injury was observed at MC of below $7.2\%$ and of above $12.1\%$, respectively. Therefore, MC of ginseng seeds need to be controlled with a range of $8{\~}10\%$ to avoid damages from both desiccation and freezing.

  • The volatile and semi-volatile compounds of 5 accessions of domestic scented and 25 accessions of introduced scented were extracted by solid phase microextraction (SPME) and analyzed by gas chromatographymass spectrum (GC-MS). A total of 156 volatile and semivolatile compounds were identified from 30 accessions of aromatic rice, including 32 alcohols, 25 acids, 25 ketones, 21 hydrocarbon, 18 esters, 16 aldehydes, 4 ethers, 5 amines, 2 phenols, 2 bases, and 8 miscellaneous compounds. By UPGMA/Neighbor-join tree analysis, the thirty accessions of aromatic rice could be classified into seven groups according to the major odor or aroma compounds. Group I included indica type of Basmati varieties. Group II and Group IV included japonica type introduced scented. Group III consisted only Hyangmibyeo1ho in domestic scented. Group V and Group VII included indica type of Basmati and non-Basmati varities. Group VI included four of domestic scented of seven accessions excepted Basmati6129, Basmati 6311, and Seratus Malam.

Ore Minerals, Fluid Inclusions, and Isotopic(S.C.O) Compositions in the Diatreme-Hosted Nokdong As-Zn Deposit, Southeastern Korea: The Character and Evolution of the Hydrothermal Fluids (다이아튜림 내에 부존한 녹동 비소-아연광상의 광석광물, 유체포유물, 유황-탄소-산소 동위원소 : 광화용액의 특성과 진화)

  • The Weolseong diatreme was temporally and spatially related to the intrusion of the Gadaeri granite, and was -mineralized by meteoric aqueous fluids. In the Nokdong As-Zn deposit, pyrite, aresenopyrite and sphalerite are the most abundant sulfide minerals. They are associated with minor amount of magnetite, pyrrhotite, chalcopyrite and cassiterite, and trace amounts of Pb-Sb-Bi-Ag sulphosalts. The AsZn ore probably occurred at about $350^{\circ}C$ according to fluid inclusion and compositional data estimated from the arsenic content of arsenopyrite and iron content of sphalerite intergrown with pyrrhotite + chalcopyrite + cubanite. Heating studies of fluid inclusions in quartz indicate a temperature range between 180 and $360^{\circ}C$, and freezing data indicate a salinity range from 0.8 to 4.1 eq.wt % NaCl. The coexisting assemblage pyrite + pyrrhotite + arsenopyrite suggests that $H_2S$ was the dominate reduced sulfur species, and defines fluid parameter thus: $10^{-34.5}$ < ${\alpha}_{S_2}$ < $10^{-33}$, $10^{-11}$ < $f_{S_2}$ < $10^{-8}$, -2.4 < ${\alpha}_{S_2}$ < -1.6 atm and pH= 5.2 (sericte stable) at $300^{\circ}C$. The sulfur isotope values ranged from 1.8 to 5.5% and indicate that the sulfur in the sulfides is of magmatic in origin. The carbon isotope values range from -7.8 to -11.6%, and the oxygen isotope values from the carbonates in mineralized wall rock range from 2 to 11.4%. The oxygen isotope compositions of water coexisting with calcite require an input of meteoric water. The geochemical data indicate that the ore-forming fluid probably was generated by a variety of mechanisms, including deep circulation of meteoric water driven by magmatic heat, with possible input of magniatic water and ore component.

Microbiological Quality and Safety Assessment of Commercial Ready-to-Eat Side Dishes Sold in Gyeonggi-do

  • We aimed to analyze the microbiological quality of the ready-to-eat (RTE) side dishes collected from traditional markets, supermarkets, and cafeterias in Gyeonggi-do in 2019. A total of 108 samples were analyzed for total aerobic bacterial counts, coliforms and foodborne pathogens depending on place of purchase and cooking methods. Results show that Bacillus cereus was detected in 14 (12.9%) out of 108 samples of side dishes, while no other foodborne pathogens were detected. The mean detected level (range) of total aerobic bacteria depending on place of purchase was 5.8 log CFU/g (3.0 to 8.2 log CFU/g) for traditional markets, 4.3 log CFU/g (2.4 to 7.8 log CFU/g) for supermarkets, and 3.80 log CFU/g (0.0 to 6.8 log CFU/g) for cafeterias, indicating that there was a significant (P<0.05) difference in total aerobic bacterial counts among places of purchase. Among the samples, the highest counts of total aerobic bacteria and coliforms were detected in saengchae (raw vegetables), followed by namul (seasoned herbs, vegetables), bokkeum (stir-fried foods), and jorim (foods cooked in soy sauce). The growth of total aerobic bacteria in seasoned soybean sprouts was inhibited when the sprouts were stored at 4℃ up to 24 h, whereas bacteria rapidly grew at 20 and 35℃ after 3 and 6 h, respectively. These results reveal that storage temperature might play a significant role for the microbiological quality of seasoned soybean sprouts when they are sold in markets. Thus, this study suggests that RTE side dishes should be stored at refrigerated temperature when being sold at markets as well as after purchasing to improve their microbiological quality.

Localization and characteristics of lactate and palate dehydrogenase in the sparganum and adult worm of Spirometra erinacei (만손열두조충에서 젖산탈수소효소와 말산탈수소효소의 분포와 특징)

  • This study investigated the enzyme histochemical localization and characteristics of lactate (LDH) and malate dehydrogenase (MDH) related with the oxidation-reduction metabolism in the sparganum and adult of 5. erinacei. By enzyme histochemical assay, activity of LDH was strong in the tegument and subtegumental muscle layers of the adult and sparganum. Activity of MDH was strong in the tegument of the sparganum and subtegumental muscle layers of the adult. However it was weak in the tegument of the adult. By electrophoresis, 45 kDa band was major and common in LDH of adults and spargana. The 150 kDa molecule was the major and common band in MDH of adults and r -spargana (from experimentally infected rats) . By isoelectrofocusing, isoelectric points (Pl) or 4 MDH isogyme from adult worm were 6.0.6.5, 6.7 and 7.1, respectively. Pl 6.0 was the major band. The active range of pH for MDH was about pH 6-8 and the optimum pH was pH 7 The effective temperature on the MDH was about $30^{\circ}C$$50^{\circ}C$ and the optimum temperature was about 40℃ in spargana md adult worm. In the stability against heat, when MDH was heated at 85℃ for 10 seconds, the activity was denatured perfectly. Maximum activity or MDH was 19.4 unit in the s-sparganum (from snakes), 24.5 unit in the r-sparganum (from rats) and 108.0 unit in the adult worm. The maximum activity was higher in adults than in spargana. The present result showed us that the nutrients absorbed through the tegument were transferred into inner tissues and were utilized as the source of metabolism. According to the habitat of the parasite, the isozymes of LDH and MDH are activated differently, and by this different activation the sparganum and adult can adapt themselves to parasitic circumstances.

  • The formation and dissociation rates of $Zn^{2+}$ Complexes with l,4,7,10-tetraaza-13,16-diox-acyclooctadecane-N,N',N",N'"-tetraacetic acid (1), 1,4,7,10-tetraaza-13,16- dioxacyclooctadecane-N,N',N",N'"-tetramethylacetic acid (2), and 1,4,7,10-tetraaza-13,16- dioxacyclooctadecane-N,N',N",N'"-tetrapropionic acid(3) have been measured by stopped-flow and conventional spectrophotometry. Observations were made at 25.0$\pm$0.1 $^{\circ}C$ and at an ionic strength of 0.10 M NaClO$_4$. The formation reactions of $Zn^{2+}$ ion with 1 and 2 took place by the rapid formation of an intermediate complex (ZnH$_3L^+$) in which the $Zn^{2+}$ ion is incompletely coor-dinated. This might then lead to be a final product in the rate-determining step.ln the pH range 4.76-5.76, the diprotonated (H2L2-) form is the kinetically active species despite of its low concentration. The stability con-stants (log$K_{(ZnH$_3$3$L^+$)}$) and specific water-assisted rate constants (koH) of intermediate complexes have been deter-mined from the kinetic data. The dissociation reactions of $Zn^{2+}$ complexes of 1,2, and 3 were investigated with $Cu^{2+}$ ions as a scavenger in acetate buffer. All complexes exhibit acid-independent and acid-catalyzed con-tributions. The effect of buffer and $Cu^{2+}$ concentration on the dissociation rate has also been investigated. The ligand effect on t dissociation rate of $Zn^{2+}$ complexes is discussed in terms of the side-pendant armsand the chelate ring sizes of the ligands.

  • The degradation of humic acid using $TiO_2$ coatings was studied, $TiO_2$ coatings were prepared by dip-coating method. Sol solutions for coating were prepared by mixing the gel, which can be produced by the reaction of $TiOCl_2$ and $NH_4OH$ solution, and hydrogen peroxide solution, and hydrolysis of titanium tetraisopropoxide (TTIP). It was shown from XRD that coatings from sol aged at $100^{\circ}C$ for 18h with titanium peroxo solution were crystallized to anatase in the range of temperatures of $25^{\circ}C$ to $500^{\circ}C$. In contrast, those coated from TTIP were crystallized to anatase at temperature above $400^{\circ}C$. So the sols originated from $TiCl_4$ can be applied for not only on the heat-resistance substrates but on the plastic substrates. Thickness and the quality of the films were dependent on the withdrawing speed, the concentration of sol, and the number of coating. The films showed various interference colors depending on the thickness of them. In the case that the films coated 2 times at withdrawing speed of 2.5cm per minute by 0.2M sol, the films had a transparent light blue color with thickness of around 50nm. It was known from the result of photo-degradation by $TiO_2$ coatings using humic acid that the removal efficiency of $COD_{cr}$ was over 85% after illumination of $UV/H_2O_2$ for 40min. and that of UV/VIS absorbable materials was over 95%.
