Genotoxicity Studies on Corn Silk Extract Containing High Maysin (고메이신 함유 옥수수수염 추출물의 유전독성학적 안전성 연구)

  • In this study, a battery of genetic-toxicity studies on corn silk extract with high maysin content were performed according to internationally accepted protocols. In a mutation test using Salmonella Typhimurium TA1535, TA1537, TA98, and TA100, the number of mutant colonies did not significantly increase up to a maximum concentration of $5,000{\mu}g/plate$ in the presence or absence of the S9 metabolic activation system. In the chromosome aberration test using Chinese hamster lung fibroblasts, negative results were observed in the concentration up to $1,250{\mu}g/mL$ of corn silk extract. In the micronucleus test using ICR mice, incidence of polymorphonuclear erythrocytes with a maximum concentration of 2,000 mg/kg corn silk extract did not show any significant difference compared to the negative control group. Based on these results, the test substance, con silk extract, did not influence genotoxicity.

Microbiological and Genotoxicological Safety of Gamma-Irradiated Chicken (감마선조사 닭고기의 미생물학적 및 유전독성학적 안전성 평가)

  • Gamma irradiation (1-10 kGy) was applied to chicken for the evaluation of their microbiological safety and possible genotoxicity. In 3 kGy-irradiated sample, the growth of psychrophile was inhibited about 1.5 log cycles and no cells were recovered in total microbial counts. All kinds of contaminated microorganism were sterilized by 7 kGy-irradiation. Also, irradiation followed by freeze-storage at the same time was very effective in inhibiting bacterial growth. The genotoxicity of 10 kGy-irradiated chicken was evaluated by Salmonella Typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. The results were negative in the bacterial reversion assay with S. Typhimurium TA98, TA100, TA1535, and TA1539. Clastogenic effects were not shown in vivo mouse micronucleus assay at 10 kGy-dose tested.

Genotoxicity evaluation of balanced nutritional food for patients pasteurized by gamma irradiation at 4 kGy (4 kGy로 감마선 살균처리된 환자용 균형영양식의 유전독성 평가)

  • This study was conducted to evaluate the genotoxicity of balanced nutritional formular for patients containing various ingredients after gamma irradiation at 4 kGy. Since viable bacteria were not observed within the detection limit of 1 log CFU/g, a dose of 4 kGy was appropriate for the pasteurization of the formular. In a bacterial reverse mutation assay, both hot water and methanol extracts of the formular exhibited dose-independent responses, which was similar to those obtained from that of the negative control (distilled water or dimethyl sulfoxide). In a chromosomal aberration test using lung fibroblast cells of Chinese hamster, the numbers of normal chromosomes were comparable to those observed in the negative control, regardless of the treatment dose and metabolic activation system. Furthermore, no significant increases in the frequency of micronucleated polychromatic erythrocytes were observed relative to the control, when mice were fed with the formular at doses up to 2,000 mg/kg body weight. Therefore, the balanced nutritional formular for patients did not exhibit genotoxicity when pasteurization by gamma irradiation at 4 kGy.

Establishing the Genotoxicological Safety of Gamma-irradiated Egg White and Yolk (감마선 조사 계란의 유전독성학적 안전성 평가)

  • The genotoxicological safety of gamma-irradiated egg white and yolk was examined to ensure that required safety parameters were met, and in an effort to further apply gamma-irradiation for improvement of the hygienic qualities of eggs. Egg white and yolk were irradiated at 20 kGy, much higher than the legally approved dose (less than 5 kGy), and possible genotoxicity was evaluated using in vitro and in vivo tests. The SOS chromotest employing Escherichia coli PQ37, and a chromosomal aberration test in cultured Chinese hamster lung (CHL) cells, were performed in vitro with or without metabolic activation (S9). An in vivo micronucleus development test was conducted using mouse bone marrow cells. Negative results were obtained in the SOS chromotest. The incidence of chromosomal aberration in CHL cells and the frequency of micronuclear developmentin mouse bone marrow cells treated with irradiated samples were not significantly different from those of non-irradiated controls. Thus, it may be concluded that up to 20 kGy of gamma irradiation applied to egg white and yolk did not show any genotoxic effects under our experimental conditions.

A Chromosome Aberration Test of HMC05 on Cultured Chinese Hamster Lung Cells (HMC05의 배양 Chinese Hamster Lung 세포를 이용한 염색체이상 시험)

  • Objectives : We investigated genetic toxicity of HMCO5 in relation to chromosome aberration test on Cultured Chinese Hamster Lung (CHL) in the presence and absence of S-9 mix. Methods : Experimental groups were divided into two groups: with S-9mix (+S) or without S-9 mix (-S). -S group was also divided 2 series by treatment hours (6 hr: 6-S; or 24 hr; 24-S). Each group treated with vehicle only (complete culture medium), HMCO5 (1,250, 2,500, $5,000\;{\mu}g/ml$), and cyclophosphamide monohydrate (CPA) and ethylmethanesulfonate (EMS), respectively. Results : HMC05 did not show any aberrant metaphase. However, there were significant (p < 0.01) aberrant metaphases with CPA in S+ and with EMS in S-. Conclusions : These results indicate that HMC05 formula does not show any toxicity in chromosome aberration test.

Genotoxicological Safety of Gamma-Irradiated Salted and Fermented Anchovy Sauce (감마선 조사된 멸치액젓의 유전독성학적 안전성 평가)

  • Gamma irradiations at 5 or 10 kGy were applied to salted and fermented anchovy sauce, for improving the hygiene Quality and evaluating the genotoxicological safety. In vitro genotoxicological safety of irradiated sauces was evaluated by Salmonella Typhimurium (TA98, TA100, TAI535 and TAI537) and E. coli WP2 uvrA, reversion assay, SOS chromotest (Escherichia coli PQ37), and chromosome aberration test (Chinese hamster lung fibroblast cells) in the absence or presence of an exogenous metabolizing system (S9 mix). The gamma-irradiated samples were not significantly different from nonirradiated-control for three in vitro tests (p<0.05). :In vivo micronucleus test using ICR mice (male) was not significantly different from the control at p<0.05. The salted and fermented anchovy sauce exposed to 5 or 10 kGy-gamma ray revealed negative results in these three in vitro mutagenetic tests and in vivo micronucleus test upto 50,000 $\mu$g/plate, respectively. The results indicated that 5 or 10 kGy gamma-irradiated salted and fermented anchovy sauces did not show any mutagenicity.

Chromosomalanomaly test of Sipjeondaebo-tang extract using the Chiness hamster lung (Chiness hamster lung cell(CHL)에서 십전대보탕 염색체 이상 시험)

  • Objectives : This study was to assessment the toxicity of Sipjeondaebo-tang(Shiquan dabu-decoction) by Chromosomalanomaly test. Methods : Sipjeondaebo-tang(Shiquan dabu-decoction) water-extract in vivo Chromosomalanomaly test was performed using chiness hamster lung cell line. Results : Sipjeondaebo-tang water extract was negative in Chromosomalanomaly test at the doses of 0, 625, 1250 and $2500{\mu}g/m\ell$ at 6h and 24h.(S9- fraction). Chromosomalanomaly test(S+fraction) was also negative at the doses of 0, 1250, 2500 and $5000{\mu}g/m\ell$. Conclusions : It was concluded that Sipjeondaebo-tang extract did not induce Chromosomalanomaly in the chiness hamster lung cell.

  • Gamma irradiation at 5 kGy was applied to beefs for evaluation of their possible genotoxicity and acute oral toxicity. The genotoxicity of 5 kGy irradiated beef was evaluated by Salmonella typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. The results were negative in the bacterial reversion assay with S. typhimurium TA98, TA100, TA1535, TA1537. Clastogenic effects were not shown in vivo mouse micronucleus assay at 5 kGy dose tested. In an acute toxicity test, 5 kGy-irradiated beef was administrated orally at a dose level of 313 to 5,000 mg/kg, and then number of deaths, clinical signs, body weights, and pathological examinations were examined daily for 14 days post-administration. The results indicate that 5 kGy irradiated beef did not show any toxic effect on mice and oral $LD_{50}$ value was over 5,000 mg/kg on ICR mice.

  • Guh Sung Y.L.S-95 (GS95) is a kind of polyacidic solution, which contains acetic acid as a main component. We investigated in the present study tile genetic toxicity of GS95 according to the standard operation procedure from Korean Institute of Toxicology. In the Salmonella typhimurium reverse mutation assay using TA1535, TA1537, TA98 and TA100, GS95 did not induce mutation up to $5,000{\mu}g/plate$. GS95 did not induce chromosome aberration in Chinese hamster lung fibroblast in the concentration range between 1.25 and 5 mg/mL. In the rodent micronucleus assay, the frequency of micronucleated polychromatic erythrocyte in GS95 treated mice were not increased up to 5,000 mg/kg compared to the vehicle treated mice. Taken all these data together, GS95 was proven to be nongenotoxic in the concentration ranges tested.

  • There are $10{\sim}30%$ polyphenol and $2{\sim}4%$ caffeine in green tea. Caffeine is a kind of alkaloid containing nitrogen which cause stimulation, impatience, headache, insomnia, low birth weight infant. Because of these negative effect, decaffeined beverage came out and decaffeined coffee already have a big market since 1970s. Having proving the physiologic functions of green tea, high consumption of coffee is shifting to green tea. Because of the carcinogenic effect of the organic solvents, decaffeine processing with supercritical carbon dioxide has industrialized and have an advantage in environment-friendly and minimized flavor loss. Decaffeined green tea using supercritical carbon dioxide is considered to be safe but there are not enough study, We investigated the chromosome aberration test with mammalian cell line, CHL. When the cells were treated with 5000, 2000, 1000 ${\mu}g/ml$ and compared with the negative controls, there were no significant (P>0.05) increased chromosome aberration. Same results was observed when adding S9 mixture or not. As a result, water extract of decaffeined green tea using supercritical carbon dioxide does not induce chromosome aberration.
