The Study on the Methylmercury Analysis and the Monitoring of Total Mercury and Methylmercury in Fish (어류 중 메틸수은 분석법 확립 및 모니터링)

  • Procedure for analysis of methylmercury in fish was developed, involving addition of HCl, extraction with toluene, and clean-up using L-cystein solution. Obtained extract is analyzed by gas chromatography with electron capture detector using Ulbon HR-Thermon-Hg column. Detection limit and recovery of the method were 0.005mg/kg (expressed as Hg), 98-107 (103%), respectively. Total mercury and methylmercury concentrations in 175 commercial fish samples ranged from [mean-max (mean), unit: mg/kg]: 0.014-1.200 (0.270) and 0.006-0.901 (0.168) in tuna-fish, 0.020-0.934 (0.323) and 0.012-0.553 (0.149) in martin-fish, 0.082-0.782 (0.391) and 0.040-0.436(0.201) in shark, 0,023-0.031 (0.026) and 0,013-0.018 (0.015) in salmon, 0.098-0.193 (0.133) and 0.031-0.015(0.090) in tilefish, and 0,031-0.214 (0.089) and 0.016-0.093 (0.042) in canned tuna respectively. No sample of analyzed fish exceeded 1.0mg/kg wet wt., limit for methylmercury established by Codex. In all species examined, estimated weekly intake was lower than Provisional Tolerable Weekly Intake recommended by the JECFA (the Joint FAO/WHO Expert Committee on Food Additives).

Improving a Korean Spell/Grammar Checker for the Web-Based Language Learning System (웹기반 언어 학습시스템을 위한 한국어 철자/문법 검사기의 성능 향상)

  • The goal of this paper is the pedagogical application of a Korean Spell/Grammar Checker to the web-based language learning system for Korean writing. To maximize the efficient instruction of our learning system \\`Urimal Baeumteo\\` we have to improve our Korean Spell/Grammar Checker. Today the NLP system\\`s performance defends on its semantic processing capability. In our Korean Spell/Grammar Checker. the tasks accomplished in the semantic level are: the detection and correction of misused derived and compound nouns in a Korean spell-checking device and the detection and correction of syntactic and semantic errors in a Korean grammars-checking device. We describe a common approach to the partial parsing using collocation rules based on the dependency grammar. To provide more detailed semantic rules. we classified nouns according to their concepts. and subcategorized verbs referring to their syntactic and semantic features. Improving a Korean Spell/Gl-Grammar Checker makes our learning system active and intelligent in a web-based environment. We acknowledge the flaws in our system: the classification of nouns based on their meanings and concepts is a time consuming task. the analytic unit of this study is principally limited to the phrases in a sentence therefore the accurate parsing of embedded sentences remains a difficult problem to solve. Concerning the web-based language learning system. it is critically important to consider its interface design and structure of its contents.

Yolk absorption and growth of churn salmon, Oncorhynchus keta alevin (연어, Oncorhynchus keta전기자어의 난황흡수 및 초기성장)

  • The process of yolk absorption and the growth pattern of churn salmon, Oncorhynchus keta alevin were studied. Matured males and females of chum salmon were sampled from the Namdaechun River in the east coast of Korea, and then an artificial fertilization was carried out using their eggs and sperms. Fertilized eggs were incubated in controlled water temperature of $13^{\circ}C$ and samples of 40 alevins were randomly taken at just hatching and at every three days between 12 days alter hatching and yolk absorption. The means of total length and total weight of hatched alevins were respectively 1.97cm and 1.85g and the yolk absorption took about 33 days after hatching, based on the morphological measurement. Growth curves of total length, total weight and somatic weight were fitted well with the Gompertz growth model. However, the shrinking in yolk-sac length, yolk-sac weight, yolk-sac height and yolk-sac volume revealed a linear phenomenon until the absorption of yolk was completed. The relative growth of chum salmon alevin, such as the relationship between total length and total weight, or between total weight and yolk-sac weight, was also studied.

Inhibition of Foodborne Pathogens on Polystyrene, Sausage Casings, and Smoked Salmon Using Nonthermal Plasma Treatments (비열 플라즈마 처리를 이용한 polystyrene, 소시지 케이싱, 그리고 훈제연어에서의 식중독균 저해)

  • The effects of nonthermal plasma treatments against Salmonella Typhimurium, Escherichia coli O157:H7, and Listeria monocytogenes living on polystyrene (PS), sausage casings, and smoked salmon were investigated. Inoculated PS, casings, and salmon were treated with nonthermal plasma generated with helium (5 L/min) or with both helium (5 L/min) and oxygen (100 mL/min) at 60 Hz and 30 kV/cm for 2, 5, or 10 min. S. Typhimurium exhibited the highest sensitivity to the helium-used treatment. The greatest reduction (3.9${\pm}$0.8 log$CFU/cm^2$) was observed with L. monocytogenes on PS after the treatment with the mixed gas for 5 min. The treatment with the mixed gas inhibited L. monocytogenes on casings and salmon by 0.5${\pm}$0.3 log$CFU/cm^2$ and 1.0${\pm}$0.3 log CFU/g, respectively. Different treatment times did not result in different reductions of L. monocytogenes on both casings and salmon. The types of treatment gas and material of contamination need to be considered for evaluating the antimicrobial effects of nonthermal plasma treatments.

Evaluation of Conventional Culture Methods and Validation of Immunoassays for Rapid Detection of Listeria monocytogenes in Dairy and Processed Foods (유제품 및 가공식품에서 Listeria monocytogenes 검출을 위한 배지법과 신속 검사키트의 유효성 검증)

  • Listeria monocytogenes is a foodborne pathogen inducing listeriosis in human. We compared two different culture methods for detection of L. monocytogenes and validated two commercial kits, $VIDAS^{(R)}$ and $REVEAL^{(R)}$ for Listeria. L. monocytogenes was inoculated into various food samples to generate partial positive samples. The inoculated samples were enriched in half-Fraser broth for 48 hr at $30^{\circ}C$. The enriched samples were streaked onto Oxford agar at 24 and 48 hr postincubation followed by biochemical confirmation and concurrently analyzed by using the two commercial kits for comparison. When the enrichment period was extended from 24 to 48 hr, the numbers of positive samples were dramatically increased from 6 to 52 out of 80 samples tested using the culture method. With the commercial kits, the numbers of positive samples were also significantly increased from 10 to 18 and 1 to 18, respectively, when the enrichment period was extended from 48 to 72 hr. There was no statistical difference between the 24 hr culture method and $VIDAS^{(R)}$ or $Reveal^{(R)}$ with 48 hr enrichment. In conclusion, the 24 hr for the culture method was insufficient to detect L. monocytogenes in various foods. The commercial kits could be adequate means for presumptive screening of L. monocytogenes in food.

  • 자연언어 처리에 있어 문장의 성분 구조를 파악하는 통사적 해석에서는 애매성 있는 결과가 많이 생성된다. 한국어의 경우 어순 등의 통사적 특성뿐 아니라 상황과 의미, 문맥이 문장의 분석에 더 중요한 역할을 하기 때문에 문맥 자유 문법에 의한 접근 방법만으로는 중의적 구조의 해결이 어렵다. 이는 또한 의미 분석시 애매성을 증가시키는 원인이 된다. 이러한 통사적, 의미적 중의성 해결을 위해 용언 중심의 하위범주화 사전을 구축하였다. 본 논문에서는 용언에 따라 제한될 수 있는 하위범주 패턴을 정의하고 패턴에 따라 하위범주 사전을 구축하였다. 하위범주화 사전에는 명사의 시소러스와 정합하여 보어를 선택 제한(Selectional Restriction)할 수 있도록 용언과 명사와의 의미적 연어 관계에 따라 의미마커를 부여했다. 말뭉치를 통해 수집된 용언 12,000여개를 대상으로 25,000여개의 하위범주 패턴을 구축하였고 이렇게 구축한 하위범주화 사전이 120,000여 명사에 대한 의미를 갖고 있는 계층 시소러스 의미 사전과 연동하도록 하였다. 또한 논문에서 구현된 하위범주화 사전이 구문과 어휘의 중의성을 어느 정도 해소하는지 확인하기 위해 반자동적으로 의미 태깅(Sense Tagging)된 2만여 문장의 말뭉치를 통해 검증 작업을 수행하고, 의존관계와 어휘의 의미를 포함하고 있는 말뭉치에 하위범주 패턴이 어느정도 정합되는지를 분석하여, 하위범주 패턴과 말뭉치의 의존관계만 일치하는 경우와 어휘의 의미까지 일치하는 경우에 대해 평가한다. 이 과정에서 하위범주 패턴에 대한 빈도 정보나, 연어 정보를 수집하여 데이터베이스에 포함시키고, 각 의미역과 용언의 통계적 공기 정보 등을 추출하는 방법도 제시하고자 한다.을 입증하였다.적응에 문제점을 가지기도 하였다. 본 연구에서는 그 동안 계속되어 온 한글과 한잔의 사용에 관한 논쟁을 언어심리학적인 연구 방법을 통해 조사하였다. 즉, 글을 읽는 속도, 글의 의미를 얼마나 정확하게 이해했는지, 어느 것이 더 기억에 오래 남는지를 측정하여 어느 쪽의 입장이 옮은 지를 판단하는 것이다. 실험 결과는 문장을 읽는 시간에서는 한글 전용문인 경우에 월등히 빨랐다. 그러나. 내용에 대한 기억 검사에서는 국한 혼용 조건에서 더 우수하였다. 반면에, 이해력 검사에서는 천장 효과(Ceiling effect)로 두 조건간에 차이가 없었다. 따라서, 본 실험 결과에 따르면, 글의 읽기 속도가 중요한 문서에서는 한글 전용이 좋은 반면에 글의 내용 기억이 강조되는 경우에는 한자를 혼용하는 것이 더 효율적이다.이 높은 활성을 보였다. 7. 이상을 종합하여 볼 때 고구마 끝순에는 페놀화합물이 다량 함유되어 있어 높은 항산화 활성을 가지며, 아질산염소거능 및 ACE저해활성과 같은 생리적 효과도 높아 기능성 채소로 이용하기에 충분한 가치가 있다고 판단된다.등의 관련 질환의 예방, 치료용 의약품 개발과 기능성 식품에 효과적으로 이용될 수 있음을 시사한다.tall fescue 23%, Kentucky bluegrass 6%, perennial ryegrass 8%) 및 white clover 23%를 유지하였다. 이상의 결과를 종합할 때, 초종과 파종비율에 따른 혼파초지의 건물수량과 사료가치의 차이를 확인할 수 있었으며, 레드 클로버 + 혼파 초지가 건물수량과 사료가치를 높이는데 효과적이었다.\ell}$ 이었으며 , yeast extract 첨가(添加)하여 배양시(培養時)는 yeast extract

Study of the Most Common Allergic Foods in Korea (국내 주요 알레르기 원인 식품에 대한 조사)

  • Prevalence of food allergic disease was examined by identifying the most common foods implicated in allergic reactions in Korea. Patients were subjected to test determining the amount of specific IgE antibody in serum against food allergens by CAP system. A total 9054 CAP analyses on egg white, egg yolk, cow milk, ${\alpha}-lactalbumin,\;{\beta}-lactoglobulin$, casein, wheat, rice, buckwheat, soybean, peach, crab, shrimp, pork, beef, chicken, tuna, salmon, mackerel, and food mix were undertaken. The results were considered to be positive when CAP value was same and/or greater than +2 (0.7 U/mL). Positive results of CAP analyses were 11.3% (1022/9054 cases), consisting of 336 on egg white, 266 on cow milk, 95 on egg yolk, 76 on soybean, 69 on ${\alpha}-lactalbumin$, 61 on casein, 58 on ${\beta}-lactoglobulin$, 39 on buckwheat, 12 on wheat, 3 on beef, 2 on crab, and 1 on rice, shrimp, pork, chicken or mackerel, and 0 on peach, tuna or food mix. Egg, cow milk, soybean, buckwheat, and wheat were identified as the most common allergic foods in Korea, showing an average of two different food sources for allergy per patient.

Preparation of Snack Using Residues of Fish Gomtang (생선 곰탕 잔사를 이용한 스낵의 제조)

  • For the use of salmon frame in zero emission, the snack using residues of salmon Gomtang was prepared and investigated on the food component characterization. According to the results of volatile basic nitrogen, water activity and sensory evaluation, the optimal substitution ratio of residues was 15% based on the mix. Total amino acid content was higher in the snack (14.8 g/100 g) with 15% residues than in snack (9.8 g/100 g) without residues. The major amino acids of the residues-added snack were aspartic acid (9.9%), glutamic acid (14.7%) and proline (9.5%). The snack with 15% residues were enriched in the mineral (calcium and phosphorus) and polyunsaturated fatty acid (20:5n-3 and 22:6n-3) compared to those of snack without residues.

A study on the improvement of import conditions for salmonid fish to reduce the possibility of entry of infectious salmon anaemia virus (전염성연어빈혈증(Infectious salmon anaemia) 바이러스의 국내 유입 가능성 저감을 위한 연어과 어류 수입조건 개선안)

  • As Korea declared that the entire country is free from ISA (Infectious salmon anaemia) based on the OIE aquatic code in 2019, it is necessary to improve the import quarantine system that meets the level of ISA management in Korea. Currently, Korea imports Atlantic salmon and rainbow trout from countries which have history of ISA outbreak such as Norway and the United States, so there is a constant possibility that trade with these countries will bring ISAV (Infectious salmon anaemia virus) into Korea. Therefore, some amendments to the Aquatic Life Diseases Control Act (ALDCA) are needed to prevent the entry of ISAV into the Korea through international trade. The amendment to the ALDCA should contain a list of countries, zones or compartments that the Korean competent authority has allowed or banned imports of ISA-susceptible fish. In addition, the subordinate statutes or administrative rules of the ALDCA need to be revised so that on-site inspections/audit can be conducted for the evaluation of the ISA management system in exporting countries. After the revision of the subordinate statutes or administrative rules, it is necessary to strengthen the present import conditions in line with those that meet the level of ISA management in Korea. As for the strengthened import conditions, the competent authorities of exporting countries must export only salmonid fish produced in zones or compartments declared freedom of ISA to Korea, and must prove through lab-testing that ISAV should not be detected in the fish exporting to Korea. In addition, official veterinarians/fish health professionals of the exporting country should check the health status of the fish within 72 hours prior to export. Also, competent authorities of the exporting countries must attest that fish storage containers and water, ice and other equipment used for transportation should be cleaned and disinfected to avoid contamination with pathogens, including ISA. Therefore, the proposed measures presented here will further improve the current import condition for salmonid fish and assist decision-making on strategies to reduce the risk of ISA introduction into Korea. Also, it is expected to contribute to maintain the status of ISA-free country.

산천어의 생식선자극호르몬 유전자발현에 미치는 에스트로젠의 영향과 에스트로젠수용체 cDNA cloning

  • 어류의 번식활동은 뇌-뇌하수체-생식소로 이어지는 내분비기관이 주로 담당하고 있으며, 그 중에서 뇌하수체에서 만들어지는 생식선자극호르몬 (GTH)은 생식소 발달에 매우 중요한 역할을 한다. 한편, GTH는 생식소에서 생성되는 스테로이드호르몬에 의해 유전자의 발현이 조절된다. 본 연구에서는 산천어 (Oncorhynchus masou)의 GTH 유전자발현에 미치는 estradiol-17$\beta$ (E2)의 영향을 생체내에서 먼저 조사하고, 유전자발현조절의 메카니즘을 구명하기 위하여 에스트로젠 수용체 (ER)의 cDNA를 cloning하였다. 또한 내분비교란물질로 알려져 있으며 에스트로젠 수용체와의 유사결합작용이 알려진 노닐페놀 (NP)의 영향도 아울러 조사하였다. 미성숙 산천어를 E2 및 NP (각각 5 mg/kg 및 10 mg/kg 체중)로 처리하면 72 시간째에 뇌하수체내의 $\beta$-actin mRNA에 비해 GTH (FSH$\beta$, LH$\beta$) mRNA가 상대적으로 증가하는 경향을 나타내었다. 뇌하수체에서 분리한 ER의 기본적인 구조는 DNA결합도메인 (Zf-C4), 호르몬결합도메인 (hormone_rec), 기타 전사활성화도메인으로 구성되어 있었으며 (그림 1), 기존에 보고된 에스트로젠 수용체중에서 무지개송어 및 대서양연어의 ER과 각각 98%, 96%의 높은 상동성이 관찰되었다. 향후, 준비된 ER을 재료로 하여 GTH 유전자발현의 호르몬의존성을 조사할 예정이다.
