Screening for Antimutagenic Compounds in Unused Marine Resources by the Modified Ames Test (수산 미이용자원 중에 존재하는 항변이원성 물질의 검색)

  • To detect naturally occurring antimutagenic substances from unused marine resources, the screening for the antimutagenic compounds containing in various solvent extracts of eight unused marine resources by modified Ames test was carried out. The tested unused marine resources were Asterina pectinifera, Halocynthia roretzi skin, Nototodarus sloani ink, Anthocidaris crassispina skin, Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata. The ether extracts of Halocynthia roretzi skin showed the only antimutagenic effect among several solvent extracts of tested samples. The effect increased with increasing concentration of the ether extracted ones.

Extraction Efficiencies of Organochlorine Pesticides Spiked in Fish Tissues by Supercritical Carbon Dioxide (초임계이산화탄소에 의한 어류조직 중 유기염소계 농약의 추출수율)

  • A simple, fast, inexpensive method hal been developed to extract organochlorine pesticide spiked in fish tissues by supercritical carbon dioxide. Following static supercritical fluid extraction (SFE), pesticides are collected by rapidly depressurizing the carbon dioxide effluent through a 0.79 mm i.d. metering valve into a capped screw-top tube. Percent recovers of standard organochlorine pesticides mixture (SOPM) increased with the decrease of the extraction temperature at 103 bar. Maximum average percent recovery of SOPM was 98% at $40^{\circ}C/172\;bar$ Average recoveries of SOPM spiked in Pacific cod, flatfish and common squid were 85,74,and 83% respectively at $40^{\circ}C/172\;bar$ Application of supercritical carbon dioxide ectraction offers and attractive alternative to the use of organic solvents for extraction of organochlorine pesticides from fish tissues.

  • To detect naturally occurring antioxidative components from unused marine resources, the screening test for the antioxidants containing in starfish, ascidian skin, cuttlefish ink, echinoid skin, Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata was carried out. The ether, acetone and methanol fractions successively extracted from Odonthalia corymbifera were very effective on prevention of the nonenzymatic lipid oxidation and their $IC_{50}$, (a concentration to inhibit 50% of control lipid oxidation rate) were $30,\;100\;and\;200{\mu}g/ml$, respectively. The water and methanol extracts obtained from cuttlefish ink also revealed strong antioxidant effect and their $IC_{50}$, were $390,\;100{\mu}g/ml$, respectively.

Monitoring of Crude Fat and Trans Fatty Acids Contents of Take-Out Foods in Daejeon, Chungcheong Province (대전, 충청 지역에서 판매되는 즉석식품의 트랜스지방 및 조지방 함량 모니터링)

  • This study was designed to determine the trans fatty acid (tF A) contents of 41 take-out food items in Daejeon and Chungcheong Province. Total fatty acid composition and tFA contents of samples were determined by gas chromatography (GC) after fat extraction (Folch method). Maximum fat content among the observed samples was obtained from theater popcorn (13.74${/cdot}$38.09%) while minimum content was observed from the glutinous rice doughnut. 31${/cdot}$5.44%). The contents of tFA (g/100 g food) in toasts, glutinous rice doughnut and ggwabaegi (twisted doughnut sold in streets) ranged from 0.02 to 0.56 g while those in fried sweet potatostick (from highway resting place), fried squid and theater popcorn ranged from 0.05 to 3.08 g/100 g. As a result, most samples showed the tFA content as less than 1 g (g/100 g food) except one sample from theater popcorn which contained 3.08 g (g/100 g food) as tFA content.

  • The colorimetric estimation, by dithiol method, of the solubility of tin dissolved from the container the tin plate in some canned marine foods has been made on 200 cans of sample on the market. Some of them contained more than 150 ppm of Sn concentration were as follows; (1) ca. 50% of squid can, (2)ca. 30% of mackerel can, (3) ca. of saury can, (4) naught of crab, whale, clam(Japanese hard clams), fish meat ball, and Turbo cornutus one. (1) It had the tendency of increasing Sn concentration according to lowering or arising the value of pH. (2) No significant difference in Sn concentration was caused by the variation of the sort of tinning as H-D or E-T. (3) The longer the period of the storage the higher Sn concentration was found, and the concentration more than 150 ppm of Sn was generally found among the cans stored more than twelve months. (4) The concentration of Sn in the liquid was higher than in solid portion, and most of dissolved Sn was distributed in the liquid portion.

  • Tocopherol and tocotrienol contents of commonly consumed foods in Korea were determined by saponification or direct solvent extraction followed by normal phase liquid chromatography. All samples were locally obtained in the year of 2002 and 2003. The study included 13 meats, 15 fishes and shellfishes, 4 seaweeds, 7 mushrooms, 19 milk and milk products, 6 legumes, 12 nuts, 17 processed foods and 17 Korean traditional foods. All of the vitamin E isomers were quantitated and the results were expressed as $\alpha-tocopherol$ equivalent $(\alpha-TE)$. The relatively higher amount of vitamin E was found in beef boiled in soy, dried squid, toasted seaweed, milk powder, soritae, sunflower seeds, ramyon (instant noodle), and kochujang from meats, fishes, seaweeds, milk products, legumes, processed foods, and traditional foods, respectively. This study provided reliable vitamin E data in commonly consumed foods in Korea for the nutritional information and food composition database.

Purification of Eicosapentaenoic Acid (EPA) and Docosahexaenoic Acid (DHA) Esters from Squid Oil by Silver Ion Chromatography (은 이온 크로마토그래피에 의한 오징어유로부터 eicosapentaenoic acid(EPA) 및 docosahexaenoic acid(DHA)의 분리농축)

  • EPA and DHA extracted from methyl esterified squid oil were purified by silver exchanged resin, silver nitrate-impregnated silica gel, silver exchanged zeolite, and silica gel column chromatography, among which column chromatography using mixture of silver exchanged resin and silica gel (10% by weight) showed the best result. By this simple purification method, EPA and DHA were concentrated from 12.5 to 27.9% (yield, 86,0%) and from 21.7 to 49.5% (yield, 87.3%), respectively. Silver exchanged resin had additional advantages of outstanding reusability and simple recovery of silver.

Concentration of Polybrominated Diphenyl Ethers and Their Composition in Octopus minor Collected from Seosan Intertidal Zone (낙지(Octopus minor)에서의 브롬계화합물(Polybrominated diphenyl ethers, PBDEs)의 잔류농도와 조성특성)

  • Octopus(Octopus minor), benthic cephalopod, were collected from intertidal zone in Seosan, Choongnam and analyzed for polybrominted diphenyl ethers(PBDEs). PBDEs concentrations ranged from 29 to 109 ng/g lipid wt(mean; 54 ng/g) in mantle and from 12 to 89 ng/g lipid wt(mean; 48 ng/g) in internal organ. PBDEs concentrations in octopus internal organ were lower about two times than that in common squid collected in Yellow Sea, indicating Seosan is relatively less contaminated with PBDEs. Major congener was BDE 206, occupying 72% and 49% of total PBDE concentration in mantle and in internal organ, respectively, which is very different from PBDE composition in common squid with major congeners of BDE 47 and 99. From PBDE composition, octopus seemed to concentrate higher brominated BDE rather than lower brominated BDE. This PBDE composition in octopus may be affected by sediment with extremely high contribution of deca-BDE to total PBDE concentration.

Acoustic Target Strength of Live Japanese Common Squid(Todarodes pacifica) for Applying Biomass Estimation (살오징어 (Todarodes pacifica)의 음향 반사강도 측정)

  • Target strength (TS) of Japanese common squids (Todarodes pacificus) were measured using 38 and 120 kHz split beam scientific echosounders under the live condition. For the TS measurement of an individual, a total of 3 squids (mantle length (ML): 22.8, 25, and 27 cm) were used using small fishhook method, whereas for measurement of swimming angle, a total of 8 squids (ML: 21-27 cm) were used under live condition, confined with net cage with 2 m diameter At the same time, two underwater video cameras enabled continuous monitoring of squid behavior. Considering normal behavior, the mean TS at 38 and 120 kHz varied from -48.6 to -45.9 dB, and from -46.5 to -44.6 dB, respectively In both frequencies, mean TS at 120 kHz is relatively higher than that of 38 kHz, approximately 1.3-2.5 dB. From free living condition, the mean swimming angle of the squlds was $-24^{\circ}$. The results of the measurement will be provided basic information for conducting acoustic surveys of the squid.

Effects of Heating Condition and Additives on Rheology of Squid Meat Paste Products (오징어 연제품의 물성에 미치는 가열조건과 첨가제의 영향)

  • A squid meat has not been utilized for gel products because of its lower gel forming ability. The objectives of this study were as followed; 1) the optimum heating condition on squid meat paste products and 2) the optimum added level for jelly strength of squid meat paste products. Optimum heating conditions of squid meat kamaboko were as followed; setting (pre-heating) at 15$^{\circ}C$ or 55$^{\circ}C$ for 2 hours and heating at 90$^{\circ}C$ for 60 minutes. Effect for jelly strength of starch additives wheat starch, potato starch and com starch were examined. The jelly strength of heat induced gels differed from the levels of additives. In case of adding starch, potato starch was resulted in the superior jelly strength than the other starchs, wheat starch and corn starch, at any levels. Optimum concentration was 10%(w/w) at every additives. Folding test value was B at added 10% and this value was mean good product. Data of jumbo and flying squid meat paste products added potato starch, corn starch and wheat starch of 10% were shown below, jelly strengths were 858${\pm}$34∼1020${\pm}$37gㆍcm and 966${\pm}$33∼l148${\pm}$45gㆍcm and moisture contents were 72.43∼73.04% and 71.61∼72.78%, respectively. To adding edible agar and sea tangle, showed the highest jelly strength (edible agar>sea tangle, flying squid>jumbo squid) at added 0.5%(w/w) concentration.
